ABSTRACT Sponge Aaptos aaptos is a marine biota that has great potential, which can be applied, in the pharmaceutical field because of the presence of large compounds in inhibiting microbial growth. This study aims to determine the inhibitory activity of extracts and fractions of sponge Aaptos aaptos on microbial growth of Escherichia coli, Staphylococcus aureus, and Candida albicans. The samples were extracted by maceration with 96 % ethanol and fractioned with n-hexane, choloroform and methanol. Testing is done using the Disc Diffusion Agar method. Crude ethanol extract and fraction of sponge Aaptos aaptos showed the greatest antimicrobial activity against Staphylococcus aureus and categorized as strong, with an average value of 20.32 mm for ethanol extract with strong categories, chloroform fraction 13,28 mm with medium category and methanol fractions 18,48 mm strong category. Keyword: Aaptos aaptos, antimicrobial activity, Escherichia coli, Staphylococcus aureus, Candida albicans.  ABSTRAK Spons Aaptos aaptos merupakan biota laut yang memiliki potensi sebagai antimikroba yang dapat diterapkan di bidang farmasi dengan kandungan senyawa yang besar dalam menghambat pertumbuhan mikroba. Penelitian ini bertujuan untuk mengetahui aktivitas daya hambat dari ekstrak dan fraksi spons Aaptos aaptos terhadap pertumbuhan mikroba Escherichia coli, Staphylococcus aureus, dan Candida albicans. Sampel diekstraksi secara maserasi dengan etanol dan difraksinasi dengan pelarut n–heksan, kloroform dan metanol. Pengujian dilakukan dengan menggunakan metode Disc Diffusion Agar. Ekstrak kasar etanol dan fraksi dari Spons Aaptos aaptos menunjukkan aktivitas antimikroba paling besar terhadap Staphylococcus aureus dan dikategorikan kuat, dengan nilai rata – rata 20,32 mm untuk ekstrak etanol dengan kategori kuat, fraksi kloroform 13,28 mm, kategori sedang dan fraksi metanol 18,48 mm kategori kuat.Kata Kunci : Aaptos aaptos, aktivitas antimikroba, Escherichia coli, Staphylococcus aureus, Candida albicans

This study aims to determine the antimicrobial activity of red fruit paste against Staphylococcus aureus FNCC-0047 and Eschericia coli FNCC-0091 by using ethanol and n-hexane as well as the total plate count value of the patty with red fruit paste added on different observations day. The concentration of paste extract for antimicrobial activity test was 6.25%, 12.5%, 25%, 50%. The calculation of the total value of the burger patty plate is calculated on the 3rd, 7th and 14th d with the red fruit paste content in the formula as much as 0%, 5%, 10%, 15%. The results showed that the fatty acids of red fruit paste were dominated by oleic acid and linoleic acid, which are unsaturated fatty acids and palmitic acid, which are saturated fatty acids. The yield of red fruit paste in n-hexane solvent was higher than ethanol solvent. There was a significant difference (P <0.05) from the use of different solvents to the mean clear zone of the two bacteria tested. The antibacterial activity shown by the ethanol extract and n-hexane extract of red fruit paste was in the inactive category at the 6.12% level, the moderate category at the 12.5% to 25% level and the strong category at the 50% level. There was a significant difference (P <0.05) from the total microbial value in each treatment where the higher the red fruit paste content in the patty, the lower the total microbial value found on each d of observation. The use of red fruit paste at a level of 10% to 15% can withstand the rate of microbial growth

