The influences of dietary fumonisin B1(FB1), a metabolite ofFusarium verticillioides, on the onset of puberty, semen quality, fertility rates and testicular morphology in male rabbits (bucks) were studied. Forty male rabbits were randomly assigned and fed four diets containing 0.13, 5.0, 7.5 and 10.0 mg FB1/kg, constituting diets 1 (control), 2, 3 and 4 respectively, for a period of 175 days in a completely randomized design. During the last week of the feeding trial, two untreated female rabbits were mated to each of the four treated bucks per treatment to assess the fertility rate of the treated bucks. Onset of puberty in animals fed diets 3 and 4 was significantly (P<0.05) delayed by some 9–12 days. The weight at puberty, sperm concentration and total sperm/ejaculate were not significantly influenced by the dietary FB1. Sperm mass activities, motility and live spermatozoa of the rabbits' semen significantly (P<0.05) declined with an increase in the dietary FB1. The highest sperm cell abnormalities were recorded in the animals fed 10.0 mg/kg FB1, while the least was observed in the control animals. The conception rate, litter size and embryo survival rate were statistically the same among the dietary treatments. Embryo mortality was significantly (P<0.05) higher in rabbits fed diets 3 and 4 than in others. Testicular elements were significantly (P<0.05) impaired by the toxin in rabbits fed 7.5 and 10.0 mg FB1/kg. This suggests that LOAEL of 7.50 mg/kg FB1delayed puberty, impaired semen quality and spermatogenesis and induced embryo mortality without a statistically adverse effect on the fertility rates of male rabbits.

<p class="MDPI17abstract"><strong>Objective: </strong>Reproduction and nutritional status is closely related. Nutritional deficiency or insufficiency directly affects reproductive efficiency. Deficiency of nutrition could affect the sperm quality and the ability to fertilize. The objective of this study was to evaluate the effect of feed additive (Minoxvit) on semen quality of buck.</p><p class="MDPI17abstract"><strong>Methods: </strong>This study used 6 sexually mature bucks, aged 1.5 years old. The bucks were fed daily with 3 kg of freshly chopped king grass, 1 kg of Legume (<em>Calliandra</em> sp.), and 500 g of concentrate. Three bucks were given Minoxvit by 1.25 g/day in the concentrate, while three other bucks were considered as control. Semen was evaluated once a week for 49 days. Semen was evaluated macroscopically and microscopically. The macroscopic evaluation consisted of volume, color, and consistency of semen, while microscopic evaluation consisted of mass motility, sperm motility, viability, and sperm concentration. Data were analyzed using Completely Randomized Design with Tukey test.<strong></strong></p><p class="MDPI17abstract"><strong>Results: </strong>The result showed significantly different (<em>P</em>&lt;0,05) in which bucks semen in Minoxvit addition group had higher semen volume (0.75 ml vs 0.54 ml), mass motility (3.32 vs 2.67), sperm motility (70% vs 58 %), sperm viability (86.67% vs 79.19%), and sperm consentration (2,6x10<sup>9</sup> mL vs 1,7x10<sup>9</sup> mL).<strong></strong></p><p><strong>Conclusions: </strong>This study concludes that the addition of Minoxvit has a positive influence on the quality of buck sperms providing volume, mass motility, individual motility, viability, and concentration of the sperm.</p>

Bisphenol A (BPA) is considered an endocrine disruptor and has been associated with deleterious effects on spermatogenesis and male fertility. Bisphenol F (BPF) and S (BPS) are structurally similar to BPA, but knowledge of their effects on male fertility remains limited. In this cross–sectional study, we investigated the associations between exposure to BPA, BPF, and BPS and semen quality in 556 men 18–20 years of age from the Fetal Programming of Semen Quality (FEPOS) cohort. A urine sample was collected from each participant for determination of BPA, BPF, and BPS concentrations while a semen sample was collected to determine ejaculate volume, sperm concentration, total sperm count, sperm motility, and sperm morphology. Associations between urinary bisphenol levels (continuous and quartile–divided) and semen characteristics were estimated using a negative binomial regression model adjusting for urine creatinine concentration, alcohol intake, smoking status, body mass index (BMI), fever, sexual abstinence time, maternal pre–pregnancy BMI, and first trimester smoking, and highest parental education during first trimester. We found no associations between urinary bisphenol of semen quality in a sample of young men from the general Danish population.

