Experimental study on disorders in the activity of macrophages in the acute period of different severities of cerebral stroke

BACKGROUND: The development of immunosuppression in the acute phase of cerebral stroke may lead to infectious and inflammatory complications. However, data on the functional activity of macrophages are insufficient to understand the pathogenesis of stroke. AIM: This study aimed to investigate the functional activity of macrophages in the acute period of experimental cerebral stroke (ECS) of different severities. MATERIALS AND METHODS: In an experimental study on 45 Wistar rats, the functional and spontaneous metabolic activity of macrophages in the spleen and abdominal cavity and the induced metabolic activity of peritoneal macrophages were evaluated through the reduction of nitro blue tetrazolium via spectrophotometry. The spontaneous and induced adhesive activity and the inhibition of the modified adhesive activity of peritoneal macrophages were examined via crystal violet staining by using a colorimetric method. ECS was modeled by the rotational movements of mandrel-knife inserted into the region of capsula interna (c.i.) on the left by guiding a needlecannula with the subsequent damage of vessels and the formation of mild (local incision of vessels in the region of c.i.), moderate (additional introduction of autologous blood in the area of c.i.), and severe (cutting of vessels from c.i. to pia mater with the subsequent introduction of autologous blood into the damaged area) hemorrhage. Data were evaluated in 72 h after different severities of ECS were modeled. RESULTS: In 72 h after ECS with different severities was modeled, the functional and spontaneous metabolic activity of splenic macrophages significantly decreased compared with those in the control group. The functional, spontaneous, and induced metabolic activity of peritoneal macrophages significantly reduced compared with those in the control group. The adhesive activity of peritoneal macrophages significantly increased compared with that of the control group. These disorders exacerbated in animal models with a more severe degree of stroke. CONCLUSION: In the acute period of ECS, the functional activity of macrophages decreased, whereas their adhesive activity increased. These phenomena enhanced in more severe models of stroke.

The objective: to reveal the features of the parameters of the blood coagulation system in women in dynamics – from the acute period of ovarian apoplexy to the long-term postoperative period.Materials and methods. The study involved 82 women (main group, n=82), operated on for ovarian apoplexy, and 30 patients in the control group (n=30), who had no history of this pathology. All patients underwent a comprehensive examination using clinical, instrumental and laboratory research methods. The groups were homogeneous in age (average age was 26,1±2,3 years) and statistically comparable.Results. During the study, it was found that in the acute period of ovarian apoplexy, pathological changes were found in the plasma, vascular-platelet components and the fibrinolytic system of hemostasis, as evidenced by the lengthening of the thrombin time to 22,3±1,32 seconds, an increase in the concentration of fibrinogen to 6,73±0,64 g/l, SFMC up to 0,7±0,03 g/l and platelet hyperaggregation, stimulated by ADF, ristocetin and collagen, and reaching 80%. The extremely high content of D-dimer in the abdominal cavity in patients with ovarian apoplexy indicated increased thrombus formation in the damaged ovary, followed by activation of the fibrinolytic system and lysis of fibrin clots. Genetic thrombogenicity was found in general in 80,4% of patients, including those with a predominant lesion of the folate cycle enzymes - in every second patient.Positive dynamics with a tendency to normocoagulation was observed in the distant postoperative period. In the reference range were SFMC indices in 86,6% of cases, fibrinogen and thrombin time in 89,1% of cases, ristocetin-stimulated aggregatogram in 95,1% of cases (p<0,05).Conclusions. In women who underwent ovarian apoplexy, in the acute period of the disease, pathological changes in the vascular-platelet link of the hemostasis system were observed. The diagnosed thrombophilic disorders could lead to a deterioration in the rheological properties of blood, a violation of the processes of oxygenation and tissue trophism. The dynamic study of the hemostasis system in the acute and late postoperative periods of patients with ovarian apoplexy, as well as the data obtained on the prevalence of genetic thrombogenicity in the examined patients, should be taken into account when managing women from this risk group.

