Many studies have demonstrated that oxidative stress plays an important role in several ailments including neurodegenerative diseases and cerebral ischemic injury. Previously we synthesized some carbazole compounds that have anti-oxidant ability in vitro. In this present study, we found that one of these 22 carbazole compounds, compound 13 (3-ethoxy-1-hydroxy-8- methoxy-2-methylcarbazole-5-carbaldehyde), had the ability to protect neuro2a cells from hydrogen peroxide-induced cell death. It is well known that neurite loss is one of the cardinal features of neuronal injury. Our present study revealed that compound 13 had the ability to induce neurite outgrowth through the PI3K/Akt signaling pathway in neuro2a cells. These findings suggest that compound 13 might exert a neurotrophic effect and thus be a useful therapy for the treatment of brain injury.

The present study evaluates the anti-oxidative stress activity of Vaccinium bracteatum Thunb. fruit extract (VBFW) to identify the mechanisms responsible for its antidepressant-like effects. To evaluate the antidepressant and anti-oxidant effects of VBFW, malondialdehyde (MDA), serotonin transporter (SERT), and monoamine oxidase A (MAO-A) levels were measured in a mouse model of chronic restraint stress (CRS). The underlying mechanisms preventing oxidative stress and neuronal apoptosis were investigated using in vitro models of hydrogen peroxide (H2O[Formula: see text]-induced neuronal damage. The results showed that VBFW treatment (200[Formula: see text]mg/kg) significantly reduced MDA, SERT, and MAO-A levels in the prefrontal cortex of CRS mice. Furthermore, VBFW (30[Formula: see text][Formula: see text]g/mL) exhibited protective effects against H2O2-induced cell death via inhibition of the H2O2-induced increase in Bax and decrease in Bcl-2 levels within the mitochondria of SH-SY5Y cells. Furthermore, VBFW (10 and 30[Formula: see text][Formula: see text]g/mL) exerted protective effects against H2O2-induced cell death through inhibition of key mitochondria-associated apoptotic proteins such as cytochrome c, caspase-3 and PARP. Additionally, VBFW (10 and 30[Formula: see text][Formula: see text]g/mL) could improve the activity of anti-oxidant enzymes (such as SOD and catalase) in H2O2-treated SH-SY5Y cells. These results suggest that the antidepressant and anti-oxidant effects of VBFW might be mediated by the regulation of SERT and MAO-A, and possibly associated with regulation of oxidative stress-induced apoptosis.

High-fat diet (HFD) usually induces oxidative stress and astaxanthin is regarded as an excellent anti-oxidant. An 8-week feeding trial was conducted to investigate the effects of dietary astaxanthin supplementation on growth performance, lipid metabolism, antioxidant ability, and immune response of juvenile largemouth bass (Micropterus salmoides) fed HFD. Four diets were formulated: the control diet (10.87% lipid, C), high-fat diet (18.08% lipid, HF), and HF diet supplemented with 75 and 150 mg kg−1 astaxanthin (HFA1 and HFA2, respectively). Dietary supplementation of astaxanthin improved the growth of fish fed HFD, also decreased hepatosomatic index and intraperitoneal fat ratio of fish fed HFD, while having no effect on body fat. Malondialdehyde content and superoxide dismutase activity were increased in fish fed HFD, astaxanthin supplementation in HFD decreased the oxidative stress of fish. The supplementation of astaxanthin in HFD also reduced the mRNA levels of Caspase 3, Caspase 9, BAD, and IL15. These results suggested that dietary astaxanthin supplementation in HFD improved the growth performance, antioxidant ability and immune response of largemouth bass.

Background: Oxidative stress plays an important role in the development of diabetic complications. Objective: This study evaluated the impact of pre- and post-treatment with Lactobacillus acidophilus ATCC 4356 on the oxidant and anti-oxidant factors of testis and epididymis in streptozotocin-induced diabetic rats. Materials and Methods: Thirty male Wistar rats (10 wk old) weighing 220-230 g. were divided into five groups (n = 6/ each): 1- normal group, 2- normal lactobacillus group, 3- diabetic group, 4- diabetic + lactobacillus before (DLB) group, and 5- diabetic + lactobacillus after (DLA) group. The normal and diabetic groups received daily 1 mL normal saline for 6 wk. Normal lactobacillus group received daily L. acidophilus for 6 wk. Group DLB received daily L. acidophilus for 2 wk before diabetes and for 4 wk after diabetes. Group DLA received daily 1 mL normal saline for 2 wk before diabetes and L. acidophilus for 4 wk after diabetes. The dose of L. acidophilus was 1 × 109 CFU/mL. Results: The administration of L. acidophilus worsened blood glucose level and reduced the levels of Malondialdehyde (p = ≤ 0.0001) and Hydrogen peroxide (p ≤ 0.0001) and, Catalase and Glutathione peroxidase activity increased in the testis. In epididymis, Glutathione peroxidase and Catalase (p = 0.013) activity increased and Hydrogen peroxide concentration reduced, while Malondialdehyde concentration did not show any changes compared to the diabetic rats. Also, there was no significant difference between DLB and DLA groups, in these markers. Conclusion: Data obtained suggests that L. acidophilus has anti-oxidant effects on the testis and sometime in the epididymis in diabetic rats.
