Changes in Carbohydrate Content during Wheat Maturation—What is Measured by near Infrared Spectroscopy?

The role of bread, pasta and related products produced from milled wheat seeds is important to the human diet, so monitoring changes of starch content in developing grain is essential. Immature wheat grains are also used as a functional food, particularly as a source of water-soluble carbohydrates. The amount and variation in content of different carbohydrates changes considerably during maturation and these changes were non-destructively monitored in developing grain using near infrared (NIR) spectroscopy. Characteristic changes in three carbohydrate absorption bands [1585–1595 nm (Carbohydrate I), 2270–2280 nm (Carbohydrate II) and 2325–2335 nm (Carbohydrate III)] were identified and it was concluded that the different dynamics of carbohydrates (starch accumulation as well as synthesis/decomposition of water-soluble carbohydrates) could be followed sensitively by monitoring these three different regions of NIR spectra. Carbohydrate I represents the effect of starch accumulation during maturation based on the vibrations of intermolecular hydrogen bonded O–H groups in polysaccharides. Carbohydrate II is the manifestation of O–H stretching and C–C stretching vibrations existing unengaged in water-soluble carbohydrates while Carbohydrate III describes the changes in C–H stretching and deformation band of poly- and mono-oligosaccharides. NIR spectroscopic techniques are shown to be effective in monitoring plant physiological processes and the spectra have hidden information for predicting the stage of growth in wheat seed.

Three white clover cultivars of differing geographical origin, namely AberHerald, Grasslands Huia and Sandra, were grown in binary mixtures with perennial ryegrass (cv. Preference) for a period of 3 years at Uppsala (59° 49′ N, 17° 39′ E) where the length of the main growing season is c. 200 days. Changes in growing point numbers, stolon length and stolon weight from autumn until late spring were followed by four to six destructive harvests each season. Changes in carbohydrate content (starch and water-soluble carbohydrates) were also determined on six or seven occasions during the same time. Cold hardening and dehardening were followed by artificial freezing tests on stolons removed from the plots on seven occasions during the last study season. The rate of node production was followed in the last spring. Temperatures in the stolon environment were recorded during the last two study seasons.All cultivars suffered a loss of stolon length during the winters, with Huia being most affected. The extent of recovery during spring varied between years. Late spring frosts checked the growth of both AberHerald and Huia more than that of Sandra. The accumulation of starch appeared to depend on autumn temperatures and there was a decline in starch content during the winter. Sandra maintained a higher content of water-soluble carbohydrates during the winter and the spring than the other cultivars. Sandra also achieved a higher degree of freezing tolerance and dehardened later than both AberHerald and Huia. However, temperatures in the stolon environment always exceeded the degree of freezing tolerance attained irrespective of cultivar.It is concluded that the accumulation and utilization of non-structural carbohydrates are important in determining the extent to which a white clover cultivar survives the winter and spring in a harsh climate. Freezing tolerance per se does not seem to be the most limiting trait, but in a climate where frequent spring frosts occur, a late dehardening seems to be valuable.

