Neospora caninum is a intracellular protozoan parasite and is one of the major causes of repeated abortions, foetal malformations, pre-term deliveries, stillbirth and possible loss of milk yield in livestock. The presence of specific antibodies against N. caninum in the blood serum of dairy cows is investigated in the present study. A total of 184 blood serum samples of dairy cows were examined in Bursa province in the Marmara Region. N. caninum antibodies were measured using an indirect enzyme-linked immunosorbent assay (ELISA) (The Svanovir Neospora-Ab ELISA). From the total sample, antibodies to N. caninum were detected in 62 of the 184 examined cows (33.3%) and neurological findings were seen in a calf.

Neospora caninum is a protozoan parasite that causes abortion in ruminants, including goats. The objective of the present study was to determine the seroprevalence of N. caninum in native Korean goats (Capra hircus coreanae). A commercial enzyme-linked immunosorbent assay kit was used to analyse 464 serum samples for the presence of N. caninum antibodies. Four samples (0.9%, 95% confidence intervals – CI: 0.0–1.7) were found to be positive for N. caninum antibodies. The seroprevalence was analysed according to age (less than to one year, young; more than or equal one year, adult; and unknown), sampling season (April to September, warm; October to March, cold), and region (northern, central, and southern). However, there were no statistically significant differences in seroprevalence according to age, season, and region (P > 0.05). This is the first report on the seroprevalence of N. caninum in native Korean goats. The results of this study indicate a nationwide distribution of N. caninum among goats, with a relatively low prevalence. Therefore, the implementation of integrated control strategies as well as measures for prevention and control of N. caninum infection among goats is recommended.

Abstract. Sera samples from 89 dairy cows with repeat breeder and 94 healthy pregnant dairy cows (controls) from Elazig province of eastern Turkey were tested for presence of Neospora caninum antibodies by use of a competitive enzyme linked immunosorbent assay (cELISA). Fifteen out of 183 cows tested (8.19%) were found to be seropositive to N. caninum antibody. Overall, seroprevalence of N. caninum was significantly higher (P<0.05) in dairy cows with repeat breeder (13.48%, 12/89) than in dairy cows with healthy pregnant (3.19%, 3/94). There was no statistically significant relationship between seroprevalence and breed and age groups in cows with repeat breeder (P>0.05). In conclusion, the current study highlights the importance of N. caninum in cows with repeat breeder.

AbstractNeospora caninum, an obligate intracellular protozoan parasite, is the causative agent of neosporosis, recognized as a major cause of bovine abortion around the world. Thailand is a developing agricultural country located in Southeast Asia. Livestock developments particularly in dairy cows of this country have been hampered by low productivity including milk and slow growth rate due to the impact of many pathogens including N. caninum. Currently, there is no effective method for control of neosporosis since there is less information regarding current status of infections. The objective of this study was to investigate the seroprevalence of neosporosis in dairy cows of the northern part of Thailand. During 2006–2007, the sera of 642 cows from 42 small farm holders with the top three highest consensus of dairy farms in the northern provinces, such as Chiang Rai, Chiang Mai and Lumpang were collected and performed tests. Antibodies to N. caninum were assayed by enzyme-linked immunosorbent assay (ELISA) with recombinant N. caninum surface antigen 1 (NcSAG1) and indirect fluorescent antibody test (IFAT). The overall prevalence of N. caninum infection in this study was 46.9% (301/642) by ELISA and 34.3% (220/642) by IFAT.

SUMMARYToxoplasma gondii and Neospora caninum are closely related protozoan parasites which cause lowered production and increased abortion in dairy cows. The aim of the present study was to determine the seroprevalence of T. gondii and N. caninum infection in dairy cows in the Guangxi Zhuang Autonomous Region (GZAR), subtropical southern China. In total, 875 serum samples were collected from the tail veins of dairy cows in 6 main dairy cow-rearing districts of 4 administrative cities in GZAR. The samples were surveyed for T. gondii antibody using the Indirect Haemagglutination Test (IHA), and 365 of the serum samples were examined for N. caninum antibody by indirect Enzyme-Linked Immunosorbent Assay (ELISA). The overall seroprevalence of T. gondii in dairy cows was 13·71% (120/875), and the average seroprevalence of N. caninum was 15·07% (55/365). There were significant differences in the seroprevalence of N. caninum infection between different districts (P = 0·002, χ2 = 9·261). The highest prevalences of T. gondii and N. caninum were found in cows older than 8 years and those that had completed 5–6 pregnancies. Five cows (1·37%) presented antibodies against both T. gondii and N. caninum, and dairy cows with both T. gondii and N. caninum antibodies had higher abortion rates. The present results indicate widespread exposure of dairy cows to T. gondii and N. caninum in GZAR, subtropical southern China.

