Despite several studies on genetic markers and differently expressed genes related to ribeye area (REA) and tenderness traits in beef cattle, there is divergence in the results regarding the genes associated with these traits. Thirteen genes that had been associated or have biological functions that may influence such phenotypes were included in this study. A total of five genes for REA (IGF-1, IGF-2, MSTN, NEDD4, and UBE4A) and eight genes for meat tenderness (CAPN1, CAPN2, CAST, HSPB1, DNAJA1, FABP4, SCD, and PRKAG3) were selected from previously studies in beef cattle. Genes and its respective proteins expression were validated in a commercial population of Nellore cattle using quantitative real-time PCR (RT-qPCR) and advanced mass spectrometry (LC / MS-MS) techniques, respectively. MSTN gene was upregulated in animals with low REA. CAPN1, CAPN2, CAST, HSPB1, and DNAJA1 genes were upregulated in animals with tougher meat. The proteins translated by these genes were not differentially expressed. Our results could confirm the potential of some studied genes as biomarkers for carcass and meat quality in Nellore cattle.

The aim of this study was to identify genomic regions that associated with beef tenderness in Nellore cattle. Phenotypes were obtained according to the standard USDA Quality Grade (1999). Data from 909 genotyped Nellore bulls were used in the Genome-Wide Association Study (GWAS) undertaken using a single-step approach including also a pedigree file composed of 6276 animals. The analyses were performed using the Blupf90 software, estimating the effect of genomic windows of 10 consecutive markers. The GWAS results identified 18 genomic regions located on 14 different chromosomes (1, 4, 6, 7, 8, 10, 18, 19, 20, 21, 22, 25, 26 and 29), which explained more than 1% of the total additive genetic variance; several candidate genes were located in these regions including SLC2A9, FRAS1, ANXA3, FAM219A, DNAI, AVEN, SHISA7, UBE2S, CDC42EP5, CNTN3, C16orf96, UBALD1, MGRN1 and SNORA1 With the single-step GWAS, it was possible to identify regions and genes related to meat tenderness in Nellore beef cattle.

The activities conducted in Maros regency Tanralili district Purnakarya village in chickens and beef cattle groups Cedde Jaya. The main source of livestock feed cows during the dry season are dry forage, rice straw, corn straw and hay crops of nuts which happened to be planted at the time of drought. Besides, leaf  Gamal is also a main feed in dry  season. Number of cattle per family ownership only 2-3  head extensively maintained. Range chicken as well as extensive maintenance, ownership on average 2-5 chicken each household group.  To increase the productivity of cattle, the team provides counseling, coaching, training and mentoring as well as providing some main of facilities such as cattle shed, biogas digester, planting banana. Results are expected in the presence of  activities is the avaibility of green forage mainly during the dry season, so that the cattle population will increase, the biogas as a fuel  alternative to kerosin and LPG gas, liquid and solid fertilizer from biogas by-product process, the increasing population of chicken qualified (meat and eggs as well as the day old chick).  Besides the problem of production is also done on the farm management. The activities is expected that will increase the incomes of farmers further improve the welfare of Maros regency. For the sustainability of the program it is necessary supervision and guidance are routinely performed by the related department along with the college.

Glycosyl monomers for the assembly of multivalent ligands are typically synthesized using carbohydrates with biological functions and polymerizable functional groups such as acrylamide or styrene introduced into the carbohydrate aglycon, and monomers polymerized using a radical initiator. Herein, we report the acryloylation of 6-aminohexyl α-mannoside and its conversion into the glycosyl monomer bearing an acrylamide group. The general acryloylation procedure afforded the desired N-hexyl acetylmannosyl acrylamide monomer as well as an unexpected compound with a close Rf value. The compounds were separated and analyzed by nuclear magnetic resonance spectroscopy and mass spectrometry, which revealed the unknown compound to be the bivalent N,N-bis(hexyl α-d-acetylmannosyl) acrylamide monomer, which contains two hexyl mannose units and one acrylamide group. To the best of our knowledge, this side reaction has not previously been disclosed, and may be useful for the construction of multivalent sugar ligands.

