The study was designed to evaluate the phenolic, flavonoid contents and antioxidant and antimicrobial activities of onion (Allium cepa), garlic (Allium sativum), mint (Mentha spicata), thyme (Thymus vulgaris), oak (Quercus), aloe vera (Aloe barbadensis Miller), and ginger (Zingiber officinale). All extracts showed a wide range of total phenolic contents, that is, 4.96 to 98.37 mg/100 g gallic acid equivalents, and total flavonoid contents, that is, 0.41 to 17.64 mg/100 g catechin equivalents. Antioxidant activity (AA) was determined by measuring reducing power, inhibition of peroxidation using linoleic acid system, and 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging activity. Different extracts inhibited oxidation of linoleic acid by 16.6–84.2% while DPPH radical scavenging activity (IC50 values) ranged from 17.8% to 79.1 μg/mL. Reducing power at 10 mg/mL extract concentration ranged from 0.11 to 0.84 nm. Furthermore the extracts of these medicinal herbs in 80% methanol, 80% ethanol, 80% acetone, and 100% water were screened for antimicrobial activity by disc diffusion method against selected bacterial strains, Staphylococcus aureus, Escherichia coli, Bacillus subtilis, and Pasteurella multocida, and fungal strains, Aspergillus niger, Aspergillus flavus, Rhizopus solani, and Alternaria alternata. The extracts show better antimicrobial activity against bacterial strains as compared to fungal strains. Results of various assays were analyzed statistically by applying appropriate statistical methods.

In the present study, we report for the first time the antioxidant and antimicrobial activities of Withania frutescens (L.) Pauquy roots and leaves. Total phenolic content was determined using the Folin-Ciocalteu method and antioxidant activity was evaluated by the DPPH free radical scavenging and reducing power methods. Antimicrobial activity tests were carried out against ten bacterial species involved in nosocomial infections and two opportunistic clinical yeast isolates. The ethyl acetate and n-butanol leaf fractions exhibited the highest DPPH radical scavenging activity with IC50 = 4.53 ± 0.12 and 8.49 ± 0.46 μg/mL, respectively. The n-butanol root fraction showed the greatest reducing power comparable with that of quercetin at 0.4 mg/mL. The dichloromethane leaf fraction exhibited the highest antibacterial activity against both Gram-positive and Gram-negative bacteria with MIC values ranging between 50 and 400 μg/mL, depending on the tested bacteria. However, none of the examined extracts exhibited anticandidal activity. The polyphenol and glycowithanolide constituents appeared to be responsible for the antioxidant capacity of W. frutescens, whereas the observed antimicrobial activity may be due to the presence of withanolides.

Objective: This present study is planned to reveal about the pharmacognostical details, phytochemistry, antimicrobial activity, and antioxidant activity of aqueous and alcoholic extract of the berries of Solanum torvum Swartz. belonging to the family Solanaceae, which is a very good anti-inflammatory medicine in traditional system of therapies.Methods: The physicochemical parameters such as ash values, extractive values, and total fiber content, qualitative preliminary phytochemical studies, quantitative estimation for total phenolic contents and flavonoid content, anjd inorganic minerals such as sodium, calcium, and potassium were estimated. Flavonoids present in the methanolic extract were isolated and its Rf values were calculated by high-performance thin-layer chromatography method and compared with that of the standard epicatechin. The antioxidant activity was evaluated by Reducing power ability, 1, 1-diphenyl-2-picrylhydrazyl method, and hydrogen peroxide methods. The antibacterial and antifungal activities of the aqueous and methanolic extracts are evaluated by disc diffusion method comparing the zone of inhibition with that of the standards such as amikacin and Ketoconazole.Results: It contains a number of potentially pharmacologically active chemicals including the sapogenin steroid, chlorogenin, alkaloids, flavonoids, phenols, and tannins. Both the extracts showed good activity, but the alcohol extract is more active toward all bacterial organisms and also toward fungi organism. Methanol extract shows good antioxidant activity. Antimicrobial activity and antioxidant activity of the berries are due to the presence of phytoconstituents present in it.Conclusion: These berries can be used for new formulation in nutraceuticals and polyherbal natural formulations for reducing the oxidative stress-related diseases and different therapies.

