The objective of this research was to verify the presence of spermatic abnormalities on semen of Brycon orbignyanus after cryopreservation. Semen was collected from ten four-year-old males who presented secondary reproductive characteristics for migrating fish. Sperm was evaluated for motility, vigor and spermatic morphology before and after cryopreservation. A cryoprotectant solution was made of 20 mL of yolk egg, 5.0 g of glucose and dimethyl sulfoxide diluted in distilled water (10 mL: 90 mL). The diluted semen (1:3, semen:solution) was submitted to nitrogen steam for 24 hours and then to liquid nitrogen (-196 ºC) for 60 days. Cryopreservation decreased the percentage of normal spermatozoa from 62.20% to 54.60%. Consequently, the percentage of spermatozoa with secondary abnormalities increased from 8.50% to 15.00%. However, there was no difference in primary abnormalities. Both spermatic motility and vigor were decreased in cryopreserved semen compared with fresh semen. In conclusion, cryopreservation of semen of B. orbignyanus increased the percentage of secondary abnormalities and decreased the spermatic motility and vigor.

Results are reported on the fertilization (to 18%) of steelhead trout (Salmo gairdneri) eggs with cryo-preserved sperm. Fresh semen was progressively diluted to 1: 4 with an extender containing 5 or 12.5% dimethyl sulfoxide, sealed in 1.0-ml ampoules, frozen in the vapor of liquid nitrogen, and subsequently thawed in a 4 C water bath. The fertilized eggs and alevins produced appeared normal when compared with those from fresh spermatozoa.

The objective of this study was to evaluate the effects of two cryoprotectants followed by thawing on the physiological potential of Genipa americana L. seeds. Two experiments testing 12 treatments were conducted, one for each cryoprotectant, both in a factorial scheme of 6 × 2 (cryoprotectant concentrations × thawing methods). We tested 0, 5, 10, 15, 20, and 25% for dimethyl sulfoxide, and 0, 0.25, 0.5, 0.75, 1.0, and 1.25 M for sucrose. The thawing methods were fast (38 °C for 30 min) and slow (25 °C for 4 h). The seeds were immersed in the cryoprotectant solution for 3 h, stored for 120 h, and then thawed. The seeds were then sown in substrate (sand and vermiculite, 1:1). Emergence percentage along with speed index, length, fresh and dry matter mass of seedlings were evaluated. Dimethyl sulfoxide and sucrose can be used as cryoprotectants in G. americana seeds. Thawing should be slow when treating seeds with dimethyl sulfoxide.

This paper summarizes historical and current attempts to cryopreserve the gametes of fish, and suggests future uses of cryogenics in fishery biology. Particular attention is given to the evaluation of procedures for collecting, freezing, thawing, and testing spermatozoa. Best methods entail manual stripping for semen, rapid freezing in the vapor of liquid nitrogen, rapid thawing of sperm cells, and fertilization tests with fresh ova. Assessment of the cryoprotective influence of common constituents of fish sperm extenders and life protectors is given. Based on current studies, a simple extender containing two salts and lecithin seems as adequate as more complex diluents. Dimethyl sulfoxide repeatedly provides best protection of salmomd spermatozoa from damage by freezing. Considering current developments, recommendations are made for improving cryoprotective procedures. The paper concludes with speculation on cryopreserving eggs, and on possible areas for further research.

SummaryHuman platelets frozen at −195° C (liquid nitrogen) retain their morphological integrity and ability to promote clot retraction when 5% dimethyl-sulfoxide and 5% dextrose are added to the suspending plasma medium. Slow freezing was more effective than direct immersion in the liquid nitrogen. Although similar results may be achieved with dimethylsulfoxide alone with rigidly controlled freezing rates, the addition of sugars may permit freezing under less critical conditions.Dimethylsulfoxyd und 5% Dextrose dem Plasmamilieu hinzugefügt werden. Das langsame Einfrieren ist effektiver als das direkte Eintauchen in flüssigen Stickstoff. Alternaria dauci (J.G. Kühn) J.W. Groves et Skolko and A. radicina Meier, Drechsler et E.D. Eddy are important seed-transmitted pathogens of carrot. The aim of this study was to evaluate the effects of acetic acid treatments on the quality of stored carrot seeds. Seeds of two samples were soaked for 30 min in 0.5, 1 and 2% acetic acid. Controls included untreated seeds, seeds soaked in distilled water and seeds treated with fungicide Zaprawa Nasienna T 75 WS/DS (a.i. thiram 75%). Germination, vigour and health of untreated and treated seeds were evaluated before and after 5 and 12 months of storage at 4 and 20 °C. Seeds of both samples treated with 0.5 and 1% acetic acid were characterized by higher germination capacity after storage than untreated seeds. However, treatments with 1 and 2% acetic acid negatively affected seed vigour. Generally, seeds of both samples treated with acetic acid were characterized by lower infestation with A. alternata and A. radicina after storage than untreated seeds and seeds soaked in distilled water. Moreover, acetic acid often controlled these fungi more effectively than the fungicide. Regardless of the storage duration, infestation with fungi was higher if seeds of both samples were stored at a lower temperature.

