Abstract Pot experiments were conducted in 2017, 2019, and 2020 to examine the effects of potassium nutrition on the nutritional components of vegetable soybeans with different K efficiency at immature and mature stages. Two vegetable soybean varieties with higher K efficiency and two varieties with lower K efficiency were studied in the low available K soil under the condition of no K and normal K fertilization. The results indicated that almost all nutritional components in vegetable soybean were affected by K, genotypes, inter-annual differences, and their interactions. In general, no K fertilization increased protein and amino acid concentrations but decreased oil, soluble sugar, sucrose, K, Mg, and Fe concentrations in immature and mature vegetable soybean. The sensitivity of nutritional components to K nutrition differed among varieties. For instance, K high-efficiency varieties generally exhibited higher protein and amino acid concentrations without K application. K high-efficiency vegetable soybeans are low-K tolerance varieties to isoflavones. The results of this study provide insights for high yield and quality vegetable soybean breeding against soil K deficiency.

The combined intake of citrulline (CIT) and leucine (LEU) can stimulate protein synthesis. Therefore, this study aimed to investigate the effect of combined intake of CIT and LEU accompanied by exercise for 20 weeks on body composition, physical activity (PA), and amino acid concentrations in older Japanese women with low body mass index (BMI) (16 to 21 kg/m2) using a randomized, double-blind, placebo-controlled design. The supplement was administered twice a day for 20 weeks (Ex (exercise) + CIT·LEU group, n = 10: mainly 0.8 g CIT and 1.6 g LEU; Ex + Placebo group, n = 13: mainly 3.5 g carbohydrate). Additionally, both groups exercised (weight-bearing exercise, square stepping exercise) once a week for 75 min. Body composition, PA, and amino acid concentrations in the plasma were measured. Body weight, BMI, body mass, household PA, total PA, and phenylalanine significantly increased in the Ex + CIT·LEU group (p < 0.05) post intervention. This study suggests that the combined intake of CIT and LEU accompanied by exercise can improve body weight, BMI, body mass, and PA in older women with low BMI, which may prevent sarcopenia and frailty.

Abstract Plant-based proteins are considered to be less effective in their capacity to stimulate muscle protein synthesis when compared with animal-based protein sources, likely due to differences in amino acid contents. We compared the postprandial muscle protein synthetic response following the ingestion of a lysine-enriched plant-based protein product with an isonitrogenous amount of chicken. Twenty-four men (age: 24±5 y; BMI: 22.9±2.6 kg·m−2) participated in this parallel, double-blind, randomised controlled trial and consumed 40 g protein as a lysine-enriched wheat and chickpea protein product (Plant, n=12) or chicken breast fillet (Chicken, n=12). Primed, continuous intravenous L-[ring-13C6]-phenylalanine infusions were applied while repeated blood and muscle samples were collected over a 5h postprandial period to assess plasma amino acid responses, muscle protein synthesis rates, and muscle anabolic signalling responses. Postprandial plasma leucine and essential amino acid concentrations were higher following Chicken (P<0.001), while plasma lysine concentrations were higher throughout in Plant (P<0.001). Total plasma amino acid concentrations did not differ between interventions (P=0.181). Ingestion of both Plant and Chicken increased muscle protein synthesis rates from post-absorptive: 0.031±0.011 and 0.031±0.013 to postprandial: 0.046±0.010 and 0.055±0.015%∙h−1, respectively (P-time<0.001), with no differences between Plant and Chicken (P-interaction=0.068). Ingestion of 40 g protein in the form of a lysine-enriched plant-based protein product increases muscle protein synthesis rates to a similar extent as an isonitrogenous amount of chicken in healthy, young men. Plant-based protein products sold as meat replacers may be as effective as animal-based protein sources to stimulate postprandial muscle protein synthesis rates in healthy, young individuals.

The objective of this study was to investigate the impacts of dietary crude protein (CP), fishmeal and sorghum on nutrient utilisation, digestibility coefficients and disappearance rates of starch and protein, amino acid concentrations in systemic plasma and their relevance to growth performance of broiler chickens using the Box-Behnken response surface design. The design consisted of three factors at three levels including dietary CP (190, 210, 230 g/kg), fishmeal (0, 50, 100 g/kg), and sorghum (0, 150, 300 g/kg). A total of 390 male, off-sex Ross 308 chicks were offered experimental diets from 14 to 35 days post-hatch. Growth performance, nutrient utilisation, starch and protein digestibilities and plasma free amino acids were determined. Dietary CP had a negative linear impact on weight gain where the transition from 230 to 190 g/kg CP increased weight gain by 9.43% (1835 versus 2008 g/bird, P = 0.006). Moreover, dietary CP linearly depressed feed intake (r = -0.486. P < 0.001). Fishmeal inclusions had negative linear impacts on weight gain (r = -0.751, P < 0.001) and feed intake (r = -0.495, P < 0.001). There was an interaction between dietary CP and fishmeal for FCR. However, growth performance was not influenced by dietary inclusions of sorghum. Total plasma amino acid concentrations were negatively related to weight gain (r = -0.519, P < 0.0001). The dietary transition from 0 to 100 g/kg fishmeal increased total amino acid concentrations in systemic plasma by 35% (771 versus 1037 μg/mL, P < 0.001). It may be deduced that optimal weight gain (2157 g/bird), optimal feed intake (3330 g/bird) and minimal FCR (1.544) were found in birds offered 190 g/kg CP diets without fishmeal inclusion, irrespective of sorghum inclusions. Both fishmeal and sorghum inclusions did not alter protein and starch digestion rate in broiler chickens; however, moderate reductions in dietary CP could advantage broiler growth performance.

