Abstract This study aimed to determine the rumen fluid’s in-vitro dry matter digestibility (IVDMD), in-vitro organic matter digestibility (IVOMD), and pH of different species Acacia seed pods as a single feed ingredient. The Acacia species in this study were Acacia mangium, Acacia auriculiformis, and Acacia crassicarpa. The method used was in vitro two-stages with the first 48 hours of incubation (the rumen) and the second 48 hours of incubation (post-rumen), and analyses of pH. The research design used was a completely randomized design (CRD) with 3 treatments and 5 replications based on the Acacia species. The data were analysed with oneway analysis of variance (ANOVA) followed by Duncan’s multiple range test (DMRT). The results showed that IVDMD stage one was 34.59-37.59%; IVOMD stage one was 38.66-43.06%. Stage two of IVDMD was 58.02-59.23%; stage two of IVOMD was 51.67-55.01% and rumen fluid pH were 6.58-7.02. Different Acacia seed pods had significant differences in IVDMD and IVOMD stage one in the rumen and stage two in the post-rumen (P<0.05), but not in the pH value (P>0.05). We concluded that different acacia seed pods had different digestibility values in the rumen and post-rumen. However, it does not affect the rumen acidity.

Abstract In vitro digestibility methods have been developed to overcome problems in the in vivo digestibility measurement, but its accuracy should be tested in a local setting. In vitro methods developed by Tilley and Terry (T2), Theodorou (T3) and Sutardi (T4) have been compared to in vivo method (T1) in a block randomized design study. Four heifers FH (337.50 ± 45.87 kg BW) were used in T1, and two fistulated FH bulls (510 ± 20 kg BW) were used as inoculant sources in the in vitro methods. Dairy cattle ration consisted of 54.0% Napier grass and 46.0% concentrate with 58.8% DM, 12.1% ash, 10.0% CP, 3.3% EE, 26.5% CF, and 61.1% TDN. The observed parameters were ration fermentability (pH, NH3, and VFA concentration) and digestibility (DMD and OMD). The data were analyzed using analysis of variance (ANOVA) followed by the Tukey test. The correlation was made before regression analysis to estimate the in vivo parameters from the in vitro. The results showed that pH values are in the normal range (6.7 – 6.8), and insignificantly different between treatments (P>0.05). The concentration of NH3 and VFA were significantly different between the treatments (P<0.05), but T2 produced similar NH3 and VFA concentrations to T1. Similar results were also found in the DMD and OMD. Correlation analysis showed that pH value of T3 correlated significantly with T1, while DMD value of T4 correlated to T1. The T1 DMD (Y) could be estimated from T4 DMD (X) using formula Y (%) = y = -0.091x2 + 9.1632x - 168.4. It is concluded that tropical dairy feedstuffs in vitro digestibility using Tilley and Terry’s method produced similar result to in vivo digestibility method, but in vivo dry matter digestibility can be estimated accurately by in vitro dry matter digestibility using Sutardi method.

In a study that included C4 tropical grasses, C3 temperate grasses and C3 pasture legumes, in vitro dry matter digestibility of extrusa, measured as in vitro dry matter loss (IVDML) during incubation, compared with that of the forage consumed, was greater for grass extrusa but not for legume extrusa. The increase in digestibility was not caused by mastication or by the freezing of extrusa samples during storage but by the action of saliva. Comparable increases in IVDML were achieved merely by mixing bovine saliva with ground forage samples. Differences were greater than could be explained by increases due to completely digestible salivary DM. There was no significant difference between animals in relation to the saliva effect on IVDML and, except for some minor differences, similar saliva effects on IVDML were measured using either the pepsin–cellulase or rumen fluid–pepsin in vitro techniques. For both C4 and C3 grasses the magnitude of the differences were inversely related to IVDML of the feed and there was little or no difference between extrusa and feed at high digestibilities (>70%) whereas differences of more than 10 percentage units were measured on low quality grass forages. The data did not suggest that the extrusa or saliva effect on digestibility was different for C3 grasses than for C4 grasses but data on C3 grasses were limited to few species and to high digestibility samples. For legume forages there was no saliva effect when the pepsin–cellulase method was used but there was a small but significant positive effect using the rumen fluid–pepsin method. It was concluded that when samples of extrusa are analysed using in vitro techniques, predicted in vivo digestibility of the feed consumed will often be overestimated, especially for low quality grass diets. The implications of overestimating in vivo digestibility and suggestions for overcoming such errors are discussed.

