1. trans-4-Hydroxy-l-proline was found to occur in the bound state in the leaves of sandal (Santalum album L.), in which large amounts of free cis-4-hydroxy-l-proline are also present. 2. Bound trans-4-hydroxy-l-proline was present, associated mainly with a `wall' fraction and a `soluble' fraction roughly in equal proportions. 3. Bound proline is present only in small amounts in the `soluble' fraction but is mostly associated with the `wall' fraction and the other sedimented fractions. 4. In the free amino acid fraction more than 98% of the hydroxyproline had the cis-configuration, whereas in the `wall' and `soluble' fractions more than 90% of the bound hydroxyproline was in the trans-configuration. 5. Various extraction procedures indicated heterogeneity of the hydroxyproline-containing components. Hot 5% (w/v) trichloroacetic acid extracts about 25% of hydroxyproline and m-NaOH extracts an additional 25%. 6. Incorporation of [14C]proline into the bound hydroxyproline was demonstrated. The hydroxyproline component of the `soluble' fraction does not appear to be the precursor of that of the `wall' fraction.

Abstract Latex rubber which has been purified to the point at which it contains an insignificant amount of nitrogen can be separated by fractional dissolution in a mixture of petroleum and acetone into a series of hydrocarbon fractions of decreasing solubility and increasing molecular magnitude. All these fractions except the highest are soluble in petroleum and in benzene. Crepe rubber, on the other hand, appears invariably to contain a small, most-soluble fraction of oxygenated rubber, and a small similar quite insoluble fraction of material of high molecular weight. Between these extremes the rubber can be divided into fractions of increasing molecular weight, although, up to the present, about 70 per cent of the total rubber has appeared in a single fraction. It may be possible later, by judicious choice of another pair of solvents, to resolve this major fraction into a series of subfractions. Kemp and Peters refer to the effect of polar nonsolvents in reducing the viscosity of rubber solutions and also in assisting to bring gel rubber into solution, phenomena to which the polar molecules conceivably contribute by countering the forces of association between the rubber molecules. The present series of fractionations was conducted throughout in the presence of a polar nonsolvent (acetone), and hence may be considered to approach towards a separation of true rubber molecules as distinct from molecular aggregates. It is found, however, that, whereas the more soluble fractions of acetone-extracted crepe rubber contain small proportions of nitrogen, the least soluble fractions contain substantial proportions. Any effect which the nitrogenous material may have in assisting to link together hydrocarbon molecules to which it is attached, i. e., in contributing to the high-molecular condition of a portion of natural rubber, remains at present uncertain in character. The fractions of rubber, and especially the higher ones, show a strong tendency to become insoluble when they have once been freed from the last traces of solvent. It seems doubtful whether the decreased solubility is due to oxygen as it would require to be effective at exceedingly low concentrations.

L-Methionine-1-14C was fed through the roots to 6-day-old pea seedlings. After 10 h over 80% of the absorbed radioactivity was recovered in the free amino acid fraction, 8% in the extracted proteins, 5% as carbon dioxide, and 2.2% in the organic acid fractions. Analysis of the amino acids revealed that only 24% of the total activity was recovered as methionine, 40% as homoserine, 10% as isoleucine, and smaller but significant amounts of aspartate, O-acetylhomoserine, and cystathionine. Dilution values strongly imply that aspartate is not an intermediate in the extensive conversion of methionine to homoserine; rather a more direct pathway is involved.

Amino acids, both free and combined as protein, and some other nitrogenous constituents of Calanus finmarchicus have been examined. Seventy-six per cent of the total nitrogen in C. finmarchicus was present in protein amino acids, 14% in the free amino acid fraction, 6% in trimethylamine oxide, and 1·5% in betaine. These findings are discussed in relation to previous work on nitrogenous constituents of Calanus.The free amino acid fraction of Calanus is compared with that of higher Crustacea and it is suggested that this fraction may be important in the adaptation of the animal to dilute sea water.Amino acid nitrogen made up 40% of the total nitrogen of particulate material from Clyde sea water. This finding is compared with other recent analyses of the phytoplanktonic nitrogen and total particulate nitrogen of inshore waters.

