Lettuce ( Lactuca sativa) is an important leaf vegetable crop worldwide. The total phenolic content was higher in the red skirt methanol extracts with a level of 64.9 ± 0.3 mg GA/g, and phenolic content of green skirt was 49.4 ± 0.3 mg GA/g. The total flavonoid content in the methanol extract of red skirt showed the highest concentration (291.6 ± 9.0 mg rutin/g), followed by green skirt (223 ± 9.8 mg rutin/g). The red skirt showed a higher content of anthocyanin (23.7 ± 0.8 mg/100 g) compared to the green skirt (7.4 ± 0.9 mg/100 g). Seven carotenoids i.e., lutein, β–carotene, 9 Z–β–carotene, 13 Z–β–carotene, violaxanthin, zeaxanthin, and α–carotene were detected in both cultivars. The polar metabolites are strongly associated with the metabolites that participate in the TCA cycle. Correlations between primary and secondary metabolites were easily achieved by gas chromatography-time-of-flight mass spectrometry (GC-TOFMS)-based metabolite profiling method. Red skirt methanol extract showed 77.5 ± 0.4% 2,2-diphenyl-1-picrylhydrazyl (DPPH) activity, 50.8 ± 0.8% hydrogen peroxide radical scavenging assay and absorbance value (0.052) reducing power assay at 250 μL/mL, having significantly higher activity than the green skirt extract. The red lettuce extract also showed higher levels of phenolic compounds, flavonoids, and anthocyanins, and it possesses more antioxidant properties compared to the green skirt extract. In this study we compared two cultivars of lettuce, red skirt and green skirt, for various nutritional parameters and to understand the association of secondary metabolites with their corresponding primary metabolites. From the overall analysis, it is confirmed that GC-TOFMS-based metabolite profiling is an appropriate method for determining biochemical reactions and also acts as a combined resource to understand primary and secondary metabolism.

Antioxidant and free radical scavenging activity of methanol, ethanol, ethyl acetate and chloroform extracts of aerial parts of Lamium purpureum L. was determined by DPPH, ABTS, FRAP and TRP assays. Contents of flavonoids and phenols were also investigated. The total phenolic content in the extracts, determined using Folin?Ciocalteu assay, ranged between 8.57 to 128.00 mg GAE/g d.e. while concentrations of flavonoids in the extracts varied from 24.20 to 39.80 mg QuE/g d.e. The highest phenolic content was found in methanol extract (128.00 mg GAE/g d.e.). The highest content of total flavonoids was identified in the methanol extract (39.80 mg QuE/g d.e.) and the lowest was in the chloroform (24.30 mg QuE/g d.e.). DPPH scavenging of the extracts was determined and obtained IC50 values ranged from 0.12 to 3.12 mg/mL of solution. The values of ABTS radical scavenging activity ranged from 0.35 to 1.80 mg AA/g. The highest ABTS antiradical activity was registered for methanol extract. The FRAP value was found within the range 0.08 to 1.04 ?mol Fe/mg. The best radical scavenger was methanol (1.04 ?mol Fe/mg). In reducing power assay different extracts of L. purpureum showed increasing of activity with increased concentration, and all extracts possessed substantial dose dependent antioxidant activity. The best reducing capacity was obtained with methanol extract of L. purpureum (0.0132 mg AA/mL). The results in this study confirmed that L. purpureum possesses moderate antioxidant properties.

A comparative study of antioxidant properties on two varieties of Phyllanthus (Phyllanthus niruiri L. & Phyllanthus urinaria L. leaves) growing in Bangladesh was done in the present study. Five complimentary test methods namely DPPH free radical scavenging activity, reducing power assay, total antioxidant capacity, total phenolic and flavonoid contents determination were used for the evaluation of antioxidant properties of two species of Phyllanthus. In the present investigation for the evaluation of antioxidant activities the concentration is expressed by the ratio of crude sample per solvent volume instead of extract weight per solvent volume. Based on these concentrations the investigated P. niruri L. and P. urinaria L. leaves extracts exhibited significant results. At 2 mg/ml concentration DPPH radical scavenging capacity of the methanol extract of both the plants was found to show significant (>90%) activity which is comparable to Ascorbic acid and BHA. In case of reducing power tests for both the extract, the activity of P. urinaria is comparable to that of BHA and AA and that of P. niruiri is comparable to BHA. P. niruiri showed better antioxidant potential than that of P. urinaria based on the observed results of their corresponding methanol extract. Ruta angustifolia was used in this study in order to evaluate the antimicrobial activity and antioxidant properties and its correlation with the polyphenolic content. Two Gram-positive bacteria (Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778) and two Gram–negative bacteria (Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 8739) were used to determine the antibacterial activity. Aqueous maceration extract was used for antioxidant activities and methanolic maceration extract was used for antibacterial activity. The antioxidant properties and activities were evaluated by using total phenolic content (TPC), total flavonoid content (TFC), DPPH free radical scavenging activity and beta-carotene bleaching method. Whereas, the antibacterial activity was examined using disc diffusion method against selected microorganism at concentration 1.0 mg/disc. The results showed the phenolic content of R. angustifolia extract was 18.89 g GAE/100 g extract while the flavonoid content was 14.170 g QE/100 g extract. R. angustifolia exhibited good radical scavenging with IC50 value of 2.04 mg/ml. The result for disc diffusion method showed no inhibition zone against all the strains of bacteria at 1.0 mg/disc concentration of the extract. Based on the results, it can be concluded that the R. angustifolia aqueous extract has the antioxidant properties and there is correlation between polyphenolic content of the extract with its antioxidant activity. However, R. angustifolia methanolic extract did not show any antibacterial activity.

