The efficacy of Truong Xuan Cao Bang (TXCB), a Vietnamese herbal medicine prescription, on the quality of semen, serum testosterone (TES), and malondialdehyde (MDA) concentrations in rabbits whose reproductive ability were impaired by fluconazole was investigated. Mature male rabbits were randomly assigned into control (G1), fluconazole (G2), fluconazole and 180 mg/kg/day (G3), and 360 mg/kg/day TXCB (G4) groups. The reaction time (RT), ejaculate volume (EV), sperm concentration (SC), total sperm output (TSO), total sperm motility (TSM), progressive sperm motility (PSM), nonprogressive sperm motility (NPSM), sperm immobility (SIM), dead sperm (DS), and abnormal sperm (AS) levels were determined after TXCB administration. After 60 days, G3 showed a significant increase in serum TES and a significant decrease in the concentrations of serum MDA (SMDA) and seminal plasma MDA (SPMDA) compared with G1, in addition to increase in EV, SC, TSO, TSM, PSM, and serum TES, and decrease in RT, SIM, DS, AS, SMDA, and SPMDA compared with G2. G4 showed a steep rise in EV, SC, and serum TES and a downturn in DS, SMDA, and SPMDA compared with G1, and, compared with G2, a significant increase in EV, SC, TSO, TSM, PSM, and serum TES and a significant decrease in RT, SIM, DS, AS, SMDA, and SPMDA. Taken together, TXCB rehabilitates and simultaneously enhances spermatogenesis in rabbits whose reproductive ability was impaired by fluconazole.

Issues related to the reproduction of animals have been and remain one of the most complex and relevant problems of biology and are constantly finding a direct and effective way out into livestock farming practice. The rational use of breeding sheep as producers is limited by the lack of standard, objective methods and methods for the timely assessment of their reproductive ability. The article presents the results of evaluating the sperm production of rams-producers. For the first time, the freshly obtained sperm of rams of Moldavian type producers of the Karakul breed was evaluated using the macroscopic method - ejaculate volume, color and smell, as well as the microscopic method - motility, sperm concentration in the ejaculate, total sperm count in the ejaculate sperm movements (VAP; VSL and VCL) as well as the percentage of abnormal forms of sperm. The experiments were carried out on sheep producers grown on a pedigree farm of the experimental farm of the Moldavian Scientific and Practical Institute of Biotechnology in Animal Husbandry and Veterinary Medicine. As a result of the studies, it was found that the ejaculate volume on average was 0.99 ± 0.04 ml, motility was 0.95 ± 0.02 and sperm concentration 1.51 ± 0.14 billion/ml. The percentage of pathological forms in sperm averaged 13.72 ± 0.61, an indicator that characterizes the high quality of sperm. The average quality indicator of ejaculates obtained from ram-producers of the Moldavian type of the Karakul breed corresponds to physiological standards for the Karakul breed. The average percentage of pathological forms of sperm found in ejaculates indicates that the rams were in good conditions of feeding and keeping. According to research, we can say that the Moldovan type of Karakul rams can be successfully used to obtain high quality ejaculates suitable for freezing sperm at 196°C.

