The occurrence and severity of grass toxicoses in Australian alpaca (Vicugna pacos) herds

A survey of 108 alpaca producers registered with the Australian Alpaca Association examined the occurrence and severity of ‘staggers’ (a colloquial term embracing various diseases of the nervous system in animals, characterised by neck tremors and head nodding in the milder alpaca cases and a lack of coordination in moving, a staggering gait and frequent falling in severe cases) in Australian alpaca and the presence of four pasture grasses, during three production seasons in 2004–2006. There have been few studies on the susceptibility of alpaca to staggers and its effect on productivity and animal welfare. The survey found that 23% of alpaca producers had observed staggering animals, with Victoria and South Australia being the most severely affected states. Clinical signs of staggers were most frequent in January–March, with a mean duration of 3 months. Some animals showed clinical signs lasting up to 12 months. A strong correlation was found between the presence of perennial ryegrass (P < 0.001) and phalaris (P < 0.003) and the occurrence of staggers. Based on grass presence and the timing when staggers was observed, it was concluded that perennial ryegrass toxicosis was the main cause of staggers in alpaca. About 12% and 9% of alpaca grazing pasture containing perennial ryegrass exhibited staggers in the two full seasons for which data were collected. Herds with staggering animals were correlated with those exhibiting possible subclinical effects – heat sensitivity and ill-thrift (P < 0.01) and reduced fertility (P < 0.05) – of perennial ryegrass endophyte toxins. Results indicate that weanlings and cria are more likely to stagger than adults. Thirteen of the 15 mixed farms with staggering alpaca did not observe staggers in other livestock, suggesting that alpaca may be more sensitive to the causal toxins than sheep and cattle.

Abstract Objective To test the hypotheses that blood biomarkers for nervous system injury, serum concentrations of neurofilament light chain protein (NfL) and glial fibrillary acidic protein (GFAp) can serve as biomarkers for disease severity in COVID-19 patients. Methods Forty-seven inpatients with confirmed COVID-19 had blood samples drawn on admission for assessing serum biomarkers of CNS injury by Single molecule array (Simoa), NfL and GFAp. Concentrations of NfL and GFAp were analyzed in relation to symptoms, clinical signs, inflammatory biomarkers and clinical outcomes. We used multivariate linear models to test for differences in biomarker concentrations in the subgroups, accounting for confounding effects. Results In total, 21% (n = 10) of the patients were admitted to an intensive care unit, and the overall mortality rate was 13% (n = 6). Non-survivors had higher serum concentrations of NfL (p < 0.001) upon admission than patients who were discharged alive both in adjusted analyses (p = 2.6 × 10–7) and unadjusted analyses (p = 0.001). The concentrations of NfL in non-survivors increased over repeated measurements; whereas, the concentrations in survivors were stable. The GFAp concentration was also significantly higher in non-survivors than survivors (p = 0.02). Conclusion Increased concentrations of NfL and GFAp in COVID-19 patients on admission may indicate increased mortality risk. Measurement of blood biomarkers for nervous system injury can be useful to detect and monitor CNS injury in COVID-19.

The acute oral and dermal toxicity of two new ethyl-carbamates (ethyl-4-bromophenyl-carbamate and ethyl-4-chlorophenyl-carbamate) with ixodicide activity was determined in rats. The oral LD50of each carbamate was 300 to 2000 mg/kg, and the dermal LD50of each carbamate was >5000 mg/kg. Clinically, the surviving rats that had received oral doses of each carbamate showed decreased weight gain (P<0.05) and had slight nervous system manifestations. These clinical signs were evident from the 300 mg/kg dose and were reversible, whereas the 2000 mg/kg dose caused severe damage and either caused their death or was motive for euthanasia. At necropsy, these rats had dilated stomachs and cecums with diffuse congestion, as well as moderate congestion of the liver. Histologically, the liver showed slight degenerative lesions, binucleated hepatocytes, focal coagulative necrosis, and congestion areas; the severity of the lesions increased with dosage. Furthermore, an slight increase in gamma-glutamyltransferase, lactate dehydrogenase, and creatinine was observed in the plasma. The dermal application of the maximum dose (5000 mg/kg) of each carbamate did not cause clinical manifestations or liver and skin alterations. This finding demonstrates that the carbamates under study have a low oral hazard and low acute dermal toxicity.

The carbohydrate (CHO) fraction of pasture grasses is a major source of energy for many domestic herbivores. However, the amounts, and types, of the water–soluble carbohydrate (WSC) fraction (i.e. glucose, fructose, sucrose, and polymers of sucrose and fructose, the fructans) present in such grasses, varies with species and environmental conditions. As the WSC constitute a highly digestible, energy yielding fraction of grasses, it is important to be able to measure their levels in a sward so that the diets of pastured animals may be designed to elicit optimal health and productivity. The aim of this study was to characterise the WSC profile of six UK pasture grasses, and to develop a technique for extracting the fructan portion of the WSC.Six species of UK pasture grasses [Cocksfoot (C), Timothy (T), Meadow Fescue (M), Italian Ryegrass (IR), Perennial Ryegrass (PR) and Hybrid Ryegrass (HR)] were grown in experimental field plots at IGER.