Flavor instability of beer is affected by the rise of aroma-active aldehydes during aging. Aldehydes can be either released from bound-state forms or formed de novo. This second part of our study focused on the de novo formation of aldehydes during the Maillard reaction, Strecker degradation, and oxidation reactions. Key precursor compounds for de novo pathways are free amino acids. This study varied the potential for reactions by varying free amino acid content in fresh beer using different proteolytic malt modification levels (569–731 mg/100 g d. m. of soluble nitrogen) of the used malt in brewing trials. Overall, six pale lager beers were produced from three malts (different malt modification levels), each was made from two different barley varieties and was naturally and forcibly aged. It was found that higher malt modification levels in fresh beer and during beer aging increased amino acid and dicarbonyl concentrations as aging precursors and Strecker aldehyde contents as aging indicators. Dicarbonyls were degraded during aging. Advanced glycation end products as possible degradation products showed no consistent formation during aging. Therefore, Strecker reactions were favored during beer aging. No alternative oxidative formation of Strecker aldehydes from their corresponding alcohols could be confirmed. Along with the preceding part one of our investigation, the results of this study showed that de novo formation and release occur simultaneously. After 4 months of natural aging, aldehyde rise is mainly accounted for by de novo formation.

Pembenihan ikan kerapu dihadapkan pada rendahnya sintasan, terutama fase awal hidupnya. Rendahnya sintasan diduga berhubungan dengan belum berkembangnya organ pencernaan larva dan tidak tersedianya pakan yang siap diserap tubuh. Pemberian asam amino bebas seperti taurin dan glutamin melalui rotifer diharapkan dapat meningkatan ketersediaan nutrien yang dapat diserap dengan cepat oleh larva, sehingga sintasan larva tinggi dan dapat tumbuh lebih cepat. Penelitian bertujuan mengukur pengaruh glutamin atau taurin terhadap sintasan dan pertumbuhan larva kerapu bebek Cromileptes altivelis. Larva umur 1 hari (10 ekor.L-1) dipelihara dalam bak fiberglass volume 500 L yang diisi air laut sampai 330 L. Larva diberi pakan rotifer diperkaya 0,5 g glutamin, 0,5 g taurin, atau tanpa glutamin dan taurin. Larva dipelihara selama 16 hari dengan pemberian pakan 2 kali  per hari. Satu jam setelah pemberian pakan dilakukan pengamatan terhadap jumlah rotifer dalam saluran pencernaan larva ikan. Hasil penelitian menunjukkan jumlah rotifer dalam saluran pencernaan larva meningkat seiring dengan bertambahnya umur. Namun, jumlah rata-rata rotifer yang dikonsumsi larva pada perlakuan 0,5 g glutamin atau 0,5 g taurin lebih banyak dibanding perlakuan tanpa taurin dan glutamin. Larva yang diberi rotifer hasil pengkayaan dengan 0,5 g glutamin atau 0,5 g taurin memiliki sintasan yang tidak berbeda (P>0,05), masing-masing 19,2±2,25%, dan 24,5±1,20%, dan secara signifikan lebih banyak dibanding larva di perlakuan tanpa glutamin dan taurin yang nilainya 13,0±1,70%. Larva yang diberi rotifer hasil pengkayaan dengan 0,5 g taurin secara signifikan lebih panjang (5,5±0,18 mm) dari ikan di perlakuan 0,5 g glutamin (5,3±0,12 mm). Demikian juga, larva di perlakuan glutamin lebih panjang dari pelakuan tanpa glutamin dan taurin (5,0±0,16 mm). Pengkayaan rotifer menggunakan asam amino bebas taurin atau glutamin dapat meningkatkan sintasan dan pertumbuhan larva kerapu bebek. Namun, taurin lebih baik dibanding glutamin karena memberi efek ke pertumbuhan ikan yang lebih cepat. Kata kunci: taurin, glutamin, kerapu bebek, Cromileptes altivelis. The hatchery production of grouper faced to the problem of low surival rate, especially during the first feeding period. During this period, the digestive organ of larvae was underdeveloped, and the nutrient in live food was not readily absorbed by larvae. Feeding on free amino acids, such as taurine or glutamine, enriched rotifer would be enhanced the nutrient readily absorbed thereby resulting to the improvement of growth and survival rate of larvae. A duplicate experiment was conducted to evaluate the effect of rotifer enriched with free amino acid of glutamine or taurine, on the growth and survival rate of larval grouper Cromileptes altivelis. One day old larvae were cultured in 500 l fiberglass containing 330 l of sea water with a density of 10 ind.L-1. Larvae were fed on rotifer enriched with either 0.5 g glutamine, 0.5 g taurine, or without glutamine nor taurine. Larvae were fed on rotifer twice a day at 08.00 and 14.00, and cultured for 16 days. One h after feeding, rotifer content in the gut of larvae were observed. Results showed that larvae fed on rotifer enriched with 0.5 g taurine or 0.5 g glutamie had higher rotifer content in the gut than that in the other group. Larvae fed on rotifer enriched with 0.5 g glutamine or 0.5 g taurine had the same survival rate (P>0.05), 19.2±2.25% and 24.5±1.20%, respectively; these values were significantly higher than that in the other group which was 13.0±1.70%.  The total length of larvae fed on rotifer enriched with 0.5 g taurine (5.5±0.18 mm) significantly was the biggest compared than others; while those fed on rotifer enriched with 0.5 g glutamine (5.3±0.12 mm) was bigger than fish fed on rotifer without glutamine nor taurine (5.0±0.16 mm). Thus, it can be concluded that survival rate and growth of larvae were improved when feeding on rotifer enriched with taurine or glutamine. Nevertheless, taurine was more efective than glutamine due to its effect resulted the highest growth performance of larval grouper. Keywords: taurine, glutamine, grouper, Cromileptes altivelis

