Axillary buds of in vitro microshoots were successfully frozen at –196 °C by the one-step freezing method using the protective vitrification solution 2 (PVS2). Microshoots were taken from 11 transgenic lines and three wild type lines. Influence of different explant pretreatments were analyzed from the point of their influence towards recovery after cryopreservation. It was found out that the use of axillary buds as explants after removal of the apical one increases recovery on average by 8%. The cultivation on growth medium of higher density insignificantly raises the regenerants survival rate. Pretreatment of the osmotic fluid (OF) shows the greatest influence on the survival rate. It leads to the increase in survival rate by 20%. The cryopreservation technology providing regenerants average survival rate of 83% was developed. It was based on the experimental results obtained with explant pretreatment. Incubation time in liquid nitrogen did not affect the explants survival rate after thawing. After six months cryostorage of samples their genetic variability was analyzed. Six variable simple sequence repeat (SSR) loci were used to analyze genotype variability after the freezing-thawing procedure. The microsatellite analysis showed the genetic status identity of plants after cryopreservation and of the original genotypes. The presence of the recombinant gene in the transgenic lines after cryostorage were confirmed so as the interclonal variation in the growth rate under greenhouse conditions. The developed technique is recommended for long-term storage of various breeding and genetically modified lines of aspen plants, as it provides a high percentage of explants survival with no changes in genotype.

In 1974 three members of the transplant team from the Western Infirmary in Glasgow moved to the new medical school in Leicester. The initial experience with 33 patients transplanted in Glasgow was published in 1972 and this paper compares the results of that series with the first 21 patients grafted in Leicester. Despite improvements in tissue typing, better quality donor kidneys and fewer complications, there has been a failure to improve on the levels of graft survival. The overall one year graft survival rate in the Glasgow series was 79 per cent compared to 52 per cent in Leicester. In these two series the difference in results appears to be explained by blood transfusion. All the Glasgow patients had been poly-transfused but of the Leicester patients the 10 transfused pre-transplant had a one year graft survival of 90 per cent while in the 11 non-transfused patients the one year graft survival was only 18 per cent.

4129 Background: Combinations of gemcitabine (GEM)/5-FU, GEM/oxaliplatin (LOHP) or 5-FU/LOHP work synergistically in pancreatic and/or colorectal malignancies, and have non-overlapping safety profiles. This phase II-study was designed to evaluate the efficacy and safety of the triple combination GEM/LOHP/5-FU in patients (pts) with advanced or metastatic carcinoma of the gallbladder. Methods: One-stage, multicentre phase II study. Eligibility criteria: chemonaive pts with histologically proven advanced, recurrent or metastatic gallbladder carcinoma (ECOG 0–1; expected survival >3 months; measurable disease; adequate renal, hepatic and bone marrow function). According to the results of our previous phase I-study (Proc ASCO 2003, # 1298), pts were treated with GEM 900mg/m2 as a 30-min infusion, followed by LOHP 65 mg/m2 (2-hr infusion) after a 30 min rest and 5-FU 1500 mg/m2 (24-hr-infusion) on d 1, 8, every 3 weeks. Planned sample size: 35 response evaluable patients. The primary endpoint was tumor response, secondary endpoints were toxicity, median survival, the one-year-survival rate, clinical benefit and quality of life. Results: At time of abstract submission, median follow-up of 35 enrolled pts is 9.8 months. Pt. characteristics: m/f: 11/24, median age 61 (range 42–81), ECOG 0/1: 24/11 (69/31%) pts, locally advanced/metastatic disease 1/32 (3/91%) pts. Analysis of tumor response is still pending. Grade III/IV (NCI-CTC) toxicities occurred in 36/3% of 191 cycles and were: leucopenia 3/1%, neutropenia 4/1%, thrombocytopenia 4/1%, anemia 2/0%, nausea 1/0%, sensory neuropathy 4/0%, asthenia 1/0%, elevated bilirubin 2/0%, AP 4/0%, or elevated SGOT/SGPT 1/0%, edema 1/0%, infection 1/0%, dyspnoe 1/1%. Median survival of all pts is 9.9 months (95% CI: 7.5–11.5), the one-year-survival-rate is 30 % (95% CI: 16–47). Conclusions: GEM/LOHP/5-FU combination therapy is tolerated well in patients with gallbladder cancer. The promising survival data has to be confirmed in a phase III study. By a modification of the DeLury method an estimate is made of the stock of lemon sole (Parophrys vetulus) on the fishing grounds in Hecate Strait, British Columbia. The method is based on (1) the trend in catch of tagged fish per unit of effort in relation to accumulated catch of tagged fish, and (2) the trend in catch of untagged fish per unit of effort in relation to accumulated catch of untagged fish. At the start of the experiment 4.72 million pounds are estimated to have been present, while during the experiment 3.74 million pounds entered the area of fishing, 3.26 million pounds emigrated from it, and 2.54 million pounds were caught.A Petersen-type estimate based on the ratio of tagged to untagged fish was 4.70 million pounds present at the start of the experiment—practically identical with the one derived from catch-effort information. Total population for the year (1950) is estimated as 9.8–12.2 million pounds, including catch and possible emigration prior to the experiment, the stock on the fishing grounds at the start of the experiment, and the immigration subsequently.The average annual survival rate of age VII – age IX lemon soles in Hecate Strait has decreased from 0.770 to 0.614 during the period of growth of the fishery, 1944–1951. From this difference the average rate of exploitation is estimated as 16 to 20 per cent and the average population as 8.9 to 11.5 million pounds. Since this range is nearly the same as the range described above, it is concluded that almost the whole of the Hecate Strait stock was accessible to fishing in 1950, which was a year of unusually high production.

