This research has an aim to observe the production of lettuce (Lactuca sativa L.) on hydroponic technology of floating system with red and blue LED (Light Emitting Diode) at night. This research was conducted inside the Screenhouse in which located on the top of Griya Kads House building, Bulusan Village, Tembalang Sub-district from May to June 2017. The research was conducted by 3x3 factorial experiments in Nested Design with 3 replications. The first factor was 0, 1.5 and 3 hours irradiation of red and blue LED lights at night. The second factor was 3, 5 and 7 ml/liter of AB mix nutrient concentration which was nested in the first factor. The data collected were fresh weight, dry weight and root canopy ratio. The results of the study showed that the treatment of AB mix nutrient concentration had no significant effect on all parameters. The production of lettuce with level of 5 ml concentration of AB mix was quite good, yet 3 ml and 7 ml levels didn’t improve lettuce production. The duration of red and blue LED irradiation only resulted in an increase in the production of canopy fresh weight. This research can be concluded that 3 hours was sufficient to give the good results to improve the results of lettuce plants production. The goal of this research was to investigate the effect of electrical conductivity (EC) levels of the nutrient solution on the fresh weight, chlorophyll, and nitrate content of hydroponic-system-grown lettuce. The selected cultivars are the most representative commercial varieties grown for European markets. Seven cultivars (‘Sintia,’ ‘Limeira,’ ‘Corentine,’ ‘Cencibel,’ ‘Kiber,’ ‘Attiraï,’ and ‘Rouxaï’) of three Lactuca sativa L. types’ (butterhead, loose leaf, and oak leaf) were grown in a phytotron in rockwool, meanwhile the EC level of the nutrient solutions were different: normal (<1.3 dS/m) and high (10 dS/m). The plants in the saline condition had a lower yield but elevated chlorophyll content and nitrate level, although the ‘Limeira’ and ‘Cencibel’ cultivars had reduced nitrate levels. The results and the special characteristic of the lollo-type cultivars showed that the nitrate level could be very different due to salinity (‘Limeira’ had the lowest (684 µg/g fresh weight (FW)) and ‘Cencibel’ had the highest (4396 µg/g FW)). There was a moderately strong negative correlation (−0.542) in the reverse ratio among the chlorophyll and nitrate contents in plants treated with a normal EC value, while this relationship was not shown in the saline condition. Under the saline condition, cultivars acted differently, and all examined cultivars stayed under the permitted total nitrate level (5000 µg/g FW).

In this study, we investigated optimal light conditions for enhancement of the growth and accumulation of glucosinolates and phenolics in the sprouts of canola (Brassica napus L.). We found that the shoot lengths and fresh weights of red light-irradiated sprouts were higher than those of sprouts exposed to white, blue, and blue + red light, whereas root length was not notably different among red, blue, white, and blue + red light treatments. The accumulations of total glucosinolates in plants irradiated with white, blue, and red lights were not significantly different (19.32 ± 0.13, 20.69 ± 0.05, and 20.65 ± 1.70 mg/g dry weight (wt.), respectively). However, sprouts exposed to blue + red light contained the lowest levels of total glucosinolates (17.08 ± 0.28 mg/g dry wt.). The accumulation of total phenolic compounds was the highest in plants irradiated with blue light (3.81 ± 0.08 mg/g dry wt.), 1.33 times higher than the lowest level in plants irradiated with red light (2.87 ± 0.05 mg/g dry wt.). These results demonstrate that red light-emitting diode (LED) light is suitable for sprout growth and that blue LED light is effective in increasing the accumulation of glucosinolates and phenolics in B. napus sprouts.

