Here, we tested whether combined contrast-enhanced magnetic resonance imaging (CCE-MRI), using a mixture of gadolinium- and iron oxide-based contrast agents, can segment the bladder wall from the bladder lumen. CCE-MRI relies on the differences in particle size and contrast mechanisms of two agents for improved image contrast. Under isoflurane anesthesia, T1-weighted imaging of adult female Sprague-Dawley rat bladder was performed using standard turbospin echo sequences at 7 Tesla, before and after transurethral instillation of 0.3 ml of single-contrast MRI or CCE-MRI composed of 0.4–64 mM of gadolinium chelate (Gd-DTPA/Gadavist) and 5 mM ferumoxytol. Bladder wall contrast was assessed in the control group exposed to saline and in the bladder injury group exposed to 0.5 ml of protamine sulfate (10 mg/ml) for 30 min. CCE-MRI following instillation of 0.4–4 mM Gd-DTPA and 5 mM ferumoxytol mixture achieved segmentation between the bladder lumen and bladder wall. Hyperintensity in the bladder wall combined with hypointensity in the lumen is consistent with the increased diffusion of the dissolved Gd-DTPA and simultaneous localization of the larger nanoparticles of ferumoxytol in the lumen. The normalized hyperintense signal in the bladder wall increased from 0.46 ± 0.07 in control group to 0.73 ± 0.14 in the protamine sulfate-exposed group ( P < 0.0001). CCE-MRI following instillation of contrast mixture identifies bladder wall changes likely associated with bladder injury with improved image contrast.

Abstract Amikacin (AK) is frequently used in the treatment of gram-negative and some gram-positive infections. However, its use is limited due to nephrotoxicity due to the increase in reactive oxygen radicals. The aim of this study was to investigate the role of carvacrol (CAR) against AK-induced nephrotoxicity in rats. Thirty-two Sprague Dawley rats were randomly divided into four groups as control (Vehicle), AK (400 mg/kg), CAR + AK (80 mg/kg CAR + 400 mg/kg AK), and AK + CAR (400 mg/kg AK + 80 mg/kg CAR) groups. AK and CAR were administered via intramuscular and per-oral for 7 days, respectively. Blood and kidney tissue samples were taken at the end of the experiment. Renal function and histopathological changes were compared, and the relevant parameters of oxidative stress and inflammation were detected. Histopathological findings (necrotic changes and dilatation and inflammatory cell infiltration) significantly increased in the AK group compared to the control group. Also, the rats in the AK group lost weight significantly. It was found that CAR treatment before and after AK significantly improved nephrotoxicity histopathologically (p < 0.05). However, this improvement was not detected biochemically. These results show that CAR treatment before and after AK improves nephrotoxicity in the histopathological level.

<p>Centella asiatica is considered herbal plant for increasing memory performance. Brain-derived neurotrophin factor (BDNF) has a significant role in memory formation process, while stress causes memory impairment. Objective: This study aimed to investigate the effects of ethanol extracts of Centella asiatica leaf on serum BDNF concentration of rats that was taken serially before and after chronic electrical stress. Materials and Methods : Twenty male rats (Sprague Dawley) were divided into four groups: control/aquades group and groups treated with different doses (mg/kg) of Centella asiatica :150 (CA150), 300 (CA300) and 600 (CA600). Each rat underwent memory exercise for nine days before and after electrical stress and oral administration of ethanol extracts of Centella asiatica for twenty-eight days. Blood sampling was taken serially from rats’ tail for four times : (1) before memory exercise, (2) after memory exercise (before stress), (3) after chronic stress, and (4) after memory exercise (following chronic stress). Concentration of serum BDNF was assessed using ELISA. Results: There was no significant difference in serum BDNF concentration between groups in first and second serum sampling, which was prior to chronic stress and administration of different treatments. However, there was significant difference in third and fourth serum sampling between groups. Mean concentration of serum BDNF (ng/ml) in third and fourth sampling for control group, CA150, CA300, and CA600, respectively were 1.88+0.21 &amp;1.93+0.24; 2.29+0.13 &amp; 2.01+0.22; 2.29+0.08 &amp;1.86+0.11; 2.71+0.70 and 2.99+0.27 (p&lt;0.05). Conclusion: Ethanol extracts of Centella asiatica leaf increases serum BDNF concentration in rats after chronic stress. <br /><strong></strong></p><p><strong>Keywords</strong>: stress, memory, Centella asiatica, BDNF</p>