The contractility of the thread model prepared from the KCl-soluble proteins of the egg and in vivo factors for the contraction are investigated in Hemicentrotus, Anthocidaris, and Pseudocentrotus eggs. The contractility of the thread model induced by metal ions or cystine changes during development in the characteristic pattern of high at the metaphase and low at the monaster and the interkinetic stages. The change in contractility is paralleled by the change in the —SH content of the protein. The water-soluble fraction of the eggs has activity in causing contraction of the thread model. This activity changes during development in the same way as the contractility itself. The contraction of the thread induced by the water-soluble fractions is accompanied by a decrease in the —SH content of the thread. The activity of the water-soluble fraction in inducing the contraction is proportional to its ability to decrease the number of —SH groups. On boiling, the activity is largely destroyed. The activity is due to two components, one being non-dialyzable and the other dialyzable. Separately each component has little effect, but when mixed, the activity of the original sample is completely restored.

The effect of the protein precipitants acetone, acetonitrile, perchloric acid and trichloroacetic acid on free amino acid concentrations in supernatants from ovine plasma and bovine serum albumin solutions was determined. The organic precipitants decreased (P < 0.05) free amino acid concentrations in a protein concentration dependent manner while the acid precipitants had no effect. The effect of storage at subzero temperatures (-5°C, -12°C, and -20°C) on hake ( Merluccius merluccius L.) muscle free-amino acid fraction was evaluated. A significant increase in free aspartic acid, serine, threonine, arginine, β-alanine, tyrosine, methionine, valine, phenylalanine, isoleucine, leucine, and lysine was found at -5°C, whereas at -12°C, a significant decrease in free glutamic acid, glycine, methyl-histidine, β-alanine, taurine, alanine, and leucine was the most noticeable. No changes in the free amino acid fraction were observed at -20°C. Activity of different kind of enzymes, aminopeptidases, aminoacid deaminases, and decarboxylases might be involved in the changes observed at -5°C and -12°C.
