ABSTRACT The protozoa Cryptosporidium spp. and Giardia spp. are etiological agents responsible for the transmission of gastroenteritis, mainly due to the consumption of contaminated water. Their (oo)cysts are resistant to adverse environmental conditions, as well as to most conventional water treatment processes. Thus, the objective of this work was to evaluate the occurrence of (oo)cysts of these protozoans in surface water collected for human consumption in the state of Goiás, Brazil. Fifteen samples of raw water were collected to assess the occurrence of (oo)cysts of the protozoa using the Membrane Filtration method, in addition to turbidity and pH analyses. Recovery rates in tests with ultrapure water reached the USEPA (2012) criteria for Giardia (78.1% ± 0%) and for Cryptosporidium (60.6% ± 32.6%); however, recovery in raw water was lower due to turbidity. All samples (n = 15) were positive for Cryptosporidium spp. oocysts, with a maximum concentration of 250 oocysts/L, demonstrating that these protozoa are disseminated in the aquatic environment of the state of Goiás and pose a risk to public health. Due to the use of water sources for public consumption, it is recommended that public authorities and sanitation companies act to preserve and maintain water courses, carry out periodic monitoring of treatment plants that supply the Cerrado, Santana and São Manoel streams and improve existing treatment technologies. The results did not allow to infer whether animal load and grazing area promote an increase in contamination of the lotic aquatic systems.

Surface water contaminated by domestic sewage discharges is a potential source of pathogens, including protozoa. During 2005–2006, the source water (Atibaia River) of the Surface Water Treatment Plant (WTP) of Campinas city, São Paulo, Brazil was sampled to obtain an assessment of Cryptosporidium oocyst and Giardia cyst concentrations. Calcium carbonate flocculation (CCF) and membrane filtration (MF) concentration techniques, with and without purification by immunomagnetic separation (IMS) were evaluated. The cysts and oocysts were detected by immunofluorescence assay (IFA) and confirmed by differential interference contrast (DIC). Membrane filtration method generally produced higher recovery efficiency. Giardia spp. was detected in 87.5% of the water samples analyzed with densities ranging from 2.5 to 120 cysts per L. Cryptosporidium spp were detected in 62.5% and the concentrations ranged from 15 to 60 oocysts per L. Cryptosporidium oocyst and Giardia cyst concentrations detected in this study were elevated and are associated with discharge of untreated sewage in Atibaia River. Measures should be taken to protect surface water from sources of contamination.

The purpose of this study was to investigate the occurrence of Cryptosporidium spp. and Giardia spp. in a public water-treatment system. Samples of raw and treated water were collected and concentrated using the membrane filtration technique. Direct Immunofluorescence Test was performed on the samples. DNA extraction using a commercial kit was performed and the DNA extracted was submitted to a nested-PCR reaction (n-PCR) and sequencing. In the immunofluorescence, 2/24 (8.33%) samples of raw water were positive for Giardia spp.. In n-PCR and sequencing, 2/24 (8.33%) samples of raw water were positive for Giardia spp., and 2/24 (8.33%) samples were positive for Cryptosporidium spp.. The sequencing showed Cryptosporidium parvum and Giardia duodenalis DNA. In raw water, there was moderate correlation among turbidity, color and Cryptosporidium spp. and between turbidity and Giardia spp.. The presence of these protozoans in the water indicates the need for monitoring for water-treatment companies.

Introduction. In order to standardize the quality indicators during the expected shelf life of the developed drug naltrexone hydrochloride in the form of a nasal spray containing a high concentration of poloxamer and benzalkonium chloride as a preservative, a microbiological study was carried out. The possibility of using the membrane filtration method for testing prototypes recommended by the State Pharmacopoeia XIV.Aim. Study and selection of test conditions for the «microbiological purity» indicator of nasal spray samples containing naltrexone hydrochloride.Materials and methods. As an object of research, a naltrexone hydrochloride nasal spray was used. When analyzing the microbiological purity, the membrane filtration method recommended in the State Pharmacopoeia XIV.Results and discussion. As part of the study, it was found that the samples of the drug meet the requirements for a microbiological indicator for drugs of category 2. Testing the suitability of the method for samples of the dosage form showed that the antimicrobial effect of the drug was completely removed by washing the filter, which was proved by inoculation of indicator test microorganisms, the quantitative and qualitative nature of growth, which did not differ from the control without the drug.Conclusion. As a result of the studies carried out, the optimal test conditions for the «Microbiological purity» indicator for the nasal spray containing naltrexone hydrochloride were selected and substantiated.

