Abstract Histone modifications and their functional readout serve as an important mechanism for gene regulation. Lysine benzoylation (Kbz) on histones is a recently identified acylation mark associated with active transcription. However, it remains to be explored whether putative readers exist to recognize this epigenetic mark. Here, our systematic binding studies demonstrated that the DPF and YEATS, but not the Bromodomain family members, are readers for histone Kbz. Co-crystal structural analyses revealed a ‘hydrophobic encapsulation’ and a ‘tip-sensor’ mechanism for Kbz readout by DPF and YEATS, respectively. Moreover, the DPF and YEATS family members display subtle yet unique features to create somewhat flexible engagements of different acylation marks. For instance, YEATS2 but not the other YEATS proteins exhibits best preference for Kbz than lysine acetylation and crotonylation due to its wider ‘tip-sensor’ pocket. The levels of histone benzoylation in cultured cells or in mice are upregulated upon sodium benzoate treatment, highlighting its dynamic regulation. In summary, our work identifies the first readers for histone Kbz and reveals the molecular basis underlying Kbz recognition, thus paving the way for further functional dissections of histone benzoylation.

SummaryInteractions between tranexamic acid and protein were studied in respect of the antifibrinolytic actions of tranexamic acid. Tranexamic acid did neither show any interaction with fibrinogen or fibrin, nor was incorporated into cross-linked fibrin structure by the action of factor XIII. On the other hand, tranexamic acid bound to human plasmin with a dissociation constant of 3.5 × 10−5 M, which was very close to the inhibition constant (3.6 × 10−5 M) for this compound in inhibiting plasmin-induced fibrinolysis. The binding site of tranexamic acid on plasmin was not the catalytic site of plasmin, because TLCK-blocked plasmin also showed a similar affinity to tranexamic acid (the dissociation constant, 2.9–4.8 × 10−5 M).In the binding studies with the highly purified plasminogen and TLCK-plasmin preparations which were obtained by affinity chromatography on lysine-substituted Sepharose, the molar binding ratio was shown to be 1.5–1.6 moles tranexamic acid per one mole protein.On the basis of these and other findings, a model for the inhibitory mechanism of tranexamic acid is presented.

Incubation of cultured cells of Glycine max with trans-resveratrol gave its 3-O-β-D- and 4′- O-β-D-glucosides. Cultured Gossypium hrsutum cells glycosylated trans-resveratrol to its 3-O-β-D-, 4′- O-β-D- and 3,4′- O-β-D-diglucosides. On the other hand trans-resveratrol was converted into cis-resveratrol 4′- O-β-D-glucoside together with trans-resveratrol 3- O-β-D-glucoside and trans-resveratrol 4′- O-β-D-glucoside by Eucalyptus perriniana.

Reducing the internal pH of cultured cells by several different protocols that block endocytosis is found to alter the structure of clathrin lattices on the inside of the plasma membrane. Lattices curve inward until they become almost spherical yet remain stubbornly attached to the membrane. Also, the lattices bloom empty "microcages" of clathrin around their edges. Correspondingly, broken-open cells bathed in acidified media demonstrate similar changes in clathrin lattices. Acidification accentuates the normal tendency of lattices to round up in vitro and also stimulates them to nucleate microcage formation from pure solutions of clathrin. On the other hand, several conditions that also inhibit endocytosis have been found to create, instead of unusually curved clathrin lattices with extraneous microcages, a preponderance of unusually flat lattices. These treatments include pH-"clamping" cells at neutrality with nigericin, swelling cells with hypotonic media, and sticking cells to the surface of a culture dish with soluble polylysine. Again, the unusually flat lattices in such cells display a tendency to round up and to nucleate clathrin microcage formation during subsequent in vitro acidification. This indicates that regardless of the initial curvature of clathrin lattices, they all display an ability to grow and increase their curvature in vitro, and this is enhanced by lowering ambient pH. Possibly, clathrin lattice growth and curvature in vivo may also be stimulated by a local drop in pH around clusters of membrane receptors.

Estrogen-related receptors (ERRs) are founding members of the orphan nuclear receptor (ONR) subgroup of the nuclear receptor superfamily. Twenty-seven years of study have yet to identify cognate ligands for the ERRs, though they have firmly placed ERRα (ESRRA) and ERRγ (ESRRG) at the intersection of cellular metabolism and oncogenesis. The pace of discovery for novel functions of ERRβ (ESRRB), however, has until recently been somewhat slower than that of its family members. ERRβ has also been largely ignored in summaries and perspectives of the ONR literature. Here, we provide an overview of established and emerging knowledge of ERRβ in mouse, man, and other species, highlighting unique aspects of ERRβ biology that set it apart from the other two estrogen-related receptors, with a focus on the impact of alternative splicing on the structure and function of this receptor.