PURPOSE: To elucidate the biologic association between germ cell neoplasia and testicular dysfunction, through investigation of Leydig cell function and semen quality in men with carcinoma-in-situ (CIS) of the testis. PATIENTS AND METHODS: We examined two groups of men, unilaterally orchidectomized for testicular cancer. Biopsy of the contralateral testis had showed CIS in a group of 24 patients and no evidence of CIS in the other group of 30 patients. Semen quality and serum levels of testosterone, luteinizing hormone (LH), and follicle-stimulating hormone (FSH) were compared in these two groups of men after orchidectomy but before further treatment for testicular cancer. RESULTS: Significantly higher LH levels (median, 8.1 IU/L v 4.8 IU/L; P < .001) and generally lower testosterone levels (median, 12.5 nmol/L v 15.5 nmol/L; P = .13) were found in the CIS group. The proportion of patients with Leydig cell dysfunction was higher in the group of patients with CIS (11 of 24) than in the group of patients without (two of 30) (P = .01). Sperm concentration and total sperm count were significantly lower (P < .001) in patients with CIS (median, 0.03 × 106/mL and 0.10 × 106, respectively) than in patients without (median, 9.1 × 106/mL and 32 × 106, respectively), whereas the levels of FSH were significantly higher (P < .001) in the former group of men (median, 19.6 IU/L v 9.0 IU/L). CONCLUSION: Not only spermatogenesis but also Leydig cell function is impaired in testes with CIS. This impairment could be due to common factors in the pathogenesis of germ cell neoplasm and testicular dysfunction. Alternatively, CIS cells may have a negative impact on Leydig cell function.

Abstract Study question What is the cause of semen quality impairment in oncological patients during fertility preservation programs? The cancer type and stadiation or the resulting inflammatory state? Summary answer The inflammatory state seems to be related to the decrease of sperm concentration, motility, morphology and viability due to the worsening of oxidative stress microenvironment. What is known already Fertility preservation acquired a great importance in the last decades due to increase survival of oncological patients, boost of diagnosis under 40 years and postponement of paternal age. At the time of cryopreservation, only one third of these males are normozoospermic. Tumor itself or other factors, added to psychological reasons, may be involved but there is no clear evidence. An imbalance of ROS (reactive oxygen species) in semen can compromise its quality. However, the correlation between cancer-related generalized stress state and fertility is poorly investigated. Inflammatory conditions induced by infections and pathologies, including cancer, increase ROS. Study design, size, duration Retrospective observational analysis was performed on 45 patients (29.0 ± 6.9 yrs) recruited during their fertility preservation program between 2016 and 2019 with written consent on use of their clinical data for research purpose. Patients presented several oncological diagnoses. Semen samples obtained from multiple collections (N = 58) were analyzed before applying standard freezing protocol. Data on semen parameters, inflammatory indices, hematological values and type/stage of tumors were collected. No exclusion criteria were applied. Participants/materials, setting, methods Routine semen analysis was performed according to the WHO standards. Sperm concentration and motility were evaluated on Makler Chamber, whereas eosin stain and Diff-quick slides were used for viability and morphology, respectively. Lymphoma was present in 72% of cases, leukemia in 8%, seminoma in 7% and other cancers in 13%. Correlations (Pearson/Spearman tests) among principal semen parameters and hematological values (leukocytes, erythrocytes, hemoglobin, RDW, albumin, etc.) were calculated with a P-value &lt;0.05 considered statistically significant. Main results and the role of chance The majority of semen samples showed a severe impairment, with one or more parameters under lower reference limits (WHO): 48.3% had sperm concentration under 15 millions/ml, 43.1% had a progressive motility under 32%, 41.4% had viability under 58% and 91.4% had abnormal morphology (under 4%). The role of potential inflammatory state was analyzed by correlating semen parameters and some hematological values. No correlation was found with cancer type. Negative association resulted between progressive motility (%PR) and leukocytes (p = 0.041) or RDW% (p = 0.015), but positive one with albumin (p = 0.012). Even sperm count, total motility (%PR+NP) and morphology were significantly correlated to RDW% (p = 0.003, p = 0.032, p = 0.034, respectively). These findings suggest a possible role of inflammation and ROS related generation in semen quality impairment. Indeed, albumin exerts a protective action, but leukocytes are known to cause ROS increase. Cancer-induced oxidative stress state may alter red blood cells homeostasis and vitality and increase erythrocytes turnover resulting in high RDW values. It is likely semen is worse when blood values indicate more severe cancer-induced inflammatory condition. Limitations, reasons for caution Significant correlations with type/stage of cancer were not found due to small number of each diagnosis, in spite our study considered 3 years of patients inclusion. Moreover, we lack to analyze the same patient before the cancer onset to avoid the influence of inflammatory state generated by the tumor itself. Wider implications of the findings: Understanding the influence of cancer-induced inflammatory state on semen quality could increase the awareness that clinicians should direct patient to the fertility preservation as soon as possible, even if diagnosis is still ongoing. It should be evaluated whether offering specific treatments may reduce oxidative stress conditions. Trial registration number Not applicable

An experiment was carried out with the objective of evaluating the addition of 25-hydroxycholecalciferol (25(OH) D3) in diets of broiler breeder hens. The experiment used Cobb 500 broiler breeder hens and was allotted to a complete randomized design with four treatments and eight replications of twenty females and two males each. The treatments consisted of vitamin premixes with 2,000 and 3,400 IU/kg diet vitamin D3 as the only source of vitamin or 2,000 IU D3 plus 35 or 69 mg/t of 25(OH) D3. Results of this experiment indicated that 25(OH) D3 had no significant effect on egg production parameters from 32 to 67 weeks. The supplementation of 25(OH) D3 resulted in better quality egg shells evaluated by the specific gravity at 60 weeks of age, regardless of the dosage. No significant differences were observed for hatchability of broiler breeder fertile eggs at 54 and 64 weeks. At 64 weeks, the hatch residue breakout showed less embryo mortality at the third week for treatments receiving 2,000 UI D3 in the diet and less embryo mortality at the second week of development from hens aged 67 weeks and supplemented with 2,000IU D3 and 2,000IU D3+ 69 mg 25(OH)D3. It was concluded that the supplementation with 25-hydroxycholecalciferol with cholecalciferol had similar effects as the diets with vitamin D3 as the only source on the productive performance of broiler breeder hens.

