Abstract: The aim of the present study was to determine the most suitable pretreatment to enhance the tetrazolium test in seeds of the Epidendrum genus. Initially, mature capsules were harvested at El Escorial village, in the municipality of Pamplona, Colombia. Subsequently, the seeds were subjected to five pretreatments: deionized water, 0.5% NaClO, 1.0% NaClO, 10.0% sucrose and a control group. Using the syringe method with cloth filter, the seeds were rinsed with distilled water and subjected to two concentrations of tetrazolium solution (0.25%, 1.0%) and three exposure times (6 h, 24 h and 48 h). Finally, the tetrazolium viability test results were correlated with the in vitro germination test. It was found that the use of deionized water improves the efficiency of the tetrazolium test in seeds of Epidendrum fimbriatum and E. microtum; as in seeds of E. elongatum when using 1.0% tetrazolium for 24 h. Similarly, a high correlation was found between viability and germination, using deionized water and 10.0% sucrose, with homogeneous results with each other.

Proposition: The purpose of this study was to evaluate in vitro the color changes of esthetic orthodontic elastomeric ligatures of different shades when exposed to four food colorings commonly found in the diet of patients. Materials and Methods: The sample consisted of esthetic orthodontic elastomeric ligatures in the colors pearl, pearl blue, pearl white and colorless, which were immersed for 72 hours in five different solutions: distilled water (control group), coffee, tea, Coca-Cola ® and wine. The color changes of the esthetic orthodontic elastomeric ligatures were measured with the aid of a spectrophotometer, at T1 - as provided by the manufacturer; and T2 - after colorings process. Results: The results indicated that the esthetic orthodontic elastomeric ligatures of all initial hues are susceptible to pigmentation. Among the evaluated colors, all changed the finished look and the color of the samples tested. In ascending order, the color of the samples was as follows: distilled water, Coca-Cola®, black tea, wine and coffee. Conclusion: The substances that have a greater potential for pigmentation in esthetic orthodontic elastomeric ligatures were black tea, wine and coffee, respectively. All shades of esthetic orthodontic elastomeric ligatures are susceptible to color change.

Introduction: Acidulated phosphate fluoride (APF) gels are commonly used as preventive caries materials in paediatric dentistry while glass ionomer cements (GICs) are widely used as a restorative material. The purpose of this study was to determine the effect of acidulated phosphate fluoride application towards the microhardness of GIC. Methods: This study was an experimental laboratory study where ten specimens were prepared from high viscosity GIC, which was Fuji IX. Specimens were stored in 50 ml distilled water at first 24 hours at 37°C, and then specimens were divided into fluoride group which was immersed in 25 ml of 1.23 % APF gel and control group which was stored 25 ml distilled water for 24 hours at 37°C. The Micromet II Microhardness Tester, Buehler, IL, USA that was standardised for Vickers hardness test was used to test the specimens at 100-gram load. Data were analysed using the t-test comparison test. Results: The level of microhardness of the fluoride group (14.34) was much lower compared to the control group (43.21) with a highly significant difference (p<0.01). Conclusion: The application of 1.23% APF gel on high viscosity GIC reduces the microhardness level compared to the control group.

INTRODUCTION: The purpose of this study was to compare the frictional forces between 0.014-in NiTi wires (Aditek) with 4 mm horizontal deflection and brackets with different archwire ligation systems. METHODS: Four types of self-ligating brackets (Damon MX, Easy Clip, Smart Clip and In-Ovation), a triple bracket (Synergy) and a twin bracket with 8-shaped ligature (Tecnident) were tested. Twin brackets with conventional elastomeric ligatures (Morelli) were used as control group. Tests were repeated 10 times for each bracket/archwire combination. Frictional forces were measured in an Instron universal tensile machine at 3 mm/minute speed and a total displacement of 6 mm. Statistical analysis comprised ANOVA and Dunnett's multiple comparison post hoc test. RESULTS: Deflection-induced frictional (DIF) forces increased in the following order: Synergy, Damon, 8-shaped Ligature, Easy Clip, In-Ovation, Smart-Clip and conventional ligatures. The differences among groups were significant, with the exception of the 8-shaped ligature groups which was equal to the Damon and Easy Clip groups. CONCLUSIONS: Compared to conventional ligatures, all ligation systems tested reduced frictional forces. However, such reduction varied according to the ligation system employed.