AbstractSeaweeds are able to produce a great variety of secondary metabolites that are characterized by a broad spectrum of biological activities. Two seaweeds species, namelyHypnea musciformisandEnteromorpha intestinalis werestudied to evaluate the phytochemical constituency and antimicrobial activities. First of all, crude extracts of both sea weeds were prepared by two different extraction methods (soaking and water bath) using different solvents. Phytochemicals profiling results revealed the presence of bioactive compounds (flavonoids, alkaloids, tannin, saponin and phenols) in both seaweed extracts. Quantification results for ethanolic extracts ofH. musciformis and E. intestinalisestimated 51 mg and 43 mg tannins in per gram of dried samples and flavonoids contents were found 67 mg and 57 mg/g mg QE/g respectively. Total phenolic contents were determined in terms of gallic acid equivalent (GAE).H. musciformisexhibited higher amount of phenolics (59 ± 0.0002 mg GAE/g) thanE. intestinalisextracts (41 ± 0.0002 mg GAE/g). In antimicrobial activity test, ethanol extractsof H. musciformisandE. intestinaliswere found 10 mm of inhibition diameter against all of the bacterial strains. Besides, methanol extracts ofE. intestinaliswere more susceptible toStaphylococcus aureus and Pseudomonaswhich was close to the inhibition diameter (>15 mm) of the mainstream antibiotic; Gentamicin. Moreover,Klebsiella sp. was found more susceptible to ethanol and methanol extracts of Hypnea musciformisas it showed inhibition zone greater than 15 mm. Both Seaweed extracts possessed higher amount of phytochemicals and showed promising antimicrobial activities when compared with the standards.

The phytochemical contents of Salvia virgata mericarps and the antioxidant and antimicrobial activities of its ethanol extract were studied for the first time. S. virgata mericarps were collected in the province of Trabzon, Turkey. Chemical analyses were performed using chromatographic methods. The total phenolic and flavonoid contents and antioxidant potential were measured using Folin-Ciocalteu, Al(NO3)3, and DPPH radical scavenging activity methods, respectively. The antimicrobial activity was evaluated using the microdilution method for all the tested bacterial and fungal strains, with the exception of Mycobacterium tuberculosis for which the resazurin microtiter plate method was applied. The mericarps were rich in glutamic acid (3934 mg/100 g), potassium (12578.8 ?g/g), calcium (12092.0 ?g/g), and dietary fibers (35.565 g/100 g). The total phenolic and flavonoid contents were 2.50 ?g GAE/mg extract and 0.34 ?g QE/mg extract, respectively. The most effective DPPH free radical scavenging activity determined for the highest applied concentration was 92.44%. The ethanol extract obtained from the mericarps was found to be as effective as the reference drug ampicillin (MIC value = 125 ?g/mL) against the nosocomial pathogen Acinetobacter baumannii. In conclusion, S. virgata mericarps provide good nutritional value with low amounts of carbohydrates and high dietary fibers, amino acids, minerals and total phenolic and flavonoid contents and medicinal properties.

Background: Synthetic azo compounds and their derivatives have been studied extensively due to their biological and pharmacological activities. Pyranopyridines, pyranopyrimidines and tetrazoles derivatives have emerged as a promising and attractive scaffold in the development of potent biological and pharmacological agents. Objectives: To design a series of new benzochromeno(pyridine/pyrimidine/tetrazole) derivatives and evaluate their antimicrobial activity against some bacterial strains (Gram-positive and Gram-negative) and some fungal strains. Materials and Methods: The (E)-7-(4-chlorophenyl)-5-(phenyldiazenyl)-10-thioxo-7,9,10,11-tetrahydro-8Hnaptho[ 1,2-b]pyrano[2,3-d]pyrimidin-8-one (4) was synthesized by the reaction of 4H-naphtho[1,2-b]pyran-3- carbonitrile (3) with carbon disulfide in alcoholic potassium hydroxide solution. Reaction of 3 with sodium azide in DMF and in presence of ammonium chloride afforded 6-(phenyldiazenyl)-3-(1H-tetrazol-5-yl)-4Hbenzo[ h]chromen-2-amine (7) while with malononitrile, thiourea or urea gave chromeno[2,3-b]pyridine-9- carbonitrile (8), chromeno[2,3-d]pyrimidine-10-thione (9) and chromeno[2,3-d]pyrimidin-10-one (10), respectively. The assignment structures were established on the basis of spectral data. Results: In this study, the antimicrobial activity of the synthesized compounds 3-12 was examined for their in vitro antimicrobial activity by using agar diffusion method such as Mueller-Hinton agar medium for bacteria and Sabouraud’s agar medium for fungi. Ampicillin and mycostatine were included in the experiments as reference drugs. Conclusion: A series of new benzochromeno(pyridine/pyrimidine/tetrazole) derivatives were synthesized in this work. All compounds were evaluated in antimicrobial activities.

