Protective effect of coenzyme Q10 against bisphenol-A-induced toxicity in the rat testes

The present study was conducted to investigate the antioxidant, histomorphometric, histochemical, immunohistochemical, biochemical, and cytological effects of coenzyme Q10 (CoQ10) against bisphenol-A (BPA)-induced testicular toxicity in rats. A total of 40 adult male Wistar rats were divided into five equal groups. The control group remained untreated. The vehicle control group was administered corn oil (2 ml/kg/day), the BPA group was given BPA (100 mg/kg/day), the CoQ10 group was supplemented with CoQ10 (10 mg/kg/day), and the rats in the CoQ10-BPA group received CoQ10 (10 mg/kg/day) followed by BPA (100 mg/kg/day) 1 h later. The treatments were administered by oral gavage for 14 days. Results showed that the seminiferous tubule diameters (STDs) and seminiferous epithelium heights (SEHs) at stages VII–VIII and XII–XIV, number of undifferentiated embryonic cell transcription factor-1 (UTF-1) positive cells per tubule, UTF-1 positive tubules (%), plasma glutathione (GSH), and serum superoxide dismutase activities, testicular GSH activity and sperm viability (%) decreased whereas the number of terminal dUTP nick end labeling (TUNEL) positive cells per tubule, TUNEL positive tubules (%), testicular and serum malondialdehyde (MDA) levels, and the rate of mid-piece sperm abnormality increased in the BPA administered group. However, while the STDs at stages VII–VIII and XII–XIV, SEHs at stages VII–VIII, plasma GSH, and serum SOD activities increased, serum MDA level decreased in the CoQ10-BPA group. In conclusion, these results suggest a protective effect of CoQ10 against BPA-induced testicular toxicity in rats.

Background and Objective.Bisphenol A (BPA) is an abundantly used xenoestrogenic chemical which may cause various disorders in body. In the present study, we sought to investigate the effects of various doses of BPA on hepatic oxidative stress-related gene expression in rats.Methods.Male Wistar rats weighing 150–200 g were used in this study. Three doses of the BPA (5, 25, and 125 μg/kg) in corn oil were administered as gavage during 35 consecutive days. After the experiment, the rats were expired and the livers were removed and stored at −80°C freezer for RNA extraction.Findings.The Real Time PCR showed increased expression of HO-1 in the rats receiving BPA doses compared to the control group. This effect was dose-dependent and higher at doses of 25 and 125 μg/kg than 5 μg/kg of body weight (p<0.05). It was also demonstrated that various doses BPA can increase GADD45B gene expression compared to control group. That expression was significantly dominant in the lowest dose (5 μg/kg) of the BPA (p<0.05). The final body weights (168.0±10.0 gr) in the treatment group [BPA (125 μg/kg)] showed a significant decrease compared to control group (191.60±6.50 gr).Conclusion.These findings demonstrate that BPA generated ROS and increased the antioxidant gene expression that causes hepatotoxicity.

Pak-wanban (Sauropus androgynus L. Merr.), a popular Thai vegetable, has been found to have a high content of Coenzyme Q10 (CoQ10), which is a powerful antioxidant. This study investigated the quantitation, absorption and tissue distribution of CoQ10 from raw and stir-fried Pak-wanban and its antioxidant activities in rats. Male Wistar rats (seven weeks old) were randomly grouped as follows: (1) control, (2) raw Pak-wanban powder of 0.5 mg CoQ10/kg/day, (3) stir-fried Pak-wanban powder of 0.5 mg CoQ10/kg/day, (4) stir-fried Pak-wanban powder of 1.0 mg CoQ10/kg/day, and (5) commercially CoQ10 supplement groups of 0.5 mg CoQ10/kg/day. The results found that stir-fried cooking did not significantly reduce the content of CoQ10 in the Pak-wanban leaves. After 3 weeks of experimentation, the level of CoQ10 in the plasma, liver and spleen was increased in all Pak-wanban groups when compared to the control group. The level of CoQ10 in the stir-fried Pak-wanban group was significantly higher than the raw Pak-wanban group but slightly lower than the CoQ10 supplement group.  Liver alpha-tocopherol concentrations were markedly increased in rats that consumed a high dose of CoQ10 from stir-fried Pak-wanban of 1 mg of CoQ10/kg/day when compared with the control group. Plasma antioxidant activities (ORAC: FRAP: DPPH) were significantly increased in both groups of stir-fried Pak-wanban when compared with the control group. We concluded that CoQ10 in  Pak-wanban could be well absorbed and improved the plasma antioxidant activities. Furthermore, cooking oil may increase the bioavailability of CoQ10 from vegetables. Therefore, it would be useful for vegetarian people.