Xylopia aromatica is a native species from Brazil's "Cerrado", recommended for restoration ecology and also as a medicine. Its seeds have embryos with morphophysiological dormancy, making nursery propagation difficult. The objective of this study was to verify the efficiency of X-ray and tetrazolium tests for evaluating the viability of three seed lots, stored for different periods. All seeds were X-rayed (13 kV, 350 seconds) and samples used for tetrazolium and germination tests. In the tetrazolium test, seeds were submitted to six treatments at two temperatures (25 and 30 °C) with imbibition in distilled water and immersion in three concentrations of tetrazolium solution (0.5, 0.75 and 1%) at the two imbibition temperatures. Seeds for the germination test were placed for imbibition in distilled water and a 500 ppm Promalin® (6-Benzyladenine + GA4 + GA7) solution and later sown in sterilized sand. The embryo could not be observed with the X-ray test. However, those seeds observed with an undamaged endosperm did not differ in the percentages of seeds with firm and stained endosperms observed in the tetrazolium test for all the lots. The tetrazolium test is efficient for evaluating seed viability, principally if imbibed at 30 °C and immersed in a 0.5% solution at 30 °C.

ABSTRACT Aim This study was undertaken to study the effect of three most commonly used dentifrices for denture cleaning and to find out any positive association between number of strokes by toothbrush and surface roughness of acrylic resin. Materials and methods About 40 acrylic rectangular blocks with high finish were polished until surface roughness was calculated as 0 µm. The materials tested were distilled water as a control group, Patanjali Dant Kanti, Colgate Total Cream, and Cleansodent as dentifrices. Stroking was done with dentifrices, and surface roughness readings were recorded at 1,000, 9,000, and 18,000 strokes. Results Patanjali Dant Kanti was found to have maximum abrasive potential followed by Colgate Total Cream, followed by Cleansodent and least by distilled water. Conclusion Surface roughness was found to be directly proportional to the type, size, and fineness of the abrasive component used; the professional denture cleanser (Cleansodent) has less abrasive potential than the regular toothpastes. Among those tested, the highest abrasive potential was found in Patanjali Dant Kanti and lowest by Cleansodent (denture cleanser). Clinical significance The patient as well as dentist should choose the dentifrice with utmost care noting the size and type of abrasives used. Washing the dentures normally with water without toothbrush daily and cleaning of dentures once in 10 to 15 days with a soft toothbrush and professional denture cleanser should be the line of maintenance of dentures. The purpose of this study was to evaluate in vitro the effectiveness of the dentin bonding system All Bond 2 associated with Resinomer (Bisco), and of Vitrebond (3M) glass ionomer cement fresh-mixed, both used in the bonded amalgam technique, to prevent short-term microleakage in class II cavities restored with Dispersalloy (Dentsply), an admixed alloy. The control group utilized the Copalite (Cooley & Cooley) varnish. Forty five sound human extracted premolars were used. Class II cavity preparations were made on the mesial and distal surfaces of non-carious teeth, with the gingival margins wall established 1mm under the cementum enamel junction. The specimens were divided randomly into three groups with thirty cavities in each group. The teeth were stored in distilled water for 24 hours and were thermocyled through 500 cycles in distilled water between 5°C and 55°C with a dwell time of 15 seconds. The apices and roots of the teeth were sealed. They were placed in a 37°C bath of 0.5% basic fuchsin dye for 24 hours. The teeth were washed in tap water for 24 hours and cut. The microleakage scores per restoration were averaged and three values of various test groups were subjected to the Kruskal-Wallis and Dunn test at a significance level of p < 0.05. The All Bond 2/Resinomer showed significantly less microleakage at gingival margins than the Vitrebond and Copalite lined to amalgam restorations. The Vitrebond fresh mixed, lined to amalgam restorations, showed significantly less microleakage than the Copalite lined to amalgam restorations.