Background: The Algerian medicinal plant Thymus pallidus Coss. is traditionally known to be used as an anti-infective, expectorant, healer of wounds and against inflammatory and gastrointestinal diseases. Objective: The aim of the present study is to investigate the antioxidants and the antibacterial activities of the aerial part extracts of this plant. Methods: Polyphenols extraction was performed with methanol 85%, followed by preliminary fractionation based on the polarity of the solvent. The extraction and fractionation process gave the crude extract (CrE), chloroform extract (ChE), ethyl acetate extract (EtAE) and aqueous extract (AqE). The antioxidant activity of extracts was evaluated using in vitro assays. While the antimicrobial activity was determined using disk diffusion method. Results: Quantitative analysis of polyphenol and flavonoids showed that EtAE includes the highest amount (P≤0.001) with 495.12±1.91 μg GAE/mg of extract and 97.25±2.84 μg QE/mg of extract, for both polyphenols and flavonoids respectively. The results of the antioxidant activity of extracts showed that the EtAE exhibited the highest inhibition activity on XO (IC50 =51.6±0.2 μg/ml) compared to other extracts (P≤0.001). Moreover, the EtAE was the most effective fraction in free radical scavenging assay against ABTS radical and hydrogen peroxide (EC50= 3.21±0.03 μg/ml and EC50 =12.12±0.05 μg/ml, respectively) compared to other extracts (P≤0.01). Using β-carotene/ linoleic acid bleaching assay, the AqE and ChE recorded a considerable inhibition effect on linoleic acid oxidation with a ratio of 81.81% and 80.86%, respectively. In the antimicrobial activity evaluation, the extracts exhibited moderate antibacterial activity against bacterial strain, with inhibition zone ranging from 07 to 14 mm. In addition, the EtAE showed a good action spectrum against six bacterial strains. In conclusion, the EtAE of Thymus pallidus showed good antioxidant activity and antibacterial effect. Conclusion: The obtained results revealed the presence of a strong correlation between the antioxidant and antibacterial activities of the extract and its total phenolic content. Furthermore, they reported that the EtAE has a considerable antioxidant capacity. This can be considered as an alternative natural source of antioxidants used generally as additives in food and pharmaceutical preparation.

The present study was carried out for phytochemical screening of principle bioactive compounds and antimicrobial activity in Elaeocarpus ganitrus Roxb., Phytochemical analysis revealed the presence of saponin, terpenoid, steroid, saponin, flavonoid, tannin and alkaloid. The petroleum, ether, chloroform, methanol, acetone and aqueous extracts were subjected to antimicrobial activity against bacterial strains Staphylococcus aureus, Pseudomonas, E. coli and Bacillus subtilis against anti-fungal strains A.awamori, A.fumigatus, Rhizopus oryzae, Trichoderma viridae and C.oryzae. The antibacterial and antifungal activity was evaluated by disc-diffusion method.

Chitosan, a natural polysaccharide, has been previously proposed as an elicitor in plants to prevent pathogen infections. The present study aimed to analyze the effect of chitosan solution and chitosan nanoparticles treatment applied on the grapevine variety Sousão with respect to the phenolic composition, antioxidant potential and antibacterial activity of its individual grape components. Grapevine plants of selected lines were sprayed with chitosan solution and chitosan nanoparticles, and ethanolic extracts of stems, seeds and skins were prepared from grapevines treated and not treated with chitosan. Total phenolic, anthocyanin and tannin contents were studied, and the identification of the individual phenolic compounds was performed by HPLC-DAD. The antimicrobial susceptibility method was performed using the Kirby-Bauer disc diffusion method against multidrug-resistant bacteria. Overall, there was small increase in the concentration of phenolic compounds, antioxidant and antimicrobial activities in grape components treated with chitosan solution. Seed extracts showed the highest antioxidant and antimicrobial activities. The studied individual components obtained from chitosan-treated grapevines could represent an added value due to the increased antioxidant and antibacterial potentials. The phenolic compounds found in components may be used in food and pharmaceutical industries as natural food preservers and antibiotic adjuvants.

The ethanol crude extracts (ECE) and their subfractions fromZanthoxylum bungeanumleaves were prepared and their phytochemical profiles and antioxidant and antimicrobial activities were investigated. Moreover, the effective HPLC procedure for simultaneous quantification of twelve compounds inZ. bungeanumleaves was established. The correlation between the phytochemicals and antioxidant activity was also discussed. The ethyl acetate fraction (EAF) had the highest total phenolic (97.29 mmol GAE/100 g) and flavonoid content (67.93 mmol QE/100 g), while the greatest total alkaloid content (4.39 mmol GAE/100 g) was observed in the chloroform fraction (CF). Twelve compounds were quantified by RP-HPLC assay. EAF exhibited the highest content of quercitrin, kaempferol-3-rhamnoside, quercetin, sesamin, and nitidine chloride (125.21, 54.95, 24.36, 26.24, and 0.20 mg/g); acetone fraction (AF) contained the highest content of chlorogenic acid, rutin, hyperoside, and trifolin (5.87, 29.94, 98.33, and 31.24 mg/g), while kaempferol-3-rhamnoside, xanthyletin, and sesamin were rich in CF. EAF and AF exhibited significant DPPH, ABTS radical scavenging abilities and reducing power (FRAP), whereas CF exhibited significant antifungal activity. Moreover, EAF also showed stronger antibacterial activity. In conclusion,Z. bungeanumleaves have health benefits when consumed and could be served as an accessible source for production of functional food ingredients and medicinal exploration.