The aim of this study was to evaluate the use of total coliforms (TC) and faecal coliforms (FC) using a membrane filtration method for precise monitoring of faecal pollution in Korean surface water. The samples were collected in Korea from both main rivers and their tributaries. Presumptive TC · FC were enumerated. The ratios of presumptive FC to TC were not constant, but varied widely, and TC were difficult to enumerate because of overgrowth by background colonies. For FC this was not the case. Seven hundred and three purified strains of presumptive TC · FC and their background colonies were biotyped using API 20E. Among 272 presumptive TC, non-faecal related species, Aeromonas hydrophila dominated (34.6%) and E. coli accounted for only 5.1%. In contrast, E. coli made up 89% of the 209 presumptive FC. Furthermore, of 164 background colonies on Endo Agar LES, 54.9% was A. hydrophila, while background colonies on m-FC Agar were few (58 strains), and despite their atypical colony appearance, most of them were biotyped as enteric bacteria. These results reveal that the detection of FC rather than TC using m-FC Agar is more appropriate for faecal pollution monitoring in eutrophicated surface water located in a temperate region.

The purpose of the article. The article considers the Blockchain technology asan innovative tool. In particular, the essence and background of the developmentof blocks, the principles and specifics of the functioning of the system, as well asthe scheme of its work, are determined. The article presents the prospects forusingdistributed registry technologies (blockchain) in various socioeconomic spheresrelated to state administration. Provided examples and forecasts of the use ofblockchain technologies in the provision of state and municipal services forindividuals and legal entities in the following areas: formation of a unified registercontaining the history of the placement of the state, municipal order, as well asprocurement of corporations with state participation and / or control; registers ofdocuments (diplomas, certificates, lost and disavowed passports, policies for movableand immovable property insurance, health, etc.); database of court decisions andexecutive proceedings; public participation portals for citizens of Ukraine district- city – country. The fact that the blockchain technology is, first of all, theprinciples, and not the only possible way of implementing them, allows us to counton maximum openness and multivariate application in a dynamically changingchanging«digital world». Methodology. The research methodology is to use a combinationof methods: analytical, historical, comparative. The scientific novelty. The priorityof state blockchain systems introduction in stationary and distant voting, distributeddocument circulation, medical data registration, land resources registration,electronic auctions (auctions) in Ukraine was grounded. Conclusions. Already today,blockchain systems can change the role and participation of citizens in the conductof the state-management process, by raising the responsibility level, from thetransparent will expression in the elections to regulating the government serviceactivity in the society’s digitization conditions. The main advantages blockchainsystems using by public authorities that will increase the level of citizens trust todigital technologies using in general, namely: reliability and reliability of datastorage, transparency of transactions and virtually absolute protection of informationfrom distortion and unauthorized removal (relocation), are determined. In furtherscientific research it is proposed to consider the promising areas of the blockchaindigital technology usage: service activities of public authorities, legal proceedings,property rights management, implementation of migration control, verification ofgoods and services, registration of data on passing qualifying tests, patenting,intellectual property, digital identification, logistics , taxation, accounting ofbudget funds movement.

