In vitro Efficacy of Experimental Chitosan-Containing Solutions as Anti-Erosive Agents in Enamel

The present study evaluated the effect of chitosans with different viscosities, dissolved in an AmF/SnCl2 solution, against erosion or erosion/abrasion. A total of 192 specimens were assigned to 2 × 6 groups (n = 16 specimens each): negative control, 4 chitosan solutions (groups Ch50, Ch500, Ch1000, and Ch2000, with viscosity of 50, 500, 1,000, or 2,000 mPas, respectively, 0.5% chitosan, 500 ppm F-, 800 ppm Sn2+, pH 4.4), and positive control (500 ppm F-, 800 ppm Sn2+, pH 4.3). One half of the groups was demineralized (experiment 1, E1; 10 days, 6 × 2 min/day, 0.5% citric acid, pH 2.8) and exposed to solutions (2 × 2 min/day); the other half was additionally brushed (15 s, 200 g) with non-fluoridated toothpaste before solution immersion (experiment 2, E2). Treatment effects were investigated by profilometry, energy-dispersive X-ray spectroscopy and scanning electron microscopy (SEM). In E1, all the chitosan-containing solutions reduced enamel loss by 77-80%, to the same extent as the positive control, except for Ch2000 (p ≤ 0.05), which completely inhibited tissue loss by the formation of precipitates. In E2, Ch50 and Ch500 showed best performance, with approximately 60% reduction of tissue loss compared to the negative control group (p ≤ 0.05 compared to other groups). SEM analysis showed differences between negative control and the other groups but only minor differences amongst the groups treated with active agents. In both E1 and E2, treatment with active agents resulted in surface enrichment of carbon and tin compared to negative control (p ≤ 0.001); brushing removed parts of carbon and tin (p ≤ 0.001). Chitosan shows different properties under erosive and erosive/abrasive conditions. Under erosive conditions high viscosity might be helpful, whereas lower viscosity seems to be more effective in cases of chemo-mechanical challenges.

Physalia utriculus is one of the invertebrate marine biota that is often found in Indonesia. Some symptoms of venoming due to jellyfish stings cause pain, itching, and hemolysis. In Indonesia, 13 cases of jellyfish stings were reported in 2005-2009 with three people dying in Java, Bali, and Bangka. Cocoa beans (Theobroma cacao L.) contain fat, carbohydrates, proteins, and polyphenol compounds that are useful as antioxidants. Polyphenols in the form of epicathechins, catechins, and procyanidins serve to provide protection to hemolysis. The purpose of this study was to determine the potential of ethanol extract of cacao (Theobroma cacao L.) in inhibiting the damage of erythrocyte induced by Physalia utriculus in vitro. This study used 28 samples of erythrocytes divided into seven groups, namely the normal control group, negative controls, and treatment with cocoa ethanol extract 0.2%, 0.1%, 0.04%, and 0.02%. Each subsequent group induced venom Physalia utriculus. The results showed that the average speed of erythrocyte lysis in the treatment group by giving cocoa ethanol extract 0.2%, 0.1%, 0.04%, and 0.02% respectively (seconds ± standard deviation) was 858,25 ± 94,44; 1.000,5 ± 159,93; 678,5 ± 19,71; and 1.006 ± 159,50. The mean speed of erythrocyte lysis in the negative control group was 1,025 ± 164.63 and the positive control group with the administration of N-Acetylcystein can last up to one hour after administration of venoms. Test for normality and homogeneity shows that data is normally distributed and homogeneous. One Way Annova analysis shows the significance value of p <0.05, then a post hoc analysis test was performed with the Bonferoni method to find out the differences in significance in each group. In this study it can be concluded that the administration of cocoa ethanol extract has no potential to inhibit erythrocyte damage that has been venomed by Physalia utriculus in vitro. Abstract Background The present study compares the apical microleakage of three different root-end filling materials in which the retrograde cavity is prepared by two different burs. Methods Eighty extracted single rooted maxillary and mandibular premolars were taken. Root canal treatment was completed. Apical 3 mm of all the teeth were resected with diamond disk. The tooth were divided into four groups with two subgroups for each group containing 10 tooth (N= 10) as: Group IA (Negative Control and IB (Positive Control); Group IIA and IIB: Prepared with round carbide bur and round diamond bur respectively, filled with GIC; Group IIIA and IIIB: Prepared with round carbide bur and round diamond bur respectively, filled with MTA; Group IVA and IVB: Prepared with round carbide bur and round diamond bur, filled with Biodentine. After applying two coats of nail varnish leaving apical 3 mm (except for negative control group) all teeth were immersed in 2% methylene blue for 3 days and again in 65% nitric acid for next 3 days for extraction of dye. The obtained solution was then transferred to eppendorf tube and centrifuged in microcentrifuges at 14,000 revolution per minutes (RPM) for 5 minutes. Optical density or absorbance of the supernatant solution was measured with UV spectrophotometer at 550 nm. Results The absorbance of the supernatant solution after dye extraction is decreasing in the order of positive control> GIC> MTA> Biodentine> negative control group. The significant difference was observed between GIC and MTA (p=0.0001) and GIC and Biodentine (p=0.0001) with two different burs but statistically non-significant difference was observed between MTA and Biodentine with Carbide bur (p=0.127) and Diamond bur (p=0.496) respectively. Conclusions Within the limitations of the present study, it can be concluded that Biodentine and MTA showed less microleakage as compared to GIC. There is no significant difference between mean microleakage of MTA and Biodentine. However, the mean OD of the Biodentine was least of all evaluated materials. Preparation of the root-end using round carbide bur as well as round diamond burs showed comparable microleakage for all three filling materials.