AbstractTo quantify the urinary bladder wall T1 relaxation time (T1) before and after the instillation contrast mixture in rats previously subjected to water avoidance stress (WAS) and/or acute exposure to protamine sulfate (PS). Female Wistar rats were randomized to receive either sham (control) or 1 h of WAS for ten consecutive days before the evaluation of nocturnal urination pattern in metabolic cages. T1 mapping of urinary bladder wall at 9.4 T was performed pre- and post- instillation of 4 mM Gadobutrol in a mixture with 5 mM Ferumoxytol. Subsequently, either T1 mapping was repeated after brief intravesical PS exposure or the animals were sacrificed for histology and analyzing the mucosal levels of mRNA. Compared to the control group, WAS exposure decreased the single void urine volume and shortened the post-contrast T1 relaxation time of mucosa- used to compute relatively higher ingress of instilled Gadobutrol. Compromised permeability in WAS group was corroborated by the urothelial denudation, edema and ZO-1 downregulation. PS exposure doubled the baseline ingress of Gadobutrol in both groups. These findings confirm that psychological stress compromises the paracellular permeability of bladder mucosa and its non-invasive assay with MRI was validated by PS exposure.

The action mechanism of acupuncture, one of the alternative methods used in obesity treatment, is unknown. The purpose of this study was to investigate the effect of electro acupuncture on weight loss and whether acupuncture treatment can change circulating levels of irisin, nesfatin-l and preptin, which have been discovered in recent years and plays an important role in nutrition and energy metabolism. Irisin, a newly discovered myokine, is associated with increased energy consumption and has a potential role in obesity. Preptin, present in islet b-cells of pancreas and associated with the carbohydrate metabolism, nesfatin1 is an anorexogenic peptide composed of 82 amino acids and regulates food intake by modulating neuropeptides in feeding centers in the brain. These three new hormones regulate energy intake and expenditure and preserve energy homeostasis and composition. In this study 4 weeks old Sprague Dawley male rats (n=16) were randomly divided into 2 groups which were the control (C): normal diet and obese (0): high fat diet (HFD) groups. 105 After 16 weeks which obesity was developed, electroacupuncture (EA) was performed to rats once in every second days for 3 times weekly for four consecutive weeks. EA was applied bilaterally to Zusanli (ST36), Bai Hui (Du20), He Gu (Li4), Tianshu (ST25) and Sanyinjiao (SP6) acupoints in animals to both groups. Blood was collected before and after EA and serum irisin, nesfatin-l and preptin levels were analyzed by ELISA (Enzyme-InducedImmunosorbent Analysis) method. After four week EA treatment, there was sginificant decrease of the average body weight in both groups rats. There were increases in the serum nesfatin-l and preptin levels in the both groups after acupuncture. A decrease was observed in the irisin levels in obese group, but an increase in the control group. These findings suggest that electroacupuncture may help to weight loss due to its effects on some important metabolic hormones such as irisin, nesfatin-l and preptin only for a few weeks. It is thought that the changes of these hormones levels may playa role in beneficial effect of acupuncture used in the treatment of human obesity. The application of EA in the treatment of obesity is very effective, which may have led to weight loss by altering these hormone levels in obese people.

To determine whether the volume reflex is defective in the diabetic state, the diuretic and natriuretic responses to acute volume expansion (VE) were measured in streptozotocin (STZ)-induced diabetic (Dia) rats. Urine flow (UV) and sodium excretion (UNaV) were measured before and after VE from innervated and denervated kidneys in anesthetized (Inactin 0.1 g/kg, ip) control and Dia rats (Sprague-Dawley rats injected with vehicle or STZ 65 mg/kg ip, respectively, 2 wk before the experiment). Blood glucose levels were significantly elevated in the Dia group compared with the control group. A VE of 1.2 ml/min for 15 min produced a significantly greater diuresis and natriuresis in control rats compared with Dia rats. In addition, reducing the hyperglycemia in Dia rats (third group) by treatment with insulin reversed the blunted UV and UNaV responses to VE. Ratios of UV (innervated-denervated, I/D) before and after VE indicate significant increases in UV by the innervated kidneys, relative to the denervated kidneys in all three groups. I/D ratios of UNa V were not different between the three groups before VE, but were significantly smaller in the Dia rats compared with both control and STZ plus insulin groups after VE. This study demonstrates that 1) there is an abnormal volume reflex in the STZ-induced Dia rats; 2) the natriuresis due to renal sympatho-inhibition is blunted in response to VE in Dia rats; and 3) restoring the glucose levels to normal by insulin treatment in the Dia rats normalizes the volume reflex.

