Total Phenolics Concentration and Antioxidant Potential of Extracts of Medicinal Plants of Pakistan

Abstract Thirty-seven plant organs, traditionally used as drugs, collected in Pakistan, were extracted with 70% acetone and analyzed for their total phenolics concentration and antioxidant potential. Seven extracts showed more than 85% inhibition of lipid peroxidation in vitro as compared with blank. Butylated hydroxytoluene (BHT) (IC50 = 233.6 μg / 1 ± 28.3) was the strongest antioxidant in our test system. The IC50 results indicate that the extracts of Nymphaea lotus L. flowers, Acacia nilotica (Linn.) Delile beans, Terminalia belerica Roxb. fruits, and Terminalia chebula Retz. (fruits, brown) were stronger antioxidants than a-tocopherol, while Terminalia chebula Retz. (fruit coat), Terminalia chebula Retz. (fruits, black) and Ricinus communis L. leaves were weaker antioxidant extracts than a-tocopherol and BHT. Total phenolics concentration, expressed as gallic acid equivalents, showed close correlation with the antioxidant activity. High performance liquid chromatographic analysis with diode array detection at 280 nm, of the seven extracts indicated the presence of hydroxybenzoic acid derivatives, hydroxycinnamic acid derivatives, flavonol aglycones and their glycosides as main phenolics compounds. This information, based on quick screening methods, enables us to proceed towards more detailed chemical and pharmacological understanding of these plant materials.

Willowherb (Epilobium angustifolium L., family Onagraceae) is a well-known food and medicinal plant used after fermentation as a source of beverages with high antioxidant potential. Despite this long history of use, only a few papers have described the chemical profile and bioactivity of fermented willowherb tea in general. To understand the basic metabolic differences of non-fermented and fermented E. angustifolium leaves, we used general chemical analysis, high-performance liquid chromatography with photodiode array detection and electrospray ionization triple quadrupole mass spectrometric detection assay, and an isolation technique. As a result, the content of 14 chemical groups of compounds was compared in the two plant materials; 59 compounds were detected, including 36 new metabolites; and a new water-soluble phenolic polymer of melanoidin nature was isolated and characterized. The fundamental chemical shifts in fermented E. angustifolium leaves relate mainly to the decrease of ellagitannin content, while there is an increase of melanoidin percentage and saving of the antioxidant potential, despite the significant changes detected. The strong antioxidative properties of the new melanoidin were revealed in a series of in vitro bioassays, and a simulated gastrointestinal and colonic digestion model demonstrated the stability of melanoidin and its antioxidant activity. Finally, we concluded that the new melanoidin is a basic antioxidant of the fermented leaves of E. angustifolium, and it can be recommended for additional study as a promising food and medicinal antioxidant agent.

The aim of this study was to evaluate the antiproliferative activity, the antioxidant potential, and the chemical profile obtained from the whole fruit and from leaves of Garcinia gardneriana, a fruit tree from Brazilian Cerrado. To determine in vitro antiproliferative activity, the following neoplastic cell lines were considered, along with an immortalized nontumor cell line. The antioxidant potential was determined, and the evaluation of antiradical air activity was performed. The levels of vitamin C and carotenoids were determined. The chemical profile was analyzed by high-performance liquid chromatography coupled to a diode array detector and a mass spectrometer using electrospray ionization interface. The chloroform fraction of the leaf showed antioxidant activity. The vitamin C content had lower values in fruits and higher in leaves. The content of carotenoids for fruits and leaves was expressive. The ethanolic extract and the hexane and chloroform fractions of fruits were active in all neoplastic lines tested. The leaves showed cytotoxic activity in the hexane fraction in the breast carcinoma line. The analysis of data obtained verified the presence of dimers, monomers, and tetramers of hexoses, polycarboxylic acids, xanthones, flavonoids, biflavonoids, and benzophenones.

Schisandra rubriflora is a dioecious, underestimated medicinal plant species known from traditional Chinese medicine. The present study was aimed at characterising the polyphenolic profile composition and the related antioxidant capacity of S. rubriflora fruit, stem and leaf and in vitro microshoot culture extracts. Separate analyses of material from female and male specimens were carried out. This study was specifically aimed at detailed characterisation of the contribution of phenolic compounds to overall antioxidant activity using ultra-high-performance liquid chromatography with a photodiode array detector coupled to electrospray ionization ion trap mass spectrometry (UHPLC-DAD-ESI-MS3) and a high-performance liquid chromatography-diode array detector (HPLC-DAD). Using UHPLC-DAD-ESI-MS3, twenty-seven phenolic compounds from among phenolic acids and flavonoids were identified. Concentrations of three phenolic acids (neochlorogenic, chlorogenic and cryptochlorogenic acids) and eight flavonoids (hyperoside, rutoside, isoquercitrin, guaijaverin, trifolin, quercetin, kaempferol, and isorhamnetin) were determined using HPLC-DAD using reference standards. The highest total phenolic content was confirmed for the stem and leaf extracts collected in spring. The contents of phenolic compounds of in vitro biomasses were comparable to that in the fruit extracts. The methanolic extracts from the studied plant materials were evaluated for their antioxidant properties using various in vitro assays, namely free radicals scavenging estimation using 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH), ferric-reducing antioxidant power (FRAP) and cupric-reducing antioxidant capacity (CUPRAC) as well as QUick, Easy, New, CHEap, and Reproducible CUPRAC (QUENCHER-CUPRAC) assays. A close relationship between the content of polyphenolic compounds in S. rubriflora and their antioxidant potential has been documented.

