SummaryAn improved method for the preparation of bovine α-thrombin is described. The procedure involves the activation of partially purified prothrombin with tissue thromboplastin followed by chromatography on Sulfopropyl-Sephadex C-50. The purified enzyme is homogeneous on polyacrylamide discontinuous gel electrophoresis and has a specific activity toward fibrinogen of 2,200–2,700 N.I.H. U/mg. Its stability on storage in liquid media is dependent on both ionic strenght and temperature. Increasing ionic strength and decreasing temperature result in optimal stability. The denaturation of α-thrombin by guanidine hydrochloride was found to be a partially reversible process with the renatured species possessing properties similar to “aged” thrombin. In addition, the catalytic properties of a-thrombin covalently attached to agarose gel beads were also examined. The activity of the immobilized enzyme toward fibrinogen was affected to a much greater extent than was the hydrolysis of low molecular weight, synthetic substrates.

Some physical and catalytic properties of cerium dioxide-nickel oxide two-component catalysts have been studied over the entire composition region, employing the decomposition of hydrogen peroxide in aqueous solution as a model catalytic process. The two oxides have been found to affect each other, particularly for NiO contents of 9.1 and 96.7 mol%; the mutual influencing, the nature of which in the conditions applied remains unaffected by heat treatment of the sample or by its exposition to ionizing radiation, is manifested by the nonmonotonic dependences of the oxidation power and of the specific activity of the catalysts on their composition. This can be interpreted in terms of the concept of bivalent catalytic centres, assuming that for nickel oxide the centres consist of Ni2+-Ni3+ ion pairs, for cerium dioxide they consist of Ce3+-Ce4+ ion pairs, and that in the region of the mutual influencing , Ni2+-Ce4+ ion pairs play a major role. Within the scope of this concept, the increase in the oxidation power of all the catalysts in question and a simultaneously decrease in the specific activity of the pure nickeloxide exposed to ionizing radiation can be explained in terms of the ionization effect.

Catalytic properties of GH30 xylanases belonging to subfamilies 7 and 8 were compared on glucuronoxylan, modified glucuronoxylans, arabinoxylan, rhodymenan, and xylotetraose. Most of the tested bacterial GH30-8 enzymes are specific glucuronoxylanases (EC 3.2.1.136) requiring for action the presence of free carboxyl group of MeGlcA side residues. These enzymes were not active on arabinoxylan, rhodymenan and xylotetraose, and conversion of MeGlcA to its methyl ester or its reduction to MeGlc led to a remarkable drop in their specific activity. However, some GH30-8 members are nonspecific xylanases effectively hydrolyzing all tested substrates. In terms of catalytic activities, the GH30-7 subfamily is much more diverse. In addition to specific glucuronoxylanases, the GH30-7 subfamily contains nonspecific endoxylanases and predominantly exo-acting enzymes. The activity of GH30-7 specific glucuronoxylanases also depend on the presence of the MeGlcA carboxyl, but not so strictly as in bacterial enzymes. The modification of the carboxyl group of glucuronoxylan had only weak effect on the action of predominantly exo-acting enzymes, as well as nonspecific xylanases. Rhodymenan and xylotetraose were the best substrates for exo-acting enzymes, while arabinoxylan represented hardly degradable substrate for almost all tested GH30-7 enzymes. The results expand current knowledge on the catalytic properties of this relatively novel group of xylanases.

With the help of bio-molecule l-lysine, a seeded growth method has been developed and Pt–ZnO porous nanoribbons have been successfully obtained. For the catalytic applications of CO oxidation and the photocatalytic degradation of MO, the hybrid nanomaterial exhibits remarkably enhanced catalytic properties.

Abstract Background β-Cyanoalanine synthase plays essential roles in germinating seeds, such as in cyanide homeostasis. Methods β-Cyanoalanine synthase was isolated from sorghum seeds, purified using chromatographic techniques and its biochemical and catalytic properties were determined. Results The purified enzyme had a yield of 61.74% and specific activity of 577.50 nmol H2S/min/mg of protein. The apparent and subunit molecular weight for purified β-cyanoalanine synthase were 58.26±2.41 kDa and 63.4 kDa, respectively. The kinetic parameters with sodium cyanide as substrate were 0.67±0.08 mM, 17.60±0.50 nmol H2S/mL/min, 2.97×10−1 s−1 and 4.43×102 M−1 s−1 for KM, Vmax, kcat and kcat/KM, respectively. With L-cysteine as substrate, the kinetic parameters were 2.64±0.37 mM, 63.41±4.04 nmol H2S/mL/min, 10.71×10−1 s−1 and 4.06×102 M−1 s−1 for KM, Vmax, kcat and kcat/KM, respectively. The optimum temperature and pH for activity were 35°C and 8.5, respectively. The enzyme retained more than half of its activity at 40°C. Inhibitors such as HgCl2, EDTA, glycine and iodoacetamide reduced enzyme activity. Conclusion The biochemical properties of β-cyanoalanine synthase in germinating sorghum seeds highlights its roles in maintaining cyanide homeostasis.

Two samples of Cr2O3/Al2O3 were prepared by mixing a known mass of finely powdered Al(OH)3 with a calculated amount of CrO3 solid followed by drying at 120°C and calcination at 400°C. The amounts of chromium oxide employed were 5.66 and 20 mol% Cr2O3, respectively. The calcined solid specimens were then treated with different doses of γ-rays (20–160 Mrad). The surface and catalytic properties of the different irradiated solids were investigated using nitrogen adsorption at −196°C and the catalysis of CO oxidation by O2 at 300–400°C. The results revealed that γ-rays brought about a slight decrease in the BET surface area, SBET (15%), and in the total pore volume, Vp (20%), of the adsorbent containing 5.66 mol% Cr2O3. The same treatment increased the total pore volume, Vp (36%), and the mean pore radius, r̄ (43%), of the other adsorbent sample without changing its BET surface area. The catalytic activities of both catalyst samples were found to increase as a function of dose, reaching a maximum value at 80–160 Mrad and 40 Mrad for the solids containing 5.66 and 20 mol% Cr2O3, respectively. The maximum increase in the catalytic activity measured at 300°C was 59% and 100% for the first and second catalyst samples, respectively. The induced effect of γ-irradiation on the catalytic activity was an increase in the concentration of catalytically active sites taking part in chemisorption and in the catalysis of CO oxidation by O2 without changing their energetic nature. This was achieved by a progressive removal of surface hydroxy groups during the irradiation process.