All over the world, hundreds of plants have been identified based on researchers and experimental evidence as good sources of medicinal agents. The bioactive components (phytochemicals) of both the seeds and pulp of Cola milleni were extracted using ethanol as solvent. The bioactive components detected were alkaloids, tanins, saponins, cardiac glycosides, carbohydrates, sterols, resins and terpenes while Flavonoids, anthraquinones, anthracyanides and phenol were not detected for both the seed and pulps. Antimicrobial activity of the ethanol extract (Seed and pulp) against Staphylococcus aureus, Escherichia coli and Penicillium notatum was carried out using standard techniques. Staphylococcus aureus had the highest zone of inhibition for pulp having a range of 9.7mm±0.58mm - 19.7mm±2.52mm while Penicllium notatum had the least with 0.00mm. S.aureus also had the highest zone of inhibition range of 14.3mm±2.08mm - 21.3mm±1.53mm for the seed extract while penicillium had the least inhibition range of 5.0mm±1.00mm - 5.7mm±0.58. E.coli showed the highest minimum inhibitory concentration with ethanol extract of the pulp (160mg/ml) while penicillium notatum was not reactive. The minimum inhibitory concentration of seed against penillium notatum was the highest (160mg/ml) while staphylococcus aureus showed the lowest of 40mg/ml. The antimicrobial activity is as a result of the presence of phytochemicals detected, which suggest the use of the plant for the treatment of diseases caused by these organisms. The complete halt in the synthesis of new effective antimicrobial compounds is a global concern. Pathogenic microorganisms' virulence mechanisms seem to have a significant impact on their pathogenesis. The purpose of this study was to examine the antimicrobial activity of the ethanol and methanol fractions of Pechuel-Loeschea leubnitziae leaf extract, as well as its effect on the expression level of virulence-associated genes.The extract's fractions were evaluated for antimicrobial activity against Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Klebsiella pneumoniae (clinical), Methicillin-resistant Staphylococcus aureus, and Candida albicans ATCC 90029. The test organism's antibiogram pattern was determined. The extracts' attenuation effect on the target genes of the susceptible organisms was investigated employing relative quantification using RT-qPCR. The test organism's antibiogram pattern revealed that it was drug-resistant, intermediate, and sensitive. The extracts tested positive for antimicrobial activity against Methicillin-resistant Staphylococcus aureus and Candida albicans ATCC 90029, with zones of inhibition varying from 20.33 to 29 mm. The lowest recorded MIC value was 4.688 mg/ml, while the highest was 37.5 mg/ml. In contrast to the methanol extract, the ethanol extract had a cidal action at a lower dose. The ethanol extract's Sub-MIC (18.25 mg/ml) merely reduced the expression of the hly gene in MRSA. The MRSA virulence genes were not suppressed by the sub-MIC of methanol extract (18.25 mg/ml). Notably, the expression of als1, pbl1, and sap1 in Candida albicans ATCC 90029 was significantly attenuated when exposed to sub-MICs of ethanol extract (2,344 mg/ml) and methanol extract (9.375 mg/ml). Per the findings of this research, the leaves of P. leubnitziae could be a source of an effective antimicrobial agent in the therapy of MRSA/Candida-related disorders.

Frangipani flower (Plumeria acuminata) and bitter grape (Tinospora crispa L. Miers) stem extract alone shows antimicrobial and antioxidant activities. However, there is a limited report about the actions from the combination of both extracts. Combinations of ethanol extract of white frangipani flower (EFF) and ethanol extract of the bitter grape stem (EBS), each of 0, 1, 2, and 3% were tested for inhibition against Staphylococcus aureus growth and DPPH (1,1-diphenyl,2-picrylhydrazyl) free radicals. A yield of 3.17 and 2.62 g of EFF and EBS, respectively, were resulted from 100 g of dried powdered of the white frangipani flower and bitter grape stem using 200 mL of absolute ethanol. The antimicrobial test was assayed using a 10% stock solution of extract diluted in water. The combination of EFF and EBS showed a synergistic effect on antimicrobial activity, but it proved an antagonistic effect on antioxidant activity. The combination of EFF 3% and EBS 3% showed the highest inhibition index on the S. aureus growth , i.e. 2.02±0.06, which is higher than the inhibition index of Amoxycillin 2%, i.e. 1.85±0.09. The highest antioxidant activity showed by 1% EFF alone (57.5 ± 0.60%), while the lowest was from the combination of EFF 1% and EBS 3% (23.4±0.30%).