Yin-Deficiency (YD), representing a status of the human body under lack of nutrition and fluid in traditional Chinese medicine, is commonly seen in late stage of cancer patients. It is not known whether the severity of YD related symptoms/signs can predict the survival rate of cancer patients. This study evaluated the distribution of Yin-deficiency symptoms/signs (YDS) in cancer patients with YD, and investigated whether the severity of YDS can predict the survival rate of cancer patients with YD. From 5 January 2007 to 5 May 2007, we selected 43 cancer patients with diagnosis of YD from hospitalized patients and outpatients. The severity of YD was evaluated by a questionnaire. We further estimated the cumulative probabilities of the survival rates over 4 months since the start of study by the Kaplan-Meier product-limit method, and compared the differences among groups with various severities in each symptom/sign with the use of the log-rank test. The results revealed that, the 3 most common YDS were sleeplessness with annoyance, less or non-coated tongue with or without redness and dry mouth. In the survival rate analysis, only 2 parameters, rapidly small pulse (p = 0.002) and less-or non-coated tongue with paleness (p = 0.017), were found to be related to the decrease of cancer patients with YD. This suggests that, both rapidly small pulse and less-or non-coated tongue without redness may be used as predictors for the estimation of survival rate in cancer patients with YD.

<p>More of waste than pond aquacultutre system produced, will increase sedimentation in the bottom.  Ammonium and nitrite compounds are other forms of inorganic nitrogen in the pond. Nitrogen anorganic consist of ammonia, ammonium, nitrit, nitrat and nitrogen. Degradation of process metabolic biota culture waste can biologically be nitrat compound one of the forms that are not toxic in the nitrification process.  Five process of nitrogen biogeochemical cycle in the container cultivation is the amonification, nitrification, nitrogen assimilation, denitrification and nitrogen fixation. Nitrogen is the one of the compound in the overlay/ top stratification sediment.  Improvement of speed degradation will be success if the pond bottom on aerobic condition. Survival rate of tiger shrimp in the laboratory scale can be increase by administration of nitrification and denitrification bacteria should not just in the water kolom of pond engineering but also at the bottom pond layer at the preparation step.  Depht of the sediment 15 cm in day zero, intensive pond have been produced of nitrit and ammonium with the producing bacteria.  Application of nitrification and denitrification bacteria in the sediment and water coloum can be performed as the measurenment and evaluation nitrit, nitrat and ammonium abudance.</p> <p>Key words:  ponds, nitrogen inorganic, nitrification, denitrification</p> <p><em> </em></p> <p>ABSTRAK</p> <p>Semakin banyak limbah kegiatan yang dihasilkan dalam sistem budidaya tambak, akan meningkatkan sedimentasi dalam dasar tambak.  Senyawa amonium dan nitrit merupakan bentuk lain dari nitrogen anorganik dalam tambak. Nitrogen anorganik terdiri terdiri dari amonia (NH<sub>3</sub><sup>-</sup>), amonium (NH<sub>4</sub><sup>+</sup>), nitrit (NO<sub>2</sub><sup>-</sup>), dan nitrogen (N<sub>2</sub>). Secara biologis, proses perombakan sisa metabolisme biota budidaya dapat menjadi nitrat (NO<sub>3</sub>), suatu bentuk yang tidak berbahaya dalam proses nitrifikasi.  