The effects of different salinities on the germination of Spartina ciliata seeds were tested during 40 days in the laboratory, using six concentrations of NaCl (0,45, 80, 130, 170 and 215 mM) besides three concentrations of seawater (25,30 and 35‰). In addition, the survival and growth of seedlings were tested in five constant salinity regimes (0,45, 80 and 170 mM NaCl) as well as in five regimes of simulating seawater overwash. The results showed that seeds possed a high germination capacity over a wide range of salinities (0 to 215 mM NaCl). Although concentrations close to full-strength seawater completely inhibited seed germination, the transfer of the ungerminated seeds to freshwater immediately stimulated this. The effects of salinity on seedling growth showed that shoot height and root length were the variables most sensitive to salt stress. Frequent overwash with seawater significantly reduced the survival and growth of Spartina ciliata seedlings. The success in seed germination, survival, and growth of Spartina ciliata in salinities above those normally found in the natural environment could explain the species being distributed throughout salt-stressed foredune habitats in southern Brazil.

There are no records in Brazil for the culture of Pteridae family oysters, genus Pteria and Pinctada. The intermediate culture can be considered one of the critical stages during the oyster farming. The changes in life conditions from a semi-closed environment provided by the collector to the structures utilized for the growth represents a considerable stress factor to which the animals are subjected. The goal of the present study was to evaluate the growth and survival of the pearl oyster Pteria hirundo (Linnaeus, 1758) kept in intermediate lanterns. Seeds of P. hirundo, obtained through larviculture carried out at the Marine Molluscs Laboratory (UFSC), southern Brazil were detached from the collectors, transferred and kept in an intermediate culture system (oyster lantern). During 6 months, growth (dorso-ventral height - DVH in mm) and survival (%) were evaluated. Two size classes were tested: medium (M), between 3.4 and 4.4 mm, and large (L), bigger than 4.5 mm. The density of occupation in each floor of the lantern was 50%. At the end of the experiment, the survivals observed were 90 and 94% for the sizes M and L, respectively. Medium juveniles of Pteria hirundo had an average (Std) increase of 16.1 (±4.38) mm in the dorso-ventral height, and large juveniles an average increase of 11.4 (±3.77) mm after six months of experiment. The results observed during the intermediate stage of cultivation indicate that Pteria hirundo presents survival and growth similar to other species of pearl oysters traditionally used in the pearl industry in several countries.

Background: Nothofagus glauca (Phil.) Krasser (Nothofagaceae, “Hualo”) is an endemic tree of the Mediterranean zone of Chile. The natural forests in this area have been severely fragmented as a result of human causes such as replacement by agricultural crops and fast-growing tree species. From 1975, these forests have declined from 900,000 ha to 145,000 ha, so it is categorised on the IUCN Red List as ‘vulnerable’. In restoring this ecosystem, efforts should focus, in part, on the propagation of quality stock. However, information on propagation systems is still insufficient. Methods: We aimed to analyse the effect of different pre-germinative treatments and sowing times on seed germination, and seedling growth and quality. The pre-germinative treatments were: (i) cold stratification; (ii) soaking in gibberellic acid (GA3) and thiourea solution; and (iii) nursery cultivation, while the sowing times were July, August and September. Results: A high germination capacity was achieved by: soaking the seeds in GA3 solution irrespective of concentration; stratifying, irrespective of period; or soaking in 7.5 mg L-1 thiourea solution, values significantly varied from that of the control treatments. The sowing time was not relevant in terms of the percentage of germination or seedling development. Stratification at 5°C for 60 days produced the best quality indices for N. glauca seedlings but no significant differences were found in any of the morphological attributes tested as a result of the pre-germinative treatments. Conclusions: The pre-germinative treatments significantly improved the germination and seedlings growth of N. glauca. Cold stratification at 5°C for 60 days is recommended as it produced suitable seedlings for field establishment. Gibberellic acid and thiourea did not produce important effects on seedling growth. Our results suggest the presence of endogenous physiological dormancy of the N. glauca seeds. The results of this study provide important information on propagation and nursery techniques of N. glauca, which can be used in restoration programmes.

A study was conducted with an aim of achieving 100% seed germination and to evaluate the rate of dormancy upon exogenous application of the deproteinised leaf juice (DPJ) in various leguminous and non leguminous seeds. DPJ if inducing retardation of dormancy, the activity of enzyme was analysed to prove the presence of phytohormones. DPJ from the three non leguminous foliages of brinjal, radddish and dasheen were prepared by green crop fractionation. The seed germination results obtained by the paper towel method and the activity of enzyme protease in DPJ of radish and Colocasia leaves separately. There was variation in germination rate by different DPJ influence. Some seeds showed fast germination while some germinated late. Few seeds were not germinated by control. In some seeds, DPJ decreased the rate of germination while on the contrary there was rapid growth of seedlings by the potentiality of DPJ. DPJ found mutagenic by its influencing inhibitory expression in seedling growth in some cases. The enzyme protease released by phytohormone gibberellin during the seed germination. Positive glyoxlic test indicated presence of auxins in whey.