The article studies documents from the State Archive of the Orel Region (GAOO) as an important source for studying the sense of justice of the Oryol gubernia peasants in early 20th century. Introduction of new archival materials allows to flesh out our knowledge and to produce a true-to-life picture of the Oryol peasants’ way of life. The peasant origins of the majority of the population necessitate a comprehensive study of peasant legal consciousness. Historical legacy is pertinent to present day, and forgetting its lessons is fraught with consequences. Evolution of modern Russian statehood hedges on its historical and legal traditions. The article studies documents in the fonds of public authorities, police, gendarmerie, courts, and prosecution offices. Introduction of new materials of public authorities, police, gendarmerie, courts, and prosecution offices into the scholarship promotes the analysis of the evolution of peasant legal sense in early 20th century. The chronological framework of the article is limited to the period from 1900 to 1917, its territorial framework is limited to the Oryol gubernia in its pre-revolutionary borders. The article studies reports, dispatches, and circular letters using the comparative method. The intensification of peasant protest was incidental to the first Russian revolution of 1905-1907 – the peasants hoped to force the government to settle the agrarian question, wherein lay the crux of their interests. As peasants of the Oryol gubernia suffered from shortage of arable land, antimonarchical sentiments gained momentum and translated a growing number of trials for contempt of the Emperor. Illegal literature spreading among the peasants, further radicalized them, and the authorities grew more and more hesitant in their assessment of peasant loyalty, which is quite intelligible in the archival documents. Thus, the use of new archival documents in addition to published materials promotes the scholarship on the peasant legal sense.

The provision of water services has traditionally been considered a responsibility of the state. During the late 1980s, the private sector emerged as a key actor in the provision of public services. Mexico City was no exception to this trend and public authorities awarded service contracts to four private consortia in 1993. Through consideration of this case study, two main questions arise: First, why do public authorities establish partnerships with the private sector? Second, what are the implications of these partnerships for water governance? This article focuses, on the one hand, on the conceptual debate of water as a public and/or private good, while identifying new trends and strategies carried out by private operators. On the other hand, it analyzes the role of the state and its relationships with other actors through a governance model characterized by partnerships and multilevel networks.Spanish La provisión del servicio del agua ha sido tradicionalmente considerada como una responsabilidad del Estado. A finales de la década de 1980, el sector privado emerge como un actor clave en el suministro de servicios públicos. La ciudad de México no escapa a esta tendencia y en 1993 las autoridades públicas firman contratos de servicios con cuatro consorcios privados. A través de este estudio de caso, dos preguntas son planteadas: ¿Por qué las autoridades públicas establecen partenariados con el sector privado? ¿Cuáles son las implicaciones de dichos partenariados en la gobernanza del agua? Este artículo aborda por una parte, el debate conceptual del agua como bien público y/o privado, identificando nuevas tendencias y estrategias de los operadores privados. The City of Baltimore, Maryland is one of many US cities faced with challenges related to increasing potable water demands, diminishing fresh water supplies, and aging infrastructure. To address these challenges, the City recently undertook a $7M study to evaluate water supply and treatment alternatives and develop the conceptual design for a new 120 million gallon per day (MGD) water treatment plant. As part of this study, an innovative raw water management tool was constructed to help model source water availability and predicted water quality based on integration of a new and more challenging surface water supply. A rigorous decision-making approach was then used to screen and select appropriate treatment processes. Short-listed treatment strategies were demonstrated through a year-long pilot study, and process design criteria were collected in order to assess capital and operational costs for the full-scale plant. Ultimately the City chose a treatment scheme that includes low-pressure membrane filtration and post-filter GAC adsorption, allowing for consistent finished water quality irrespective of which raw water supply is being used. The conceptual design includes several progressive concepts, which will: 1) alleviate treatment limitations at the City's existing plants by providing additional pre-clarification facilities at the new plant; and 2) take advantage of site conditions to design and operate the submerged membrane system by gravity-induced siphon, saving the City significant capital and operations and maintenance (O&M) costs. Once completed, the new Fullerton Water Filtration Plant (WFP) will be the largest low-pressure membrane plant in North America, and the largest gravity-siphon design in the world.

Nationwide surveys of 1,4-dioxane and methyl-t-butyl ether (MTBE) levels in raw water used for the drinking water supply were conducted at 91 water treatment plants in Japan in 2001 and 2002, prior to the revision of the drinking water quality standards. 1,4-dioxane was widely and continuously detected in raw water samples and its occurrence was more frequent and its concentrations higher in groundwater than in surface water. However, its maximum concentration in raw water was much lower than its new standard value (50 μg/L), which was determined as a level of 10−5 excessive cancer risk to humans. Trace levels of MTBE were also detected in several surface water samples.