Normal human erythrocyte phosphofructokinase (ATP:c D-fructose-6, P-1- phosphotransferase, EC 2.7.1.11; PFK) has recently been shown to consist of a heterogeneous mixture of five tetrameric isozymes: M4, M3L, M2L2, ML3, and L4 (M, muscle type; L, liver type). In the light of these findings, we have investigated the molecular basis of the inherited erythrocyte PFK deficiency associated with myopathy and hemolysis (Tarui disease). The propositus, a 31-yr-old male, suffered from muscle weakness and myoglobinuria on exertion. He showed mild erythrocytosis despite laboratory evidence of hemolysis. In his erythrocytes a metabolic crossover point was found at the level of PFK; 2,3-diphosphoglycerate (2,3-DPG) was also significantly reduced. The PFK from the patient's erythrocytes consisted exclusively of the L4 isozyme, and there was a complete absence of the other four. The leukocyte and platelet PFKs from the patient showed normal activities, chromatographic profiles, and precipitation with anti-M4 antibody. These studies provide direct evidence that in Tarui disease the M-type subunits are absent; but the liver- and platelet-type subunits of PFK are unaffected. The paradox of mild erythrocytosis despite hemolysis reflects the decreased production of 2,3-DPG.

The disaster due to the novel coronavirus disease 2019 (COVID-19) around the world has made investigators enthusiastic about working on different aspects of COVID-19. However, although the pandemic of COVID-19 has not yet ended, it seems that COVID-19 compared to the other coronavirus infections (the Middle East Respiratory Syndrome [MERS] and Severe Acute Respiratory Syndrome [SARS]) is more likely to target the heart. Comparing the previous presentations of the coronavirus family and the recent cardiovascular manifestations of COVID-19 can also help in predicting possible future challenges and taking measures to tackle these issues.

<p>As a social being, interaction with others is one of human’s essential needs. These interaction and socialization in a society is often conducted in a settlement. A settlement as a community ecosystem has an important role in shaping people’s characters in a society. The condition of settlement can be an indication of the condition of its community. On the other hand, Islam as a <em>rahmatan lil 'alamin </em>religion actually provides its followers a set of values on islamic aspects in  the house and settlement. Islam gives some lessons of how a house could become not only as a gathering place for family members, but could also be a place of education and learning for  its  inhabitants.  Islam  also  gives us lessons of  how a house is  closely related to its settlement  and environment, and how every elements in a house should be in harmony and unity with its social and natural environment. This paper tries to analyze many aspects of slums based on some aspects of islamic housing. This research aims to analyze the request strategies used in an American TV Series entitled Full House season 7 episodes 1-12. The main characters are family members from three different age groups (adult, teenager, and children). This research attempts to see if there is any difference on the choice of request strategies used by the three age groups. Blum-Kulka and Olshtain’s (1984) theory on request directness level is used to classify the requests. Relative power and social distance are also studied to see how they influence the characters in making their requests. The results show that mood derivable is the most frequent strategy used by all age groups. Specifically, direct request is highly used by the adult age groups. On the other hand, indirect request is mostly used by the younger age groups. The results also show that all age groups tend to use direct strategy when the social distance is negative. However, when the social distance is positive, the choice of strategy depends on the authority of the speaker.

Titus Flavius Clemens was a philosopher and Christian theologian from the period of the 2nd–3th century. The aim of this paper is to present his view on the subject of wine and his recommendations on wine consumption as described in his work entitled Paedagogus. In this work Titus Flavius Clemens focuses primarily on the moral side of drinking wine. He is a great supporter of the ancient principle of moderation, or the golden mean (μεσότης). We also find its traces in his recommendations regarding the drinking of wine. First of all, he does not require Christians to be abstinent. Although he considers water as the best natural beverage to satisfy thirst, he does not make them reject God’s wine. The only condition he sets, however, is to maintain moderation in drinking it. He recommends diluting wine with water, as the peaceful Greeks always did, unlike the war-loving barbarians who were more prone to drunkenness. On the other hand, Titus Flavius Clemens warns the reader against excessive dilution of wine, so that it does not turn out to be pure water. He severely criticizes drunkenness, picturesquely presenting the behavior of drunks, both men and women. Wine in moderation has, in his opinion, its advantages – social, familial and individual. It makes a person better disposed to himself or herself, kinder to friends and more gentle to family members. Wine, when consumed in moderation, may also have medicinal properties. Clemens is well aware of this fact and in his work he cites several medical opinions on the subject. Unfortunately, in Paedagogus we find little information about wine as a food product / as an everyday bevarage. The input on the subject is limited to the list of exclusive, imported wines. What is worth noting, Titus Flavius Clemens appears to be a sommelier in this way.
