Целью работы стало исследование содержания белка p23 в фибробластах дермы человека от 20 нед беременности до 85 лет, а также определение участия p23 для возрастных изменений численности и пролиферации фибробластов в дерме человека. p23, ядерный антиген пролиферирующих клеток (PCNA), маркер фибробластов виментин выявляли в срезах кожи непрямым иммуногистохимическим методом. В дерме человека от 20 нед беременности до 85 лет наблюдали планомерное увеличение доли фибробластов, имеющих положительную окраску на p23. Возрастное изменение численности p23позитивных фибробластов в дерме достоверно коррелирует с возрастным уменьшением общей численности и доли PCNA-позитивных фибробластов в дерме человека, что свидетельствует об участии p23 в возрастных изменениях общей численности и доли пролиферирующих фибробластов в дерме человека. The aim of this work was to examine the content of p23 in fibroblasts of human dermis from 20 weeks of pregnancy until 85 years old, and defining of a role of p23 in age-dependent changes in the number and proliferation of fibroblasts in the dermis. p23, proliferating cells nuclear antigen (PCNA), fibroblasts marker vimentin were detected with indirect immunohistochemical technique. Results showed that portion of fibroblasts with positive staining for p23 in the dermis is gradually increased from 20 weeks of pregnancy until 85 years old. Age-related increase in a portion of fibroblasts with positive staining for p23 is significantly correlated with an age-related decrease in total number and percent of PCNA positive fibroblasts in human dermis. Age-related increase in the content of p23 in fibroblasts is involved in an age-dependent decrease in their total number and proliferation in the dermis.

Changes in root elongation are important for the acquisition of mineral nutrients by plants. Plant hormones, cytokinins, and abscisic acid (ABA) and their interaction are important for the control of root elongation under changes in the availability of ions. However, their role in growth responses to supra-optimal concentrations of nitrates and phosphates has not been sufficiently studied and was addressed in the present research. Effects of supra-optimal concentrations of these ions on root elongation and distribution of cytokinins between roots and shoots were studied in ABA-deficient barley mutant Az34 and its parental variety, Steptoe. Cytokinin concentration in the cells of the growing root tips was analyzed with the help of an immunohistochemical technique. Increased concentrations of nitrates and phosphates led to the accumulation of ABA and cytokinins in the root tips, accompanied by a decline in shoot cytokinin content and inhibition of root elongation in Steptoe. Neither of the effects were detected in Az34, suggesting the importance of the ability of plants to accumulate ABA for the control of these responses. Since cytokinins are known to inhibit root elongation, the effect of supra-optimal concentration of nitrates and phosphates on root growth is likely to be due to the accumulation of cytokinins brought about by ABA-induced inhibition of cytokinin transport from roots to shoots.

Her-2 (ErbB-2) is an oncogene frequently overexpressed in breast and gastric adenocarcinomas, and anti-Her-2 targeted therapy can be given to such patients. Her-2 overexpression and role of anti-Her-2 targeted therapy in cases of gallbladder adenocarcinomas (GBC) are still debatable. Scoring protocols for Her-2 expression in breast and gastric carcinomas are standardized, however, not for carcinomas arising in other body organs like the gallbladder. This study is conducted to evaluate the expression of Her-2 in patients with GBC, which may benefit from targeted therapy. It is a cross-sectional study conducted on patients with GBC (n=63; 53 women and ten men). An automated immunohistochemical technique was used with an anti-ErbB2 antibody. Scoring was conducted according to the CAP (College of American Pathologists) criteria for breast cancer. Positive (3+) Her-2 staining was observed in 8/63 (12.7%). Nine cases (14.3%) showed equivocal staining (2+) pattern. All of the tumors showing Her-2 overexpression were moderately differentiated. This study indicates that a significant number of GBC cases show Her-2 overexpression. This subgroup may benefit from inhibitors of the Her-2 pathway. Standardization of scoring protocol for Her-2 expression in GBC is needed to better evaluate the predictive potential of Her-2 for the treatment of these tumors.