Background and Objectives: Radiation enteritis is a common side effect after radiotherapy for abdominal and pelvic malignancies. The aim of the present study was to investigate the protective effect of melatonin, known for its free radical scavenging ability, against radiotherapy-induced small intestinal oxidative damage. Materials and Methods: Thirty male Wistar rats were randomly assigned to six groups (5 rats in each) as follows: Group I (control group) rats received neither radiation nor melatonin; group II rats received only 8 Gy single dose of gamma radiation to their abdomen and pelvis regions; group III (administered with only 50 mg/kg melatonin); group IV (administered with only 100 mg/kg melatonin); group V (50 mg/kg melatonin + 8 Gy radiation), group VI (100 mg/kg melatonin + 8 Gy radiation). All rats were sacrificed after 5 days for biochemical assessments of their intestinal tissues. Results: Treatment with melatonin post irradiation significantly reduced malondialdehyde (MDA) levels as well as increased both superoxide dismutase (SOD) and catalase (CAT) activities of the irradiated intestinal tissues. In addition, melatonin administration with different doses pre irradiation led to protection of the tissues. Moreover, the 100 mg/kg dose was more effective compared to 50 mg/kg. Conclusions: The results of our study suggest that melatonin has a potent protective effect against radiotherapy-induced intestinal damage, by decreasing oxidative stress and increasing antioxidant enzymes. We recommend future clinical trials for more insights.

Abstract Background The objective of this study was to evaluate protective effect of grape and apple juices against toxicity induced by cadmium in the kidney of rats. Methods A total of 20 male-Wistar rats were distributed into four groups (n=5): Control group: animals received an intraperitoneal (i.p.) injection of 0.9% saline solution and after 15 days, 1 mL of water was administered for 15 days, via gavage; Cadmium group: animals received an intraperitoneal injection of cadmium chloride (1.2 mg/kg) and after 15 days, 1 mL of water was administered for 15 days via gavage; Cadmium+Grape Juice: animals received an i.p. injection of cadmium chloride (1.2 mg/kg), and after 15 days, 0.8 mL of grape juice was administered for 15 days, via gavage; Cadmium+Apple Juice: animals received i.p. injection of cadmium chloride (1.2 mg/kg) and after 15 days, 1.0 mL of apple juice was administered for 15 days, via gavage. Results Histopathological analysis revealed severe tubular lesion and necrosis in the group exposed to cadmium, while animals exposed to grape or apple juices showed a significant reduction of tissue injury. 8-OHdG immunoexpression, DNA damage, cytochrome C and catalase gene expressions and Toll like signaling pathway (TLR2, and pIKKα/β) decreased in animals treated with grape juice when compared to cadmium group. Conclusion Taken together, we conclude that grape and apple juices had a protective effect by means of antioxidant, antigenotoxic actions and for promoting tissue regeneration in the kidney of rats following cadmium intoxication.

Diabetes mellitus is the most common endocrine disorder worldwide and it is usually along with complications such as retinopathy, neuropathy, nephropathy, and cardiovascular disease. The prevalence of diabetes is increasing and its treatment has created concerns in society. The use of herbal medicine can be helpful in the management of diabetes mellitus. The aim of this study was to investigate the protective effect of marigold hydroalcoholic extract under diabetes condition. A total of 36 male Wistar rats were randomly divided into four groups: normal control group, diabetic control group, and groups treated with 250 or 500 mg/kg hydroalcoholic extract of marigold flower during four weeks. At the end of the study, the rats were anesthetized with ketamine/xylazine, and sampling was performed through cardiac puncture. The results showed that treatment with marigold improved body weight. In addition, we determined that marigold normalized the level of CK-MB, total CK, amylase, and lipase in a dose-dependent manner. Probably these effects resulted from antioxidant property of marigold; thus we suggest that marigold flower can be useful for reduction of diabetes complication.