The aim of research was to reveal the dynamic changes of the level of excitatory and inhibitory neuroamino acids in patients with the primary cerebral ischemic stroke depending on postapoplectic spasticity presence at the end of the early recovery period. For this aim was studied the concentration of excitatory and inhibitory neuroamino acids in the blood plasma in first 72 hours in 97 patients with the primary ischemic cerebral stroke depending on postapoplectic spasticity on the sixth month after ischemic event. The control group included 15 patients with diagnosed chronic cerebral ischemia. In the result of research we revealed that the common sign for the two groups (with spasticity on the sixth month and without it) was the reliable rise of the level of excitatory amino acids comparing with the control. In patients without spasticity the heightened level of excitatory neurotransmitters in the most acute period of ischemic cerebral stroke was attended with the heightened level of inhibitory neuroamino acids. The distinctive feature of patients with postapoplectic spasticity was the decreased or stable level of transmitters of inhibitory action. During 6th moth after ischemic stroke was detected the rise of all studied neuroamino acids in patients with spasticity unlike to the ones without spasticity who were characterized only with the rise of taurine level and decrease of glycine and aspartate levels. So, the received results allow assume the insufficient activation of the inhibitory neuroamino acids system in the most acute period of the ischemic stroke in certain category of patients that in future are inclined to the spasticity development after stroke.

Ocular chemical burns are among the most critical and true ocular emergencies in dogs and cats, with destructive consequences on the ocular surface and intraocular tissues. Platelet alpha granules are an important reservoir of growth factors that can stimulate chemotaxis of inflammatory cells, mitosis, migration and differentiation of cells. On the other hand, tetracyclines and their derivatives exhibit non-antimicrobial properties, such as affecting inflammation, immunomodulation, cell proliferation and angiogenesis. The objective of this study is to investigate the effect of intrastromal platelet rich plasma (PRP) in combination with oral doxycycline in the healing process of the cornea. Thirty six New Zealand rabbits were used in this experimental study. Alkali burns were created by applying a round filter paper soaked in NaOH. PRP was prepared by centrifugation of autologous blood. The rabbits were allocated to four groups (9 animals each). Group 1 served as the control group without any treatment. Group 2 received doxycycline (5mg/kg b.w.) orally once a day throughout the study. Group 3 received an intrastromal injection of PRP (0.5ml) in the cornea, and group 4 received intrastromal PRP combined with oral doxycycline. Treatments had varied levels of success, with PRP combined with oral doxycycline producing the best results regarding corneal healing. Animals in the control group had epithelial defects for the whole duration of the study, and 3 animals presented corneal perforation. Groups 3 and 4 had smaller mean defect area, compared to groups 1 and 2. On the 7th day, neovascularisation was lower in treatment groups compared to the control group. Groups 3 and 4 also had less corneal oedema compared to the control group on day 3 and 7. Group 4 exhibited the best wound healing, with less neovascularization and better collagen arrangement, as shown by the histopathological evaluation. To the authors’ knowledge this is the first experimental study that intrastromal PRP is combined with oral doxycycline for the management of corneal chemical burns. This combination is a simple, safe and economical therapeutic approach that promotes corneal healing.

Актуальность. Изучение факторов риска развития инфекционно-воспалительных осложнений и путей их снижения при инсульте является актуальной неврологической проблемой. Цель: оценка функциональной активности клеток естественных киллеров в остром периоде экспериментального инсульта различной степени тяжести. Методы. В проведенном экспериментальном исследовании на животных (крысы линии Вистар, массой 200-220 г, n = 50) изучали функциональная активность естественных киллеров селезенки, при моделировании монополушарного левостороннего экспериментального инсульта (ЭИ) различной степени тяжести. Результаты. Показано, что у животных на 7-й день инсульта отмечается статистически значимое снижение функциональной активности естественных киллеров селезенки: ЭИ легкой степени - 20 (19; 23)%, р = 0,001, ЭИ средней степени - 19 (17; 21)%, р < 0,001, ЭИ тяжелой степени - 18 (16; 22)%, р < 0,001. Нарушения функциональной активности естественных киллеров более выражены на модели ЭИ средней и тяжёлой степени тяжести. Заключение. Показано, что при ЭИ наблюдается снижение функциональной активности естественных киллеров селезёнки, которое нарастает при увеличении степени тяжести инсульта. Background. Studying risk factors for development of infectious and inflammatory complications and ways to reduce them in stroke is a modern challenge to neurology. Aim: Evaluating the functional activity of natural killer (NK) cells in the acute period of experimental stroke (ES) of different severity. Methods. In this experimental study, the functional activity of spleen NK cells was studied in Wistar rats weighing 200-220 g (n = 50) with ES of different severity. Results. On the 7th day of stroke. the functional activity of spleen NK was significantly decreased: in mild ES, 20 (19; 23)%, р = 0.001, in moderate ES, 19 (17; 21)%, р <0.001, and in severe ES, (16; 22)%, р < 0.001. Disorders of the NK functional activity were more pronounced in moderate and severe ES. Conclusion. ES was associated with impaired functional activity of NK cells, which progressed with increasing grade of ES severity.

