Bacterial resistance to antimicrobials is recognized by the WHO as a major health threat of the 21st century. Antimicrobial resistance threatens the effective prevention and treatment of an ever-increasing range of infections. Infections such as tuberculosis and septicaemia which decimated populations are once again responsible for mortality in epidemic proportions. Bacterial resistance is driven by the continued use of antimicrobials and it is unlikely that the danger of resistance can be effectively diminished by the discovery of new antimicrobials. The problem lies with the patent duration (20 years) and billion dollar investments of the pharmaceutical companies in producing a new antimicrobial. We have used, or are using, our so-called drugs of last resort. There is nothing left in the armamentarium and investments in developing new antimicrobials are missing, we are moving into the post-antimicrobial era.

Apropos to the article by Dr Bali, titled “Mupirocin resistance in clinical isolates of methicillin-sensitive and resistant Staphylococcus aureus in a tertiary care centre of North India” (1), the authors have raised important issue of emerging antimicrobial resistance (AMR). Antimicrobial resistance is an increasingly serious threat to global public health that requires action across all government sectors and society. As per WHO, AMR lurks the effective prevention and management of an ever-increasing spectrum of infections caused by bacteria, parasites, fungi and viruses. Novel resistance mechanisms are emerging and spreading globally, threatening the man’s ability to treat common infectious diseases.

Conclusion: Stroke is a complex disease caused by genetic and environmental factors, and its etiological mechanism has not been fully clarified yet, which brings great challenges to its effective prevention and treatment. MAPK signaling pathway regulates gene expression of eukaryotic cells and basic cellular processes such as cell proliferation, differentiation, migration, metabolism and apoptosis, which are considered as therapeutic targets for many diseases. Up to now, mounting evidence has shown that MAPK signaling pathway is involved in the pathogenesis and development of ischemic stroke. However, the upstream kinase and downstream kinase of MAPK signaling pathway are complex and the influencing factors are numerous, the exact role of MAPK signaling pathway in the pathogenesis of ischemic stroke has not been fully elucidated. MAPK signaling molecules in different cell types in the brain respond variously after stroke injury, therefore, the present review article is committed to summarizing the pathological process of different cell types participating in stroke, discussed the mechanism of MAPK participating in stroke. We further elucidated that MAPK signaling pathway molecules can be used as therapeutic targets for stroke, thus promoting the prevention and treatment of stroke.

UNSTRUCTURED Abstract This scientific perspective of mode of transmission of COVID-19 is to aid scientific community in generating hypothesis. The inadequate evidence on SARS-COV-2 transmission has hindered development of effective prevention strategy and resulted in continues pandemic of the COVID-19. Therefore, in this perspective existing evidences are discussed, hypothesis are generated regarding COVID-19 mode of transmission and recommendations are forwarded based on existing body of knowledge. Two meter (2m) physical distance is not completely safe even for large droplets and wearing face mask is a key in prevention of SARS-COV-2 in public areas and confined space.

Mental health treatment, child protection and forensic services for criminality need major reorganisation in conceptualisation and service provision. This need results from the failure of current diagnostic methods, administrative organisations and treatment approaches to reduce the prevalence of mental illness, child maltreatment or criminal behaviour. We propose that defining these problems as individual deficits and responding to them by category of harm (to self, progeny and others, respectively) stands in the way of effective prevention and treatment. We address four topics: (1) the common basis of all of these problems in unprotected and uncomforted exposure to danger, (2) the developmental process of psychological maladaptation that occurs interpersonally in endangered families, (3) the behavioural expression of psychological development as protective strategies that can be adaptive or maladaptive, depending upon the context in which they are used, and (4) proposals for systemic change that could improve prevention and treatment. These proposals include using functional formulations to guide treatment planning, single portal entry to assessment and services, integrated universal transdisciplinary training followed by specialisation for all mental health professionals, delivering customised treatment through transitional attachment relationships and consolidating disparate disciplines in ‘departments of human adaptation’.

AbstractTransition metal carbides and nitrides (MXenes), which comprise a rapidly growing family of two-dimensional materials, have attracted extensive attention of the scientific community, owing to its unique characteristics of high specific surface area, remarkable biocompatibility, and versatile applications. Exploring different methods to tune the size and morphology of MXenes plays a critical role in their practical applications. In recent years, MXenes have been demonstrated as promising nanomaterials for cancer therapy with substantial performances, which not only are helpful to clarify the mechanism between properties and morphologies but also bridge the gap between MXene nanotechnology and forward-looking applications. In this review, recent progress on the preparation and properties of MXenes are summarized. Further applications in cancer therapy are also discussed. Finally, the current opportunities and future perspective of MXenes are described.

Background: Acanthamoeba castellanii is a pathogenic free-living amoeba responsible for blinding keratitis and fatal granulomatous amoebic encephalitis. However, treatments are not standardized but can involve the use of amidines, biguanides, and azoles. Objectives: The aim of this study was to synthesize a variety of synthetic tetrazole derivatives and test their activities against A. castellanii. Methods: A series of novel tetrazole compounds were synthesized by one-pot method and characterized by NMR and mass spectroscopy. These compounds were subjected to amoebicidal, and cytotoxicity assays against A. castellanii belonging to the T4 genotype and human keratinocyte skin cells respectively. Additionally, reactive oxygen species determination and electron microscopy studies were carried out. Furthermore, two of the seven compounds were conjugated with silver nanoparticles to study their antiamoebic potential. Results: A series of seven tetrazole derivatives were synthesized successfully. The selected tetrazoles showed anti-amoebic activities at 10µM concentration against A. castellanii in vitro. The compounds tested caused increased reactive oxygen species generation in A castellanii, and significant morphological damage to amoebal membranes. Moreover, conjugation of silver nanoparticles enhanced antiamoebic effects of two tetrazoles. Conclusions: The results showed that azole compounds hold promise in the development of new formulations of anti-Acanthamoebic agents.