Background: New methodologies have been developed seeking to maximize pregnancy rate in female dogs created in commercial kennels, and also in order to maintain the quality of canine semen after dilution, refrigeration or freezing. One of the main factors that generate damage to sperm is oxidative stress, to minimize sperm damage, selenium and antioxidants like vitamin E are administered, by oral administration, seeking to improve the quality of semen. The objective was to study the effect of vitamin E and selenium, by oral administration, in the quality of fresh, refrigerated and frozen semen in adult dogs French Bulldog breed.Materials, Methods & Results: Semen samples were collected from 5 adult dogs, French Bulldog breed, being 2 semen drawing before the daily oral supplementation with vitamin E and selenium (ESE®) and semen drawing at 20, 40 and 60 days after the beginning of oral supplement. The ejaculated samples were diluted in TRIS - fructose citric acid (3.28 g TRIS-hydroxy-methyl-amino-methane, 1.78 g of citric acid monohydrate and 1.25 g of D - fructose, dissolved in 100 mL of distilled water and added of 20% egg yolk and 6% of glycerol. The characteristics evaluated in fresh semen were: volume (mL), color, appearance, concentration (x106 / mL), sperm motility (%), sperm strength (1 to 5) and morphology (%). For refrigerated and frozen semen were analyzed: sperm motility (%), sperm strength (1-5) and morphology (%). Diluted semen samples were centrifuged at: 1500 g/10 min and “pellets” formed by sperm of each ejaculated, detached from the tube wall were diluted homogeneously in the diluent TRIS type up to the final volume of 1.5 mL. After that, packaged in 0.5 mL French straws, kept under refrigeration at 5ºC/4 h, placed in nitrogen vapor at -120ºC/15 min, and dipped in liquid nitrogen at -196ºC and then stored on identified rachis and stored in liquid nitrogen container until the time of thawing in  water bath at 37°C/30 s for semen microscopic analysis. Data from fresh, refrigerated and frozen semen were statistically analyzed by analysis of variance and the average compared by 5% of Tukey test. Fresh semen sperm concentration differed (P < 0.05) between the samples, rising after 40 days after the beginning of oral supplementation with selenium and vitamin E. For the spermatic strength, better score (P < 0.05) was observed at collection 4, in 40 days after the beginning of oral supplementation to dogs. For fresh and refrigerated semen, the total defects, defects of head, acrosome and tail did not differ (P > 0.05) between the samples. Total sperm defects and minor head and tail defects did not differ (P > 0.05) between the samples in post-thawing. Regarding the acrosome defects after thawing, there was a significant reduction (P < 0.05) in samples performed 40 and 60 days after the beginning of oral supplementation with selenium and vitamin E.Discussion: Attention should be paid for what purpose the extenders within the refrigeration or freezing biotech will be used. The managed supplement, by oral administration, containing selenium and vitamin E, influenced beneficially raising the sperm concentration in fresh semen and decreasing the acrosome defects in frozen semen. Oral administration of supplementation with selenium and vitamin E is recommended for improving the quality of fresh and frozen semen in dogs.

Tambaqui (Colossoma macropomum) is a native freshwater fish that is of great importance for Brazilian aquaculture. Because of this importance, several techniques have been developed to improve the reproduction of this species in captivity. One of these techniques is the cryopreservation of sperm. In an effort to increase the efficiency of cryopreservation protocols, researchers have tried to determine suitable diluting solutions and freezing methods, which will provide a better post-thaw sperm quality. Thus, this study aimed to evaluate the efficiency of different diluents and freezing methods for the cryopreservation of tambaqui (C. macropomum) sperm. Samples of fresh semen were diluted in different treatments (Glucose 5% + 10% Dimethyl sulfoxide – DMSO, Glucose 5% + 10% Methyl glycol – MG, BTS + 10% DMSO and BTS + 10% MG) at a 1:9 dilution rate and frozen in a programmed freezing machine and a dry shipper. The semen samples were thawed and evaluated for vitality, sperm morphology and kinetics. Cryopreserved semen with DMSO and using the programmed freezing machine provided a greater percentage of motile sperm (15.44 ± 1.04%) after thawing compared to the dry shipper (3.99 ± 0.55%), regardless of the diluent. Additionally, DMSO showed better sperm velocities than MG regardless of the freezing method and the extender employed. A higher percentage of living spermatozoa was observed when glucose (37.28 ± 1.32%) (regardless of the freezing method and cryoprotectant) and DMSO (37.98 ± 1.25%) was used in the programmed freezing machine. For morphology, a greater amount of normal spermatozoa (46.10 ± 1.82%) was observed when the semen was cryopreserved using a freezing machine programmed with DMSO as the cryoprotectant and Glucose or BTS (38.16 ± 1.9% and 39.26 ± 1.87%, respectively) as extenders. Therefore, we suggest the use of the DMSO (10%) cryoprotectant in association with the Glucose (5%) extended in the programmed freezing machine for cryopreservation of C. macropomum semen.