The aim of this study was to compare the surface roughness and gloss of polymer-infiltrated ceramics after simulated in vitro toothbrushing in different storage mediums. Four polymer- infiltrated ceramics were evaluated, Lava ultimate (LU), Vita enamic (EN), Shofu (SH), and Crystal ultra (CU). The control group was a feldspathic ceramic, Vita Mark II (VM). One hundred and twenty specimens (12 × 14 × 2.5 mm) were prepared using a precision saw. For each material (n = 24), the specimens were allocated into two groups, polished and stained. The specimens of each group were stored (for 7 days) in either citric acid (0.2N) or distilled water. Data for surface gloss (ΔE*SCE-SCI) and roughness (Ra) were evaluated before (baseline) and after simulated toothbrushing. For toothbrushing simulation, a toothpaste slurry containing a toothpaste of 100 relative dentin abrasion (RDA) and 0.3 mL distilled water was used for 3650 cycles (7300 strokes) for each specimen. Data were analyzed using t-test and ANOVA. A p-value of ≤ to 0.05 was considered significant. The highest mean value of surface gloss was identified in CU (stained—water) (4.3 (0.47)) (ΔE*) and EN (stained—acid) (4.3 (1.00)) (ΔE*) specimens, whereas the lowest mean value was shown by SH (stained—acid) (2.04 (0.42)) (ΔE*) samples. The highest mean value of surface roughness was observed in SH (0.40 (0.99)) Ra (stained—acid) whereas the lowest in VM (0.13 (0.039)) Ra (polished—water). A significant difference (p < 0.05) was observed in surface roughness and gloss between the materials with simulated toothbrushing, except in VM and LU, respectively. Therefore, it can be concluded that simulated toothbrushing impacts on surface roughness and gloss, irrespective of the storage medium.

ABSTRACT The viability of T. aurea seeds is evaluated through the germination test, taking 21 days, which makes it difficult to obtain quick information about the viability of its seeds. In this context, using the tetrazolium test as an evaluation method would be appropriate because it provides faster and more reliable information for several species. In view of the above, the objective of this study was to adapt the tetrazolium test methodology to evaluate the viability of T. aurea seeds. Initially, the imbibition curve was constructed to determine the appropriate hydration period for the seeds and the germination test was conducted, both for evaluating the initial quality and for comparison with the tetrazolium test results. For the tetrazolium test, the seeds were hydrated for 24 hours, then their coats were removed and the seeds were immersed in tetrazolium solutions. The concentrations used were 0.05, 0.075 and 0.1% for three staining periods 2, 4 and 6 hours, at 35 and 40 °C, in the absence of light. The experimental design was completely randomized in a 3 x 3 + 1 factorial scheme (three concentrations of tetrazolium solution x three staining periods + one control = germination test) with four replicates of 25 seeds. Tetrazolium test was adequate to evaluate the viability of T. aurea seeds using the concentration of 0.05% for four hours at 40 °C.