Abstract The aim of the present study was to investigate the occurrence of N. caninum associated with abortions of dairy cattle from Santa Catarina state, southern Brazil by using enzyme-linked immunosorbent assay (ELISA), immunohistochemistry (IHC), and polymerase chain reaction (PCR). Blood from dairy cows that aborted along with intrathoracic fluid and tissue samples (brain, heart, liver, and lung) from their fetuses were collected and used for serology; PCR, histopathological, and immunohistochemistry (IHC) evaluations were also conducted. Twenty-one cows (51.2%) out of 41, and eight fetuses (26.7%) out of 30 were ELISA (HerdCheck, IDEXX) positive for N. caninum. Dams > 36 months of age had a higher risk of being serum positive than younger animals. PCR and IHC revealed that 38.8% (14/36) and 25.0% (9/36) of the fetuses were positive for N. caninum, respectively for each of the tests. Seropositive cows had a higher frequency of fetuses that were also positive by either intrathoracic fluid, PCR, or IHC. In summary, the present study observed a high frequency of N. caninum in abortions from dairy cows from southern Brazil, with a higher N. caninum prevalence found in cows that were older than 36 months. In addition, serology, PCR, and IHC should be used all together for better diagnosis of neosporosis in cattle.

Abstract Background Reproductive problem is one of the main constraints of livestock genetic improvement efforts in tropical countries. The aim of this study was to determine the prevalence of major infectious causes of reproductive problems of dairy cattle in selected dairy farms in central Ethiopia. Overall 86 serum samples were collected from October 2018 to February 2019 from animals with history of reproductive problems. The collected serum was tested for antibody titer against Brucella species, Neospora caninum, Bovine Viral Diarrhea (BVD), Infectious Bovine Rhinotracheitis (IBR) and Q-fever using rose-bengal and enzyme-linked immunosorbent assay (ELISA) tests. Result Among the animals with the history of reproductive disordered; abortion, still birth and repeat breeding cases were found in 61.6%, 19.8% and 18.6%, respectively. The prevalence of IBR, BVD, Neospora caninum and Coxiella brunetti was found to be 79.1%, 38.4%, 3.5% and 1.2%, respectively. The combined infection of both BVD and IBR were detected in 21% of animals. Out of the total animals examined in this study, 95.9% of Jersey breeds were found seropositive to IBR than Boran-Friesian crosses (57.7%). The incidence of BVD was significantly higher in Boran-Friesian crossbred cattle than in Jersey which was found to be 69.3% and 14.3, respectively. The prevalence of IBR and BVD was directly proportional with age of the animal and parity. Conclusion Vaccination against IBR and BVD is not practiced in Ethiopia, the rising level of those diseases in dairy sector needs regular surveillance and control program.

ABSTRACTObjectiveToxoplasma gondii is a worldwide protozoan parasite that could infect virtually all warm-blooded animals, including humans. Our study aimed to investigate the prevalence of T. gondii infection in college students at Anhui province, China. Moreover, growing studies demonstrated the association between T. gondii infection and host behavioral changes. We also studied the linkage between T. gondii and scores of college students.Methods2704 serum samples of medical school students attending physical education lessons were collected from September 2017 to September 2019 and evaluated for T. gondii IgG antibodies using an enzyme-linked immunosorbent assay (ELISA). We also analysed PE scores of T. gondii infected students and T. gondii uninfected students.ResultsThe overall seroprevalence of T. gondii was 11.5%. The main risk factors related to T. gondii infections were cat in the household and gardening or agriculture activity. Furthermore, in basketball group and football group, scores of T. gondii seropositive students were significantly higher than that of seronegative students, while in other sports there is no difference between scores of T. gondii infected students and T. gondii uninfected students.ConclusionThis is the first report of T. gondii seroprevalence in college students in Anhui province, China.

