AbstractDetermining the accurate age of malaria vectors is crucial to measure the risk of malaria transmission. A group of fluorescent chemicals derived from a pyrimidine-pyrazine ring structure known as pteridines from the head, thorax and whole body of adult female Anopheles stephensi were identified and evaluated as a tool for chronological and physiological age determination of malaria vectors. The female mosquitoes were collected from an insectary colony at an interval of every 5 days, up to 30 days, and the pteridines of head, thorax and whole body were detected fluorometrically by high-pressure liquid chromatography (HPLC) using excitation and emission wavelengths of 365 and 455 nm, respectively. In addition, alteration of the pteridines compounds was compared between blood and sugar fed mosquito groups. Although four pteridines including pterin-6-carboxylic acid, biopterin, xanthopterin and isoxanthopterin were detected, some of them were absent in the head or thorax of mosquitoes. Levels of all four pteridines were similarly decreased in a linear manner throughout 30 days. No significant difference in alteration of pteridine compounds was observed between the two groups of blood or sugar fed mosquitoes. This result indicates that diet has a little effect on pteridines alteration. Age determination based on pteridines, as an age-grading technique, could be used for field collected mosquitoes, which have either sugar or blood meal. In addition, analyzing total pteridine fluorescence from only whole body could be a convenient method to estimate the age.

AbstractA method for physiological age determination in blackflies is described from studies on Simulium equinum (Linnaeus), S. ornatum Meigen, S. erythrocephalum (De Geer), and S. nigrum (Meigen) in Russia, and S. vittatum (Zetterstedt), and S.verecundum-venustum supercomplex in Canada. The method uses neutral red to stain the ovariolar structures which indicate preceding gonotrophic cycles. Two different processes can occur in the ovariole during the gonotrophic cycle: egg development with ovulation, or degeneration of a terminal follicle. Egg-sacs are resorbed and form zones of granulation which accumulate in the ovariole at each gonotrophic cycle. Following degeneration of the terminal follicle, a follicular relic (═gonotrophic dilatation) forms. The sum of zones of granulation plus follicular relics in each ovariole can indicate the number of completed gonotrophic cycles. However the number of follicular relics alone does not correspond to the number of preceding gonotrophic cycles and cannot reliably be used for determination of the parity of a female. Female flies collected in nature were categorized as: nulliparous; 1-parous; or multiparous. In multiparous females, most ovarioles have no more than two zones of granulation or a combination consisting of one zone of granulation and a follicular relic. Females in this category are considered to be 2-parous. In some ovarioles of parous females the germarium and terminal follicles were found to have disappeared.

Stable flies (Stomoxys spp.) are common pests of livestock in the peri-urban rangelands of Vina Division of the Adamawa Plateau. No documented information is available on their diurnal dynamics in relation to physiological age and landscape. The main aim of this study was to determine the trap apparent density (ADT) of Stomoxys and associate it to their diurnal activity rhythm and landscape. Vavoua traps (n=12) were used for Stomoxys collection. Trapping was carried out for seven days i.e. three days consecutively in August and four days consecutively in October, 2016. Three traps were pitched in each of the three biotopes (river, cattle corral and gallery forest) of the four study sites (Galim, Mbidjoro, Velambai and Soukourwo). All female Stomoxys captured underwent ovarian dissection for the determination of their physiological ages (parous or nulliparous). A total of 218 Stomoxys were caught and identified into four species (S. niger niger, S. omega, S. calcitrans, and S. xanthomelas) with an overall ADT of 2.59 flies per trap per day and Stomoxys niger niger (1.13 s.n.n. per trap per day) recorded the highest ADT. Species richness was study site dependent. Higher catches of females 113 (51.83%) were made than that of their male 105 (48.17%) counterparts. Galim recorded the highest Stomoxys apparent density (4.90) as compared to other sites with a statistically significant difference (P<0.05). The proportion of parous 87 (59.30%) was higher than that of their nulliparous 46 (40.71%) counterparts. Parity differed with landscape and capture periods. S. n. niger was the most frequent in all biotopes and Galim recorded the highest ADT. The population of female flies was made up of adults (parous) with a bimodal diurnal activity rhythm whereas males had a unimodal activity rhythm.

Disuse can induce numerous adaptive alterations in skeletal muscle. In the present study the effects of hindlimb unloading on muscle mass and biochemical responses were examined and compared in adult (450 g) and juvenile (200 g) rats after 1, 7, or 14 days of whole body suspension. Quantitatively and qualitatively the soleus (S), gastrocnemius (G), plantaris (P), and extensor digitorum longus (EDL) muscles of the hindlimb exhibited a differential sensitivity to suspension and weightlessness unloading in both adults and juveniles. The red slow-twitch soleus exhibited the most pronounced atrophy under both conditions, with juvenile responses being greater than adult. In contrast, the fast-twitch EDL hypertrophied during suspension and atrophied during weightlessness, with no significant difference between adults and juveniles. Determination of biochemical parameters (total protein, RNA, and DNA) indicated a less rapid rate of response in adult muscles. This was corroborated by assessment of muscle alpha-actin mRNA levels, which indicated a rapid (within 1 day) and significant (P less than 0.05) effect in juveniles but not in adults. The results of this investigation indicate 1) a qualitatively similar differential effect of unloading on muscles of adults and juveniles, 2) a quantitatively reduced and less rapid effect of suspension on adult muscles, and 3) a close similarity of adult and juvenile muscle responses during suspension and spaceflight, suggesting that this ground-based model simulates many of the unloading effects of weightlessness.

