Is There a Minimum Number of Landmarks That Optimizes the Geometric Morphometric Analysis of Mosquito (Diptera, Culicidae) Wings?

Abstract Culicids are the most significant arthropods affecting human health. Thus, their correct identification is critical. The use of Geometric Morphometrics (GM) has been recently incorporated into mosquito taxonomy and has begun to complement classic diagnostic techniques. Since sampling size depends on the number of Landmarks (LMs) used, this study aimed to establish the minimum number of wing LMs needed to optimize GM analysis of mosquito species and/or genera from urban and peri-urban areas of Argentina. Female left wings were used for the optimization phase, in which 17 LMs were reduced to four by iterative LM exclusion. To verify its efficiency, Principal Component Analysis (PCA), Discriminant Analysis (DA), and Canonical Variate Analysis (CVA) were performed. Additionally, a phenogram was constructed to visualize the results. We observed that five LMs for the PCA, CVA, and phenogram and nine for the DA enabled discrimination and/or clustering of almost all species and genera. Therefore, we tested the LM selection by using nine LMs and adding new species. The resulting PCA showed little overlap between species and almost all species clustered as expected, which was also reflected in the phenogram. Significant differences were found between wing shape among all species, together with a low total error rate in the DA. In conclusion, the number of LMs can be reduced and still be used to effectively differentiate and cluster culicids. This is helpful for better exploitation of available material and optimization of data processing time when classic taxonomy methods are inadequate or the material is scarce.

Culex is the largest subgenus within the genus Culex that includes important vectors of diseases. The correct identification of mosquitoes is critical for effective control strategies. Wing geometric morphometrics (WGM) has been used to identify mosquito species alongside traditional identification methods. Here, WGM was used for eleven Culex species from São Paulo, Brazil, and one from Esquel, Argentina. Adult mosquitoes were collected using CDC (Centers for Disease Control) traps, morphologically identified and analyzed by WGM. The canonical variate analysis (CVA) was performed and a Neighbor-joining (NJ) tree was constructed to illustrate the patterns of species segregation. A cross-validated reclassification test was also carried out. From 110 comparisons in the cross-validated reclassification test, 87 yielded values higher than 70%, with 13 comparisons yielding 100% reclassification scores. Culexquinquefasciatus yielded the highest reclassification scores among the analyzed species, corroborating with the results obtained by the CVA, in which Cx. quinquefasciatus was the most distinct species. The high values obtained at the cross-validated reclassification test and in the NJ analysis as well as the segregation observed at the CVA made it possible to distinguish among Culex species with high degrees of confidence, suggesting that WGM is a reliable tool to identify Culex species of the subgenus Culex.

The genetic diversity was assessed in 42 Brassica rapa genotypes for identifying parental genotypes having distant relationship for hybridization. All the genotypes were analyzed through principal component analysis, principal coordinate analysis canonical variate analysis and non-hierarchical clustering which compute Mahalanobis’s D2 statistics. The genotypes under study were grouped into six clusters. The cluster V contained the maximum numbers of genotypes (15) and cluster III had the minimum number of genotypes (3). The highest intra-cluster distance was observed in cluster V that represented maximum variability within the cluster and Cluster VI had the lowest intra-cluster distance. The maximum inter cluster distance was noticed between the clusters II and III and the lowest between clusters II and VI. Cluster with large distances were considered more diverged than those with small distances. Yield and yield contributing characters such as days to 1st flowering, number of siliquae on main racemes, number of siliquae per plant, number of seeds per siliqua, 1000 seeds weight and seed yield per plant contributed more towards genetic divergence considering diversity pattern. In 2018–2019, we conducted mosquito collections in a municipal vehicle impound yard, which is 10 km from the Serra do Mar Environmental Protection Area in Santo André, SP, Brazil. Our aim is to study arboviruses in the impound yard, to understand the transmission of arboviruses in an urban environment in Brazil. We captured the mosquitoes using human-landing catches and processed them for arbovirus detection by conventional and quantitative RT-PCR assays. We captured two mosquito species, Aedes aegypti (73 total specimens; 18 females and 55 males) and Ae. albopictus (34 specimens; 27 females and 7 males). The minimum infection rate for DENV-2 was 11.5 per 1000 (CI95%: 1–33.9). The detection of DENV-2 RNA in an Ae. albopictus female suggests that this virus might occur in high infection rates in the sampled mosquito population and is endemic in the urban areas of Santo André. In addition, Guadeloupe mosquito virus RNA was detected in an Ae. aegypti female. To our knowledge, this was the first detection of the Guadeloupe mosquito virus in Brazil.