Fibrillary Glomerulonephritis (FGN) is a rare glomerulopathy, corresponding to less than 1% of all renal biopsies performed. The pathogenesis mechanisms of FGN are still not completely understood and most cases are considered idiopathic. The clinical presentation is characterized by proteinuria, hematuria, arterial hypertension and renal failure. The evaluation of FGN is a diagnostic challenge in the practice of the pathologist, because there are no morphological characteristics with highly specific or pathognomonic lesion. The use of a sensitive and specific biomarker can make an important contribution to renal pathology and the differential diagnosis of diseases with fibrillar deposits. In this contexto, DNAJB9 can be investigated by immunohistochemical technique and is useful for the diagnostic definition of FGN.

The aim of this research was to analyze the profile of superoxide dismutase (SOD) and malondialdehyde (MDA) on the liver tissue of hypercholesterolemic rats which were given Holothuria nobilis polysaccharides (HNP). A total of 15 male rats strain Sprague Dawley were divided into prevention and curative groups. Prevention group consisted of negative/non-hypercholesterolemic group (K-), positive/hypercholesterolemic group (K+), and hypercholesterolemic prevention group which were given 1% cholesterol diet and HNP at dose of 400 mg/kg bw (PCh). The treatments were given for 28 days. The curative group was consisted of the hypercholesterolemic group, which was given 1% cholesterol diet for 28 days, then followed by standard diet for 28 days (Ch), and the hypercholesterolemia curative group which was given 1% cholesterol diet for 28 days, then followed by 400 mg/kg bw HNP for 28 days (ChP). The antioxidant activity of HNP was analyzed by DPPH method. At the end of study the liver tissue was collected and analyzed for MDA, SOD while Cu,Zn-SOD was analyzed by immunohistochemical technique. The results showed that the antioxidant activity of HNP was weak. The MDA level (µg/g) in K-, K+, PCh, Ch, and ChP groups were 1.19±0.6; 3.37±0.79; 0.29±0.14; 9.11±0.72; and 3.14±1.06, respectively. The SOD activities (U/g) in K-, K+, PCh, Ch, and ChP groups were 2141.11±83.88; 1541±211.69; 2096.67±166.66; 1063.33±88.19; 1685.55±167.77, respectively. The immuno reactivity of Cu,Zn-SOD showed that HNP could increase Cu,Zn-SOD in the liver tissues of both groups. This study concluded that the HNP increased SOD activity, Cu,Zn-SOD antioxidant content, and decreased MDA levels in the liver tissues of hypercholesterolemic rats in both preventive and curative groups.

The study aimed to assess the number, localization and distribution of interleukin 6 (IL-6) positive cells in healthy pulp, acute and chronic pulpitis. The study group included 48 patients aged between 18-72, treated in University of Medicine and Pharmacy Grigore T. Popa Iasi, Romania. The pulpectomy was performed on 42 patients diagnosed with acute and chronic pulpitis. The other 6 patients, without signs of dental caries or periodontal disease, were submitted to extractions of teeth for orthodontic purposes, with pulpectomy performed before extraction. The pulp samples were examined with optic microscope. The detection and assessment of IL-6 were performed using immunohistochemical technique. Data were statistically analysed using non-parametric tests. According to morphopathological criteria, 42.85% were classified as acute pulpitis and 57.14% as chronic pulpitis. The pulp samples in control group were not associated with IL-6 positive cells. The analysis of all samples with acute and chronic pulpitis identified 73.80% samples with IL-6 and 26.20% associated with the absence of IL-6. The highest frequency of IL-6 positive cells was recorded in rich-cell zone of crown dental pulp. The systemic distribution of IL-6 positive cells was mostly diffused without well-defined orientation. IL-6 release in acute and chronic pulpitis is significantly higher comparing with healthy pulp tissue.
