Comparative molecular docking studies on creatine and guanidinoacetic acid, as well as their phosphorylated analogues, creatine phosphate, and phosphorylated guanidinoacetic acid, are investigated. Docking and density functional theory studies are carried out for muscle creatine kinase. The changes in the geometries of the ligands before and after binding to the enzyme are investigated to explain the better binding of guanidinoacetic acid and phosphorylated guanidinoacetic acid compared to creatine and creatine phosphate.

Abstract Creatine stores high-energy phosphate bonds in muscle and is synthesized in the liver through methylation of guanidinoacetic acid (GAA). Supplementation of GAA may therefore increase methyl group requirements, and this may affect methyl group utilization. Our experiment evaluated the metabolic responses of growing cattle to postruminal supplementation of GAA, in a model where methionine (Met) was deficient, with and without Met supplementation. Seven ruminally cannulated Holstein steers (161 kg initial body weight [BW]) were limit-fed a soybean hull-based diet (2.7 kg/d dry matter) and received continuous abomasal infusions of an essential amino acid (AA) mixture devoid of Met to ensure that no AA besides Met limited animal performance. To provide energy without increasing the microbial protein supply, all steers received ruminal infusions of 200 g/d acetic acid, 200 g/d propionic acid, and 50 g/d butyric acid, as well as abomasal infusions of 300 g/d glucose. Treatments, provided abomasally, were arranged as a 2 × 3 factorial in a split-plot design, and included 0 or 6 g/d of l-Met and 0, 7.5, and 15 g/d of GAA. The experiment included six 10-d periods. Whole body Met flux was measured using continuous jugular infusion of 1-13C-l-Met and methyl-2H3-l-Met. Nitrogen retention was elevated by Met supplementation (P < 0.01). Supplementation with GAA tended to increase N retention when it was supplemented along with Met, but not when it was supplemented without Met. Supplementing GAA linearly increased plasma concentrations of GAA and creatine (P < 0.001), but treatments did not affect urinary excretion of GAA, creatine, or creatinine. Supplementation with Met decreased plasma homocysteine (P < 0.01). Supplementation of GAA tended (P = 0.10) to increase plasma homocysteine when no Met was supplemented, but not when 6 g/d Met was provided. Protein synthesis and protein degradation were both increased by GAA supplementation when no Met was supplemented, but decreased by GAA supplementation when 6 g/d Met were provided. Loss of Met through transsulfuration was increased by Met supplementation, whereas synthesis of Met from remethylation of homocysteine was decreased by Met supplementation. No differences in transmethylation, transsulfuration, or remethylation reactions were observed in response to GAA supplementation. The administration of GAA, when methyl groups are not limiting, has the potential to improve lean tissue deposition and cattle growth.

Ninety barrow piglets weaned at 21 days old were used to evaluate the effects of guanidinoacetic acid (GAA) supplementation on their performance and blood creatinine and creatine kinase (CK) levels during the nursery phase (21 to 63 days old). The piglets were distributed in a randomized block design with five treatments (0, 0.05, 0.10, 0.15, and 0.20% GAA inclusion), six repetitions, and three animals per repetition. The experimental rations were formulated to meet the nutritional requirements for the phases of 21 to 32, 33 to 42 and 43 to 63 days of age. The experimental diets and water were offered ad libitum throughout the experimental period. At 42 and 63 days old, blood was collected from one animal in each experimental unit for creatinine and creatine kinase analysis. The analysed variables were daily weight gain (DFG), daily feed intake (DFI), and feed conversion (FC) in the 21 to 42 days old and 21 to 63 days old of the nursery phase. All variables were subjected to analysis of variance and regression analysis. We adopted ? = 0.05, and considered a trend to be present when ? was between 0.05 and 0.10. In the pre-initial period (21 to 42 days old), there was an increasing trend in daily weight (P = 0.069), which increased to the level of 0.97% of GAA inclusion. The DFI and FC were not significantly affected by the addition of different GAA levels in the diet. Creatinine levels in the blood at 42 and 63 days old were not significantly influenced by the treatments. The level of CK at 42 days old showed an increasing trend (P = 0.077) that occurred quadratically to the level of 0.085% of GAA inclusion, with no significant difference for this variable at 63 days old. The use of GAA did not promote improvements in performance or blood creatinine and CK levels in piglets in the nursery phase.

The effect of using different levels guanidinoacetic acid in compound feeds on the growth of young quails meat direction of productivity. It is proved that adding to the basic diet of different levels guanidinoacetic acid (0.06, 0.12, 0.18 %) increases live weight of quails on 3.9 – 13.5 g and improves the absolute, average daily and relative gains compared to a bird in the control group, which were fed the main diet. The data show that at the age of 42 days of live weight was the highest in the diet of quail which was administered 0.12% GAA and dominant control of 13.5 g However, in 21 days live weight of quail fed 0.06 and 0.12 % guanidinoacetic acid was greater than control respectively 9.2 and 7.6 grams (P < 0.01). In absolute increments quail, fed GAA prevailed bird control group. The largest absolute increase was in birds that consumed the main diet of GAA level of 0.12% and prevailed controls 6.5 g (P < 0.001). A similar situation was of average daily gain. Young animals that received feed containing 0.12% guanidinoacetic acid had the highest average growth, which exceeded 0.9 g (15.5%) bird control group. As for the relative performance increases here were different. The lowest relative growth observed in birds which were administered GAA 0.06%, it lagged behind the controls 0.1 g in the same period quail treated with 0.12 and 0, 18% guanidinoacetic acid superior control respectively 2 and 1.1 g.