Abstract Coyotes are ubiquitous in habitats across North America, including in urban areas. Reviews of human–coyote encounters are limited in scope and analysis and predominantly document encounters that tend to be negative, such as human–wildlife conflict, rather than benign experiences. The objective of our study was to use citizen science reports of human–coyote interactions entered into iNaturalist to better understand the range of first person accounts of human–coyote encounters in Madison, WI. We report 398 citizen science accounts of human–coyote encounters in the Madison area between October 2015 and March 2018. Most human–coyote encounters occurred during coyote breeding season and half of all encounters occurred in moderate development land cover. Estimated level of coyote aggressiveness varied significantly, with 90% of citizen scientists scoring estimated coyote aggression as a 0 and 7% scoring estimated aggression as a 1 on a 0–5 scale (with 0 being calm and 5 being aggressive). Our best performing model explaining the estimated distance between the human observer and a coyote (our proxy for a human–coyote encounter) included the variables distance to nearest paved road, biological season of the year relative to coyote life history, and time of day/night. We demonstrate that human–coyote interactions are regularly more benign than negative, with almost all first-hand reported human–coyote encounters being benign. We encourage public outreach focusing on practices that can foster benign encounters when educating the public to facilitate human–coyote coexistence.

Purpose The purpose of this paper is to analyse the spatial disparity between rural and urban areas in Ghana using the Ghana Living Standards Survey’s (GLSS) rounds 5 and 6 data to advance the assertion that an endowed rural sector is necessary to promote agricultural development in Ghana. This analysis helps us to know the factors that contribute to the depravity of the rural sectors to inform policy towards development targeting. Design/methodology/approach A multivariate principal component analysis (PCA) and hierarchical cluster analysis were applied to data from the GLSS-5 and GLSS-6 to determine the characteristics of the rural-urban divide in Ghana. Findings The findings reveal that the rural poor also spend 60.3 per cent of their income on food, while the urban dwellers spend 49 per cent, which is an indication of food production capacity. They have low access to information technology facilities, have larger household sizes and lower levels of education. Rural areas depend a lot on firewood for cooking and use solar/dry cell energies and kerosene for lighting which have implications for conserving the environment. Practical implications Developing the rural areas to strengthen agricultural growth and productivity is a necessary condition for eliminating spatial disparities and promoting overall economic development in Ghana. Addressing rural deprivation is important for conserving the environment due to its increased use of fuelwood for cooking. Absence of alternatives to the use of fuelwood weakens the efforts to reduce deforestation. Originality/value The application of PCA to show the factors that contribute to spatial inequality in Ghana using the GLSS-5 and GLSS-6 data is unique. The study provides insights into redefining the framework for national poverty reduction efforts.

Intercellular communication governs multicellular interactions in complex organisms. A variety of mechanisms exist through which cells can communicate, e.g., cell-cell contact, the release of paracrine/autocrine soluble molecules, or the transfer of extracellular vesicles (EVs). EVs are membrane-surrounded structures released by almost all cell types, acting both nearby and distant from their tissue/organ of origin. In the kidney, EVs are potent intercellular messengers released by all urinary system cells and are involved in cell crosstalk, contributing to physiology and pathogenesis. Moreover, urine is a reservoir of EVs coming from the circulation after crossing the glomerular filtration barrier—or originating in the kidney. Thus, urine represents an alternative source for biomarkers in kidney-related diseases, potentially replacing standard diagnostic techniques, including kidney biopsy. This review will present an overview of EV biogenesis and classification and the leading procedures for isolating EVs from body fluids. Furthermore, their role in intra-nephron communication and their use as a diagnostic tool for precision medicine in kidney-related disorders will be discussed.