AbstractAccumulating evidences indicate that long non-coding RNA nuclear paraspeckle assembly transcript 1 (NEAT1) promotes the progression of glioma. In this study, we postulated that NEAT1 may act as a miR-128-3p sponge. Relative levels of NEAT1 and miR-128-3p expression in human glioma samples and GBM cells were detected using quantitative real-time PCR. By means of CCK-8 assays, transwell assays, and flow cytometric analysis, the biological functions of miR-128-3p and NEAT1 were investigated in U87MG and U251MG human GBM cell lines with stable miR-128-3p and NEAT1 knockdown or overexpression. The luciferase reports, RNA pull-down assay, and RNA immunoprecipitation assay were conducted to determine the relevance of NEAT1 and miR-128-3p in glioma. As a result, high expression of NEAT1 and lack of miR-128-3p were observed in glioma specimens and cells. By binding to anti-oncogene miR-128-3p in the nucleus, NEAT1 enhanced tumorigenesis and glioma development. Further experiments suggested that ITGA5 expression was increased in glioma tissues and was found to be connected with miR-128-3p. Additionally, NEAT1 facilitated ITGA5 expression via competitively binding to miR-128-3p. For this reason, ITGA5 would not be decomposed by miR-128-3p and could activate FAK signaling pathway, thereby promoting cell growth. Collectively, these results indicated that the NEAT1/miR-128-3p/ITGA5 axis was involved in glioma initiation and progression, and might offer a potential novel strategy for treatment of glioma.

The objective of this study was to estimate and assess trends in enteric methane (CH4) emissions from the Manitoba beef cattle population from the base year of 1990 to 2008 using mathematical models. Two empirical (statistical) models: Intergovernmental Panel on Climate Change (IPCC) Tier 2 and a nonlinear equation (Ellis), and two dynamic mechanistic models: MOLLY (v3) and COWPOLL were used. Beef cattle in Manitoba were categorized in to 29 distinct subcategories based on management practice, physiological status, gender, age and production environment. Data on animal performance, feeding and management practices and feed composition were collected from the literature as well as from provincial and national sources. Estimates of total enteric CH4 production from the Manitoba beef cattle population varied between 0.9 and 2.4 Mt CO2 eq. from 1990 to 2008. Regardless of the type of models used, average CH4 emissions for 2008 were estimated to be 45.2% higher than 1990 levels. More specifically, CH4 emissions tended to increase between 1990 and 1996. Emissions were relatively stable between 1996 and 2002, increased between 2003 and 2005, but declined by 13.2% between 2005 and 2008, following the same trend as that observed in the beef cattle population. Models varied in their estimates of CH4 conversion rate (Ym, percent gross energy intake), emission factor (kg CH4 head−1 yr−1) and CH4 production. Total CH4 production estimates ranged from 1.2 to 2.0 Mt CO2 eq. for IPCC Tier 2, from 0.9 to 1.5 Mt CO2 eq. for Ellis, from 1.3 to 2.1 Mt CO2 eq. for COWPOLL and from 1.5 to 2.4 Mt CO2 eq. for MOLLY. The results indicate that enteric CH4 estimates and emission trends in Manitoba were influenced by the type of model and beef cattle population. As such, it is necessary to use appropriate models for reliable estimates for enteric CH4 inventory. A more robust approach may be to integrate different models by using mechanistic models to estimate regional Ym values, which may then be used as input for the IPCC Tier 2 model.