Background: High level of total cholesterol and tryglicerides in the blood can cause atherosclerosis. Curry leaves are often used by the society to reduce total cholesterol and trygliceride levels in the blood, but no studies have examinded the effective dosage of curry leaf extract to reduce total cholesterol and triglyceride levels.Aim: This study aims to determine the effect of giving curry leaf extract (Murraya koenigii) on cholesterol and triglyceride levels of male Sprague-Dawley rats induced by high fat feed.Methods: This study was a true experimental study with a pre-post test with randomized control group design. The subjects of the study were male Sprague-Dawley rats induced by high fat feed for 10 days. Cholesterol and triglyceride levels were measured by enymatic methods. Then, the data were analyzed using paired t test and for P3 group total cholesterol using the Wilcoxon test to analyze the differences in total cholesterol and triglyceride levels between before and after given the curry leaf extract, Repeated Anova test to analyzed the differences between groups which was then followed by post hoc bonferoni and One Way Anova to analyze the differences in total cholesterol and triglyceride levels between groups.Results: There was no significant differences in total cholesterol levels between before and after given the curry leaf extract at a dose of 200 mg/KgBW, 300 mg/KgBW, and 400 mg/KgBW (p= 0.641 for the P1 group, p= 0.385 for the P2 group and p= 0.398 for the P3 group). In contrast to triglyceride levels, there were differences in triglyceride levels before and after administration of curry leaf extrcat at a a dose of 300 mg/KgBW (p= 0.012), while the other groups were not significant.Conclusion: The stratified doses of curry leaf extract (Murraya koenigii) in this study could not reduce cholesterol level and triglyceride levelKeywords: Curry leaf extract; Dyslipidemia; Total Cholesterol; Triglyceride

An atherogenic diet induces oxidative stress leading to hypercholesterolemia. This condition causes atherosclerosis followed by increased LDL and MDA. Sorghum tempeh contains fiber and antioxidants that can protectively improve LDL and MDA levels. Therefore, this research aims to determine the effect of sorghum tempeh on LDL and MDA levels in atherogenic diet-induced rats compared to sorghum flour. It used a randomized pre-post test with a control group design. The test subjects were 30 male Sprague Dawley rats, consisting of 6 normal conditioned rats (C1), and 24 that were induced by an atherogenic diet (C2, T1, T2, T3) for 2 weeks. Sorghum flour was administered at a dose of 4.095 g (T1) and the sorghum tempeh at 3.041 g (T2) and 6.081 g (T3) for 4 weeks. Furthermore, C2 was constantly induced through an atherogenic diet. Total cholesterol and LDL levels were then analyzed using the CHOD-PAP method, and MDA levels, using the ELISA method. Meanwhile, statistical analysis for these variables was carried out using IBM SPSS Statistics 21 software. The results showed that the administration of sorghum flour and tempeh significantly reduced total cholesterol, LDL, MDA levels in each group (p = 0.001). Furthermore, it showed that there was a significantly strong correlation between LDL and MDA levels before and after treatment (r = 0.610, r = 0.805, and p = 0.001). The administration of sorghum tempeh at a dose of 6.081 g caused the greatest reduction (∆) in LDL levels at -44.19 ±2.58 mg.dL-1, although, it was not the same as normal control. Meanwhile, sorghum flour at a dose of 4.095 g was the most influential in reducing MDA levels to the same as normal control with delta (∆) at -7.67 ±0.37 ng.mL-1. In conclusion, sorghum tempeh and flour were the most effective at reducing LDL and MDA levels, respectively.

