Introduction. In order to standardize the quality indicators during the expected shelf life of the developed drug naltrexone hydrochloride in the form of a nasal spray containing a high concentration of poloxamer and benzalkonium chloride as a preservative, a microbiological study was carried out. The possibility of using the membrane filtration method for testing prototypes recommended by the State Pharmacopoeia XIV.Aim. Study and selection of test conditions for the «microbiological purity» indicator of nasal spray samples containing naltrexone hydrochloride.Materials and methods. As an object of research, a naltrexone hydrochloride nasal spray was used. When analyzing the microbiological purity, the membrane filtration method recommended in the State Pharmacopoeia XIV.Results and discussion. As part of the study, it was found that the samples of the drug meet the requirements for a microbiological indicator for drugs of category 2. Testing the suitability of the method for samples of the dosage form showed that the antimicrobial effect of the drug was completely removed by washing the filter, which was proved by inoculation of indicator test microorganisms, the quantitative and qualitative nature of growth, which did not differ from the control without the drug.Conclusion. As a result of the studies carried out, the optimal test conditions for the «Microbiological purity» indicator for the nasal spray containing naltrexone hydrochloride were selected and substantiated.

Objective: The main objective of this research was to evaluate the growth parameters and sanitary quality of Solanum nigrum L. irrigated with polluted water from Avo’o river in Nomayos. Methods: Faecal sludge, water from Avo’o River and Solanum nigrum L., were analysed with respect to microbiological and parasitological parameters. Four points (S0: Control site, located in Mbankomo at 3 km away from faecal sludge discharge area; S1: located at 810 m before the faecal sludge discharge area; S2: located at 100 m away from the faecal sludge discharge area; and S3: located at 350 m after the faecal sludge discharge area) were considered for sampling events. For faecal indicator (FC: Faecal coliforms and FS: Faecal Streptococci), the membrane filtration method was used. Helminthes eggs were determined using the method of Bailenger, modified by Rodier. The growth parameters of S. nigrum L., measured in different site mentioned above, were plant height, number, length and width of fresh leaves. Results: High concentration of FC (6857 ± 5180) and FS (2500 ± 1876 CFU/100 ml), as well as helminths eggs (273 ± 186 eggs/L) were obtained in Avo’o water samples. The strong correlations (r=0.9) clearly show that the waters of the Avo’o stream influence the growth of S. nigrum L. Conclusion: Despite the best growth of Solanum Nigrum L. irrigated with water from Avo’o River, high concentration of parasites was obtained on their leaves (43 eggs/100 g) highlight the necessity of a better management of faecal sludge in this city.

Introduction. Naltrexone, an antagonist of µ-opioid receptors, is promising for the treatment of various autoimmune and oncological diseases when used in doses of 1.5–5 mg/day. To date, there are no medications that provide such dosages of naltrexone.Aim. Development and validation of a method for the quantitative determination of naltrexone hydrochloride in a nasal spray by high performance liquid chromatography (HPLC).Materials and methods. As an object of research, a naltrexone hydrochloride nasal spray was used. The quantitative determination of naltrexone in the test sample was developed using a Dionex UltiMate 3000 high-performance liquid chromatograph (Thermo Scientific, USA) equipped with a diode-matrix detector.Results and discussion. The possibility of using isocratic and gradient chromatographic modes for the quantitative determination of naltrexone hydrochloride in the nasal spray was studied. Based on these results, a new method of determination using the gradient mode is proposed, which allows minimizing the influence of the polymer component in the test sample on the analysis results.Conclusion. A new technique of high-performance liquid chromatography (HPLC) is proposed that allows identification and quantification of naltrexone hydrochloride in a nasal spray containing a high concentration of water-soluble heat-sensitive poloxamer as a thickener. The developed method was validated according to the parameters: correctness, precision, specificity, linearity.

ABSTRACT The protozoa Cryptosporidium spp. and Giardia spp. are etiological agents responsible for the transmission of gastroenteritis, mainly due to the consumption of contaminated water. Their (oo)cysts are resistant to adverse environmental conditions, as well as to most conventional water treatment processes. Thus, the objective of this work was to evaluate the occurrence of (oo)cysts of these protozoans in surface water collected for human consumption in the state of Goiás, Brazil. Fifteen samples of raw water were collected to assess the occurrence of (oo)cysts of the protozoa using the Membrane Filtration method, in addition to turbidity and pH analyses. Recovery rates in tests with ultrapure water reached the USEPA (2012) criteria for Giardia (78.1% ± 0%) and for Cryptosporidium (60.6% ± 32.6%); however, recovery in raw water was lower due to turbidity. All samples (n = 15) were positive for Cryptosporidium spp. oocysts, with a maximum concentration of 250 oocysts/L, demonstrating that these protozoa are disseminated in the aquatic environment of the state of Goiás and pose a risk to public health. Due to the use of water sources for public consumption, it is recommended that public authorities and sanitation companies act to preserve and maintain water courses, carry out periodic monitoring of treatment plants that supply the Cerrado, Santana and São Manoel streams and improve existing treatment technologies. The results did not allow to infer whether animal load and grazing area promote an increase in contamination of the lotic aquatic systems.