AbstractBackground Ascaris lumbricoides eggs had very thick walls consisted of three layers, they were albuminoid layer, hyaline layer and vitelin layer. These layers were impermeable causing the Ascaris lumbricoides eggs resistant to less supportive environmental factors. Formalin 10% was a preservative that was often used to preserve faeces containing parasites such as protozoa and worm eggs. However, without heating, formalin 10% was not effective to preserve the Ascaris lumbricoides eggs because they would keep developing to become infective (containing larvae). Objective This study aims to prove whether there is an effect of adding 10% formalin which is heated at 60 C, 70 C and 80 C to the development of Ascaris lumbricoides eggs.Methods The design of this research was an experimental study with The Randomized Posttest Control Group Design. Result The Ascaris lumbricoides eggs were still growing into infective eggs in faeces although they were heated by formalin 10% at 60C, 70C and 80C. conclusion Based on the results of the study, there was no effect of heating temperature of formalin 10% on the development of Ascaris lumbricoides egg.Keywords: Formalin 10%, Ascaris lumbricoides, Heating, Soil Transmitted Helminth

Spending colostrum in post-cesarean section mothers is difficult in the first week postpartum. Efforts to overcome colostrum disruption can use non-pharmacological methods, namely breast care and endorphin massage. This study aims to determine the expenditure of colostrum by giving breast care and endorphin massage to post-cesarean mothers. This research is a quasi-experimental study with a nonequivalent design posttest control group design. This research was carried out in the midwifery room of the second floor of Grand Medistra Lubuk Pakam Hospital during the June-July 2019 period. There were 48 participants selected using purposive sampling, with 18 assigned in breast care, endorphin massage and combination of breast care and endorphin massage. The time of spending colostrum expenditure is assessed every day while the amount of colostrum expenditure is assessed after intervening. Analysis of the data used is one-way Annova. The results showed that the combination of breast care and endorphin massage was the most effective action in the time of spending colostrum and the amount of colostrum with a value of p = 0,000. The combination of breast care and endorphin massage could be applied as an alternative therapy post-cesarean mothers

In recent years, serious games offer great opportunities for learning processes at schools. However, it is unclear whether this type of proposals can offer differentiated answers among the students according to their gender. In this context, the aim of this paper is to know the possible differences that occur in primary school classrooms according to gender, with serious games designed for the development of mathematical fluency, and to examine to what extent these games contribute to the overall school performance. We carried out a quasi-experimental study, including pretest and posttest, without control group and with several experimental groups, and the participation of 284 students from first to fourth grade. The results show that the software benefits boys and girls equally, compared to the previously followed methodology that benefited boys. A clear relation between the results achieved and the performance in the overall students’ grades has also been observed. The conclusions show the potential of serious games in school settings, and the opportunity to approach performance differences based on the gender.

Early childhood is a formative period during which healthy habits are developed, including proper hand hygiene practices. The aim of this quasi-experimental study was to determine the effectiveness of a 4-week series of educational sessions that consider the cognitive developmental stage of children on increasing their knowledge and promoting hand hygiene practices. The intervention group (n = 33) observed the hand hygiene program, whereas another group served as the waitlist control (n = 20). Creative activities were planned for the illustration of hand hygiene concepts in terms of “right moments”, “right steps”, and “right duration”. Hand sanitizer coverage was evaluated using a hand scanner. After the intervention, the experimental group had higher knowledge level toward hand hygiene than the control group (p < 0.001). Significant improvements in hand hygiene performance at the left palm and dorsum (p < 0.05), right palm (p < 0.05), and overall hand coverage (p < 0.05) were observed in the experimental group. The study demonstrated that the knowledge and proper hand hygiene (HH) practice of children can be positively influenced by the use of an age-appropriate education program. The results of this study have implications for school health educators and parents for promoting HH practices among children at home and at the school level.
