FT-IR spectra were recorded of <i>Escherichia coli</i> cell mass with the aim of obtaining spectral traits possibly useful in a rapid detection and characterisation of this indicator bacterium. A well differentiated spectrum was obtained from the cell mass harvested in a stationary phase of growth, e.g., after 24 h, from a minimum nutrient broth. The cell mass, harvested either earlier or grown in nutrient solutions which contained an enhanced carbon or nitrogen concentrations delivered somewhat different IR spectra, apparently due to a higher content of nucleic acid components as related to other structural constituents of bacterial cells. Consequently, the FT-IR spectra of <i>E. coli</i>, although rather rapidly to collect, seem only capable of delivering useful and reproducible information if the cell mass is obtained under standardised cultural conditions.

The adsorption of 1,5-pentanedithiol (1,5-PDT) and 1,6-hexanedithiol (1,6-HDT) in liquid phases on NaA (or 4A-type), CaA (or 5A-type) and NaY zeolites has been studied by using infrared spectroscopy. From the IR spectra it is found that the peak positions of the symmetric as well as the antisymmetric modes of the methylene (CH2) groups are observed at almost the same band values for the title dithiolates adsorbed on the A-type and NaY zeolites. On the other hand, the weak SH stretching vibration, observed for all samples, can be attributed to the sulphure atoms of 1,5-PDT and 1,6-HDT coordinatively adsorbed on cationic sites of the zeolites.

Dynamic FT-IR spectra in the amide I region were measured for silk fibroin cast films. Phase analyses of the dynamic spectra were carried out in order to separate them into several components which are due to the secondary structures of fibroin. These observations showed that peak positions of the amide I bands were in good agreement with those obtained by Fourier self-deconvolution procedures. In conclusion, phase analysis of dynamic FT-IR spectra is a powerful technique for separation of highly overlapping bands.

The IR spectra of 1,3-diaminopropane adsorbed on NaA (type 4A), CaA (type 5A), NaX (type 13X) and NaY zeolites are reported. From the IR spectral data it can easily be stated that the characteristic NH vibration bands of aliphatic amine groups play an important role in the adsorptions of 1,3-diaminopropane on the mentioned zeolites.

Ganoderma lucidum is an important medicinal mushroom widely used in pharmaceuticals for their antiviral, antibacterial, antifungal, anticancer and immunoregulatory properties and also used in agriculture as an antiviral and antibacterial agent. Fourier Transform Infrared (FT-IR) spectroscopy is a tool widely used in the researches for the identification of organic compounds in the organism. In our study, we used FT-IR Spectroscopy for determination the chemical nature of Ganoderma lucidum, and their standards Squalene and Ganoderic acid A. In the FT-IR spectrum for the G. lucidum obtained in the region of 3782.69 cm-1 to 568.898 cm-1 the absorptions peak represents the alcohols, halogens, silicon and phosphorus groups present in the sample. In the squalene standard, ketones, amides, and guanidine, nitrile and azine stretches were found in the wavelength 1655-1550 cm-1. In the Ganoderic acid A sulfur compounds with weak stretching intensity were obtained in the wavelength 500-400 cm-1. We have concluded that FT-IR spectroscopy is an effective method to analyze the chemical nature of the organic groups present in the samples.