A fluorogenic β-glucuronidase assay comprising membrane filtration followed by selective enumeration on m-FC agar at 44.5°C and further confirmation using tlie 4-metliylumbelliferyl-β-D-glucuronide (MUG) containing medium was evaluated for the detection of Escherichia coli in water. A total of 200 typical blue and non-typical blue colonies were isolated from sea and fresh water samples using initial selective enumeration on m-FC agar. Pure cultures of the selected colonies were further tested using the MUG assay and identified using the API 20E method. Of the colonies tested which were shown to be positive using the MUG assay 99.4% were Escherichia coli. The results of this study indicate the combination of the m-FC method followed by the MUG assay to be highly efficient for the selection and confirmation of E. coli from a wide range of environmental waters.

AbstractOn the basis of investigations with samples from a fertilization trial started in 1980, some parameters of food storage ability are evaluated. Microbial infestation of the product during incubation seems to be the most reliable parameter, but the circumstances of infestation and the optimal test conditions are unknown. There are no reliable correlations among the results of degradation tests, storage tests under optimal conditions, and chemical contents of the product. The concept of product vitality (a product-oriented quality referring to a product full of vigor) is based on results of degradation tests. Ultimately, however, food quality standards are based on human priorities (human-oriented). Therefore, not only product characteristics, but also social, psychological, and environmental criteria should be considered.

Surface water samples in Vietnam were collected from the Saigon River, rural and suburban canals, and urban runoff canals in Ho Chi Minh City, Vietnam, and were processed to enumerate Escherichia coli. Quantification was done through membrane filtration and quantitative real-time polymerase chain reaction (PCR). Mean log colony-forming unit (CFU)/100 ml E. coli counts in the dry season for river/suburban canals and urban canals were log 2.8 and 3.7, respectively, using a membrane filtration method, while using Taqman quantitative real-time PCR they were log 2.4 and 2.8 for river/suburban canals and urban canals, respectively. For the wet season, data determined by the membrane filtration method in river/suburban canals and urban canals samples had mean counts of log 3.7 and 4.1, respectively. While mean log CFU/100 ml counts in the wet season using quantitative PCR were log 3 and 2, respectively. Additionally, the urban canal samples were significantly lower than those determined by conventional culture methods for the wet season. These results show that while quantitative real-time PCR can be used to determine levels of fecal indicator bacteria in surface waters, there are some limitations to its application and it may be impacted by sources of runoff based on surveyed samples.

The reuse of treated wastewater to augment natural drinking water supplies is receiving serious consideration. Treatment of secondary and tertiary effluent by membrane filtration was investigated by assessing nanofiltration (NF) membrane and ultrafiltration (UF) membranes in bench-scale experiments. It was found that secondary and tertiary effluent contained high concentration of effluent organic matter (EfOM), contributing EfOM-related fouling. Flux decline and EfOM rejection tests were evaluated, using a dead-end stirred cell filtration unit. Surface charge and molecular weight cut-off (MWCO) of membranes were significant factors in membrane performance including permeability and EfOM-rejection.

Fifteen stations, in two estuaries, along the Northumberland Strait of Nova Scotia were examined between June and September 1981 for a relationship between the concentrations of commonly monitored fecal indicator bacteria and the potential pathogens Candida albicans, Pseudomonas aeruginosa, and Vibrio parahaemolyticus. Increased densities of these three organisms were usually associated with high densities of indicator bacteria. Whereas C. albicans and P. aeruginosa occur in human fecal wastes, V. parahaemolyticus is indigenous to the marine environment and positively responds to elevated nutrient levels in sewage. There is also some evidence that these bacteria survive as long or longer in marine waters than the common indicator bacteria. While membrane-filtration techniques for the enumeration of C. albicans and P. aeruginosa proved satisfactory, a V. parahaemolyticus membrane-filtration method lacked specificity and was supplemented by a most-probable-number method. In marine recreational and shellfish waters, these three organisms could complement fecal coliforms and fecal streptococci as indicators of human fecal contamination.
