Glyphosate (GLY) is worldwide one of the most used active substances in non-selective herbicides. Although livestock might be orally exposed via GLY-contaminated feedstuffs, not much is known about possible hepatotoxic effects of GLY. As hepatic xenobiotic and nutrient metabolism are interlinked, toxic effects of GLY residues might be influenced by hepatic nutrient supply. Therefore, a feeding trial with lactating dairy cows was conducted to investigate effects of GLY-contaminated feedstuffs and different concentrate feed proportions (CFP) in the diets as tool for varying nutrient supply to the liver. For this, 61 German Holstein cows (207 ± 49 days in milk; mean ± standard deviation) were either fed a GLY-contaminated total mixed ration (TMR, GLY groups, mean GLY intake 122.7 μg/kg body weight/day) or control TMR (CON groups, mean GLY intake 1.2 μg/kg body weight/day) for 16 weeks. Additionally, both groups were further split into subgroups fed a lower (LC, 30% on dry matter basis) or higher (HC, 60% on dry matter basis) CFP resulting in groups CONHC (n = 16), CONLC (n = 16), GLYHC (n = 15), GLYLC (n = 14). Blood parameters aspartate aminotransferase, γ-glutamyltransferase, glutamate dehydrogenase, cholesterol, triglyceride, total protein, calcium, phosphorus, acetic acid and urea and histopathological evaluation were not influenced by GLY, whereas all mentioned parameters were at least affected by time, CFP or an interactive manner between time and CFP. Total bilirubin blood concentration was significantly influenced by an interaction between GLY and CFP with temporarily elevated concentrations in GLYHC, whereas the biological relevance remained unclear. Gene expression analysis indicated 167 CFP-responsive genes, while seven genes showed altered expression in GLY groups compared to CON groups. Since expression changes of GLY-responsive genes were low and liver-related blood parameters changed either not at all or only slightly, the tested GLY formulation was considered to have no toxic effects on the liver of dairy cows.

The study was planned to evaluate the effect of exogenous fibrolytic enzymes (EFE) supplementation for 56 days @ 240 mg/kg total mixed ration (TMR) on digestibility of dry matter and nutrients in dairy cows. Six dry non-pregnant cows were assigned in each treatment with and without EFE. The digestibility trial of seven days was conducted after 49 days of feeding. Dry matter and nutrients intake of cows was not influenced by EFE. The supplementation of EFE had improved digestibility of dry matter, organic matter, crude fiber, neutral detergent fiber, cellulose (p less than 0.01), as well as digestibility of nitrogen-free extract and acid detergent fiber, was also higher (pless than 0.05). The body weight gain of cows was higher on the supplementation of EFE in TMR. The study concluded that feeding exogenous fibrolytic enzymes (240 mg/kg) supplemented TMR improved digestibility of dry matter and nutrients, which was reflected as higher body weight gain in dry non-pregnant Gir and crossbred dairy cows.