The increase in drought events due to climate change have enhanced the relevance of species with greater tolerance or avoidance traits to water restriction periods, such as Bromus valdivianus Phil. (B. valdivianus). In southern Chile, B. valdivianus and Lolium perenne L. (L. perenne) coexist; however, the pasture defoliation criterion is based on the physiological growth and development of L. perenne. It is hypothesised that B. valdivianus needs a lower defoliation frequency than L. perenne to enhance its regrowth and energy reserves. Defoliation frequencies tested were based on B. valdivianus leaf stage 2 (LS-2), leaf stage 3 (LS-3), leaf stage 4 (LS-4) and leaf stage 5 (LS-5). The leaf stage development of Lolium perenne was monitored and contrasted with that of B. valdivianus. The study was conducted in a glasshouse and used a randomised complete block design. For Bromus valdivianus, the lamina length, photosynthetic rate, stomatal conductance, tiller number per plant, leaf area, leaf weights, root growth rate, water-soluble carbohydrates (WSCs) and starch were evaluated. Bromus valdivianus maintained six live leaves with three leaves growing simultaneously. When an individual tiller started developing its seventh leaf, senescence began for the second leaf (the first relevant leaf for photosynthesis). Plant herbage mass, the root growth rate and tiller growth were maximised at LS-4 onwards. The highest leaf elongation rate, evaluated through the slope of the lamina elongation curve of a fully expanded leaf, was verified at LS-4. The water-soluble carbohydrates (WSCs) increased at LS-5; however, no statistical differences were found in LS-4. The LS-3 and LS-2 treatments showed a detrimental effect on WSCs and regrowth. The leaf photosynthetic rate and stomatal conductance diminished while the leaf age increased. In conclusion, B. valdivianus is a ‘six-leaf’ species with leaf senescence beginning at LS-4.25. Defoliation at LS-4 and LS-5 was optimum for plant regrowth, maximising the aboveground plant parameters and total WSC accumulation. The LS-4 for B. valdivianus was equivalent to LS-3.5 for L. perenne. No differences related to tiller population in B. valdivianus were found in the different defoliation frequencies.

Near-infrared spectroscopy (NIRS) was used to predict the nutritive value of forage species available to the critically endangered northern hairy-nosed wombat (Lasiorhinus krefftii). Nutritive attributes of the forage successfully estimated included total nitrogen concentration, fibre (including neutral detergent fibre, acid detergent fibre and acid lignin), organic matter, water soluble carbohydrates and in vitro dry matter digestibility. The reported results demonstrate the seasonal variability of the forage resource available to L. krefftii in its tropical savanna habitat. Multivariate modelling of the spectra enabled the nutritive value of forage samples to be estimated with coefficients of determination (r2) of 0.770–0.995 and standard errors of the cross-validation of 0.070–2.850 using a modified partial least-squares analysis technique. The standard error of the laboratory was 0.02–1.42. This study demonstrates that broad-based NIRS predictive equations can be used to predict the nutritive value of a number of plant types available to a herbivore over time. By using NIRS the analyst can rapidly analyse large numbers of samples with limited reduction of precision, thereby enabling large-scale ecological applications that may have previously been impeded by time and costs.

Submergence stress at early vegetative stage is one of the most important constraints in the productivity of rice in Bangladesh. Submergence causes yield loss of rice at Aman season in Bangladesh and therefore, it is necessary to develop submergence tolerant rice cultivars. A pot experiment was conducted at the net house of Department of Crop Botany, Bangladesh Agricultural University, during Aman season from July to December, 2017 to evaluate the changes in root porosity and water soluble carbohydrates (WSCs) associated with submergence tolerance in rice. The experiment consisted of two factors—(i) Rice cultivars (Binadhan-11, Binadhan-12, BRRI dhan51 and BRRI dhan52 as tolerant and BRRI dhan49 as susceptible) and(ii) Submergence stress: Submergence for 14 days at vegetative stage and control. Submergence stress was imposed by dipping of pots into a water tank with about 90 cm depth of water while the control plants are maintained in the pot house of the field laboratory. The plants were sampled at seven days interval during submergence to determine the changes in root porosity and to examine the contribution of shoot reserves for their survival. The root porosity was measured by pycnometer method and water soluble carbohydrate was measured by the anthrone method. Tolerant cultivars showed greater root porosity development in both control and stress condition but the susceptible cultivar showed significantly lower root development in stress condition. Higher root porosity might help tolerant cultivars to survive in submergence stress more efficiently. Tolerant rice cultivars had high initial soluble carbohydrate than the susceptible one. Under submergedcondition, the tolerant cultivars showed slow depletion of water soluble carbohydrate compared to susceptible cultivar. Higher carbohydrate contents in tolerant cultivars might act as buffer stock during submergence for their better survival and growth. J Bangladesh Agril Univ 17(4): 539–544, 2019