Investigations were carried out to study the effects of light-emitting diode (LED) lights on growth and development of isosteroidal alkaloids in embryogenic calli of Fritillaria cirrhosa D. Don, an important traditional Chinese medicine herb. Calli were cultured in glass bottles, each containing 100 mL of Murashige and Skoog’s basal medium supplemented with 2% sucrose and 0.4% gellan gum powder, a gelling agent. These bottles were incubated in a specially designed plant growth chamber equipped with eight different LED lights consisting of single or combinations of four different light spectra emitting blue (450 nm), green (525 nm), red (660 nm), and far-red (730 nm) light. After three months of incubation, morphological changes in embryogenic calli were recorded, and LC-MS/MS analysis of cultures was carried out for peimisine, sipeimine, peiminine, and peimine. The highest number of somatic embryos and the maximum fresh weight was recorded in calli incubated under red (9R), infrared (9IR), and a combination of red+blue+infrared (3R3B3IR), respectively, in decreasing order. The highest contents of peimisine, peiminine, and peimine were recorded under red (9R) and infrared (9IR) lights, respectively. Eight LED lights had significant effects on the morphogenesis of embryogenic calli of F. cirrhosa D. Don and contents of isosteroidal alkaloids.

Monitoring movements of larval Drosophila with electrical detection allows one to record the behaviors without the use of lights and cameras. This is a suitable technique when studying the use of light-sensitive proteins in optogenetic studies. Electrical measures are feasible to use in determining when a larva starts to move or continues to move after a light induced activation of channelrhodopsin. We have developed a technique using an electrical measure of the media as an index of larval movement. As a proof of concept, recordings with an infrared camera of the larval movement were simultaneous made with electrical measures. The two techniques parallel each other in their ability to index larval movements. Bright light-emitting diode (LED) lights used in optogenetic experiments tend to saturate the detectors of the camera unless filters are used and different filters maybe necessary depending on the LED spectrum and sensitivity of the camera. Impedance measures are independent of the type of LED or brightness. We also assessed the use of a non-solvent based glue (3M Vetbond) to hold larvae in place while measuring synaptic function of neuromuscular junctions, cardiac function and influence of modulators, or activation of light-sensitive channels.

ABSTRACT Handroanthus ochraceus (yellow lapacho) is a medicinal, ornamental and timber tree which can be propagated by in vitro culture. Conventional methods use fluorescent lighting (FL), whereas light emitting diode (LED) has been used for this purpose only recently. The aim of this work was to evaluate the effects of FL and high-power LED (HP-LED) on the in vitro multiplication and rooting of yellow lapacho at different irradiances (15 to 60 µmol m-2s-1). Epicotyls obtained from half-siblings was multiplicated in WPM (Woody Plant Medium) supplemented with 20 µM benzilaminopurine and 1 mM IBA (indolebutiric acid). For rooting, shoots were cultured for 3 days in ½WPM supplemented with 50 µM IBA and for 42 days in auxin-free ½WPM under HP-LED or FL lighting. Under HP-LED, the multiplication rate of shoots increased significantly (61%) from 20 to 40 µmol m-2s-1 respect to FL. Differences in abaxial stomatal density and size were observed between light sources at 20 µmol m-2s-1. High HP-LED irradiance produced the highest rooting percentage. In the rooting stage, the marginal means of treatments without factors interaction showed that HP-LED irradiances significantly increased shoot length by 20%, shoot fresh weight by 77% and shoot dry weight by 30% in comparison to the values under FL. The maximum values calculated from the regression curves were around 50 µmol m-2 s-1 for HP-LED for all parameters except root lenght whereas were around 20 µmol m-2 s-1 for FL for all parameters except fresh and dry weigth of shoot. Here we show that HP-LED lighting improve in vitro culture of H. ochraceus, reduced 81% energy consumption respect to FL and uses only a multispectral LED instead of different single color LEDs. Therefore, HP-LED could be useful for the micropropagation of tree species contributing to sustainable agriculture and ecological restoration of degraded areas.

This paper describes an experiment to investigate discomfort glare caused by white light-emitting diode (LED) lights having different spectral power distributions. It included two groups: a ‘Metamerism’ group and a ‘correlated colour temperatures (CCT)’ group. In the former group, it was found that white lights at 7000 K constructed from different blue LEDs and the same red and green LEDs gave about the same glare perception. In the latter group, there was a significant difference in glare perception between white lights having different CCTs. Finally, glare models, including unified glare rating (UGR) and the newly derived QUGRspd, and mUGRspd models, were tested using the data from the experiment. All of them gave quite accurate predictions of the data.