In two experiments, lactating cows (26 in exp. 1; 32 in exp. 2) were fed an 11.3% crude protein (CP) diet, a 14.7% CP diet containing untreated soybean meal (SBM) or a 14.6% CP diet containing formaldehyde-treated SBM (FSBM), all three fed as a complete blended diet with control corn silage (CCS) (exp. 1); a 12% CP diet, a 13.4% CP diet containing SBM or a 13.3% CP diet containing FSBM, all fed as a complete blended diet with urea-treated corn silage (UCS) (exp. 2). In both experiments concentrates and CCS or UCS were mixed (50:50 dry matter) daily and fed ad lib for a 16-wk period. The increase of CP over the low CP negative control by addition of untreated SBM resulted in a significant (P < 0.05) increase in milk production in both experiments. The addition of FSBM had no significant effect. When comparing solids-corrected milk (SCM), addition of SBM, whether treated or not, significantly increased SCM in both experiments. In neither experiment did FSBM improve milk production over untreated SBM. All animals fed CCS lost body weight but there was no difference between treatments. For animals fed UCS the low CP group lost significantly (P < 0.05) more body weight than did the group fed FSBM. Dry matter (DM) intake was significantly (P < 0.05) lower for the FSBM fed group than for the group fed untreated SBM in exp. 1 (CCS). DM intake was significantly (P < 0.05) increased by either FSBM or SBM over the low CP control fed UCS. Milk protein was the only component significantly (P < 0.05) decreased by FSBM in exp. 1. Isobutyric and isovaleric acids were significantly (P < 0.05) lower in rumen fluid of animals on FSBM plus CCS. Rumen microbial protein content seemed to be more adversely affected by FSBM with CCS than UCS. Amino acid content of FSBM and SBM demonstrated that lysine and tyrosine were lost from SBM after treatment with formaldehyde. The lack of a response in milk production to FSBM is discussed in terms of a lysine and tyrosine deficiency for milk protein synthesis and the adverse effect that formaldehyde protection of SBM may have on the rumen microbial population. AbstractA feeding trial with growing sheep was conducted on mixed roughage-supplement diets, to test whether roughage intake could be estimated from a known intake of a supplement and an alkane-based estimate of the supplement and roughage proportions in the diet (diet composition) without the need for separate dosing with external marker alkanes. Diets consisted of chaffed Trifolium subterraneum hay (SC), or a mix of SC and beeswax-labelled cottonseed meal (CSM) in the proportions of 7: 1, 6: 2, 5: 3 and 4: 4 (air-dry matter basis). Six sheep were given the pure SC diet and three sheep were given each of the mixed diets. Additionally, the CSM was also labelled with octatriacontane (C38) to investigate its use for labelling supplements. Due to unexpectedly low recoveries, C38 was not included in any estimation of diet composition or digestibility.The inclusion of beeswax-labelled CSM in a SC diet resulted in lower faecal alkane recoveries (as proportions of alkane ingested; P ≤ 0·05). Within mixed diets, recoveries decreased significantly with increasing proportion of CSM, but this effect was significant (P < 0·05) only for alkanes consisting of 25, 26, 27 and 29 carbon-atoms. Estimates of diet composition were close to measured values. Daily SC intake and organic matter digestibility (OMD) differed by −3·7 to 7·2% and −2·0 to 5·7% from measured values if a mean faecal recovery, across diets, was applied for each alkane. Accuracy increased greatly if estimates were based upon faecal alkane recoveries for individual diets, instead of mean recoveries across diets (−0·3 to 0·9% and −0·2 to 0·0% for SC intake and OMD, respectively).The results indicate that roughage intake can be estimated from a combination of known supplement intake and an estimate of diet composition, which obviates the need for separate alkane dosing to estimate intake. The method should be applicable in situations in which animals either normally receive supplements (e.g., dairy cows) or could readily be given supplements.

The aim of this study was to evaluate energy metabolism and lipid mobilization via ultrasonic measurements (USM), considering inner fat depots, in lactating dairy cows differing in body condition score (BCS) and fed rations with low (35% at dry matter basis; C35) or high (60% at dry matter basis; C60) concentrate feed proportions postpartum. Sixty pluriparous German Holstein cows were arranged in a 2 × 2 factorial design from d 42 antepartum (relative to calculated calving) until d 120 postpartum. Animals were divided into a group with a lower (initial BCS = 3.1 ± 0.38 SD; BCSL) and a group with a higher (initial BCS = 3.83 ± 0.41 SD; BCSH) BCS. Due to higher dry matter intake C60 groups reached the positive energy balance earlier, whereas C35 groups had a more pronounced negative energy balance. Although this would suggest a more pronounced mobilization of C35 groups the USM revealed no differences between feeding groups. Differences in BCS between both BCS groups remained almost the same over the trial. This was not reflected in ultrasonic data, as lipid mobilization was higher in higher conditioned cows. These findings demonstrate the extended possibilities of USM to depict metabolic processes.