The supplementation peel coffee beans in the ration Aceh cattle on in vitro digestibilityABSTRACT. This research was carried out at the Laboratory of  Feed and Animal Nutrition,  Department of Animal Husbandry Faculty of Agriculture Syiah Kuala University for  73 days from December 26, 2011 to March 7, 2012. The purpose of this experiment was to find out the effects of the coffee peeling on in vitro digestibility of Aceh beef cattle. Completely Randomized Design (CRD) was applied in this experiment consisting of 4 treatments  namely levels of  coffee peeling on basal diet   (R0=0%, R1=10%, R2=20%, R3=30%). Each treatment consists of 5 replications. Parameters observed in this research were dry and organic matter digestibility. The results showed that dry matter digestibility was not significantly different (P0,05)amongst treatments  54,42%, 49,32%, 45,97%, 42,08%) for R0, R1, R2 and R3 respectively.  On the other hand, organic metter digestibility was significantly different (P0,05) in which R2 treatment was the highest and R0 treatment was the lowest (54,59%).

Abstrak: Penelitian ini telah dilaksanakan di Laboratorium Ilmu Nutrisi dan Makanan Ternak Program Studi Peternakan Fakultas Pertanian Universitas Syiah Kuala dan dilanjutkan dengan pengiriman sampel penelitian ke Laboratorium Ternak Perah Institut Pertanian Bogor (IPB) untuk pengujian kecernaan secara in vitro. Tujuan dari penelitian ini adalah menguji kecernaan secara in vitro complete feed berbahan dasar ampas sagu dengan teknik fermentasi yang berbeda.Pembuatan complete feed disusun sesuai dengan kebutuhan ternak domba. Penelitian ini menggunakan Rancangan Acak lengkap (RAL) dengan 3 perlakuan dan masing-masing perlakuan terdiri atas 5 ulangan. Perlakuan 1 (kontrol) adalah complete feed tanpa penambahan Saus Burger Pakan (SBP) dan tanpa fermentasi, perlakuan 2 adalah ampas sagu difermentasi dengan SBP selama 14 hari kemudian dicampur menjadi complete feed dan difermentasi hingga 21 hari, sedangkan perlakuan 3 adalah ampas sagu + complete feed kemudian difermentasi dengan SBP selama 21 hari. Parameter yang diamati dalam penelitian ini adalah pH, Bahan Kering (BK), Bahan Organik (BO), Koefisien Cerna Bahan Kering (KCBK), dan Koefisien Cerna Bahan Organik (KCBO). Hasil pengamatan menyimpulkan bahwa, teknik fermentasi yang berbeda berpengaruh sangat nyata (P0,01) terhadap semua parameter yang diamati, yaitu pH, BK, BO, KCBK dan KCBO. Dari hasil penelitian dapat disimpulkan bahwa nilai kecernaan complete feed berbahan dasar ampas sagu dipengaruhi oleh perbedaan teknik fermentasi. Digestibility Evaluation In Vitro Complete Feed Fermentation Sago Residues Based with Different Fermentation Techniques Abstract: This research was conducted at the Laboratory of Nutrition and Feed Studies, Program study Animal Husbandry, Faculty of Agriculture, University of Syiah Kuala and continued with the delivery of sample to the Laboratory of Dairy Cattle Institute Pertanian Bogor (IPB) for testing of in vitro digestibility. The purpose of this study was to test in vitro digestibility of complete feed made from  the residues of sago with different fermentation techniques. Complete feed was arranged based on the needs of sheep. This study used a complete randomized design (CRD) with 3 treatments and each treatment consisted of 5 replicates. Treatment 1 (control) was a complete feed without the addition of Saus Burger Pakan (SBP) and without fermentation, treatment 2 was sago residu  fermented with SBP for 14 days and then mixed into a complete feed and fermented up to 21 days, whereas treatment 3 was the sago residue + complete feed then fermented with SBP for 21 days. The parameters observed in this study were pH, Dry Matter (BK), Organic Matter (BO), Dry Matter digestibility coefficients (KCBK), and Organic Matter Digestibility Coefficient (KCBO). The result influence showed that, different fermentation technique was significantly (P 0.01) all parameters, namely pH, BK, BO, KCBK and KCBO. It can be concluded that the digestibility of complete feed made from sago residue influenced by differences in fermentation techniques