Introduction: In Japan, the number of automated external defibrillators (AEDs) placed in public areas has climbed up to the estimated 530,000. We reported previously that a half of out-of-hospital cardiac arrests occurred within 100 m from a public AED in urban areas. However, shocks for defibrillation were given to less than 15% of those patients. The objectives of this study was to identify the limiting factors against the use of AEDs by bystanders. Methods: A prospective survey on out-of-hospital cardiac arrest was conducted in the city of Osaka (Dec 2016 through Mar 2017) and the city of Nagoya (Dec 2016 through Nov 2017) searching for patient demographics and AED settings including; absence/presence of a public AED on the scene, attachment of AED pads, delivery of shocks before the time of EMS arrival. Results: Of 558 cases of cardiac arrest registered during the survey period, an AED had already been delivered at the patient side by the time of EMS arrival in 92 case (16.5%). Of those, pads had been attached in 89 cased (96.7%). Shocks had been advised in 35 cases (39.3%) and to all but one of those cases at least 1 shock had been given by the bystanders. There were no statistically significant differences between the groups of cases with and without AEDs at the patient side regarding the patient age (65 years [IQR: 48-74] vs 63 years [IQR49-75], p=0.84), gender (p=0.68) and the time from call to EMS arrival (7 min [IQR: 6-9] vs 7 min [IQR: 6-9], p=0.74). Cases occurring indoors had more chances of AEDs being delivered on the patient side than cases occurring outdoors (24.0% vs 7.8%, p<0.001). Conclusions: Our study indicates that once an AED is delivered at the patient side, pads are successfully attached and shocks are given if indicated in almost all cases. This suggests that finding and delivering an AED onto the patient side are the limiting factors against the use of AEDs by bystanders. In order to popularize the use of AEDs by bystanders, informing people with AED placement and clear direction to the AED location seem to be the key steps.

Abstract. Aerosol mass concentrations are affected by local emissions as well as long-range transboundary (LRT) aerosols. This work investigates regional and local variations of aerosols based on Distributed Regional Aerosol Gridded Observation Networks (DRAGON). We constructed DRAGON-Japan and DRAGON-Osaka in spring of 2012. The former network covers almost all of Japan in order to obtain aerosol information in regional scale over Japanese islands. It was determined from the DRAGON-Japan campaign that the values of aerosol optical thickness (AOT) decrease from west to east during an aerosol episode. In fact, the highest AOT was recorded at Fukue Island at the western end of the network, and the value was much higher than that of urban areas. The latter network (DRAGON-Osaka) was set as a dense instrument network in the megalopolis of Osaka, with a population of 12 million, to better understand local aerosol dynamics in urban areas. AOT was further measured with a mobile sun photometer attached to a car. This transect information showed that aerosol concentrations rapidly changed in time and space together when most of the Osaka area was covered with moderate LRT aerosols. The combined use of the dense instrument network (DRAGON-Osaka) and high-frequency measurements provides the motion of aerosol advection, which coincides with the wind vector around the layer between 700 and 850 hPa as provided by the reanalysis data of the National Centers for Environmental Prediction (NCEP).

Genetic divergence of 40 parental lines comprising 30 restorer and 10 maintainer lines were studied through Mohalanobis's D2 and principal component analysis for eleven characters. Genotypes were grouped into five different clusters. Cluster V comprised maximum number of genotypes (thirteen) followed by cluster I and II. The inter-cluster distance was maximum between clusters I and V (13.495) indicating wide genetic diversity between these two clusters followed by the distance between cluster I and 11 (9.489), cluster IV, and cluster V (8.969) and cluster I and cluster III (8.039). The minimum inter-cluster distance was observed between cluster II and cluster III (3.034) followed by cluster 111 and cluster IV (3.834) and cluster II and cluster V (4.945) indicating that the genotypes of these clusters were genetically close. The intra cluster distance in the entire five clusters was more or less low which indicated that the genotypes within the same cluster were closely related. Among the characters panicle weight contributed most for divergence in the studied parental lines. Difference in cluster means existed for almost all the characters studied. Highest mean value for number of effective tillers (7.8), days to 50% flowering (95.5), panicles/m2 (192.6), panicle weight (2.9), spikelet fertility (84.8), number of grains/panicle (177.8), days to maturity (123.6), and grain yield/plot (1065.5) were observed in cluster I indicated the parental lines fallen in this cluster having the genetic potentiality to contribute better for yield maximization of hybrid rice. Leaves are plant structures that express important traits of the environment where they live. Leaf description has allowed identification of plant species as well as investigation of abiotic factors effects on their development, such as gases, light, temperature, and herbivory. This study described populations of Dalbergia ecastaphyllum through leaf geometric morphometrics in Brazil. We evaluated 200 leaves from four populations. The principal component analysis (PCA) showed that the first four principal components were responsible for 97.81% of variation. The non-parametric multivariate analysis of variance (NPMANOVA) indicated significant difference between samples (p = 0.0001). The Mentel test showed no correlation between geographical distances and shape. The canonical variate analysis (CVA) indicated that the first two variables were responsible for 96.77 % of total variation, while the cross-validation test showed an average of 83.33%. D. ecastaphyllum leaves are elliptical and ovate.