Abstract Background Mahanintangtong is listed in the Thailand’s National List of Essential Medicines (NLEM). It is used to treat non-specific fevers and illnesses such as pharyngitis and chickenpox. In this study, we investigated the biological activities of the different medicinal plants used in the Mahanintangtong formula. Methods The plant materials were extracted by maceration and decoction. Antimicrobial activity, assessed by disc diffusion method, the minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) were compared with commercially available standard antibiotics. To elucidate the anti-inflammatory mechanisms, inhibition of nitric oxide (NO), tumor necrosis factor α (TNF-α), and interleukin 6 (IL-6) production was tested by Griess and ELISA techniques. Antioxidant activity was measured by ABTS and DPPH scavenging assays. Results The extracts with the best antimicrobial activities were carbonized Tectona grandis showing against Streptococcus pyogenes, Staphylococcus aureus, Methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa. The ethanol extract of Dracaena loureiroi wood exhibited the highest NO and IL-6 inhibitory activity with IC50 values of 9.42 ± 1.81 and 12.02 ± 0.30 μg/mL, respectively. The ethanol extract of Pogostemon cablin had the highest TNF-α inhibitory with IC50 values of 10.68 ± 0.02 μg/mL. In anti-free radical testing, the ethanol extract of D. loureiroi displayed high antioxidant activity by both ABTS and DPPH assays. Conclusion The ethanol extracts from carbonized T. grandis and Mahanintangtong showed good antimicrobial activity, especially against S. pyogenes, and good anti-inflammatory activity. These findings are relevant to the pathogenesis of pharyngitis and justify additional studies to see if Mahanintangtong could have clinical utility.

An AgNPs solution was synthesized by chemical reduction, characterized, and tested againstCandida glabrata,Candida tropicalis,Staphylococcus aureus, and methicillin-resistantStaphylococcus aureus(MRSA). Minimum inhibitory (MICs) and minimum fungicidal/bactericidal concentrations (MFC/MBC) were determined on planktonic cells. Also, total biofilm mass was determined by crystal violet (CV) staining and morphological changes by scanning electron microscope (SEM). MICs forC. glabrata,C. tropicalis,S. aureus, and MRSA were 15.63, 3.91, 1.95, and 1.95 µg/mL, respectively. MFC forC. glabratawas 62.5 µg/mL and forC. tropicalis15.63 µg/mL The same MBC (3.91 µg/mL) was observed forS. aureusand MRSA. CV assay showed that the AgNPs (1000 μg/mL) promoted reductions in biofilm mass of ~60% forC. glabrataand ~35% forC. tropicalis. A reduction of ~20% inC. tropicalisbiomass was also observed at the concentration of 3.91 µg/mL. No significant effect on total biomass was found forS. aureusand MRSA. SEM images revealed thatC. glabrataandC. tropicalisbiofilm cells, exposed to the AgNPs (1000 μg/mL), had an irregular and shriveled appearance. AgNPs solution exhibited considerable antimicrobial activity against important fungal and bacterial pathogens, associated with several oral and systemic diseases, and has potential as an antimicrobial agent.

Multidrug resistance has increased globally in the communities. Bacterial infections associated with health care have weakened the existing antimicrobial therapy and demand the search for alternative therapies. In the present investigation, the medicinal plant Pulicaria gnaphalodes from Quetta, Pakistan, has been screened for antimicrobial potential. In vitro antimicrobial efficacy of P gnaphalodes extracts (methanol and ethanol) was quantitatively evaluated on the basis of zone of inhibition against different bacteria and minimum inhibitory concentration (MIC). In vivo, antihypercholesterolemic activity is determined in different rat groups. The results of the study indicated that the ethanol extract of P gnaphalodes showed maximum zone of inhibition for Bacillus subtilis of 12.1 ± 1.1 mm from all others. The methanol extract showed maximum zone of inhibition for Staphylococcus aureus of 11.9 ± 1.0 mm and rifampicin showed maximum zone of inhibition of 23.1 ± 0.9 mm. The results of ethanol and methanol extract of P gnaphalodes against different bacteria revealed that this plant has greater antimicrobial activity. However, the plant extract shows nonsignificant antihypercholesterolemic activity. The extract of this plant can be utilized as medicine to inhibit several infections caused by some bacterial pathogens found in human body.