AbstrakIndonesia memiliki banyak tumbuhan berpotensi obat salah satunya bisa dijadikan sebagai obatkontrasepsi tradisonal yang biasa digunakan untuk menjarangkan anak atau sterilisasi sepertirebusan akar Caesalpinia bonduc (Aru). Tujuan penelitian untuk mengetahui pengaruh obatkontrasepsi tradisional air rebusan akar aru terhadap kualitas pematangan sperma epididimis mencit(Mus musculus). Mencit dipilih secara acak untuk mewakili 4 kelompok dosis yaitu : 0,0 (K0), 16,0(K1) 24,0 (K2) dan 32,0 (K3) μl/gram berat badan/hari. Setiap kelompok perlakuan dilakukandengan 5 kali replikasi. Perlakuan diberikan melalui oral dengan menggunakan jarum gavage selama10 hari berturut-turut. Pembedahan dilakukan pada hari pertama setelah perlakuan selesai.Pengamatan kualitas sperma epididimis dengan perhitungan produksi sperma, % sperm motil, %sperma hidup, % sperma abnormal,dan rangking keaktifan sperma. Untuk mengetahui ada atautidaknya perbedaan yang bermakna antar perlakuan dalam kelompok dilakukan uji Anava satu arah,bila terdapat perbedaan bermakna dilanjutkan uji BNJ untuk membandingkan angka rata-rata antarkelompok perlakuan. Pemberian air rebusan akar Caesalpinea bonduc pada mencit menunjukanpengaruh perbedaan secara signifikan pada jumlah (%) sperma abnormal antara dosis 0,0 μl dengan24,0 dan 32,0 μl serta antara 16,0 μl dengan 24,0 dan 32,0 μl; Jumlah (%) sperma hidup terdapatperbedaan antara dosis 0,0 μl dengan 16,0 , 24,0 dan 32,0 μl; Jumlah (%) sperma motil terdapatperbedaan antara dosis 0,0 μl dengan 24,0 dan 32,0 μl; Rangking keaktifan sperma terdapatperbedaan antara dosis 0,0 μl dengan 24,0 dan 32,0 μl. The aims of this study is to know the effect of root boiled water ofCaesalpinia bonduc as traditional contraception medicine on quality of sperm maturation ofepididymis of mice (Mus musculus). Mice were choosed radomly and doses gouped: 0,0 (K0 ), 16,0(K1) 24,0 (K2) and 32,0 (K3) μl/gram body weight/day. Each of group was replicated 5 times.Treatment were given by oral with using gavage needle for ten days. Surgery was carried out on firstday after completing treatments. Examination of epidymis sperm by counting number of spermproduction, percentage of motil, live, abnormal sperm, and rank of sperm motility. In order to knowthe deferences between control and treated group, was used one way anava analysis and analysedvalue for comparing between teratment group. The treatments of root boiled water of Caesalpiniabonduc have effect significantly on percentage of abnormal sperm between dose of 0,0 μl with 24,0and 32,0 μl, also between 16,0 μl with 24,0 and 32,0 μl. Percentage of live sperem is different betweendose of 0,0 μl with 16,0 , 24,0 and 32,0 μl. Percentage of motile sperm is defferent significantlybetween dose of 0,0 μl with 24,0 and 32,0 μl. Meanwhile percentage of motile rank has differencebetween dose of 0,0 μl with 24,0 and 32,0 μl. The treatment of root boiled water of Caesalpiniabonduc has effect signifiantly on increasing percentange of sperm abnormality, decreasingpercentage of life sperm, motile and rank of motile of mice sperm.

     The present study was undertaken to evaluate the effect of aqueous extract of Date Palm Pollen DPP on the testicular function and serum testosterone, FSH and LH hormones value. Thirty five male rats were divided randomly into five equal groups. Group 1: received 0.5 ml of distilled water (control group), group 2: was treated orally 0.250 p.p.m of sodium florid (NaF) (with volume of 0.5 ml / rat), Group 3: was treated with 0.250 p.p.m of NaF and 50 mg/kg. B.W. of DPP extract (0.5ml D.W \rat), Group 4: was treated with 0.250 p.p.m of NaF and 100 mg/kg. B.W. of DPP extract and Group 5: was treated with 0.250 p.p.m of NaF and 150 mg/kg. B.W. of DPP extract. The results showed  significant (P< 0.05) decrease in sperm concentration, motility and significant (P< 0.05) increases in dead and abnormal sperm in the group 2 in comparison to control, while all groups of  DPP extract showed significant (P< 0.05) increase in  sperm concentration, motility and decrease in dead and abnormal sperm. Maximum effect was observed in animals treated with a dose of 150 mg/kg of DPP extract, also the results revealed significant (P< 0.05) increase in testosterone, FSH and LH hormones in groups treated with DDP in comparison to G1andG2. Male rats received DPP for 50 days showed significant (P< 0.05) increases in body and testes weight as compared to G1andG2. In conclusion the results revealed that the aqueous extract of DPP pollen can be used as a sex enhancer and seems to cure male infertility.