Lima proses siklus biogeokimia nitrogen yang terjadi di wadah budidaya adalah amonifikasi, nitrifikasi, asimilasi nitrogen, denitrifikasi, dan fiksasi nitrogen. Nitrogen merupakan senyawa yang biasanya terletak di lapisan paling atas dalam sedimen.  Peningkatan kecepatan degradasi akan dapat dicapai jika sedimen berada dalam kondisi aerobik. Tingkat kelangsungan hidup udang pada uji skala laboratorium memperlihatkan bahwa penambahan bakteri nitrifikasi dan denitrifikasi terseleksi dapat meningkatkan kelangsungan hidup udang windu Aplikasi pemberian bakteri nitrifikasi dan denitrifikasi tidak seharusnya hanya dalam air saja, namun juga dalam pengolahan tanah dasar tambak.  Pada kedalaman sedimen 15 cm hari ke-0, tambak intensif telah terdapat bakteri penghasil nitrit dan amonium dan kelimpahannya semakin meningkat seiring dengan bertambahnya umur udang.  Aplikasi pemberian bakteri nitrifikasi dan denitrifikasi di sedimen dan kolom perairan dapat dilakukan seiring dengan pengukuran dan evaluasi kondisi nitrit, nitrat dan ammoniumnya.  Secara alami,  dalam kolom perairan telah terdapat senyawa nitrit, nitrat dan amonium, seberapapun itu perlu diimbangi dengan kebijakan dalam pemberian bakteri dari luar.</p> <p>Kata kunci: tambak, nitrogen anorganik, nitrifikasi, denitrifikasi</p>

BackgroundInfections remain a major cause of morbidity and mortality in kidney transplant (KT) recipients. This study was performed to identify the overall prevalence of early infections, prevalence of carbapenem-resistant Klebsiella pneumoniae (CRKP) infection after KT, one-year postoperative mortality in patients with early infections and risk factors for CRKP infections.MethodsWe conducted a retrospective study of all patients who received KT in our hospital between January 2017 and December 2019. We evaluated the demographic, clinical, infection characteristics and the one-year postoperative outcomes.ResultsAmong the 419 patients who received KT between January 2017 and December 2019, 150 patients had at least one infection within 90 days after KT. The total prevalence of early infections was 36.1% (150/415), the prevalence of early CRKP infections was 10.4% (43/415), and the one-year postoperative mortality was 15.3% (23/150) in patients with early infections. The risk factors independently related to one-year postoperative mortality were mechanical ventilation (MV) &gt; 48 h (Odds ratio (OR)= 13.879, 95%Confidence interval (CI): 2.265~85.035; P=0.004) and CRKP infection (OR=6.751, 95% CI: 1.051~43.369; P =0.044). MV&gt; 48 h was independently related to CRKP infection (OR=3.719, 95% CI: 1.024~13.504; P=0.046). Kaplan-Meier survival curves showed that the one-year survival rate of patients infected with CRKP in the early postoperative stage was significantly lower than that of uninfected patients.ConclusionsIn general, the prevalence of early infections after KT is high, and CRKP infection is closely correlated with poor prognosis. The effective prevention and treatment of CRKP infection is an important way to improve the one-year survival rate after KT.