The present study aimed at evaluating the influences of different concentrate feed proportions in the ration offered to dairy cows post partum with different body condition scores (BCS) before calving. Therefore, 60 pluriparous cows were divided 42 days before expected calving into two groups with a higher or an adequate BCS. After calving, both groups were further subdivided into a group fed a ration with either a low concentrate feed proportion (C, 35% at dry matter basis) or a high (60% at dry matter basis) one. It was hypothesized that different BCS would lead to different reactions concerning varying concentrate feed proportions. Isolated BCS effects were detected in the white blood profile only before calving. Neither low nor high concentrate feed proportions affected hematological, blood immune cell phenotypes and inflammatory markers consistently irrespective of BCS group. It was concluded, that the assessed BCS span covered a range in which the capability of cows to cope with different dietary post partum energy supply remained unchanged.

Abstract The objective of this study was to determine if increasing propionate alters dry matter intake (DMI), glucose clearance rate, blood metabolites, insulin concentrations and hepatic gene expression in steers fed a finishing diet. Holstein steers (n = 15; BW = 243 ± 3.6 kg) were individually fed a finishing diet ad-libitum. Steers were allocated by body weight (BW) to receive: no Ca propionate (Control), 100 g/d Ca propionate (Low), or 300 g/d Ca propionate (High) in the diet. Orts were collected and weighed daily to determine DMI. Blood samples were collected on d 0, 7, and 21, and BW recorded on d 0, 14, and 28. An intravenous glucose tolerance test (IVGTT) was conducted on d 14 and 28 of the trial. Liver biopsies were collected on d 33 for gene expression analysis. Blood samples were analyzed for whole blood glucose and lactate, plasma non-esterified fatty acids (NEFA) and insulin concentrations. Data were analyzed using a mixed model with treatment, day and their interaction included, with day and minute as a repeated measure. The control treatment had greater (P &lt; 0.01) DMI than low and high steers. Body weight was increased in control steers on d 14 and 28 compared to the steers receiving the High treatment (P = 0.03 for the interaction). Blood glucose concentrations tended (P = 0.09) to be higher on d 21 than d 0 and 7 but was not affected by treatment (P = 0.58). Plasma NEFA concentrations were lower (P = 0.05) for control steers than other treatments, and greater (P = 0.002) on d 0 than d 7 and 21. Blood lactate concentrations were greater (P = 0.05) on d 7, than d 0 and 21, but not affected by treatment (P = 0.13). High steers had greater plasma insulin concentrations in response to the IVGTT than steers on the other treatments (P = 0.001). There was no treatment (P ≥ 0.16) or day effect (P ≥ 0.36) on glucose peak, plateau, or clearance rate. High steers had greater expression of solute carrier family 16 member 1 (SLC16A1; P = 0.05) and tended to have greater hepatic expression of solute carrier family 2 member 2 (SLC2A2; P = 0.07). These data indicate that increased propionate may decrease DMI and insulin sensitivity.

The effects of supplementing low (23.9%) dry matter (DM) and medium (28.9%) DM corn silage roughage rations fed ad libitum to high-producing dairy cows, with orchard grass hay at 0.7% of their body weight on milk production and composition, was determined in a double reversal feeding trial using 24 lactating cows. Cows fed medium DM corn silage consumed 15.2% more DM/100 kg body weight daily than those cows fed low DM silage (P < 0.05). Supplemental hay fed with either low or medium DM silage resulted in equal consumption of total forage DM/100 kg body weight by cows on the two treatments. Each kilogram of hay DM consumed by cows offered low DM corn silage reduced silage DM intake by 0.47 kg, whereas hay DM intake reduced silage DM intake by 0.79 kg for cows offered medium DM silage. Daily production of 4% fat-corrected milk (FCM), percent nonfat solids (SNF), and daily body weight gain were lower for cows consuming low DM corn silage than for those on the other treatments (P < 0.05). Cows consuming low DM corn silage plus orchard grass hay at 0.7% of their body weight produced milk at the same level and of similar protein, lactose, and SNF content as those fed medium DM silage plus hay. Supplemental hay fed with medium or low DM corn silage increased FCM production over feeding silage alone, independent of DM or moisture content of the silage fed (P < 0.05).
