Abstract Aims Laser micromanipulation such as dissection or optical trapping enables remote physical modification of the activity of tissues, cells and organelles. To date, applications of laser manipulation to plant roots grown in soil have been limited. Here, we show laser manipulation can be applied in situ when plant roots are grown in transparent soil. Methods We have developed a Q-switched laser manipulation and imaging instrument to perform controlled dissection of roots and to study light-induced root growth responses. We performed a detailed characterisation of the properties of the cutting beams through the soil, studying dissection and optical ablation. Furthermore, we also studied the use of low light doses to control the root elongation rate of lettuce seedlings (Lactuca sativa) in air, agar, gel and transparent soil. Results We show that whilst soil inhomogeneities affect the thickness and circularity of the beam, those distortions are not inherently limiting. The ability to induce changes in root elongation or complete dissection of microscopic regions of the root is robust to substrate heterogeneity and microscopy set up and is maintained following the limited distortions induced by the transparent soil environment. Conclusions Our findings show that controlled in situ laser dissection of root tissues is possible with a simple and low-cost optical set-up. We also show that, in the absence of dissection, a reduced laser light power density can provide reversible control of root growth, achieving a precise “point and shoot” method for root manipulation.

AbstractSilicon is reported to reduce the toxic effects of Al on root elongation but the in planta mechanism by which this occurs remains unclear. Using seedlings of soybean (Glycine max) and sorghum (Sorghum bicolor), we examined the effect of up to 2 mM Si on root elongation rate (RER) in Al-toxic nutrient solutions. Synchrotron-based low energy X-ray fluorescence (LEXRF) was then used for the in situ examination of the distribution of Al and Si within cross-sections cut from the apical tissues of sorghum roots. The addition of Si potentially increased RER in Al-toxic solutions, with RER being up to ca. 0.3 mm h−1 (14 %) higher for soybean and ca. 0.2 mm h−1 (17 %) higher for sorghum relative to solutions without added Si. This improvement in RER could not be attributed to a change in Al-chemistry of the bulk nutrient solution, nor was it due to a change in the concentration of Al within the apical (0-10 mm) root tissues. Using LEXRF to examine sorghum, it was demonstrated that in roots exposed to both Al and Si, much of the Al was co-located with Si in the mucigel and outer apoplast. These observations suggest that Si reduces the toxicity of Al in planta through formation of Al-Si complexes in mucigel and outer cellular tissues, thereby decreasing the binding of Al to the cell wall where it is known to inhibit wall loosening as required for cell elongation.

Changes in root elongation are important for the acquisition of mineral nutrients by plants. Plant hormones, cytokinins, and abscisic acid (ABA) and their interaction are important for the control of root elongation under changes in the availability of ions. However, their role in growth responses to supra-optimal concentrations of nitrates and phosphates has not been sufficiently studied and was addressed in the present research. Effects of supra-optimal concentrations of these ions on root elongation and distribution of cytokinins between roots and shoots were studied in ABA-deficient barley mutant Az34 and its parental variety, Steptoe. Cytokinin concentration in the cells of the growing root tips was analyzed with the help of an immunohistochemical technique. Increased concentrations of nitrates and phosphates led to the accumulation of ABA and cytokinins in the root tips, accompanied by a decline in shoot cytokinin content and inhibition of root elongation in Steptoe. Neither of the effects were detected in Az34, suggesting the importance of the ability of plants to accumulate ABA for the control of these responses. Since cytokinins are known to inhibit root elongation, the effect of supra-optimal concentration of nitrates and phosphates on root growth is likely to be due to the accumulation of cytokinins brought about by ABA-induced inhibition of cytokinin transport from roots to shoots.

The diversity and degradation capacity of hydrocarbon-degrading consortia from surface and deep waters of the Eastern Mediterranean Sea were studied in time-series experiments. Microcosms were set up in ONR7a medium at in situ temperatures of 25 °C and 14 °C for the Surface and Deep consortia, respectively, and crude oil as the sole source of carbon. The Deep consortium was additionally investigated at 25 °C to allow the direct comparison of the degradation rates to the Surface consortium. In total, ~50% of the alkanes and ~15% of the polycyclic aromatic hydrocarbons were degraded in all treatments by Day 24. Approximately ~95% of the total biodegradation by the Deep consortium took place within 6 days regardless of temperature, whereas comparable levels of degradation were reached on Day 12 by the Surface consortium. Both consortia were dominated by well-known hydrocarbon-degrading taxa. Temperature played a significant role in shaping the Deep consortia communities with Pseudomonas and Pseudoalteromonas dominating at 25 °C and Alcanivorax at 14 °C. Overall, the Deep consortium showed a higher efficiency for hydrocarbon degradation within the first week following contamination, which is critical in the case of oil spills, and thus merits further investigation for its exploitation in bioremediation technologies tailored to the Eastern Mediterranean Sea.

