The regulation of body weight and composition is complex, simultaneously affected by genetic architecture, the environment, and their interactions. We sought to analyze the complex phenotypic relationships between voluntary exercise, food consumption, and changes in body weight and composition and simultaneously localize quantitative trait loci (QTL) controlling these traits. A large ( n = 815) murine advanced intercross line (G4) was created from a reciprocal cross between a high-running line and the inbred strain C57BL/6J. Body weight and composition (% fat, % lean) were measured at 4, 6, and 8 wk of age. After measurements at 8 wk of age, mice were given access to running wheels, during which food consumption was quantified and after which body weight and composition were assessed to evaluate exercise-induced changes. Phenotypic correlations indicated that the relationship between exercise and overall change in weight and adiposity depended on body composition before the initiation of exercise. Interval mapping revealed QTL for body weight, % fat, and % lean at 4, 6, and 8 wk of age. Furthermore, QTL were observed for food consumption and changes in weight, % fat, and % lean in response to short-term exercise. Here we provide some clarity for the relationship between weight loss, reduction in adiposity, food consumption, and exercise. Simultaneously, we reinforce the genetic basis for body weight and composition with some independent loci controlling growth at different ages. Finally, we present unique QTL providing insight regarding variation in weight loss and reduction in adiposity in response to exercise.

Current estimates of obesity costs ignore the impact of future weight loss and gain, and may either over or underestimate economic consequences of weight loss. In light of this, I construct static and dynamic measures of medical costs associated with body mass index (BMI), to be balanced against the cost of one-time interventions. This study finds that ignoring the implications of weight loss and gain over time overstates the medical-cost savings of such interventions by an order of magnitude. When the relationship between spending and age is allowed to vary, weight-loss attempts appear to be cost-effective starting and ending with middle age. Some interventions recently proven to decrease weight may also be cost-effective.

Background: Childhood poverty is associated with poorer food consumption patterns but longitudinal data on this association is limited. To assess if the relationship between food consumption and poverty differs depending on the child’s age and pattern of poverty, we analyzed the relationship between consumption of selected foods and poverty trajectories at various ages in a birth cohort. Methods: The 1998-2010 "Quebec Longitudinal Study of Child Development" (n=2,120) cohort was used for these analyses. Household income was measured annually with poverty defined as income below the low-income thresholds established by Statistics Canada adjusted for household size and geographic region. Frequency of children’s consumption of dairy (milk, cheese, yogurt), fruits, and vegetables were reported by parents using a food frequency questionnaire. Analyses were conducted on the 739 children with food consumption data. Trajectories of poverty at 6, 8, 10, and 12 years were characterized with latent class group analysis using maximum likelihood in a semiparametric mixture model. Multivariable logistic regression predicted the likelihood of having less than 2 servings a day of dairy, fruits and vegetables based on poverty trajectories after adjusting for age and sex. Results: The poverty trajectories were stable and fell into 1 lower exposure category (consistently low exposure (73%, n=537)) and 3 higher exposure categories (increasing: 8%, n=61; decreasing: 10%, n=73; or consistently high exposure: 9%, n=68)). Compared to children experiencing low exposure to poverty, children with increasing or high exposure to poverty were less likely to have at least two servings of fruit a day at all ages, but the results were not significant. Compared to children experiencing low exposure to poverty, children with high exposure were 55% (CI: 0.2-0.8, p=0.001), 31% (CI: 0.4-1.2, p=0.23), 67% (CI: 0.2-0.6, p<.0001), and 49% (CI: 0.3-0.8, p=0.001) less likely to have at least two servings of dairy a day at 6, 8, 10, and 12 years, respectively. Compared to children with low exposure to poverty, children with high exposure were 43% (CI: 0.3-0.9, p=0.02), 46% (CI: 0.3-0.9, p=0.02), 55% (CI: 0.3-0.8, p=0.003), and 47% (CI: 0.3-0.9, p=0.02) less likely to have at least two servings of vegetables a day at 6, 8, 10, and 12 years, respectively. Children at all ages with decreasing or increasing exposure to poverty were less likely to have at least two servings of vegetables a day, but the results were not statistically significant. Conclusion: Experiencing high exposure to poverty has consistent effects on food consumption throughout childhood. In addition, compared to children with low exposure to poverty, children with increasing or decreasing exposure were less likely to have at least 2 servings of fruits and vegetables a day, suggesting any exposure to poverty may have detrimental effects on consumption of selected foods.