Background: Gelsemium sempervirens (Gelsemium s.) is a highly toxic plant but is employed at low doses and/or high dilutions as an anxiolytic and antidepressant. Previous investigations in our laboratory [1,2] have shown a significant anxiolytic-like activity of Gelsemium s., using emotional response models in laboratory mice. Although there is some biochemical evidence of a possible role of neurosteroid metabolism [3], the cellular and molecular mechanisms involved in the effects of Gelsemium s. at the level of nervous system are largely unknown. To help determine these pathways, we used human neurocytes (SH-SY5Y cell line) treated in vitro with different dilutions of Gelsemium s. and evaluated their vitality and gene expression changes. Methods: The drugs were produced by Boiron Laboratoires, Lyon (F), starting from a whole-plant-hydroalcoholic extract of Gelsemium s. Solutions 1C, 2C, 3C, 4C, 8C and 29C (C= centesimal dilution/dynamization prepared in 30% ethanol/distilled water) were provided in 30-ml glass bottles, wrapped in aluminium foil and were stored in the dark at room temperature in a metal cupboard. Control solutions (“placebo”) were serially diluted/dynamized 30% ethanol/distilled water. Before each experiment, 0.05 ml samples of Gelsemium s. and placebo were added to 5 ml of distilled sterile and apyrogenic water in a 15 ml Falcon polystyrene plastic tube, closed and shaken in mechanical shaker DinaA for 7.5 sec (150 strokes) to obtain the final 2C, 3C, 4C, 5C, 9C and 30C succussed dilutions, with ethanol concentration of 0.3% (v/v) (final 0.03% in the assay system). Human neuroblastoma cell line SHSY5Y was grown in DMEM-F12 medium (Lonza), with 10% fetal bovine serum (FBS), penicillin (100 units/ml) and streptomycin (100 mg/ml). To assess cell viability and metabolism, 20,000 cells per well were seeded in 96 microplate wells in 200 μl of medium. After overnight incubation, 22 μl of drug or placebo were added and the plate was incubated at 37°C with 5% CO2 in a humidified atmosphere for 72 hours. Then the viability test with reagent WST-1 (Roche) was performed for 3 hours and the absorbance was detected with multiplate reader. A total of 17 experiments, each done with six replicate microwells. To make a relative measurement of protein, the cells were lysed and a Bradford assay was done directly in the plate. The Student t-test and the sign rank test for paired data were utilized for data analysis. To obtain a profile of gene expression, cells were pre preconditioned with Gelsemium s./placebo dilutions for 24 h, then RNA was isolated and analysed by microarray and RT-PCR. SHSY5Y cells were plated onto Petri dishes (day 1) and the day after the medium was replaced with the medium with 2% FBS (day 2). After 24h, 10%v/v Gelsemium s. or placebo dilutions were added to the medium (day 3) and cells were incubated for a further 24h. On day 4, cells were then harvested and the RNA extracted using the Qiagen RNAeasy Mini Kit following the manufacturer’s instructions. Microarray analysis was performed on a custom 12 x 135 k human NimbleGen microarray containing 45033 genes with 3 probes per target gene. Four biological replicates were analysed for each condition. Analysis of differentially expressed genes was performed using linear modelling and empirical Bayes methods and p-values were adjusted for multiple testing with the Benjamini and Hochberg method. A Human Neurotransmitter Receptors and Regulators RT2 Profiler PCR array (Qiagen) was performed in profiling the expression of genes involved in modulating the biological processes of neurotransmitter biosynthesis, uptake, transport and signaling through neurotransmitter receptors. Results: In viability tests, cells treated with Gelsemium s. showed slightly higher metabolic activity (3-4 %) than those treated with placebo. Overall comparison of the data for the whole sample of placebo versus that of Gelsemium s. using the Student t-test showed a small but significant difference (p < 0.001). Furthermore, a non parametric approach comparing the two treatments at the same dilution yielded a significant difference under the sign test (p < 0.01) and Wilcoxon rank test for paired data (p < 0.05), so that the values of differences were also considered. The differences between groups having the same dilution (placebo 2C versus Gelsemium s. 2C etc.) were significant in four dilutions: 2 C, 3 C, 4C (p < 0.01), 9 C (p < 0.02), while 5 C and 30 C yielded non-significant values. No changes due to Gelsemium s. were detected using protein assay, suggesting that the viability test revealed effects on metabolic activity instead of on cell proliferation. In microarray analysis, transcripts expression was analyzed and genes differentially expressed by the Gelsemium s. dilutions were selected. A gene was considered to be differentially expressed if it showed an absolute value of log-ratio greater than or equal to 0.5, an index that translates to a fold-change of 1.4 in transcript quantity. ABSTRACT Objective: The aim of this study was to evaluate the effect of agents available in whitening toothpastes (Crest® and Sensodyne®) in vitro on the force decay of elastomeric chains used in orthodontics. Materials and Methods: A total of 300 specimens of elastomeric chains were divided into five groups (n = 60) and were evaluated. These groups included (1) the regular Crest® toothpaste and distilled water solution, (2) whitening Crest® toothpaste and distilled water solution, (3) regular Sensodyne® toothpaste and distilled water solution, (4) whitening Sensodyne® toothpaste and distilled water solution, and (5) distilled water as a control group. The samples' force was measured using Instron at intervals of 0, 1, 7, 14, 21, and 28 days. Data were analyzed by SPSS software. Results: At the initial time point, the difference in the force values of elastomeric chain between any of the groups was not significant (P > 0.05). On the 1st day, there was a significant difference (P < 0.05) between all groups except the groups of whitening Crest®, regular, and whitening Sensodyne® (P > 0.05). On days 7, 14, and 28, the rate of decline for all groups was statistically significant (P < 0.05). Conclusions: According to the results obtained in the present study, it seems that toothpastes without whitening agents have less effect on force decay of elastomeric chain over time.