Leaf and stem fractions of Digitaria decumbens (pangola grass) cut at two stages of regrowth (47 and 89 days), were extracted with ether, ethanol and water (100°C) before being partially delignified with sodium chlorite in acetic acid. Delignification reduced the lignin concentrations by 52 and 72% in the 47-day-old leaf and stem, and by 69 and 87% in the 87-day-old leaf and stem. In vitro dry matter digestibility was correspondingly increased, by 17–20% (47-day material) and 28–31% (89-day material). Hemicelluloses were digested to a greater degree than cellulose in the delignified material. The reverse was found in the material prior to delignification. In the hemicelluloses arabinose was always digested to a greater extent than xylose. Polysaccharides from delignified 89-day regrowth leaf and stem tissue were still digested to a lesser extent than those from the 47-day regrowth. Maximum digestibilities estimated for zero lignin content were 96.6, 88.1 and 82.8% for young leaf, young stem-told leaf, and old stem respectively. Thus, while lignin has been shown to be the major factor limiting total digestion of the plant material, other minor factors could not be excluded, particularly in the older tissues.

This experiment investigated various substrate growth of bamboo stems microbes (BSM) in rice straw fermentation and its effect on dry matter digestibility (DMD), organic matter digestibility (OMD), NH3-N and VFA concentration. Materials were rumen fluid of beef cattle from the slaughterhouse. Basal diets consisted of rice straw and concentrate with ratio of 40 : 60 (% dry matter) were administered to five treatments namely P0 = untreated rice straw (control), P1 = Ammoniated rice straw,  fermented rice straw using microbial bamboo stems grown on ammoniated rice straw (P2), rice bran (P3) and cassava waste (P4). The experiment used in vitro method was designed with Completely Randomized Design and the data were analyzed variance. The results showed that the treatment had no significant effect on dry matter digestibility (DMD), organic matter digestibility (OMD), NH3-N and VFA concentration (P>0,05). The fermentation process of microbial bamboo stems in rice straw with a level of 10% with different substrate did not able to  increase the digestibility and products fermentation, but there tends that the rice bran and substrates can increase DMD and N-NH3 concentration

In the tropical area, productivity of ruminant has not optimized caused by the low quality of nutrition that leads to low-efficiency metabolism at the level of ruminal fermentation, post rumen digestibility, and intermediary metabolism. The study aimed to analyze effect of methionine hydroxy analog (MHA) on ruminal fermentation profiles of indigenous sheep specifically in the increase of ruminant productivity. In vitro utility test was conducted using rumen fluid of the indigenous sheep and sample of rational ration having a proportion of grass and concentrate 30%:70%, dry matter basis. The treatments implemented were three levels of MHA supplementation; T0: 0 g/day, T1: 3 g/day, and T2: 6 g/day. Variables measured were dry matter digestibility (DMD), organic matter digestibility (OMD), production of VFA, NH3, as well as total protein, and molar proportion of partial VFA of rumen fluid. Data were analyzed using analysis of variance (ANOVA) in a completely randomized design (CRD). The 0.2% MHA supplementation increased OMD with the highest production of total protein was from 28.57 mg/g (T0) to 40.49 mg/g (T2) (P<0.05). Meanwhile, the lowest ratio of acetate : propionate was from 2.74 (T0) to 2.33 (T2) (P<0.05). Supplementation of MHA up to 6 g/day concentrate increased the performance of fermentation and/or feed utility.

The research aims to evaluate calsium soap protection as feed supplement based on dry matter<br />digestibility, organic matter digestibility and pH in vitro in the rumen and post rumen. Calsium soap<br />was produced from sardine fish oil and soybean meal, through two protection methods. It was<br />saponification and microencapsulation. This research used Split Subject Repeated Measurement<br />Design with 3 treatments (calcium soap, soybean meal and pangola grass) which each treatment was<br />repeated 5 times. The observation kinetics of dry matter digestibility, organic matter digestibility, and<br />pH rumen fluid were done in incubation times (2, 4, 8, 12, 24 and 48 hours respectively). The first<br />results of the research showed that in the first step (rumen) has dry matter digestibility of calcium<br />soaps: 38.65; 40.00; 42.66; 46.32; 54.40 and 51.40% respectively; organic matter digestibility: 39.44;<br />41.27; 44, 31; 46.32; 56.62 and 52.50% respectively. Kinetics of pH rumen fluid: 6.89; 6.79; 6.78;<br />6.74; 6.71, and 6.88 respectively. In the second step (post rumen), the digestibility of calsium soap<br />production has dry matter digestibility of 74.85% and organic matter of digestibilty of 72.94%. The<br />results could be concluded that in the rumen, calcium soap is the most microbial degradation resistant<br />compared the others and it can be degraded and digested in the abomasum to the small intestine.<br />Therefore pH of rumen fluid is optimal, so it does not interfere with the fermentation in the rumen.<br />Keywords: calsium soap, in vitro, dry matter digestibily, organic matter digestibility, pH value