Abstract: Bisphenol A (BPA), commonly found in plastic packaging and cans, can induce oxidative stress. This study was aimed to prove that oral BPA could decrease superoxide dismutase (SOD) and increase F2-isoprostan levels in male Sprague Dawley rats. This was an experimental study using randomized pretest-posttest control group design. There were 14 rats (Rattus norvegicus), male, Sprague Dawley strain, 8-10 weeks old, as subjects. Rats were divided into two groups with 7 rats each. The control group (P0) was treated with placebo for 21 days and the treatment group (P1) was treated with oral BPA dose of 400mg/kg for 21 days. Before and after treatment for 21 days, blood was drained through the canthus medial sinus orbital and urine was collected as pretest and posttest data. SOD and F2-isoprostan levels were then examined by using ELISA. The results showed that before treatment (pretest), the mean SOD level in P0 group was 42.51±3.127 U/mL and in P1 group was 41.87±2.477 U/mL (P > 0.05). The mean F2-isoprostane level in P0 group was 3.18±0.599 ng/ml and in P1 group was 3.22±0.489 ng/ml (P > 0.05). However, after treatment for 21 days (posttest), the mean level of SOD in P0 group was 43.52±3.472 U/mL and in P1 group was 31.84±2.563 U/mL (P <0.01). The mean level of F2-isoprostane in P0 group was 2.94±0.500 ng/ml and in P1 group was 4.47±0.241 ng/ml (P < 0.01). The analysis of the treatment effect showed significant decreased level of SOD and increased level of F2-isoprostane in P1 group (P < 0.01). Conclusion: Oral bisphenol A could decrease superoxide dismutase (SOD) and increase F2-isoprostan levels in male Sprague dawley rats.Keywords: bisphenol A (BPA), SOD, F2-isoprostane, Sprague Dawley ratsAbstrak: Bisphenol A (BPA) banyak ditemukan dalam kemasan plastik dan kaleng dan dapat menyebabkan peningkatan radikal bebas. Penelitian ini bertujuan untuk membuktikan pemberian BPA oral dapat menurunkan kadar superoksida dismutase (SOD) dan meningkatkan kadar F2-isoprostan pada tikus (Rattus norvegicus) jantan galur Sprague Dawley. Jenis penelitian ialah eksperimental dengan randomized pretest-postest control group design yang menggunakan 14 ekor tikus (Rattus norvegicus) jantan, galur Sprague Dawley, berumur 8-10 minggu, yang terbagi menjadi 2 (dua) kelompok masing-masing berjumlah 7 ekor tikus. Kelompok kontrol (P0) diberikan plasebo selama 21 hari dan kelompok perlakuan (P1) diberikan BPA dosis 400mg/kgBB selama 21 hari. Sebelum dan sesudah perlakuan selama 21 hari, darah diambil melalui chantus medial orbitalis dan urin dikumpulkan untuk pemeriksaan pretest dan postest. Kadar SOD dan F2-isoprostan diperiksa dengan metode ELISA. Hasil penelitian menunjukkan bahwa sebelum perlakuan (pretest), rerata kadar SOD kelompok P0 42,51±3,127 U/mL dan kelompok P1 41,87±2,477 U/mL (P > 0,05). Rerata kadar F2-isoprostan kelompok P0 3,18±0,599 ng/ml dan kelompok P1 3,22±0,489 ng/ml (P > 0,05). Setelah perlakuan selama 21 hari (posttest), rerata kadar SOD kelompok P0 43,52±3,472 U/mL dan kelompok P1 31,84±2,563 U/mL (P < 0,01). Rerata kadar F2-isoprostan kelompok P0 2,94±0,500 ng/ml dan kelompok P1 4,47±0,241 ng/ml (P < 0,01). Analisis efek perlakuan menunjukkan terjadi penurunan kadar SOD dan peningkatan kadar F2-isoprostan yang bermakna pada kelompok P1 (P < 0,01). Objectives. To determine whether feeding CircuCare to rats improves blood circulation, metabolism, immune regulation, endocrine activity, and oxidative stress. Methods. 28 eight-week-old male Sprague-Dawley rats were evenly randomized into control and experimental groups. The control group was fed with ordinary drinking water, while the experimental group was fed with CircuCare at a daily dose of 93.75 mg per 300 g of body weight over eight weeks. Both groups were subjected to a swimming test, and blood samples were taken to observe any variations in various biochemical parameters before and after the test. Key Findings. The experimental group’s mean swimming exhaustion duration was 53.2% longer and had a significantly higher lactic acid removal ratio. Their mean prostaglandin E2 level and mean glucose, cortisol, and glutathione level (30 minutes after swimming test) were also significantly higher. No undesirable impacts from CircuCare relating to general blood biochemistry values and bone mineral density were reported. Conclusions. The present results show that CircuCare can be safely used to increase stamina and exercise capability, expedite the metabolism of lactic acid, accelerate muscle repair, and promote the antioxidant activity of cells in rats.

This study aimed to estimate the reduction in the minimum alveolar concentration (MAC) of sevoflurane induced by low and high doses of methadone (5 and 10 mg/kg), tramadol (25 and 50 mg/kg), butorphanol (5 and 10 mg/kg) or morphine (5 and 10 mg/kg) in the rat. A control group received normal saline. Sixty-three adult male Sprague-Dawley rats were anaesthetized with sevoflurane ( n = 7 per group). Sevoflurane MAC was then determined before and after intraperitoneal administration of the opioids or saline. The duration of the sevoflurane MAC reduction and basic cardiovascular and respiratory measurements were also recorded. The baseline MAC was 2.5 (0.3) vol%. Methadone, tramadol and morphine reduced the sevoflurane MAC (low dose: 31 ± 10, 38 ± 15 and 30 ± 13% respectively; high dose: 100 ± 0, 83 ± 17 and 77 ± 25%, respectively) in a dose-dependent manner. The low and high doses of butorphanol reduced the sevoflurane MAC to a similar extent (33 ± 7 and 31 ± 4%, low and high doses, respectively). Two rats developed apnoea following administration of high-dose butorphanol and methadone. These anaesthetic-sparing effects are clinically relevant and may reduce the adverse effects associated with higher doses of inhalational anaesthetics.