Sperm motility is known to affect fertilization; however, little is known about the relationship between frozen–thawed sperm motility and in vivo fertilization following superovulatory treatment. The objective of this study was to evaluate a sperm function test as potential predictors of embryo production following superovulatory treatment in cattle. Two to five batches of semen (Japanese black bull, n = 4, A to D) were diluted with egg york-citrate-glycerol in 0.5 mL plastic straws, and they were stored in liquid nitrogen until analyzed. Frozen–thawed spermatozoa were evaluated for motility {motile sperm concentration (MSC, million mL–1), progressive MSC (PMSC, million mL–1) and velocity (μm s–1)} using a sperm quality analyzer for bulls (SQA-Vb, Medical Electronic Systems, Caesarea, Israel). Each sample of 20 μL aspirated into the disposable capillary, was inserted into SQA-Vb. Measurements were displayed within 75 s. Intra-assay CVs of MSC, PMSC, and velocity were 14.2, 7.3 and 7.5%, respectively. Inter-assey CVs of them were 13.5, 3.9 and 4.3% respectively. Superstimulated donors (Japanese black cows, n = 161) were artificially inseminated with one dose of frozen–thawed semen (bull A = 74, B = 46, C = 21 and D = 20). The proportion of transferable embryo (IETS grade 1 to 3) was examined on day 7 (day 0 = estrus). Data were analyzed using ANOVA followed by Scheffe multiple comparison test, and Fisher’s z-transformation. MSC, PMSC and velocity values differed significantly among each bull. The values of bull A were much lower than those of the other bulls. The proportion of transferable embryos produced by bull A was significantly lower than that of other bulls (P < 0.05, Table 1). Correlations showed significant association between MSC and proportion of transferable embryos (r = 0.99, P < 0.01). We conclude that bovine sperm motility using a SQA-Vb is a useful predictor of embryo production following superovulatory treatment. Inbreeding depression, a genetic condition produced by the mating of close-related individuals, has been associated with a reduction of fertility in several species. However, a loss in sperm quality was also associated with age. In horses, the few existing reports have described a tendency of both parameters to produce a negative effect on sperm quality. However, those reports were performed using a subjective evaluation of sperm motility. In the present study, a total of 692 ejaculates from 86 Pure Spanish stallions (PRE), aged between 3 and 22 years, were evaluated using a computer-assisted methodology to determine the effect of inbreeding in four semen parameters: free-gel volume (V), sperm concentration (C, by haemocytometer), and total (TM) and progressive (PM) sperm motility (by Spermvision sperm class analyser; Minitube). The inbreeding coefficient (F) was estimated using 300 000 PRE pedigree records approximately (minimum pedigree depth, eight equivalent complete generations; range, between 1 and 30.1%). Stallion, age, ejaculate, and season of semen collection were the variables included in the statistical model (general linear model), with ejaculate and season being the variables with a major effect (by variance components analysis). Our results showed that sperm concentration (r=−0.18; P&lt;0.0001) and volume (to a lesser extent) were reduced with advancing age, both showing a major decline after 15 years of age. To the contrary, sperm motility was not affected by age of the stallion. We also found a negative correlation between the inbreeding coefficient and ejaculate volume (r=−0.14; P&lt;0.001), with a marked decrease seen when F was between 7 and 20%. Also, a negative correlation was observed in PM (r=−0.08; P&lt;0.05), although to a lower extent. Conversely, C and TM were not affected by inbreeding depression (P&gt;0.05). In conclusion, our results demonstrated that high levels of inbreeding can compromise severely the sperm quality of the PRE stallion, which, subsequently, may have a negative influence on fertility. Ongoing studies using genomic data will help to detect genetic variants associated with stallion semen quality and how it is influenced by inbreeding in specific genomic regions.

