The objective was to determine the effects of supplementing the diet with fish oil during the peri-partum period on the immune competence and the pathophysiological response to a lipopolysaccharide-induced mastitis challenge. Multiparous Holstein cows (n=30) were completely randomized to one of two treatments at 3 weeks pre-partum. Treatments differed only in the source of supplemental lipid and included either Energy Booster® or fish oil. Treatment diets were fed from −21 d relative to expected date of parturition until 10 d post partum. Treatments were fed as a bolus prior to the a.m. feeding. The dose of lipid during the pre-partum period was 250 g/d, whereas the amount of lipid supplemented post partum was adjusted to the level of intake, approximately 0·92% of the previous day's dry matter intake. Ex-vivo analyses of immune competence were measured including the antimicrobial activity of whole blood against Escherichia coli, Salmonella typhimurium and Candida albicans as well as the production of interferon-γ by peripheral blood mononuclear cultures. At 7 days in milk cows were infused with 100 μg of Esch. coli lipopolysaccharide into one rear quarter. Supplementing fish oil increased plasma concentrations of eicosapentaenoic and docosahexaenoic acids, but had no affect on the proportions of arachidonic acid at calving. Fish oil did not influence the production of interferon-γ or the antimicrobial activity of whole blood against any of the microorganisms. Furthermore, fish oil had no ameliorative effect on either the local or the systemic acute phase response following an intramammary lipopolysaccharide challenge in early lactating Holstein cows. Supplementing fish oil in the diet of peri-partum cows will not protect them from deleterious effects of an excessive acute phase response.

ABSTRACT Little is known about the morphological response of the omasum in dairy cows that consume a high-energy diet pre-partum. The aim of this study was to investigate whether a transitional diet with high grain content is able to induce changes in omasum morphology. Six weeks before the expected calving, four Holstein cows were fed a standardization diet, and four weeks before delivery, the cows were fed a diet with high grain content (HGC) or low (LGC) grain content. After calving, all of the cows were fed a high energy lactation diet. The cows that were fed the HGC diet pre-partum had higher dry matter and nutrient intake than the cows that were fed the LGC diet. The mitotic index of the omasum epithelium was higher than the mitotic index in the rumen, but apparently the response to the diet stimuli was slower. In the cows that were fed the HGC diet, the omasum papillae were taller one week before parturition and two weeks post-partum. Cows that were fed the HGC diet had a thinner epithelium due to thinner non-keratinized layers of the omasum epithelium. We conclude that the omasum mucosa of dairy cows responds to the stimuli of a pre-partum HGC diet, which was indicated by the greater height of the omasum papillae and by the reduced thickness of the omasum epithelium. It seems that the mitotic index responds a little more slowly, but the response to the diet stimuli is stronger in the omasum epithelium than in the rumen.

This study evaluated the effects of replacing corn with wheat on the lactation performance, ruminal fermentation, plasma metabolites and hormonal profiles of multiparous Holstein cows. Thirty-four multiparous cows were divided into two equal groups, based on parity, body condition score, and bodyweight. The basal diet was supplemented with 186 g/kg corn or wheat, and was offered from day -28 (±7) relative to the date of expected calving. After calving, all cows received the same diet for four weeks (day +28). The wheat diets increased (P <0.05) the body condition score in the peri-parturient period and caused greater changes in dry matter intake and bodyweight post-partum, with no differences in energy balance compared with the corn diets. Further, the wheat increased (P <0.05) the concentration of propionate in the rumen, thus reducing the ratio of acetate to propionate, and produced (P <0.05) more milk and milk protein, but had reduced levels of urea nitrogen in the milk and serum. Similarly, the wheat diets increased (P <0.05) the plasma concentrations of albumin and glucose, and reduced (P <0.05) triglycerides with no effect (P >0.10) post partum on insulin, cortisol, β-hydroxybutyric acid and aspartate aminotransferase. The replacement of corn with wheat increased (P <0.05) plasma calcium and reduced non-esterified fatty acids around calving (day 0±1) and post-partum (days 0 to +28), and reduced urinary pH during the last week relative to the date of expected calving.Keywords: calving, energy balance, grain source, peri-parturient period

AbstractThis study was undertaken to quantify the effect of episodes of subclinical hypocalcaemia on the interval from calving to first ovulation. It incorporated three separate surveys during early lactation of 44 and 60 cows at Mutdapilly Research Station and 56 cows at Wollongbar Agricultural Institute. Twenty-five, 37 and 39% of cows respectively, experienced episodes later than 3 days after calving, of non-parturient post-partum hypocalcaemia, when plasma calcium concentration fell below 2⋅00 mmolll. Episodes of subclinical hypocalcaemia did not affect the interval to first post-partum ovulation in the two experiments conducted at Mutdapilly Research Station but ovulation was significantly delayed in cows that experienced episodes of subclinical hypocalcaemia at Wollongbar Agricultural Institute.