An increasing number of studies have shown that circRNAs are closely related to the carcinogenesis and development of prostate cancer (PCa). However, little is known about the effect of the biological functions of circRNAs on the enzalutamide resistance of PCa. Through bioinformatic analysis and experiments, we investigated the expression pattern of circRNAs in enzalutamide-resistant PCa cells. Quantitative real-time PCR was used to detect the expression of circRAB3IP, and plasmids that knock down or overexpress circRAB3IP were used to evaluate its effect on the enzalutamide sensitivity of PCa cells. Mechanistically, we explored the potential regulatory effects of eIF4A3 and LEF1 on the biogenesis of circRAB3IP. Our in vivo and in vitro data indicated that increased expression of circRAB3IP was found in enzalutamide-resistant PCa, and knockdown of circRAB3IP significantly enhanced enzalutamide sensitivity in PCa cells. However, upregulation of circRAB3IP resulted in the opposite effects. Further mechanistic research demonstrated that circRAB3IP could regulate the expression of serum and glucocorticoid-regulated kinase 1 (SGK1) by serving as a sponge that directly targets miR-133a-3p/miR-133b. Then, we showed that circRAB3IP partially exerted its biological functions via SGK1 signaling. Furthermore, we discovered that eIF4A3 and LEF1 might increase circRAB3IP expression in PCa.

<p>Siwab or its extension Mandatory cattle breeding is a manifestation of government commitment in increasing beef cattle population, and as a target for meat sufficiency in 2026. The program is believed to lead Indonesia to achieve beef self-sufficiency in the next 5-10 years. Beef cattle can be maximized in order to produce calves, and become a government’s focused program on enhancing beef cattle production through artificial insemination (AI). Based on the above problems, the government hopes to develop the program, it should not fail the umpteenth time to fulfil meat needs of the country. The purpose of this review is to describe the SIWAB program and the economic value of female beef cattle produced by AI which produces calf. This study approach is done through literature reviews related to SIWAB program implementation. SIWAB program includes two main programs namely the increase of porong cattle population through artificial insemination of AI and natural mating (Inka). With the AI through prgram, the parent beef cattle can regulate the cow's birth well. The mother cow bunting AI results can increase the selling value higher and can improve the welfare of farmers. The government's policy to pursue targeted beef self-sufficiency by the year 2026 is achieved, but the program must be responded and done well. Government policy to boost short-term beef cattle population can help to meet the needs of beef consumption, and in the long run the economic impact of farmers.</p><p> </p><p>Abstrak</p><p>Program Sapi Induk Wajib Bunting (SIWAB) adalah perwujudan komitmen pemerintah dalam meningkatkan populasi sapi potong dan sebagai target untuk kecukupan daging tahun 2026. Program tersebut diyakini dapat mengantarkan Indonesia mencapai swasembada daging sapi pada 5-10 tahun ke depan. Sapi potong dapat dimaksimalkan potensinya agar dapat menghasilkan pedet, dan menjadi program pemerintah yang difokuskan untuk peningkatan produksi sapi potong melalui inseminasi buatan (IB). Berdasarkan permasalahan tersebut di atas, harapan pemerintah dengan mengembangkan program tersebut tidak boleh gagal ke sekian kalinya dalam mencukupi kebutuhan daging di dalam negeri. Tujuan tulisan review ini adalah untuk  mendiskripsikan program SIWAB dan nilai ekonomi pada usaha sapi potong betina hasil IB yang menghasilkan pedet. Kajian ini merupakan studi pustaka melalui review berbagai referensi terkait pelaksanaan program SIWAB. Program SIWAB mencakup dua program utama yaitu peningkatan populasi sapi porong melalui inseminasi buatan IB dan kawin alam (Inka). Program IB memungkinkan mengatur kelahiran anak sapi dengan baik. Sapi induk bunting hasil IB dapat meningkatkan nilai jual lebih tinggi dan dapat meningkatkan kesejahteraan peternak. Kebijakan pemerintah adalah untuk mengejar swasembada daging sapi yang ditargetkan sampai tahun 2026 bisa tercapai, namun program tersebut harus direspon dan dikerjakan dengan baik. Kebijakan pemerintah untuk menigkatkan populasi sapi potong dalam jangka pendek bisa membantu memenuhi kebutuhan konsumsi daging sapi dan dalam jangka panjang berdampak peningkatan ekonomi peternak.</p>