Sangi1), Ni Putu Purwati2) 1)Program Studi Kimia FMIPA Universitas Sam Ratulangi, Manado 95115; 2)Alumni Program Studi Kimia FMIPA Universitas Sam Ratulangi, Manado 95115 ABSTRAK Penelitian ini bertujuan untuk mengukur tingkat oksidasi lipid dalam plasma darah tikus yang diberi pakan VCO mengandung ekstrak wortel.  Tikus Wistar jantan pada penelitian ini dibagi menjadi empat kelompok, yaitu (I) pakan standar (kontrol), (II) 10% VCO tanpa ekstrak wortel, (III) 10% VCO mengandung ekstrak wortel dan (IV) 10% minyak jagung.  Setelah 4 minggu masa perlakuan, plasma darah dipisahkan dengan sentrifugasi untuk analisis tingkat oksidasi plasma dengan mengukur konsentrasi malondialdehida (MDA) menggunakan metode uji asam tiobarbiturat.  Hasil penelitian menunjukkan bahwa konsentrasi MDA tertinggi terdapat pada kelompok tikus yang diberi minyak jagung (11,253 mM), diikuti kelompok kontrol (10,167 mM), lalu kelompok tikus yang diberi VCO mengandung ekstrak wortel (8,656 mM) dan VCO (8,269 mM).  Penelitian ini menyimpulkan bahwa tingkat oksidasi lipid plasma tikus yang diberi VCO mengandung ekstrak wortel relatif sama dengan yang diberi VCO tanpa ekstrak wortel, tetapi lebih rendah daripada kontrol dan yang diberi minyak jagung. Kata kunci: malondialdehida, minyak kelapa, wortel EFFECT OF VIRGIN COCONUT OIL (VCO) CONTAINING CARROT EXTRACT ON PLASMA LIPID PEROXIDATION ABSTRACT The aim of this study was measure the level of lipid oxidation in the blood plasma of rats fed VCO containing carrot extracts. Male Wistar rats in this study were divided into four groups: (i) control, (II) 10% VCO without the carrot extract, (III) 10% VCO containing carrot extracts, and (IV) 10% corn oil. After 4 weeks of treatment, blood from each rat was taken by syringe.  Plasma is separated from whole blood by centrifugation for analysis of plasma oxidation levels by measuring the malondialdehyde (MDA) concentration using tiobarbituric acid test. The results showed that the highest concentration of MDA was found in rats fed corn oil (11.253 m M), followed by the control group (10.167 m M), then rats fed VCO containing carrot extract (8.656 m M) and VCO (8.269 m M). This study concluded that the level of plasma lipid oxidation in rats fed VCO containing carrot extracts is relatively similar to rats fed VCO without the carrot extract, but it is lower than the control and corn oil groups. The resveratrol is a polyphenol known for its health benefits, which includes the ability to interfere in the osteoblastogenesis, which may foster adverse immunomodulators effects in the host response to periodontal disease. In the present study we evaluated the appearance of periodontal tissues of rats with experimentally induced periodontitis, by using resveratrol. Twenty-four male Wistar rats were used, in which half of the animals received a ligature around the first lower molars, then forming the groups with experimental periodontitis. Next, four groups were created: 1) Control Group (CON); 2) The Ligature Group (LIG); 3) Group Resveratrol (RSV); 4) Ligature-Resveratrol Group (LIG-RSV). The animals of the Resveratrol groups were daily dosed with 10 mg/kg of body weight of polyphenol orally, during four weeks. After 105 days of experimental period, euthanasia was performed. The results showed a significantly lower alveolar bone loss (p<0.05) in animals that received resveratrol, and still, the polyphenol was able to reduce concentration of interleukin 17 (IL-17) in the groups dosed with it. Our conclusion is that dosing rats with experimental periodontitis with resveratrol could cause a protective effect on the alveolar bone loss, in addition to act positively on the IL-17.