Indonesia has  biodiversity potential to be developed as medicinal plants, such as basil leaves (Ocimum sanctum L.). Basil was reported to have a very high antioxidant activity in vitro. The aim of this study was to determine the effect of basil leaves (Ocimum sanctum L.) infusion to liver based Aspartate Aminotransferase (AST) and Alanine Aminotransferase (ALT) value in each dose. 18 rats were divided into 6 groups.,  control group,  negative control group treated with CMC Na 0.5%,  positive control group treated with Curcuma 3.6 mg / 200gBB, groups of 4-6 consecutive given a 80 mg  dose infusion basil / 200 gBB, 160 mg / 200 gBB, 320 mg / 200 gBB for 8 consecutive days, on the day of the 4th and 8th all treatment groups induced by toxic doses of paracetamol (500 mg / 200 gBB) except the normal control group 1, The research data in the form of enzyme activity of AST and ALT were analyzed using parametric and nonparametric ANOVA, and Friedman test with the level of trust  then followed by SNK test and Bnj test. The statistical test result with a 95% of level of trust that shown basil infuse with a dose of 80mg / 200gBB, 160mg / 200gBB, 320mg / 200gBB have hepatoprotective effects in rats induced by paracetamol 500mg / 200gBB. Based on the result of changes in average levels of AST on the fourth day and the eighth day of the three treatment infusion, infusion at a dose of 160 mg / 200gBB most effectively reduce average levels of AST and a group that has the average AST closest to the control group is positive, but infusion at a dose of 320mg / 200gBB the group that has the closest average ALT positive control group.Keywords : hepatoprotective, Ocimum sanctum L, Paracetamol

ABSTRACTAlthough beta-lactam antibiotics are not considered as antituberculous drugs, it has been recently shown that the combination of carbapenems and clavulanate is bactericidalin vitro. We evaluated in a murine model of tuberculosis the activity of carbapenems alone and combined with clavulanate againstMycobacterium tuberculosis. Swiss mice infected intravenously with 3 × 105M. tuberculosisH37Rv were treated for 4 weeks with clavulanate alone or imipenem, meropenem, and ertapenem alone or combined with clavulanate, whereas a positive control group was treated with isoniazid, and a negative control group was held without treatment. The combination of imipenem or meropenem plus clavulanate significantly improved survival. Among groups of mice with 100% survival, only isoniazid reduced lung CFU counts; the carbapenem-clavulanate combinations did not prevent bacterial growth. Although less active than isoniazid, the combinations of imipenem or meropenem plus clavulanate improved the survival of mice infected withM. tuberculosisand should be further evaluated.