Research was conducted to develop sperm sorting and novel sperm preservation methodologies for sex predetermination in the bottlenose dolphin (Tursiops truncatus) using artificial insemination. In Study 1, the effect of seminal plasma (SP), sperm concentration and freezing rate (FR) on in vitro sperm quality of liquid-stored, non-sorted spermatozoa was examined. There was no effect (P > 0.05) of prefreeze SP addition on post-thaw quality (progressive motility, kinetic rating, sperm motility index (SMI), viability and acrosome integrity). Post-thaw motility parameters and viability were higher (P < 0.05) for slow FR than fast FR samples. In Study 2 investigating the effects of liquid storage and sorting on sperm quality, motility and SMI after sorting and centrifugation were lower (P < 0.05) than those of the initial ejaculate. The sort rate for enrichment (91 ± 4% purity) of X- and Y-bearing spermatozoa was 3400 ± 850 spermatozoa sex−1 s−1. In Study 3, compared with a modified straw method, directional freezing resulted in enhanced in vitro quality of sorted and non-sorted spermatozoa derived from liquid-stored semen (P < 0.05). In Study 4, endoscopic insemination of three dolphins with sorted, frozen–thawed X-bearing spermatozoa resulted in one conception and the birth of a female calf. High-purity sorting of dolphin spermatozoa, derived from liquid-stored semen, can be achieved with minimal loss of in vitro sperm quality and samples are functional in vivo.

The objective of this study was to investigate the effect of using different extenders <italic>viz.</italic> Androhep, Safecell Plus and SUS during cryopreservation on quality of frozen-thawed boar semen. Semen volume, sperm motility, sperm concentration, percentage of morphologically abnormal spermatozoa, total number of spermatozoa per ejaculate and activity of the enzyme aspartate aminotransferase (AST) were assessed in fresh semen collected from 39 fertile AI boars. Semen from each boar was divided into three portions and diluted 1:1.5 in extender Androhep, Safecell Plus and SUS and keep at 17°C for 15-h holding time before cryopreservation. Then sperm was cryopreserved. Straws were thawed in a water bath at 38°C for 40s and post-thaw sperm motility with AST activity was assessed. Significant difference in post-thaw sperm motility was found between extender Androhep and Safecell Plus (P<0.05). AST activity did not differ significantly between tested extenders (P>0.05). In conclusion, the results of the study indicate that using Safecell Plus extender during holding period before cryopreservation significantly affected post-thaw sperm motility.

A study was conducted to assess the semen quality using Resazurin Reduction Test (RRT) in Jersey and Jersey cross bulls. A total 36 fresh ejaculates were collected for evaluation RRT, sperm motility, acrosomal integrity and sperm concentration. The mean value of RRT in bulls was 15.77±0.85 minutes.Statistical analysis revealed that RRT was highly significantly (p<0.01) and positively correlated with sperm concentration (r=0.769), sperm motility(r=0.858) and acrosomal integrity (r=0.515), hence it may be helpful in evaluating the quality of semen in bulls.

A total of sixteen rabbits sourced from two locations in Southeastern Nigeria were used for this study. Semen was collected from all the rabbits twice weekly for eight weeks using an artificial vagina while observation on the effect of time of collection on semen quality were also made. The results obtained indicated that time of collection had no significant effect (P > 0.05) on all the seminal characteristics measured. However, source of the animals showed significant effect (P < 0.05) on semen volume, sperm concentration, total sperm per ejaculate and proportion of abnormal sperm. Average semen volume, sperm concentration and total sperm per ejaculate were: 0.69±0.007 ml, 104.0±1.44 x 106/m1 and 71.40±0.90x106 respectively for the rabbits sourced from Port Harcourt. The corresponding values for rabbits sourced from Owerri were: 0.66±0.008m1, 96.02 ±1.63x106iml and 63.49±1.50 x 106 respectively. Abnormal sperm proportions were 11.05±0.22% and 1216 ± 032% for the rabbits sourced from Port Harcourt and Owerri respectively. There were no significant (P> 0.05) differences observed between locations in live sperm concentration and sperm motility. Live sperm concentration and sperm motility were 87.75±1.09% and 71.62±0.74% for the Port Harcourt group, while the values obtained for the Owerri group were 88.06±034% and 71.19±0.89% respectively.

Abstract The aim of the study was to compare the quality and quantity parameters of semen in male lake minnow, Eupallasella percnurus (Pallas, 1814), from six Polish populations. Substantial differences among the populations were found in milt volume (36–112 µl) and sperm concentration (4.7–8.0 × 109 ml−1). Mean values for sperm motility were similar; however, there were population-related differences in sperm velocity measured with VSL (96.3–127 µm s−1). Despite this, the quality of semen in all cases was high and appropriate for both artificial reproduction and cryopreservation.