The present study was conducted to evaluate the antioxidant, antidiarrheal and analgesic activities of Vachellia farnesiana (L.) Wight & Arn. The methanol extract of V. farnesiana and its different fractionates were subjected to in-vitro assay for the determination of total phenolic content and antioxidative potential. The ethyl acetate soluble fraction (EASF) exhibited the highest free radical scavenging capacity (IC50 value of 21.49 ± 1.04 μg/ml) as compared to that exhibited by the standard butylated hydroxyl toluene (BHT) (IC50 value of 20.41 ± 0.05 μg/ml). Such prominent antioxidative potential was further reinforced by a phenolic content of 39.26 ± 0.85 mg of gallic acid equivalent per gram of extract. The plant extract, at the dose of 400 mg/kg body weight, reduced castor oil-induced diarrhea in mice model by a statistically significant (p < 0.05) margin of 47.62%, while the standard loperamide produced 66.67% reduction of diarrheal feces. The central and peripheral analgesic activities of the crude methanol extract of V. farnesiana (MEVF) was determined by tail flick- and acetic acidinduced writhing methods, respectively, in Swiss albino mice. In the tail flick method, oral administration of MEVF at doses of 200 and 400 mg/kg body weight exhibited 221.09 and 237.09% elongation of pain response time, respectively, after 90 minutes of administration whereas the standard morphine effectuated 518.34% elongation within the same time. Furthermore, the same doses of the extract illustrated 63.27 and 69.39% reductions, respectively, in the acetic acid-induced abdominal constrictions in mice. Compared to the standard acetylsalicylic acid with 75.51% inhibition, statistically significant (p < 0.05) peripheral analgesic activity was established. The results of the present investigations suggest that methanol extract of V. farnesiana possesses antioxidant, antidiarrheal and analgesic activities which eventually indicates the presence of biologically important phytoconstituents within the plant that needs further exploration. For the functional food applications, antioxidant properties and the bioactive compounds of the 23 Curcuma species commercially cultivated in Thailand were studied. Total phenolic content and DPPH radical scavenging activity were determined. The concentrations of eight bioactive compounds, including curcumin (1), demethoxycurcumin (2), bisdemethoxycurcumin (3), 1,7-diphenyl-(4E,6E)-4,6-heptadien-3-ol (4), germacrone (5), furanodienone (6), zederone (7), and ar-turmerone (8), were determined from the Curcuma by HPLC. While the total phenolic content of C. longa was highest (22.3 ± 2.4 mg GAE/g, mg of gallic acid equivalents), C. Wan Na-Natong exhibited the highest DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) radical scavenging activity. Twenty-three Curcuma species showed characteristic distributions of the bioactive compounds, which can be utilized for the identification and authentication of the cultivated Curcuma species. C. longa contained the highest content of curcumin (1) (304.9 ± 0.1 mg/g) and C. angustifolia contained the highest content of germacrone (5) (373.9 ± 1.1 mg/g). It was noteworthy that 1,7-diphenyl-(4E,6E)-4,6-heptadien-3-ol (4) was found only from C. comosa at a very high concentration (300.7 ± 1.4 mg/g). It was concluded that Thai Curcuma species have a great potential for the application of functional foods and ingredients.

Dryopteris marginalis L. is a local plant in Bangladesh which is widely used as folk medicine. In this study, the chemical composition of essential oil of D. marginalis L. as well as the antioxidant activity, antibacterial activity and total phenolic content of various organic extracts were analyzed. The gas chromatography–mass spectrometry analysis was used to identify the chemical compositions. The antibacterial activity of different extracts of Dryopteris marginalis was examined against three gram-negative bacteria and two gram-positive bacteria. The antibacterial activity of different extracts was investigated using disc diffusion method and minimum inhibitory concentration showing important antibacterial activity and was measured by using broth dilution method. The antioxidant activity was determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and superoxide radical scavenging assay. The total phenolic portion of the extracts were ascertained by Folin-Ciocalteu in alkaline medium. Twenty-two volatile compounds were identified from the essential oil. The methanol extract exhibited significant inhibitory effect which was close to the value of standard antibiotic (Streptomycin). The zones of inhibition of different organic extracts against the tested bacteria were found in the range of (7-15) mm and the minimum inhibitory concentrations (MIC) were recorded (15.62-1000) µg/ml. The antioxidant activity of D. marginalis was (82.13+1.2) % and (59.21+0.45) % highest activity with DPPH and superoxide radical scavenging assay respectively. The methanol extract showed highest (147.43+3.1) mg/g GAE of total phenolic content. The extracts and essential oil of D. marginalis showing strong antibacterial activity can be considered as a potential source of natural medicine and the ingredient of modern therapeutic agents.