The impact of salt stress on the growth and phosphorus utilization efficiency (PUE) of two leguminous species: Retama monosperma and Acacia gummifera was studied. The effectiveness of arbuscular mycorrhizal fungi (AMF) to mitigate salt stress was furthermore assessed. Growth, N and P tissue concentrations, mycorrhizal root colonization frequency and intensity, and P utilization efficiency (PUE) in the absence or presence of AMF were evaluated under no salt (0 mM L−1) and three salt (NaCl) concentrations of (25, 50 and 100 mM L−1) using a natural sterilized soil. A significant difference in mycorrhizal colonization intensity, root-to-shoot ratio, P uptake, PUE, and N uptake was observed between the legume species. Salt stress inhibited the shoot and root growth, and reduced P and N uptake by the legume species. Mycorrhizal inoculation aided to mitigate the effects of salt stress with an average increase of shoot and root growth responses by 35% and 32% in the inoculated than in the non-inoculated A. gummifera treatments. The average shoot and root growth responses were 37% and 45% higher in the inoculated compared to the non-inoculated treatments of R. monosperma. Average mycorrhizal shoot and root P uptake responses were 66% and 68% under A. gummifera, and 40% and 95% under R. monosperma, respectively. Mycorrhizal inoculated treatments consistently maintained lower PUE in the roots. The results provide insights for further investigations on the AMF conferred mechanisms to salt stress tolerance response by A. gummifera and R. monosperma, to enable the development of effective technologies for sustainable afforestation and reforestation programs in the Atlantic coast of Morocco.

A commercial crystallization robot has been modified for use in setting up sitting-drop vapor-diffusion crystallization experiments, and for setting up protein crystallization screensin situ. The primary aim of this effort is the automated screening of crystallization of integral membrane proteins in detergent-containing solutions. However, the results of this work are of general utility to robotic liquid-handling systems. Sources of error that can prevent the accurate dispensing and mixing of solutions have been identified, and include local environmental, machine-specific and solution conditions. Solutions to each of these problems have been developed and implemented.

Soil compaction has been considered a principal form of damage associated with logging, restricting root growth and reducing productivity. The rate and extent of soil compaction on skid trails was measured at six field locations covering a range of dry and wet forests. Data was collected for up to 21 passes of a laden logging machine. A similar extent of compaction, averaging 0.17 g·cm-3 increase in total soil bulk density (BD), was recorded for all field sites despite substantial site and soil differences. On average, 62% of the compaction in the top 10 cm of the soil occurred after only one pass of a laden logging machine. The environment under which soils had formed played a major role in determining the BD of the undisturbed soil. Compaction was strongly related to the original BD, forest type, and soil parent material. Soil strengths obtained in the field fell below levels found to restrict root growth. However, reduction in macropores, and the effect of that on aeration and drainage could reduce tree growth. On the wettest soils logged, machine forces displaced topsoils rather than causing compaction in situ. Recommended logging methods and implications for the development of sustainability indices are discussed.

Concepts and results are described for the use of a single, but extremely flexible, probing tool to address a wide variety of genomic questions. This is achieved by transforming genomic questions into a software file that is used as the design scheme for potentially any genomic assay in a microarray format. Microarray fabrication takes place in three-dimensional microchannel reaction carriers by in situ synthesis based on spatial light modulation. This set-up allows for maximum flexibility in design and realization of genomic assays. Flexibility is achieved at the molecular, genomic and assay levels. We have applied this technology to expression profiling and genotyping experiments.

Abstract. In 1997 the International Maritime Organisation (IMO) adopted MARPOL Annex VI to prevent air pollution by shipping emissions. It regulates, among other issues, the sulfur content in shipping fuels, which is transformed into the air pollutant sulfur dioxide (SO2) during combustion. Within designated Sulfur Emission Control Areas (SECA), the sulfur content was limited to 1 %, and on 1 January 2015, this limit was further reduced to 0.1 %. Here we present the set-up and measurement results of a permanent ship emission monitoring site near Hamburg harbour in the North Sea SECA. Trace gas measurements are conducted with in situ instruments and a data set from September 2014 to January 2015 is presented. By combining measurements of carbon dioxide (CO2) and SO2 with ship position data, it is possible to deduce the sulfur fuel content of individual ships passing the measurement station, thus facilitating the monitoring of compliance of ships with the IMO regulations. While compliance is almost 100 % for the 2014 data, it decreases only very little in 2015 to 95.4 % despite the much stricter limit. We analysed more than 1400 ship plumes in total and for months with favourable conditions, up to 40 % of all ships entering and leaving Hamburg harbour could be checked for their sulfur fuel content.

To facilitate the updating ofin situandex situconservation strategies for wild taxa of the genusHordeumL., a combined ecogeographic survey and gap analysis was undertaken. The analysis was based on the Global Inventory of Barley Plant Genetic Resources held by ICARDA plus additional datasets, resulting in a database containing 17,131 wildHordeumaccessions. The analysis concluded that a genetic reserve should be established in the Mendoza Province of Argentina, as this is the most species-rich area globally forHordeum. A network of reserves should also be set up across the Fertile Crescent in Israel, Palestine, Syria, Jordan, Lebanon and Turkey to provide effective conservation within the centres of diversity for gene pools 1B (Hordeum vulgaresubsp.spontaneum(C. Koch) Thell.) and 2 (Hordeum bulbosumL.). The majority of the species were deemed under-collected, so further collecting missions are required worldwide where possible. Althoughex situandin situconservation strategies have been developed, there needs to be further investigation into the ecological environments thatHordeumspecies occupy to ensure that any adaptive traits expressed are fully conserved. Additionally, studies are required to characterize existing collections and test the viability of rare species accessions held in genebanks to determine whether furtherex situcollections are required alongside the proposedin situconservation.