Neospora caninum is a protozoan parasite that causes abortion and important economic losses in cattle worldwide. There are no treatments or vaccines available; disease control is based on diagnosis and herd management strategies. We developed, validated, and evaluated under field conditions a competitive inhibition ELISA based on the truncated SAG1 protein (tSAG1), expressed in Escherichia coli, and the RafNeo5 monoclonal antibody (ciELISAtSAG1). A criterion based on the 3-y sequential serologic analysis of 230 dairy cows by IFAT was used as the gold standard. The assay was validated using 860 serum samples from cows that were consistently positive or negative by IFAT throughout the study period. ciELISAtSAG1 was then used to evaluate the prevalence of neosporosis in 16 beef cow herds (22 samples per herd, 352 total samples). The results were compared with those from IFAT and a commercial cELISA (cELISAVMRD). The ciELISAtSAG1 cutoff was ≥ 29%I, with a diagnostic sensitivity of 98.7% (95% CI = 96.8–99.7%) and a diagnostic specificity of 97.9% (95% CI = 96.4–99.0%). Concordance among IFAT, cELISAVMRD, and ciELISAtSAG1 was 90.3%. The agreement (κ) between ciELISAtSAG1 and the other 2 tests was ≥ 0.81. The overall prevalence of neosporosis in the 16 beef herds was 30% (range: 5–60%). The ciELISAtSAG1 could be useful for large-scale detection of anti– N. caninum antibodies in cattle and seroepidemiologic investigations, given its appropriate sensitivity and specificity, and the simplicity of production.

ABSTRACT An immunochromatographic test (ICT) with recombinant surface antigen 1 of Neospora caninum (NcSAG1) was developed for the rapid detection of antibodies to N. caninum in cattle. The ICT was used to clearly discriminate between immunofluorescent-antibody test (IFAT)-positive bovine sera and IFAT-negative bovine sera. Serum samples collected from cattle in Yanbian, China, were examined by the ICT. Of the 96 serum samples, 23 (24.0%) were positive by the ICT, and 19 (19.8%) samples were positive by a previously developed enzyme-linked immunosorbent assay (ELISA). Eighteen of 19 ELISA-positive samples were positive according to the ICT. A good agreement was found between the results of the ICT and the ELISA. The results presented here suggest that the ICT with recombinant truncated NcSAG1 fused to glutathione S-transferase is a useful and reliable method for the detection of antibodies to N. caninum in cattle.

Relevance. Numerous virological studies prove the importance of enteroviruses in human somatic pathology. However, the etiopathogenetic role of enterovirus infection in patients with acute cerebrovascular disorder (GVMK) is not sufficiently highlighted. Objective: to establish the value of enterovirus infection as a trigger factor in the pathogenesis of acute stroke. Materials and methods. The pear blood serum of 72 patients with acute stroke (main group) and 35 patients with neurological pathology without vascular pathology (group of comparison) were screened for presence of enteroviruses using the virological method, detection of enterovirus genomes using a polymerase chain reaction and the presence of specific Ig M and Ig G to enteroviruses in the enzyme-linked immunosorbent assay (ELISA). Results. The enterovirus genomes were isolated from blood serum in 23,6±5,9 % of patients with acute stroke, that is significantly higher than in patients of the comparison group – 2,9±2,8 % (p <0,05). The enteroviruses were isolated in 11 cases of 17 PCR-positive blood serum samples of the main group. These viruses were identified as Coxsackie B viruses (serotypes 2, 3, 4) and ECHO viruses (serotypes 6, 9, 27 (two strains), 29), three strains of viruses could not be identified. The presence of specific Ig M and Ig G in blood serum of 4 patients with HPMC, as well as enterovirus genomes, has been established. It suggest that they have a recent enterovirus infection, or can indicate a recent enterovirus infection or exacerbation of chronic enterovirus infection. Only specific Ig G in the absence of Ig M were detected in blood serum of 4 PCR positive patients, that can indicate chronic enterovirus infection. Only Ig M in the absence of Ig G was detected in blood serum of 6 PCR-positive patients, that can indicate acute enterovirus infection. No Ig M or Ig G in serum from three PCR-positive patients were detected, possibly due to the presence of latent enterovirus infection. Conclusions. Acute and chronic persistent enterovirus infection plays a possible trigger role in the development of acute stroke.  The combination of PCR to detect genomes of enteroviruses, virological for the isolation and identification of viruses, and ELISA for the detection of specific Ig M and Ig G to enteroviruses should be recommended for the diagnosis of persistent enterovirus infection in patients with acute stroke.