In this field, complex disinfectants were contrasted using effective concentrations against bacteria and spore-forming microorganisms. A significant problem for the owners is the emergence of resistance of strains of microorganisms in the existing production and the same disinfectants. Investigation of trusted working concentrations of disinfectants that do not have a corrosive effect on metal structures. According to the results of research and production, contrast is an effective disinfectant for reducing E. coli and S aureus at 0,3 – 0,5% concentration at exposure for 30 min. and consumption of 100 - 400 cm3 / m2. Laboratory researches were carried out in laboratories of microbiological faculties of veterinary medicine of Sumy National Agrarian University. Disappearance gaps and disinfection on the market in Kyiv. Samples were drawn in the refrigerators from the walls, ceilings and floors. Refrigerated chambers made of stainless steel are made up of rubber and plastic elements. Metals are very vulnerable to corrosion with more concentrated acids and alkali. This was taken into account when choosing a disinfectant and its effective organizations. As a disinfectant used the drug contrast (manufacturer PE "Kronos Agro", Ukraine). The disinfectant was taken at a dose of 100 ml per 1 m2. To produce the culture was prepared basic products containing 1000 mg of the drug in 1 ml of distilled water. The experimental solutions were prepared for study with the main formulations developed. Representatives had extraordinary effects. The  disks were leaked through the disinfectant to obtain a zone of retention of high bacteria and fungi. The reliability of disinfectant destroying the micro-organisms of E. coli and S. aureus test cultures was also known. For the trusted production preparations, the contrast at the trusted enterprises was carried out by the rehabilitation of the premises of the educational laboratory of the Sumy National Agrarian University (premises for animals, dairies, refrigeration chambers). Renovation and test activities were performed at 0,1, 0,25 and 0,5 % of exposed contrast with exposure for 60, 30 and 10 min. the disinfectant was consumed 250 cm3 / m2. Upon completion of the studies, the investigated tests were investigated with distilled water (flow rate - 1000 cm3 / m2). Water that has been washed away test products, presented in specially prepared capabilities. Test results for this after disinfection were also investigated with distilled water, which was found on the disinfectant. To study the corrosion activity of the disinfectant used metal plates 1h1sm2. The metal samples were welded to the fifth mark after the commission before and after the study. 0,5; 1,0; 1,5; 2,5 % contrast solutions were used in the study. For comparison, in the experiments used 2% of productive. M. bovis mycobacterial cultures were grown on Pavlovsky's glycerol medium. The bacterial culture was transferred into sterile vials and sterile isotonic material containing 0,05 cm3 / mg was added. A large amount of delay zone in Petri dishes containing 0,5 % of S. aureus disinfectant, 3,5 times S. cholerase 1,8 S. Enteritidis 2 more than 0,5% formal form . Higher indicators of bacterial properties of the disinfectant contrast at a concentration of 0,5%.

1. The dose-response to acetylcholine has been examined in dorsal hand veins of healthy volunteers before and after removal of the endothelium. 2. Measurements were made in single dorsal hand veins during local infusions of acetylcholine. The vein was irrigated with distilled water to remove the endothelium. Dilator studies were performed in vessels preconstricted by a continuous infusion of noradrenaline. 3. In the endothelium-intact vessel the dose-response to acetylcholine was biphasic; low doses produced venodilatation with higher doses causing venoconstriction. 4. Dilatation to low doses of acetylcholine was abolished by prior irrigation with distilled water, consistent with denudation of the endothelium by this process. Irrigation augmented the constriction seen in response to higher doses of acetylcholine. 5. This is the first demonstration of an endothelium-dependent biphasic dose-response to acetylcholine in man. The results raise questions as to the possible physiological actions of endogenous acetylcholine and as to the use of the acetylcholine dose-response curve as a marker of endothelial function.