Abstract The aim of this study was to examine the use of Gizzard Contents Extracts (GCE) and Duodenum Contents Extracts (DCE) of Chicken on In Vitro digestibility measurements of Turi Leaves (Sesbania grandiflora). The study was designed using a One-way Classification of Completely Randomized Design (CRD) with 4 treatments. Each treatment was repeated three times. Types of treatment in the form of using GCE and DCE, including: (1) control treatments (without GCE or DCE), (2) GCE, (3) DCE, and (4) a mixture of GCE and DCE. The observed variables were Coefficient of Dry Matter Digestibility (CDMD), Coefficient of Organic Matter Digestibility (COMD), and Coefficient of Soluble Protein Digestibility (CSPD). The results showed that the addition of GCE, DCE, and a mixture of GCE and DCE could not increase the CDMD, COMD, and CSPD of Turi Leaves compared to the control treatment. Penelitian dirancang menggunakan Rancangan Acak Lengkap (RAL) pola searah dengan 4 perlakuan. Masing-masing perlakuan diulang tiga kali. Macam perlakuan berupa penggunaan penggunaan CEIZ dan CEID ayam, meliputi perlakuan kontrol (tanpa CEIZ maupun CEID), CEIZ, CEID, dan campuran CEIZ dan CEID. Variabel pengamatan berupa koefisien cerna bahan kering (KCBK), koefisien cerna bahan organic (KCBO), dan koefisien cerna protein terlarut (KCPT). Hasil penelitian menunjukkan bahwa perlakuan penambahan cairan ekstrak isi gizzard, duodenum dan campuran keduanya tidak dapat meningkatkan KCBK, KCBO, dan KCPT daun turi dibandingkan dengan perlakuan control. Abstract The objective of this study was to determine the in vitro dry matter digestibility (IVDMD), gas production and pH of silages of 10 maize hybrids in Iturama, Brazil (XB 6012 BT, BM 709 VT PRO2, BM 815 VT PRO2, DKB 363 VT PRO3, AG 8740 VT PRO3, SYN 555 VIP3, SX 8555 VIP3, SX 7005 VIP3, DKB 390 VT PRO3, and RB 9789 VIP3). The maize was ensiled in experimental mini silos in a completely randomized design with 3 repetitions. After 120 d, a subsample from each mini silo was taken and the IVDMD was estimated using Ankom Daisy II and F-57 bags (n = 44). Rumen fluid from two cannulated Nellore steers were mixed and placed into fermenter jars containing Ankom buffers (1:4; v/v). After 48-h incubation (39ºC), 40 mL 6 N HCl and 8 g pepsin were added to each jar and incubated for another 24 h. For gas production and pH evaluations, penicillin-type glass vials (n = 40) were filled with McDougall buffer (20 mL), rumen fluid (10 mL) and substrates (200 mg) and incubated in a shaker (39ºC) for 24 h. The cumulative pressure was measured, vials were opened, and terminal pH was measured. Data were analyzed using the MIXED procedure of SAS. The DKB 363 VT PRO3 presented the greatest IVDMD (84.9%, P = 0.02) and the lowest pH (6.60, P = 0.002). The opposite was observed for DKB 390 VT PRO3, which presented the lowest IVDMD (71.1%) and the greatest pH (6.65). The BM 709 PRO2 produced more gas than the others (246.1 mL/g DM), while SYN 555 VIP3 produced less gas than the others (167.7 mL/g DM, P = 0.0005). In conclusion, the material DKB 363 VT PRO3 presented the most interesting characteristics for use as silage for beef cattle in Iturama, Brazil.