There was estimated 150 million envenomation cases due to jellyfish stings occur globally every year. 100 from 10,000 jellyfish species in the world known to be dangerous, one them is Physalia utriculus. The aim of this study was to determine the potential of cocoa (Theobroma cacao L.) ethanol extract toward nematocyst tubul firing inhibition of jellyfish (Physalia utriculus) toxin in vitro. The method was true experimental design with post test only control group design. The study divided into 8 groups: 1 normal control, 1 positive control, 1 negative control, and 5 treatments grups by giving cocoa ethanol extract with concentration 20%, 2%, 0.2%, 0.02%, 0.002%. The observation was made by calculating the percentage number of firing nematocysts. The result showed average percentage of firing nematocyst in the normal control group was 42.50 ± 3.18, positive control group was 37.97 ± 5.57, negative control group was 52.44 ± 2.98, and treatment group which given with cocoa ethanol extract 20%, 2%, 0.2%, 0.02%, 0.002% were 48.24 ± 5.37; 40.62 ± 7.10; 29.45 ± 5.39; 37.60 ± 9.78; 41.11 ± 3.92, respectively. The One Way Annova statistical results test show significance value 0.001 (p≤0.05). The conclusion of this study was the cocoa ethanol extract with concentration 0.2% has most potential to inhibit the jellyfish (P. utriculus) nematocyst tubule firing in vitro. The aim of this research was to find out in vitro antiplasmodium activity of Malacca leaves (Phyllantus emblica) ethanolic extract against Plasmodium falciparum growth. In this study, Plasmodium culture contained 5% parasitemia in ring stage was cultured using candle jar method and antiplasmodial activity test was carried out using microculture. The treatments were divided into 7 groups with four repetitions. K1 as negative control group was given Roswell Park Memorial Institute (RPMI), while K2 as positive control group was given artesdiaquine. Groups K3, K4, K5, K6, and K7 group was added with 100 µg/mL, 75 µg/mL, 50 µg/mL, 25 µg/mL, and 5 µg/mL of Malacca leaves ethanolic extract, respectively. Antiplasmodial activity was determined by inhibition concentration of 50% parasite growth (IC50). The data were analyzed using ANOVA and followed by Duncan test. The average of parasitemia level in group K1, K2, K3, K4, K5, K6, and K7 were 55.25±15.62, 8.50±2.52, 8.50±3.00, 9.25±0.95, 9.00±2.70, 9.79±2.06, and 10.75±2.22, respectively. The average of inhibition percentage in group K1, K2; K3; K4; K5; K6; and K7 were 0.00±0.00%, 84.62±4.55%; 84.62±5.43%; 83.26±1.73%; 83.71±4,90%; 82.35±3,73%; and 80.54±6.83%, respectively (P0.01). The results showed that the administration of malacca leaves ethanolic extract significantly affect (P0.01) the inhibition of Plasmodium growth as compared to group K1 (negative control). Probit analysis reveals the IC50 value was 3.889 µg/mL. In conclusion, all doses of malacca leaves ethanolic extract used in this study was able to inhibit Plasmodium falciparum growth with IC50 value was 3.889 µg/mL.

Dental caries is a disease caused by Streptococcus mutans. The use of chlorhexidine to inhibit bacterial colonization has side effects such as tooth staining and can kill the normal flora when used long term. Epigallocatechin gallate (EGCG) is a chemical compound in the form of polyphenols from green tea catechins which have antimicrobial potency to inhibit microorganism growth and biofilm formation. Type Laboratory Experimental Research In-vitro. The group that will be studied are the negative control group in the form of S.mutans + 5% sucrose, the treatment group in the form of S. mutans + 5% sucrose and EGCG concentration of 0.125mg/ml, 0.25mg/ml, 0.375mg/ml and a positive control group is S.mutans + 5% sucrose and 0.1% chlorhexidine. Data were analyzed using the Kolmogorov-Smirnov test to determine the normality of the data, Levene's test for homogeneity of data, One Way ANOVA Post Hoc Tukey HSD Multiple Comparison to determine differences between treatments. Results: There were significant differences between the treatment groups and the negative control at test results Post Hoc Tukey HSD and the significant differences in the concentration of EGCG 0.375mg/ml with the positive control given chlorhexidine 0.1% (p <0.05). Epigallocatechin gallate (EGCG) influence on the activity of S. mutans biofilm formation and EGCG concentration of 0.375mg/ml are more effective as an antibiofilm of S. mutans compared with chlorhexidine 0.1%.

The aim of this in vitro study was to investigate the effect of probiotic and isotonic drinks on chromium (Cr) and nickel (Ni) release from dental archwires. The conventional stainless steel dental archwires were used in this study, with 0.8 cm diameter, and 5 cm long from each sample. Total samples are 54 pieces dental archwires. The samples then divided into two major groups: (1) the probiotic (T1); and (2) isotonic (T2) drinks. Each major group consist three subgroups, such as: for the T1: (1) T1,1: as a negative control group (samples were immersed in saline solution); (2) T1,2: as a positive control group (samples were 1% lactic acid); (3) T1,3: as a treatment group (samples were immersed in normal saline + probiotic drink); and for the T2: (1) T2,1: as a negative control group (samples were immersed in saline solution); (2) T2,2: as a positive control group (samples were immersed in 1% lactic acid); (3) T2,3: as a treatment group (samples were immersed in normal saline + isotonic drink). After the immersion testing, there was a significance difference in Cr and Ni release from dental archwires between the subgroup of treatments from both probiotic and isotonic drinks (Kruskal-Wallis test followed by Mann-Whitney test; P > 0.05). In conclusion, the present study demonstrated that both probiotic and isotonic drinks induced the releasing of Cr and Ni from dental archwires.