The morphology of the female Cyrtobagous salviniae Calder and Sands reproductive system is similar to other weevil species being meroistic and telotrophic. The reproductive system is composed of 2 ovaries each containing 2 ovarioles where the follicles mature. A physiological age grading system was developed where the continuum of ovarium development was divided into 2 nulliparous and 3 parous classes. This was based on the differentiation of the ovarioles, presence, and appearance of follicular relics, cuticle hardness/coloration, and fat body quantity/appearance. High correlation occurred between the parous classes and number of eggs produced where the P3 class had over 9-fold higher number of eggs in comparison with the P1 class. Mean number of eggs produced for each parous class was significantly different, however, overlap occurred. Such a system enables a determination of the past, present, and future reproductive status of field populations and mass-rearing colonies.

Studies on development of female reproductive system insects have shown its utility in physiological age grading. The irreversible charocter of ovarian alterations correlated with mandibular wear and presence of yellow body, were applied to estimate the physiological age of Passalus convexus and Passalus latifrons females, giving results comparable to other studies and indicating the value of these methods in determination of physiological age.

SummaryThe plasma tissue factor (TF) concentration was correlated to factor VII concentration (FVIIag) and factor VII activity (FVIIc) in 498 healthy volunteers ranging in age from 17 to 64 years. Immunoassays using monoclonal antibodies (mAbs) were developed for the determination of TF and FVIIag in plasma. The mAbs and the test systems were characterized. The mean value of the TF concentration was 172 ± 135 pg/ml. TF showed no age- and gender-related differences. For the total population, FVIIc, determined by a clotting test, was 110 ± 15% and the factor VIlag was 0.77 ± 0.19 μg/ml. FVII activity was significantly increased with age, whereas the concentration demonstrated no correlation to age in this population. FVII concentration is highly correlated with the activity as measured by clotting assay using rabbit thromboplastin. The ratio between FVIIc and FVIIag was not age-dependent, but demonstrated a significant difference between men and women. Between TF and FVII we could not detect a correlation.


Problems when calculating reinforced concrete structures based on the concrete deformation under compression diagram, which is presented both in Russian and foreign regulatory documents on the design of concrete and reinforced concrete structures are considered. The correctness of their compliance for all classes of concrete remains very approximate, especially a significant difference occurs when using Euronorm due to the different shape and sizes of the samples. At present, there are no methodical recommendations for determining the ultimate relative deformations of concrete under axial compression and the construction of curvilinear deformation diagrams, which leads to limited experimental data and, as a result, does not make it possible to enter more detailed ultimate strain values into domestic standards. The results of experimental studies to determine the ultimate relative deformations of concrete under compression for different classes of concrete, which allowed to make analytical dependences for the evaluation of the ultimate relative deformations and description of curvilinear deformation diagrams, are presented. The article discusses various options for using the deformation model to assess the stress-strain state of the structure, it is concluded that it is necessary to use not only the finite values of the ultimate deformations, but also their intermediate values. This requires reliable diagrams "s–e” for all classes of concrete. The difficulties of measuring deformations in concrete subjected to peak load, corresponding to the prismatic strength, as well as main cracks that appeared under conditions of long-term step loading are highlighted. Variants of more accurate measurements are proposed. Development and implementation of the new standard GOST "Concretes. Methods for determination of complete diagrams" on the basis of the developed method for obtaining complete diagrams of concrete deformation under compression for the evaluation of ultimate deformability of concrete under compression are necessary.

<p>Regulation of physiological pH is integral for proper whole-body and cellular function, and disruptions in pH homeostasis can be both a cause and effect of disease. In light of this, many methods have been developed to monitor pH in cells and animals. In this study, we report a chemiluminescence resonance energy transfer (CRET) probe Ratio-pHCL-1, comprised of an acrylamide 1,2-dioxetane chemiluminescent scaffold with an appended pH-sensitive carbofluorescein fluorophore. The probe provides an accurate measurement of pH between 6.8-8.4, making it viable tool for measuring pH in biological systems. Further, its ratiometric output is independent of confounding variables. Quantification of pH can be accomplished both using common fluorimetry and advanced optical imaging methods. Using an IVIS Spectrum, pH can be quantified through tissue with Ratio-pHCL-1, which has been shown in vitro and precisely calibrated in sacrificed mouse models. Initial studies showed that intraperitoneal injections of Ratio-pHCL-1 into sacrificed mice produce a photon flux of more than 10^10 photons per second, and showed a significant difference in ratio of emission intensities between pH 6.0, 7.0, and 8.0.</p> <b></b><i></i><u></u><sub></sub><sup></sup><br>

Wound examination is indispensable in forensic practice. The scientific field of wound age determination has advanced progressively during recent years.The purpose of this study was to determine the differences of fibronectin and TGF-β1 expression in both antemortem and postmortem wounds. This study was an experimental with completely randomized design.  The skin wounds (vital and postmortem) were taken from fourty Wistar rats and divided into 10 groups of rats. Immunohistochemical staining was performed to determine the differences between antemortem and postmortem wounds. The result showed that in 30 minutes after antemortem wound infliction, all of samples showed weak reactivity for fibronectin and TGF-β1 (100%).  In first hour after wound infliction, 3 samples (75%) showed weakly positive and 1 sample (25%) strongly positive for fibronectin and TGF-β1.  In 2 hour after wound infliction, 1 sample (25%) showed weakly positive and 3 sample (75%) strongly positive for fibronectin and TGF-β1.  In 3 and 4 hour after wound infliction, all of samples strongly positive for fibronectin and TGF-β1.  In postmortem wound, all of samples showed negativity for fibronectin and TGF-β1. In conclusion, fibronectin and TGF-β1 may be useful in the determination of wound vitality. Keywords: wound, fibronectin, TGF-β1, vitality