Objective: Antioxidants are very important for human health, and hence, external supplementation is recommended to protect cells from the deleterious effects of excessive reactive oxygen species conditions that are capable of causing direct damage to deoxyribonuclieic acid (DNA) which might lead to the cause of cancer. In our research work, we have aimed to evaluate the antioxidant potential of Terminalia chebula and pyrogallol to relate to its apoptotic nature as well as prove its non-toxicity.Methods: Investigations were carried out to determine the antioxidant activity by 2,2-diphenyl-1-picrylhydrazyl and lipid peroxidation inhibition assay of the ethanolic extract of T. chebula and pyrogallol. The apoptotic nature of tumor cells was determined by comet assay. To prove the non-toxicity of the extract as well as compound, the hemolysis assay was carried out.Results: The results of our study established the antioxidant potential of the plant which may serve as additional evidence for the antiproliferative activity of the plant. It is considered safe for human use.Conclusion: Plant-based traditional medicine system continues to play an essential role in health care. The ethanolic extract of T. chebula and pyrogallol possesses antioxidant and anticancer properties.

ABSTRACT Introduction Laghupanchamula is a compound formulation prepared by combinations of roots of five herbs. Two variants of Laghupanchamula have been described in Ayurvedic classics where beside four common herbs fifth one is either Gokshura (Tribulus terrestris L.) or Eranda (Ricinus communis L.). The objective of the study is to make comparison between two variants of Laghupanchmula with respect to important group of phytochemicals and antioxidant activity to corroborate the science behind their therapeutic utility. Materials and methods Standard methods have been followed for quantitative determination of total quantity of phenols, tannins, flavonoids, and flavonols, and in vitro antioxidant activity in variants of Laghupanchmula formulations. Qualitative high-performance liquid chromatography (HPLC) analysis has also been performed to establish presence/absence of important chemical constituents in formulations. Results Additional quantity of phenols, tannins, flavonoids, and flavonols has been observed in Laghupanchamula variant containing Eranda than the variant containing Gokshura. Greater antioxidant activity has also been found in formulation containing Eranda. The HPLC analysis revealed the presence of shikimic acid, gallic acid, catechin in both formulations, but rutin has been found only in formulation containing Eranda. Conclusion From the results of experiments it has been observed that the formulation containing Eranda has more antioxidant activity as it contains more quantities of phenol, tannins, flavonoids, and flavonols than the formulation containing Gokshura. Objective: The main objective of this study was to determine the enhancement of in vitro antioxidant potential of fruits of Terminalia chebula (TC) when used in combination with fruit extracts of Phyllanthus emblica, Ananas comosus, and Punica granatum. Hydroxyl (OH) radical scavenging and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assays were used to analyze the antioxidant potential.Method: Formulations of different combinatorial concentrations of fruits to prepare a mixture were achieved with central composite design through response surface methodology. Screening of 300 different combinations of various concentrations was done through hydroxyl radical scavenging assay followed by statistical analysis of data. Further validation of results was done by measuring the antioxidant potential of most bioactive extracts by DPPH method.Results: Screening of 300 samples of different combinations for antioxidant potential revealed the samples with highest percentage inhibition in aqueous (85.2%), ethanolic (92.9%), and aqueous-ethanolic (84.21%) extracts. Data were subjected to analysis of variance and generated a three-dimensional response surface plot for highest activity. Further subjecting these extracts to DPPH assay revealed a significant enhancement in the antioxidant potential of ethanolic extract of TC when used in mixture with other plants.Conclusion: Antioxidant activity of TC was enhanced when used in combination with other fruits extracts. These synergistic studies generating valuable interactions between various phytochemicals could lead to a momentous increase in other associated activities to fight against diseases such as cancer and cardiovascular disorders. Further research on isolation of bioactive compounds in the mixture and their potential to fight various types of cancer could lead to a significant augmentation in the activity of natural compounds.

The present study was conducted to evaluate the in vitro antioxidant activity of Ficus carica, commonly known as fig. Methanol and ethanol extracts of F. carica leaves were subjected to 2, 2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging activity assay where ascorbic acid being positive control had an IC50 value of 3.98±0.26 while methanol and ethanol fractions showed an IC50 of 101.76±1.12 and 93.12±1.17 respectively exhibiting their high antioxidant potential. DPPH assay was also performed on high performance liquid chromatography (HPLC) elutions. Most active antioxidant components in ethanol extract were eluted between 17–18 min, and those in methanol were eluted over 14–15 min and upon ultra-high performance liquid chromatography-mass spectrometery (Orbitrap Liquid Chromatography-Mass Spectrometry) were identified to be 13-Docosenamide, (Z)- for ethanol and ficusin for methanol fraction. Thus, it is concluded that these two components are most probable determinants of antioxidant potential of F. carica leaf extracts. © 2021 Friends Science Publishers

The present study describes in vitro antimicrobial and antioxidant activities of the methanol extracts which were obtained from the leaves and twigs of the plant Daphne cneorum L. The antimicrobial activity of these extracts was tested against human pathogenic microorganisms using a minimum inhibitory concentration (MIC) values. Total phenolics and flavonoids, as well as the antioxidant activity of the extracts were determined. The two tested extracts showed good antimicrobial and antioxidant activities. The results of a high performance liquid chromatographic (HPLC) method showed that 7,8-dihydroxycoumarin is one of the most abundant secondary metabolite in the tested extracts. The results of this study clearly indicated that the extracts of D. cneorum could be used as a potential source of natural antioxidant and antimicrobial agents.
