Synthesis, Anti-Arthritic, and Anti-Inflammatory Activity of N-Tosyl aza Cyclophanes

The synthesis of novel N-tosyl tetraaza cyclophanes and N-tosyl diaza cyclophane incorporating m-terphenyl as spacer units is described. Anti-arthritic activity was studied by inhibition of the protein denaturation method (bovine serum albumin). All the N-tosyl aza cyclophanes exhibit excellent anti-arthritic activity. Anti-inflammatory activity of the synthesized cyclophanes was investigated using the human red blood cells (HRBC) membrane stabilization method and some of the N-tosyl aza cyclophanes exhibited good anti-inflammatory activity.

Objective: The objective of this study was to evaluate the anti-inflammatory and antiarthritic potential of Parmelia perlata. Methods: The relative study is based on in vitro anti-inflammatory and antiarthritic activity using hydroalcoholic extract of P. perlata (HAEPP). The preliminary phytochemical tests showed the presence of various phytochemical compounds such as alkaloids, flavonoids, and glycosides since the lichen species of P. perlata has the folklore claim of anti-inflammatory activity, thus it was studied by human red blood cells membrane stabilization method, and arthritic activity was carried using protein denaturation method using diclofenac as a standard.Results: The results showed eminent anti-inflammatory and antiarthritic activity in a dose-dependent manner. The membrane stabilization showed the maximum effect at 78.54% at the concentration of 1000 μg/, and the protein denaturation was also found maximum at 1000 μg/ml concentration at 79.43%. Thus, our research states the potent anti-inflammatory activity and antiarthritic effect in P. perlata. Conclusion: The HEAPP has a potent anti-inflammatory activity and antiarthritic activity. A further study has to be conducted to establish the pharmacological evidence behind the compound and the mechanism of action of the HAEPP on the inhibition of the inflammation process.

This investigation aims to determine the chemical composition of Pinus brutia leaves essential oil and evaluate its anti-inflammatory property using Human Red Blood Cells (HRBC) membrane stabilization assay and Albumin denaturation assay. The chemical composition of essential oil (EO) obtained by hydro-distillation of leaves of Pinus brutia was investigated by GC-MS. The anti-inflammatory effect of EO was evaluated using Human Red Blood Cells (HRBC) membrane stabilization assay and Albumin denaturation assay. The main constituents of EO were α-Terpineol (66.16%), 3-Carene (4.90%), Carveol (4.55%) and cis-Verbenol (3.22%). The inhibion of hemolysis was observed at concentrations (2.5-12.5) µg/ml. Moreover, albumin denaturation test showed protection effect at concentrations (8-40) µg/ml. We concluded that, Pinusbrutia EO shows strong anti-inflammatory activity at different concentration when compared to standard drug of Diclofenac sodium. In addition, GC-MS analysis of Pinus brutia EO showed the presence of α-Terpineol as major compound in the oil. It reveals that this constituent is responsible to maximum protection of albumin denaturation and membrane stabilization assay. The future work will be determination of anti-inflammatory by in vivo models.

The methanolic extract of C. affinis was evaluated for its in-vitro anti-inflammatory activity by bovine serum albumin denaturation method, egg albumin denaturation method and protease inhibition method at different concentrations. Diclofenac sodium was used as the reference drug. The extract exhibits anti-inflammatory activity in a concentration-dependent manner. In bovine serum albumin denaturation method, the extract at concentrations of 100, 200, 400, 600, 800, 1000 µg/ml showed 25.49, 28.43, 31.37, 37.25, 41.17, 48.03% inhibition respectively. In egg albumin denaturation method, the concentrations of 50, 100, 200, 300, 400, 500 µg/ml showed 5.3, 9.89, 28.12, 32.8, 43.2, 52.8% inhibition respectively. In protease inhibition method the extract at concentration of 100, 200, 300, 400, 500 µg/ml showed 8.87, 19.32, 28.56, 43.96, 59.92% inhibition respectively. Therefore, from the results it can be concluded that the methanolic extract of C. affinis possesses anti-inflammatory activity.

A new modification reaction of ethyl p-methoxycinnamate (1) to afford a series of p-methoxycinnamamide and p-methoxy-β-nitrostyrene has been developed by using the assistance of the unmodified microwave oven. The synthesized compounds were characterized by using various spectroscopic techniques and furthermore screened for their anti-inflammatory activity by using anti-denaturation of heat bovine serum albumin (BSA) method. The result of bioassay indicated that p-methoxycinnamamide derivatives and p-methoxy-β-nitrostyrenes showed interesting anti-inflammatory activity.

A 57-year-old man with rheumatoid arthritis developed severe anemia during treatment with adalimumab plus methotrexate. Cold agglutinin disease was diagnosed because haptoglobin was undetectable, cold agglutinin was positive (1 : 2048), and the direct Coombs test was positive (only to complement). Although the cold agglutinin titer was normalized (1 : 64) after treatment with prednisolone (0.7 mg/kg/day for two weeks), the patient’s hemoglobin did not increase above 8 g/dL. When cold agglutinins were reexamined using red blood cells suspended in bovine serum albumin, the titer was still positive at 1 : 1024. Furthermore, the cold agglutinin had a wide thermal amplitude, since the titer was 1 : 16 at 30°C and 1 : 1 at 37°C. This suggested that the cold agglutinin would show pathogenicity even at body temperature. After the dose of prednisolone was increased to 1 mg/kg/day, the patient’s hemoglobin rapidly returned to the normal range. The thermal amplitude test using red blood cells suspended in bovine serum albumin is more sensitive than the standard test for detecting pathogenic cold agglutinins.

Objective: The objective of this study was to determine the anti-inflammatory activity of methanol extract of Niebuhria apetala and its possible mechanism of action. Methods: Methanol extract of Niebuhria apetala leaf (NAL) was assessed for its anti-inflammatory activity by in vitro methods. Using albumin denaturation assay, proteinase inhibitory activity, membrane stabilization, and antilipoxygenase activity at different concentrations, in vitro anti-inflammatory activity was estimated. The standard drug used for this purpose was aspirin. Results: Methanol extract NAL at a concentration range of 100–500 μg/ml significant (p<0.01) protects the heat-induced protein denaturation. At the concentration of 500 mg/ml, NAL showed significant (p<0.01) inhibition of protease inhibitory action. Heat-induced hemolysis of erythrocyte, hypotonicity-induced hemolysis, and lipooxygenase activity were significant (p<0.01) inhibited at the concentration of 500 μg/ml. Conclusion: Finally, the present study indicates that methanol extract of Niebuhria apetala can be a potential source of anti-inflammatory agent.