SynopsisA double-blind comparison was made of the effects of amantadine hydrochloride, orphenadrine hydrochloride, and placebo in the control of the effects on the extrapyramidal nervous system of fluphenazine decanoate. Patients were assessed with regard to the main symptoms of Parkinsonism, the leading clinical signs, performance in walking, writing and squeezing, changes in mood, and unwanted effects of medication. Neither of the pharmacologically active substances was superior to placebo. Attention is drawn to the paucity of evidence for the effectiveness of anti-cholinergic drugs in both Parkinson's disease and in drug-induced Parkinsonism. The special difficulties in performing trials of substances used in relieving the unwanted effects of other drugs are discussed in relationship to defects in methodology in both this and earlier trials. The findings of the present study are not conclusive but show a clear need for further research in this field. The possible theoretical and practical importance of such research is discussed.

Animal welfare legislation in South Australia underwent amendments in 2008, where all the maximum penalties for animal welfare offences were doubled. This commitment to increased penalties arguably provides evidence of the legislature’s intent with respect to penalties. Studies have speculated that the legislative intent behind the increased penalties is not being reflected in the courts. This interdisciplinary research sought to gain evidence to confirm or disprove these speculations, by quantifying the average custodial sentence and monetary fine handed down in court before and after the 2008 amendments. Furthermore, trends relating to the species of animal affected and the demographics of the offender were identified. A total of 314 RSPCA (SA) closed case files from 2006 to 2018 were converted into an electronic form. Since the amendments, the average penalties have doubled in magnitude; fines have increased from $700 to $1535, while prison sentences have increased from 37 days to 77 days. Cases of companion animal abuse were most common (75% of all cases) and the location of the offence was found to influence offending. These findings suggest that the 2008 amendments have caused the average penalties to increase. However, it is debatable whether these increases are enough to effectively punish animal abusers.

Sapu Jagad (Isotoma longiflora (L) Presl.) plant has been empirically used as traditional medicine. Some studies showthat this plant has pharmacological effects as antibiotic, anticancer, and analgetic. It is of importance to conduct studyin finding out the safetiness of this plant as herbal medicine. Therefore, we conducted study to find out lethal dose ofits leaves on mice (Mus muculus) using acute toxicity test. Several doses have been given to certain groups to find outits effect including death. The extract has been given one time per oral. Then, we recorded the clinical signs and deathof mice until 14 days. The data was analyzed using probit analysis to measure LD50. This study shows that ethanolextract of Sapu Jagad leaves has LD50 12.610 mg/kgBW and toxicity of central nervous system proven by seizureending with death. As conclusion, this extract has toxicity especially to central nervous system.

The discussion about the rights of animals is always up-to-date. The dichotomy division into philoanimalists and philohominists, although reasonable, is not satisfactory to everyone. It is too strongly associated with the division into people and things in Roman law. To avoid this association in the context of biocentric trends in ecological ethics, accomplishments of evolutionary psychology and the concept of animal welfare, it is suggested that a third moral dimension dealing with creatures with highly developed nervous system be introduced between moral objectivity of creatures with high perception and moral subjectivity of people - creatures characterized by self-awareness and reflexive awareness. Human beings on the one hand are responsible for recognizing their rights given by nature and on the other hand, they are obliged to create a law to protect themselves.

Perennial ryegrass (Lolium perenne) straw is used as a feed for livestock and horses. Some straw is infected with the endophyte, Neotyphodium lolii, which produces lolitrem tremorgens. Ingesting of the toxin produces clinical "ryegrass staggers." A 28 day feeding trail was conducted using 61 Black Angus, pure and crossbred cattle. The cattle were fed one of three levels of lolitrem B in chopped perennial ryegrass straw ranging from 0 ppb up to 3058 ppb. Four Japanese Wagyu crossbred cattle were fed chopped ryegrass straw containing 1400 ppb lolitrem B. The cattle were evaluated and scored twice a day for clinical signs of gait difficulties. Cattle consuming 1400 ppb lolitrem B did not show any overt clinical signs. After 14 days, 7 out of 15 cattle in the 1954 ppb lolitrem B group showed clinical signs of ataxia, stiffness, and tremors as seen in ryegrass staggers. All of the cattle in the positive control group of up to 3058 ppb lolitrem B with a 4 week mean of 2574 ppb developed clinical signs of staggers. None of the Wagyu cattle at 1400 ppb lolitrem B developed clinical signs. A dose response curve was developed based on the results of this study coupled with a prior study for use in the testing laboratory. Canine cryptococcosis cases are typically reported as neurologic, disseminated, or both. There have been few reports of other parenchymal organ involvement. Dogs infected with Cryptococcus spp. are likely to develop central nervous system involvement, and those that are severely affected are treated aggressively with surgery and/or amphotericin B. This report describes two cases of canine abdominal cryptococcosis: one boxer with primary alimentary cryptococcosis alone and one miniature schnauzer with pancreatic and disseminated cryptococcosis. The boxer is unique in that the dog suffered from primary alimentary cryptococcosis without dissemination, secondary anemia due to gastrointestinal losses, and is the second case to have Cryptococcus spp. identified on fecal examination as part of the diagnostic workup. Unlike previous reports, surgery was not performed in either case, and both dogs were treated with fluconazole alone. Currently, both dogs are free from clinical signs, and Cryptococcus spp. antigen titers are negative at 17 and 15 mo after initial presentation. These cases suggest fluconazole may be effective therapy alone for canine abdominal cryptococcosis, negating the need for high-risk therapy options such as surgery and/or amphotericin B in some cases.