Background: Paraquat (PQ), an herbicide, is a very poisonous compound for both humans and animals. This study was conducted to examine the protective effect of the Coenzyme Q10 (CoQ10) in newborn rats from pregnant rats pre-treated with PQ. Methods: The experiments were conducted on 25 rats, divided in five groups randomly and equally: 1. Control Group received normal saline (0.1 ml/day); 2. PQ Group received PQ only (5 mg/kg/day); 3. PQ+CoQ10 Group received PQ (5 mg/kg) and CoQ10 (10 mg/kg) daily; 4. PQ+olive oil Group received PQ (5 mg/kg) and olive oil (10 mg/kg) daily; 5. Olive oil Group received olive oil (10 mg/kg/day). All of the injections were made intraperitoneally and started on the 16th day of pregnancy through to parturition. Sixteen days after parturition, the lungs were removed from the newborn rats, paraffin sections were made and stained with hematoxylin and eosin, and analyzed histomorphometrically and stereologically. Results: The results revealed that interstitial tissue and lung alveoli had normal structures in the control and olive oil groups. In PQ and PQ-olive oil groups alveolar hemorrhage, inflammation, extensive fibrosis, decreased alveolar numbers, increased mast cells, and changes in the epithelia were observed. In PQ-CoQ10 Group there was a significant recovery in all of the histological alterations. Conclusion: Generally, Coenzyme Q10 had a protective effect against lung damages caused by PQ, but a complete recovery of the damaged lung tissue would probably take longer than 16 days after birth.

The adolescence period, marked by sexual and brain maturation, has shown sensitivity to various environmental disruptors. Exposure to the xenoestrogen bisphenol A (BPA) is known to alter physiological and behavioral responses although its role at this critical period remains largely unknown. Recent research further suggests biochemical and genomic effects of BPA to be mitigated by various natural compounds, while effects on behavior have not been examined. This study aimed to characterize (1) the effects of dietary BPA during adolescence on endogenous corticosterone (CORT) secretion, emotional behavior, and testosterone (T) in adulthood, and (2) the impact of combined exposure to BPA with hop extracts (Hop), a phytoestrogen with anxiolytic properties. To do so, four groups of male Wistar rats [postnatal day (PND) 28] were administered corn oil (control), BPA (40 mg/kg), hops (40 mg/kg), or BPA-hops by oral gavage for 21 days (PND 28–48). Blood droplets collected on PND 28, 48, and 71 served to measure CORT and T changes. As adults, rats were tested in the elevated plus maze (EPM), the social interaction test, and the forced swim test. Our findings demonstrated elevated anxiety and a trend toward depressive-like behaviors in BPA- compared to hops-exposed rats. However, BPA intake had no impact on basal CORT levels, or adulthood T secretion and sociability. Of note, BPA's anxiogenic effect manifested through decreased EPM open arm entries was abolished by hops co-supplementation. Together, our observations suggest the adolescence period to be less sensitive to deleterious effects of BPA than what has been reported upon gestational and perinatal exposure.

Background: Tacrolimus is an immunosuppressive drug broadly used to avoid rejection of tissue transplantation. Some studies suggest that tacrolimus has destructive effects on the male reproductive system due to activation of the renin-angiotensin system (RAS) and inflammation. Objectives: The present study aimed at determining the toxic effects of tacrolimus on testis and evaluating the blocking effects of captopril and losartan on angiotensin-converting enzyme (ACE) and angiotensin receptors, respectively, in rats. Methods: The present experimental study was conducted on 36 adult male Wistar rats (weighing 200 ± 20 g) randomly divided into six groups and treated based on the experimental design for 30 days. At the end of the intervention, the rats were anesthetized and their blood samples were obtained to measure the serum levels of testosterone and ACE2 enzyme. Also, both testes were removed for histopathological examinations after scarifying. Results: Tacrolimus decreased testosterone and ACE2 levels significantly. The ACE2 levels were lower in the groups treated with a combination of tacrolimus and losartan or captopril compared to the control group while testosterone level significantly increased in the groups receiving a combination of tacrolimus and captopril. The epithelial thickness of seminiferous tubules and the number of primary spermatocytes significantly decreased in the rats only receiving tacrolimus compared to the groups that received captopril and losartan, along with tacrolimus. Conclusions: According to the findings, the administration of captopril or losartan, along with tacrolimus, can decrease its testicular toxicity. Captopril and losartan act through blocking RAS but cannot improve ACE2 level; therefore, the recovery of testicle tissue might not be attributed to the ACE2 approach but to the anti-inflammatory effect.