Seed germination and seedling growth are two essential early determinants of subsequent crop yield and quality. A high germination percentage of industrial hemp (Cannabis sativa L.) seed is required to import into Australia. The viability of hemp seed can decline rapidly depending on storage and other factors; hence, the quality of imported seed is not always reliable. Here, we aimed to investigate germination and early seedling growth responses of 14 industrial hemp varieties after being imported from various countries. Germination trials were conducted with 100 seeds of 14 varieties using a soil-less Petri dish assay and a compost growth medium under glasshouse conditions. We also assessed the effect of seed pre-treatments such as gibberellic acid (500 and 1000 mg·L−1), chlorine dioxide (500 and 1000 mg·L−1) and cold temperature (4 °C for 72 h) using 300 seeds of each of the three selected varieties in compost growth medium. Hemp varieties imported from China had higher germination and better seedling growth indices than those imported from Europe. All seed pre-treatments were associated with a decreasing trend in germination, but a positive effect on early growth responses was observed. Our findings indicate that the hemp variety Han FNQ performed better than many other varieties did regarding seed germination and seedling growth. Hemp seeds sanitising with 500 mg·L−1 of chlorine dioxide might improve the germination and early growth of seedlings.

The period between seed germination and seedling establishment is one of the most vulnerable stages in the life cycle of annuals in the saline environments. Although germination characteristics of Suaeda salsa seeds have been reported, the comparative germination patterns of dimorphic seeds and seedling growth to different abiotic stresses remain poorly understood. In this study, germination responses of dimorphic seeds to light and temperature were compared. Meanwhile, responses of dimorphic seeds and thereafter seedlings of S. salsa to different concentrations of NaCl and Na2SO4 were also tested. The results showed that the light did not significantly affect germination percentage of brown seeds, but significantly promoted germination of black seeds. Brown seeds could reach high germination percentage over a wide temperature range, however, germination of black seeds gradually increased with the increase of temperature. Brown seeds had higher germination percentage and velocity than black seeds under the same salt conditions. However, black seeds had higher recovery germination than brown seeds when transferred to deionized water. Young seedlings had lower salt tolerance than germinating seeds. At the same concentrations, Na2SO4 had stronger inhibitory effect on seed germination and seedling growth than NaCl. This study comprehensively compared germination traits of dimorphic seeds and seedling growth of S. salsa, and then developed a conceptual model to explain their adaptation to harsh saline environment.

RNA methylation and demethylation, which is mediated by RNA methyltransferases (referred to as “writers”) and demethylases (referred to as “erasers”), respectively, are emerging as a key regulatory process in plant development and stress responses. Although several studies have shown that AlkB homolog (ALKBH) proteins are potential RNA demethylases, the function of most ALKBHs is yet to be determined. The Arabidopsis thaliana genome contains thirteen genes encoding ALKBH proteins, the functions of which are largely unknown. In this study, we characterized the function of a potential eraser protein, ALKBH6 (At4g20350), during seed germination and seedling growth in Arabidopsis under abiotic stresses. The seeds of T-DNA insertion alkbh6 knockdown mutants germinated faster than the wild-type seeds under cold, salt, or abscisic acid (ABA) treatment conditions but not under dehydration stress conditions. Although no differences in seedling and root growth were observed between the alkbh6 mutant and wild-type under normal conditions, the alkbh6 mutant showed a much lower survival rate than the wild-type under salt, drought, or heat stress. Cotyledon greening of the alkbh6 mutants was much higher than that of the wild-type upon ABA application. Moreover, the transcript levels of ABA signaling-related genes, including ABI3 and ABI4, were down-regulated in the alkbh6 mutant compared to wild-type plants. Importantly, the ALKBH6 protein had an ability to bind to both m6A-labeled and m5C-labeled RNAs. Collectively, these results indicate that the potential eraser ALKBH6 plays important roles in seed germination, seedling growth, and survival of Arabidopsis under abiotic stresses.

Seeds of Radermarchera xylocarpa and Dolicandrone falcata were soaked overnight in water and germinated in different substrates. In both the plants, the highest percent of germination was recorded in cocopeat (Radermarchera xylocarpa:62%; Dolicandrone. falcata: 41%) followed by MS basal medium (Radermarchera xylocarpa: 46%; Dolicandrone falcata:20%), cocopeat: sand (Radermarchera xylocarpa:30%; Dolicandrone. falcata: 8%) and least being in filter paper (Radermarchera xylocarpa: 23%; Dolicandrone falcata: 0%).Prior to germination in optimised substrate, seeds were soaked overnight in distilled water (control) and in gibberllic acid (GA3) (5 & 10 ?M). In both the plants GA3 treated seeds failed to increase percent germination and the growth of seedlings. In control the seedling growth was better with respect to all the parameter for both the species. We determined whether the chilling process (endodormancy release) was similar in peach [Prunus persica (L.) Batch cv. Johnson Elberta] seeds, seedlings (near normal to physiologically dwarfed), and mature plants (cuttings) by comparing correlation coefficients of various growth measurements following similar chilling treatments. Seed germination (10 days after forcing at 20C) and seedling emergence (15 days after forcing in the greenhouse) correlated highly with leaf and shoot growth (56 days of growth) of seedlings and terminal shoot growth of cuttings (13 days after forcing). The correlations were higher for germination than for emergence. Initial (first season) seedling growth correlated poorly with germination, emergence, budbreak, and growth of seedlings (second season) and shoot growth of cuttings. Budbreak and growth of seedlings correlated highly with shoot growth of cuttings. The abnormal leaf problem, which can cause apex abortion (common with initial seedling growth), confounded correlations with initial seedling growth. Yet, the abnormal leaf problem did not hinder correlations with the second seasons growth. Good relationships between the chilling mechanisms that promoted germination, emergence, budbreak, and growth of seedlings and shoot growth of cuttings existed, but were dependent on what was measured and when the measurement was taken. Germination (forced at 20C) was the most accurate indicator of the seed chilling status for comparisons with the responses of the other propagules.