Objective: to investigate the relationship between adiponectin level and metabolic syndrome (MS) after weight loss in patients with abdominal obesity (AO). Method: A 3-year randomized lifestyle intervention trial performed in 153 patients with AO, age 43,2±0,8 yrs, BMI 32,1±1,9 kg/m 2 . 74 patients keep hypocaloric diet (gr.1), 79 patients keep diet and performed aerobic exercise (gr.2). Adiponectin concentration, body mass (BM), waist circumference (WC), body fat (BF), BMI, the levels of BP, glucose, insulin, HOMA-IR, TC, HDL-C, LDL-C, TG, CRP were measured before and after a 3-years outpatient intervention program. Results. 100% patients with AO had some metabolic disorders and 38% had MS before the treatment. The adiponectin levels and others parameters didn’t differ between the groups before intervention (p>0,05). In 3 years 53 (71,6%) and 58 (73,4%) patients from 1 and 2 groups reduced weight. The rate of improving BM, BMI, BF, WC, HDL-C, TG and insulin was grater in patients gr.2 (p<0,05). The favorable dynamics of MS (MS didn’t appeared at the end of study or didn’t registered in patients who had it before) didn’t differ between the groups 1 and 2 (81,1% and 91,4%, p>0,05). The increasing of adiponectin level occurred more often in patients gr.2, than gr.1 (93,1% and 58,5%, p=0,001, respectively). Adiponectin level increased only in patients gr.2 (18,0±1,1mcg/ml and 23,8±1,3 mcg/ml, p= [[Unable to Display Character: &#1088;]]=0,0001), didn’t changed in gr.1 (p>0,05). It was established that in patients with combination of weight loss and increasing of adiponectin level favorable dynamics of MS occurred more often than in patients who lost weight without increasing of adiponectin level (91,7% and 69,2%, p=0,0001). In patients with favorable dynamics of MS increasing of adiponectin level had met more often, than in patients with unfavorable dynamics of MS (MS continued or appeared) (88,6% and 11,4%, p=0,0001). Increasing of adiponectin level associated with positive dynamics of the MS - OR=9,1 (4,0-20,6). Conclusion. Combination of weight loss and increasing of adiponectin level associated with favorable dynamics of the metabolic syndrome.

Scholars endeavor to understand the relationship between human evolution and climate change. This is particularly germane for Neanderthals, who survived extreme Eurasian environmental variation and glaciations, mysteriously going extinct during a cool interglacial stage. Here, we integrate weekly records of climate, tooth growth, and metal exposure in two Neanderthals and one modern human from southeastern France. The Neanderthals inhabited cooler and more seasonal periods than the modern human, evincing childhood developmental stress during wintertime. In one instance, this stress may have included skeletal mobilization of elemental stores and weight loss; this individual was born in the spring and appears to have weaned 2.5 years later. Both Neanderthals were exposed to lead at least twice during the deep winter and/or early spring. This multidisciplinary approach elucidates direct relationships between ancient environments and hominin paleobiology.

Obesity and its attendant disorders, such as type 2 diabetes, are global health problems. We previously reported that C75, an inhibitor of fatty acid synthase (FAS) and stimulator of carnitine palmitoyltransferase I (CPT I), caused anorexia and profound weight loss in lean and genetically obese mice. To approximate human obesity, we utilized a chronic C75 treatment model for diet-induced obese (DIO) mice. Chronic C75 treatment decreased food consumption and increased energy expenditure due to increased fatty acid oxidation in both DIO and lean mice. There was a substantial loss of adipose tissue and resolution of hepatic steatosis in C75-treated DIO mice. Analysis of changes in the expression of hypothalamic neuropeptides demonstrated that the reduced food consumption in C75-treated DIO mice was accompanied by an increase in cocaine and amphetamine-related transcript expression but not by changes in neuropeptide Y such as seen with acute C75 treatment of lean mice. Inhibition of FAS and stimulation of CPT I provide a means to achieve stable, sustained weight loss in DIO mice.