Abstract In order to explore the potential drying process, the effects of different drying methods (sun drying, hot-air drying, microwave-vacuum drying, and freeze drying) on the antioxidant properties and phenolic contents in white button mushroom fruit bodies were investigated. The Folin-Ciocalteu method was used to determine total phenolic content, while 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, reducing power, iron-chelating ability, and hydroxyl radical scavenging activity assays were used to elucidate antioxidant activities. Results indicated that antioxidant properties in dried white button mushroom were affected by the drying methods to different extents, and phenolic contents showed significant differences (p<0.05) among different drying methods. Microwave-vacuum drying possessed the highest antioxidant activity in DPPH radical scavenging, reducing power and iron-chelating ability, respectively; and the content of phenolic compounds was 4.945 mg gallic acid equivalents (GAE) /g dry weight (DW). Sun drying was the worst in antioxidant activity and content of phenolic compounds was 1.964 mg GAE/g DW. Additionally, significantly positive correlation was found between the total phenolic content and antioxidant activity (p<0.01). As far as antioxidant activities are concerned, microwave-vacuum drying is proposed as an appropriate drying technique for white button mushroom.

Secondary bioactive compounds of endophytes are inevitable biomolecules of therapeutical importance. In the present study, secondary metabolites profiling of an endophytic bacterial strain, Acinetobacter baumannii, were explored using GC-MS study. Presence of antioxidant substances and antioxidant properties in chloroform (CHL), diethyl ether (DEE), and ethyl acetate (EA) crude extracts of the endophytic bacteria were studied. Total phenolic content (TPC), total flavonoid content (TFC), total antioxidant capacity (TAC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and ferrous ion chelating assay were evaluated. A total of 74 compounds were identified from the GC-MS analysis of the EA extract representing mostly alkane compounds followed by phenols, carboxylic acids, aromatic heterocyclic compounds, ketones, aromatic esters, aromatic benzenes, and alkenes. Among the two phenolic compounds, namely, phenol, 2,4-bis(1,1-dimethylethyl)- and phenol, 3,5-bis(1,1-dimethylethyl)-, the former was found in abundance (11.56%) while the latter was found in smaller quantity (0.14%). Moreover, the endophytic bacteria was found to possess a number of metal ions including Fe(II) and Cu(II) as 1307.13 ± 2.35 ppb and 42.38 ± 0.352 ppb, respectively. The extracts exhibited concentration dependent antioxidant and prooxidant properties at high and low concentrations, respectively. The presence of phenolic compounds and metal ions was believed to play an important role in the antioxidant and prooxidant potentials of the extracts. Further studies are suggested for exploring the untapped resource of endophytic bacteria for the development of novel therapeutic agents.

The chemical composition of the essential oils, antioxidant activity (DPPH and β-carotene/linoleic acid assays) and total phenolic content (Foline-Ciocalteu) of the flowers and leaves of Salvia reuterana were determined. Essential oils extracted from the flowers and leaves by hydrodistillation were analyzed by GC and GC/MS. Forty-four constituents, representing 99.7-99.9% of the oils, were identified. The major components were germacrene D, benzoic acid hexyl ester, bicyclogermacrene, β-gurjunene and ishwarene, constituting 33.7-31.9% of the oils. The highest radical-scavenging activity (DPPH test) was shown by the methanol extract of the flowers (IC50=77.6 μg/mL). In the β-carotene/linoleic acid assay, the methanol extract of the leaves showed the highest inhibition (40.3%) which was only slightly lower than that shown by BHT (82.9%). The total phenolic contents of the methanol extracts of the flowers and leaves as gallic acid equivalents were 81.4 and 88.3 μg/mg, respectively. The plant also showed good antimicrobial activity against three strains of tested microorganisms.

The objectives of the study were to assess the total phenolic content and flavonoid content, and to investigate the antioxidant and antimicrobial potential of carrot peel extracted with methanol and acetone. Total Phenolic content and Flavonoid content was higher in carrot peel acetone extract (AE) i.e. 106.36 µg/mg of GAE and 27.83 µg/mg of QE respectively than methanol extract (ME). Carrot peel acetone extract showed greater antioxidant capacity for DPPH radical scavenging activity whereas methanol extract showed better result for FRAP reducing power and Phosphomolybdenum assay. Both AE and ME could inhibit the growth of Shigella flexneri, E coli, Staphylococcus aureus and Klebsiella pneumonia. This study substantiates the potential of carrot peel to be used as a novel functional food ingredient, natural antioxidant and antimicrobial agent