The effects of daily supplementation of 1000 IU of either d-α-tocopherol acetate or dl-α-tocopherol acetate to Holstein cows from drying off to 8 wk post-partum on plasma creatine kinase and selenium levels in plasma and red blood cells were studied. Supplementation with either d-α-tocopherol acetate or dl-α-tocopherol acetate showed no effect on selenium levels in both plasma and red blood cells. However, supplementation with d-α-tocopherol acetate, but not dl-α-tocopherol acetate decreased plasma creatine kinase significantly (P < 0.05) prior to parturition. Morphological changes in the placenta at periparturition and the involvement of apoptosis controlling cell populations in the placentome were evaluated in Holstein and Nelore cows. Twenty-two Holstein cows at the end of gestation with normal non-induced parturition (group I) and 10 Holstein cows with induced parturition (group II) were used in this study. In addition, groups III and IV were constituted by 10 Nelore cows at the end of gestation with normal non-induced parturition and 21 Nelore cows with induced parturition, respectivelly. Binucleate trophoblastic cells, epithelial caruncular cells and apoptotic bodies were quantified by light microscopy. Apoptosis was further evaluated by transmission electron microscopy, ELISA and DNA electrophoresis in agarose gel. The number of caruncular epithelial cells did not vary between breeds with normal or induced parturition. The number of binucleate cells was significantly different between Holstein and Nelore cows with normal non-induced parturition (groups I vs. III), but their numbers were not significantly different between breeds when parturition was induced. In cows with normal non-induced parturition, the intensity of apoptosis, as assessed by morphometric analysis and ELISA, was significantly higher in group I when compared to group III. However, no difference in the intensity of apoptosis was found between groups II and IV. Characteristic apoptotic ladder pattern of DNA fragmentation was detected by agarose gel electrophoresis. Ultra-structural features of apoptosis in maternal and fetal epithelium as well as capillary vascular endothelium were identified by transmission electron microscopy. In conclusion, in Nelore placental maturation occurs precociously when compared to Holstein. In both breeds, apoptosis is an active physiological event that seems to be required for maturation and normal post-partum release of the placenta.

Objective:Cobalamin (B12) deficiency has been reported in infants born to mothers with low cobalamin intake. Early diagnosis of vitamin B12 deficiency in infants is critical for the prevention of neurobehavioral disorders. We investigated the relationship between serum vitamin B12 level in newborns and in their healthy mothers who consumed an omnivorous diet. Anthropometry was studied longitudinally to assess the growth velocity of the infants. Urinary methylmalonic acid (MMA) excretion of 6-month old infants was compared retrospectively as the biomarker correlated with the initial serum vitamin B12 concentrations. Methods: Serum cobalamin and blood hemoglobin were determined in 84 pairs of newborns and their mothers. Urinary MMA excretion was measured in the same subjects during the first 6 months of the post partum period. Results: At birth, median serum cobalamin levels were 152.0 pmol/L in the mothers and 296.6 pmol/L in the newborns. Maternal and neonatal serum cobalamin levels had no effect on growth velocity during the first six months of postnatal life. Serum maternal and neonatal cobalamin levels were inversely associated with urinary MMA excretion. Conclusion: Early diagnosis of vitamin B12 status in neonates and infants is crucial, particularly in nutritionally deprived areas. Biochemical measurement of plasma cobalamin or its metabolic marker MMA is highly recommended. Urinary MMA measurement in cobalamin diagnostics provides an advantage in that blood sampling is not required. A vitamin B12 taskforce should be created to alleviate vitamin deficiency and its negative consequences.

Reprod Dom Anim 2019; 54: 996–1002 Zu den wesentlichen Untersuchungen post partum gehört die makroskopische Beurteilung der Plazenta. Insbesondere bei einem Abort, einer Früh- oder Totgeburt sollte die Nachgeburt sorgfältig auf Anomalien wie Unvollständigkeit, Plazentitis, Ödeme oder andere Veränderungen untersucht werden. Um pathologische Veränderungen erkennen zu können, muss dem Untersucher der physiologische Zustand einer Nachgeburt bekannt sein. Bisher fehlen jedoch ausreichende Referenzen für unauffällige Alpakaplazenten. Ziel der Studie war, physiologische Plazenten von Alpakas hinsichtlich Aussehen, Größe, Gewicht und Volumen zu untersuchen und damit künftigen Untersuchern die objektive Beurteilung von Alpakaplazenten zu ermöglichen.