Abstract Background: Iodine is an essential element for hormones synthesized by the human thyroid gland. Thyroid hormone deficiency affects all body tissues, including multiple endocrine changes that alter growth hormones, corticotrophin, glucocorticoids, and gonadal function. In many countries, subfertility is an important problem affecting about 10–15% of all couples trying to conceive. Male subfertile factors have accounted for 25–30% of all cases in their studies. The aim of this study was to study the correlation between urinary iodine concentration (UIC) and semen quality of fertile male in China.Methods: semen and urine samples were collected from males who attended a survey for the reproductive physiological constants of male and female in China. All participants were fertile and they should be free from diseases of male reproductive system and thyroid related diseases. We studied the correlation between UIC and semen quality parameters in fertile males and the effect of iodine concentration on male reproduction. Participants were divided into six groups based on UIC. We measured semen parameters and UIC. Semen parameters or time-to-pregnancy (TTP) were analyzed by Spearman correlation, linear regression and multivariable logistic regression. Results: 1089 men were recruited in the study. The mean and median age of participants were 27.46±0.12 and 27±3.88 years, respectively. UIC of 274 (25.16%) men was lower than the UIC of WHO recommended. UIC of 405 men located in the region of WHO recommended. 62.35% of them were not located in the optimal region. Semen concentration and total sperm count were found to decrease when UIC increases when the UIC≥100 ug/L. Conclusions: A male with deficient UIC or excessive UIC has more risk of lower semen quality such as sperm concentration and total sperm count. In addition, if a male with deficient UIC or excessive UIC, his wife has longer TTP than the males who had normal UIC. Deficient UIC associated with lower semen quality and TTP than excessive UIC.

AbstractPeroxisomes are ubiquitous organelles in eukaryotic cells that fulfill various important metabolic functions. In this study, we investigated the role of Docking/Translocation Module (DTM) peroxins, mainly FvPex8, FvPex13, FvPex14, and FvPex33, in Fusarium verticillioides virulence and fumonisin B1 (FB1) biosynthesis. Protein interaction experiments suggested that FvPex13 serves as the core subunit of F. verticillioides DTM. When we generated gene deletion mutants (ΔFvpex8, ΔFvpex13, ΔFvpex14, ΔFvpex33, ΔFvpex33/14) and examined whether the expression of other peroxin genes were affected in the DTM mutants, ΔFvpex8 strain showed most drastic changes to PEX gene expression profiles. Deletion mutants exhibited disparity in carbon source utilization and defect in cell wall integrity when stress agents were applied. Under nutrient starvation, mutants also showed higher levels of lipid droplet accumulation. Notably, ΔFvpex8 mutant showed significant FB1 reduction and altered expression of FUM1 and FUM19 genes. However, FvPex13 was primarily responsible for virulence, while ΔFvpex33/14 double mutant also showed virulence defect. In summary, our study suggests that FvPex13 is the core component of DTM, regulating peroxisome membrane biogenesis as well as PTS1- and PTS2-mediated transmembrane cargo transportation. Importantly, we predict FvPex8 as a key component in DTM that affects peroxisome function in FB1 biosynthesis in F. verticillioides.

Abstract The aim of the study was to determine the relationship of sperm morphology with age of males, ejaculate concentration and volume, as well as with acrosin activity determined in sperm acrosome extracts. The study used manually collected ejaculates from 9 male arctic foxes, including 6 young males aged one year and 3 older males (between 3 and 5 years of age). All of the 39 ejaculates used in the study were classified as normal based on motility exceeding 70%. The ejaculates collected from the foxes were evaluated for volume, sperm concentration and frequency of morphological changes including primary and secondary defects. The spermiograms of the male arctic foxes were classified according to a six-grade subjective scale. In addition, acrosin activity was determined in the sperm acrosome extracts. The data were analysed using the criteria of male age, sperm concentration, ejaculate volume, and acrosin activity. The morphology of arctic fox spermatozoa was dependent on the age of the male. A greater number of morphologically altered spermatozoa tended to occur in the ejaculates of young foxes, which were in their first breeding season. In addition, statistical analysis revealed positive relationships between the frequency of morphological changes in sperm and their ejaculate concentration. In contrast, there were no significant correlations between the percentages of morphologically changed spermatozoa and the ejaculate volume and the content of acrosin, which is an indicator of acrosomal integrity. Semen quality is dependent on the number of sperm in ejaculate with morphological defects which prevent oocyte fertilization. Therefore, morphological assessment of semen, which covers both the number and type of morphological changes, is highly useful when selecting appropriate males for reproduction.