<p>For mammalian oocyte maturation it is important for the oocyte to communicate with the surrounding somatic cells in a bidirectional manner. In part, this is achieved through gap junction connections between cumulus cells and the oocyte. These gap junctions permit the transfer of essential metabolites, nucleotides, amino acids and ions from the cumulus cells to the oocyte in order for growth and development to occur. It is hypothesized that one reason for post-partum infertility in dairy cows is poor oocyte quality due to an inappropriate nutrient microenvironment in the developing antral follicle. In turn, this might compromise the nutrient and/or energy delivery by cumulus cells to the oocyte and thus affect oocyte quality. The objectives of this study were to: (1) -develop a reliable in vitro measure of gap junction activity for bovine cumulus–oocyte-complexes (COC) using a fluorescent (calcein) dye technique, and (2)- assess the effects of different amino acid compositions, cholesterol and fatty acid concentrations on gap junction activity in COC. The development and validation of the bovine COC gap junction assay was undertaken using a standard commercial medium (M199). Thereafter, media were prepared representing the amino acid concentrations in follicular fluid of both non-lactating and lactating dairy cows as measured in a NZ pastoral situation. These media formed the basis for subsequent measures of COC gap junction activity with differing concentrations of cholesterol and the essential fatty acids- oleic acid, stearic acid and palmitic acid. The major findings from this study were that the different amino acid concentrations typical of that in the follicular fluid of the non-lactating and lactating dairy cow and M199 do not significantly affect the gap junction activity in COC. Given that the amino acid compositions in lactating and non-lactating cows were approximately 50% of those in M199; this indicates that the amino acid concentrations within the ovarian follicle are not a critical factor affecting oocyte quality. However, high concentrations of cholesterol or of the essential fatty acids significantly compromised gap junction activity in bovine COC. High concentrations of the fatty acids were identified in a number of lactating dairy cows during the post-partum interval. In times of negative energy balance, which is common in pasture fed NZ dairy cows during the early post-partum interval and lactation, a significant mobilisation of fatty acids occurs. In turn, this is known to result in an increase in lipid concentrations within the ovarian follicle. The results from this study support the hypothesis that the nutrient environment of the ovarian follicle during early lactation is detrimental to the quality and developmental competence of the oocyte and thus may account for post-partum infertility in some animals.</p>

Abstract A recent association between the inclusion of pulses in canine diets and taurine deficiency has become a prevalent issue in the pet food industry. Although there is not a nutritional requirement for taurine by dogs, the appearance of taurine deficiencies has been related with more serious health issues, such as dilated cardiomyopathy. The objective of this study was to determine the circulating concentrations of plasma and whole blood taurine, indispensable and dispensable amino acid concentrations in the plasma, and taurine and creatinine concentrations in the urine of adult dogs fed a green lentil diet. Twelve, adult, female beagles were randomly assigned to a diet containing 45% green lentils (GLD) or a poultry byproduct meal diet (CON) for 90 days. Fresh urine samples were collected every 30 d and analyzed for taurine and creatinine concentrations. A blood sample also was collected every 30 d and analyzed for amino acids including taurine. Animal procedures were approved by the University of Illinois Institutional Animal Care and Use Committee. All diets were formulated to meet or exceed the nutrient requirements outlined by AAFCO (2018) and all dogs remained healthy throughout the study. The concentrations of taurine in the plasma and whole blood showed no differences (P &gt; 0.05) between dietary treatments or across time points. Similarly, no differences (P &gt; 0.05) in plasma methionine concentrations were observed between treatments or across time points. A treatment effect (P &lt; 0.05) showed dogs fed GLD had higher total primary fecal bile acid (BA) excretion compared with dogs fed CON. The differential abundance of fecal microbial communities showed Firmicutes as the predominant phyla in dogs fed both GLD and CON, with Bacteroidaceae, Erysipelotrichaceae, and Lactobacillaecae as predominant families in dogs fed GLD. The α-diversity of dogs fed GLD ( P &lt; 0.05) was lower than in dogs fed CON. These data suggest that the inclusion of 45% green lentil in extruded diets does not lower whole blood and plasma taurine concentrations during a 90 d period and is appropriate for use in a complete and balanced formulation for dogs.

Feed efficiency (FE) is an important measure of productivity in the layer industry; however, little is known about how FE differs between individual hens during the egg-laying cycle and the implications for egg quality parameters. Individual 25-week-old ISA Brown hens were observed for 42 days, ranked into three FE groups (n = 48 per High (HFE), Medium (MFE) and Low (LFE) FE groups and then monitored later in the laying cycle from 35–40 weeks. The groups exhibited different feed to egg conversion ratios (p < 0.001) from 35–40 weeks. Average daily feed intake and body weight were highest (p < 0.001) in the LFE group compared to the MFE and HFE groups, while albumen height, Haugh unit and amino acid concentrations of the albumen were significantly higher in the HFE groups compared to the LFE cohort (p < 0.001). This study concludes that FE status established in early lay is a stable variable until at least 40 weeks of age, and overweight, mid-laying hens that had poor FE produced inferior egg albumen quality measurements and composition. The distinct traits of the highly efficient hens and the poor feed efficient hens may provide important information to improving productivity in egg production.