Антимикробные пептиды (АМП) системы врожденного иммунитета - соединения, играющие важную роль в патогенезе инфекционных заболеваний, так как обладают свойством инактивировать широкий спектр патогенных бактерий, обеспечивая противомикробную защиту живых организмов. В настоящее время АМП рассматриваются как потенциальные соединения-корректоры инфекционной патологии, вызываемой антибиотикорезистентными бактериями (АБР). Цель данной работы состояла в изученим механизмов антибактериального действия трех пептидов, принадлежащих к семейству бактенецинов - ChBac3.4, ChBac5 и mini-ChBac7.5Nb. Эти химически синтезированные пептиды являются аналогами природных пролин-богатых АМП, обнаруженных в лейкоцитах домашней козы Capra hircus и проявляющих высокую антимикробную активность, в том числе и в отношении грамотрицательных АБР. Методы. Минимальные ингибирующие и минимальные бактерицидные концентрации пептидов (МИК и МБК) определяли методом серийных разведений в жидкой питательной среде с последующим высевом на плотную питательную среду. Эффекты пептидов на проницаемость цитоплазматической мембраны бактерий для хромогенного маркера исследовали с использованием генетически модифицированного штамма Escherichia coli ML35p. Действие бактенецинов на метаболическую активность бактерий изучали с применением маркера резазурина. Результаты. Показано, что все исследованные пептиды проявляют высокую антимикробную активность в отношении Escherichia coli ML35p и антибиотикоустойчивых штаммов Escherichia coli ESBL и Acinetobacter baumannii in vitro, но их действие на бактериальные клетки разное. Использован комплекс методик, позволяющих наблюдать в режиме реального времени динамику действия бактенецинов в различных концентрациях (включая их МИК и МБК) на барьерную функцию цитоплазматической мембраны и на интенсивность метаболизма бактериальных клеток, что дало возможность выявить различия в характере воздействия бактенецинов, отличающихся по структуре молекулы, на исследуемые микроорганизмы. Установлено, что действие каждого из трех исследованных бактенецинов в бактерицидных концентрациях отличается по эффективности нарушения целостности бактериальных мембран и в скорости подавления метаболизма клеток. Заключение. Полученная информация дополнит существующие фундаментальные представления о механизмах действия пролин-богатых пептидов врожденного иммунитета, а также послужит основой для биотехнологических исследований, направленных на разработку на базе этих соединений новых антибиотических препаратов для коррекции инфекционных заболеваний, вызываемых АБР и являющимися причинами тяжелых внутрибольничных инфекций. Antimicrobial peptides (AMPs) of the innate immunity are compounds that play an important role in pathogenesis of infectious diseases due to their ability to inactivate a broad array of pathogenic bacteria, thereby providing anti-microbial host defense. AMPs are currently considered promising compounds for treatment of infectious diseases caused by antibiotic-resistant bacteria. The aim of this study was to investigate molecular mechanisms of the antibacterial action of three peptides from the bactenecin family, ChBac3.4, ChBac5, and mini-ChBac7.5Nb. These chemically synthesized peptides are analogues of natural proline-rich AMPs previously discovered by the authors of the present study in leukocytes of the domestic goat, Capra hircus. These peptides exhibit a high antimicrobial activity, in particular, against antibiotic-resistant gram-negative bacteria. Methods. Minimum inhibitory and minimum bactericidal concentrations of the peptides (MIC and MBC) were determined using the broth microdilution assay followed by subculturing on agar plates. Effects of the AMPs on bacterial cytoplasmic membrane permeability for a chromogenic marker were explored using a genetically modified strain, Escherichia coli ML35p. The effect of bactenecins on bacterial metabolic activity was studied using a resazurin marker. Results. All the studied peptides showed a high in vitro antimicrobial activity against Escherichia coli ML35p and antibiotic-resistant strains, Escherichia coli ESBL and Acinetobacter baumannii, but differed in features of their action on bacterial cells. The used combination of techniques allowed the real-time monitoring of effects of bactenecin at different concentrations (including their MIC and MBC) on the cell membrane barrier function and metabolic activity of bacteria. The differences in effects of these three structurally different bactenecins on the studied microorganisms implied that these peptides at bactericidal concentrations differed in their capability for disintegrating bacterial cell membranes and rate of inhibiting bacterial metabolism. Conclusion. The obtained information will supplement the existing basic concepts on mechanisms involved in effects of proline-rich peptides of the innate immunity. This information will also stimulate biotechnological research aimed at development of new antibiotics for treatment of infectious diseases, such as severe in-hospital infections, caused by antibiotic-resistant strains.

<p>This report is a continuation of previous research on the H<sub>2</sub>O<sub>2</sub>-mediated synthesis of melanin-like pigments. We synthesized and characterized L-DOPA-based pigments using air- or H<sub>2</sub>O<sub>2</sub>-mediated<sub> </sub>oxidation. We compared their physic-chemical properties and evaluated their capacity to affect the interleukin release from immune cells. The use of higher concentrations of H<sub>2</sub>O<sub>2</sub> resulted in melanin-like materials with a distinct chemical signature in their FT-IR spectra and a lighter color. All pigments enhanced the interleukin release from immune cells. The possibility that lighter-colored melanins can be generated is discussed in the context of the importance of melanin-based pigmentation in human physiology.</p>

Growth of optically transparent single crystals of thiourea succinic acid (TUSA) was grown successfully from aqueous solution by slow evaporation technique. The crystal structure was elucidated using the single crystal XRD. The various functional groups and the modes of vibrations were identified by FT-IR spectroscopic analysis. The optical absorption studies indicate that the optical transparency window is quite wide making its suitable for NLO applications. Thermal stability of the crown crystal carried out by TGA-DTA analysis.

AbstractVitreous humor (VH) is an alternative biological matrix with a great advantage of longer availability for analysis due to the lack of many enzymes. The use of VH in forensic toxicology may have an added benefit, however, this application requires rapid, simple, non-destructive, and relatively portable analytical analysis methods. These requirements may be met by Fourier transform infrared spectroscopy technique (FT-IR) equipped with attenuated total reflection accessory (ATR). FT-IR spectra of vitreous humor samples, deposited on glass slides, were collected and subsequent chemometric data analysis by means of Hierarchical Cluster Analysis and Principal Component Analysis was conducted. Differences between animal and human VH samples and human VH samples stored for diverse periods of time were detected. A kinetic study of changes in the VH composition up to 2 weeks showed the distinction of FT-IR spectra collected on the 1st and 14th day of storage. In addition, data obtained for the most recent human vitreous humor samples—collected 3 and 2 years before the study, presented successful discrimination of all time points studied. The method introduced was unable to detect mephedrone addition to VH in the concentration of 10 µg/cm3. Graphic abstract
