Background: The objective of the study was to scientifically investigate the oral hypoglycemic activity of Caesalpinia bonduc on Alloxan induced diabetic albino rats. To compare the hypoglycemic effect of Caesalpinia bonduc with that of the standard drug Glibenclamide used in the treatment of diabetes mellitus.Methods: Adult healthy albino rats of wister strain of either sex weighing 150-200gms were included in the study. The animals were divided into 4 groups namely control, diabetic control, standard and test groups with 6 animals in each group. Diabetes was chemically induced using alloxan to produce hyperglycemia in rats. Standard drug Glibenclamide suspended in gum acacia was administered for standard group. Test drug Caesalpinia bonduc was administered for test group. Morning around 9 a.m. blood glucose levels were recorded on 1st, 3rd, 7th, 14th, 21st, and 28th days.Results: The control group of rats showed no variation. The diabetic control rats showed consistent hyperglycemia. Comparing the test drug Caesalpinia bonduc to the standard drug Glibenclamide, the test drug was 1.38 times more efficacious than the standard.Conclusions: The alcoholic extract of Caesalpinia bonduc (seeds) has shown more anti diabetic activity by lowering the blood glucose levels in diabetic rats significantly. These findings suggest that hypoglycemic potential of the test compound Caesalpinia bonduc is promising and found to be more significant than the standard compound.

To evaluate the effect of telmisartan on blood glucose levels and blood lipid levels in streptozotocin induced diabetic rats. Eighteen Wistar albino rats weighing 150-200gms of either sex were randomly selected from the central animal facility, and divided into 3 groups. Diabetes was induced by injecting Streptozotocin intraperitonelly. The control group received 1% Gum acacia (oral), standard group received 0.5 mg/kg Glibenclamide (oral) and the test group received Telmisartan 7.2mg/kg body weight (oral) from 0-28 days respectively. Body weight of the individual rats were measured on the respective days before blood glucose estimation on 0, 1, 3, 7, 14, 21 & 28th day and fasting blood glucose was estimated by (ACCUCHECK) glucometer. Estimation of fasting lipid profile by lipid screening strips on 1st and 28th day. When compared to control the capillary blood glucose (CBG) levels in the Telmisartan group was less at all the intervals but comparable with that of standard drug Glibenclamide in Streptozotocin induced diabetic rats. Improved lipid profile was seen with the Telmisartan group when compared to control group in Streptozotocin induced diabetic rats. Hypoglycemic activity and improved lipid profile action was seen with Telmisartan group which is comparable to standard drug glibenclamide in streptozotocin induced diabetic albino rats.

.The main purpose of present study was to perform phytochemical screening and explore the anti-hyperglycaemic properties of Alangium salvifolium root extract in normal and alloxan induced diabetic rats. A. salvifolium root gave maximum extractive values of 6.4 % w/w with Ethanol and other parameters were within limits. The extract gave positive tests for phytosterols, triterpenes, flavonoids, carbohydrates and alkaloids. The ethanolic extract of A. salvifolium was found to be nearly as potent as tolbutamide in decreasing the blood glucose levels in normal fasting rats. In normal control group the percent reduction in blood glucose indicated that the extract is fast acting. The extract showed significant decrease in blood glucose, when compared with tolbutamide. The evaluated blood glucose levels in alloxan induced diabetic rats were significantly decreased up to 24th h compared to tolbutamide. A single oral administration of alcoholic extract at doses 100, 250 and 500 mg per kg produced a significant blood glucose reduction in a dose dependent manner in normal and diabetic rats. These data confirm the hypoglycaemic and anti-hyperglycaemic effect of alcoholic extract of A. salvifolium root in normal and diabetic rats respectively when compared with standard drug tolbutamide.Keywords: Alangium salvifolium; Ethanolic extract; Alloxan induced; Hypoglycaemic action.© 2011 JSR Publications. Objective: The present study was aimed to evaluate the antihyperglycemic activity and in vivo antioxidant effect of methanolic extract of whole plant of Psydrax dicoccos (MEPD) belonging to the family Rubiaceae.Methods: MEPD was prepared by Soxhlet extraction. Wistar rats weighing (180–200 g) were divided into six groups (n=6), with three doses of 100 mg/kg, 200 mg/kg, and 400 mg/kg of extract. Metformin was used as a standard drug. Diabetes was induced by streptozotocin (STZ) (40–50 mg/kg, i.p) in control group. The animals were treated with different doses of extracts for 21 days, and on the 22nd day, the blood glucose levels along with antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and lipid peroxidase (LPO) were determined.Results: The phytochemical screening of the extract showed the presence of carbohydrates, phenolics, flavonoids, glycosides, and tannins. The methanolic extract of MEPD at the dose of 200 mg/kg body weight showed a significant reduction in blood glucose levels (**p<0.001) with the value of 151.2 mg/dl on the 22nd day at 8 h. A promising antioxidant effect was also evident from the determination of antioxidant enzymes such as SOD, CAT, and LPO.Conclusion: The P. dicoccos extract revealed a potential effect of antihyperglycemic activity and combating nature on oxidative stress induced by STZ.

Introduction. Researchers usually use herbal combinations to explore and develop traditional medicine to obtain additional benefits in the treatment of diseases, including diabetes. This study aims to evaluate the hypoglycemic effect of the combination of Andrographis paniculata (Burm. f.) Wall ex Nees and Caesalpinia sappan Linn extract (APCSE) on diabetes-induced rats. There has not been sufficient research on this combination; however, single extract studies of these plants have been widely conducted. Materials and Methods. Male Sprague Dawley rats (160–200 g) were induced by injecting a low dose of streptozotocin (35 mg/kg BW) twice and fed with a high-fat diet containing 25% fat, whereas control animals received only standard feed. Rats were treated with APCSE at doses of 100 mg and 200 mg/kg BW for seven days and compared to the APE and CSE groups treated with the extract at 100 mg, respectively. For the control group, rats were treated with metformin with a dose of 250 mg/kg. The antihyperglycemic and antihyperlipidemic effects were determined by measuring blood glucose levels and lipid profiles (cholesterol, triglycerides, HDL, and LDL). To assess the impact of the extract on pancreatic and adipose tissue, the number of pancreatic beta cells and adipocytes was evaluated through histopathological and immunohistochemical study. Results and Discussion. In a nonfasting state, the blood glucose change in APCSE 200 mg was 18.65% and was significantly lower from the DM group. However, a single extract of APE and CSE showed lower fasting blood glucose levels compared to the combined extract. Lipid profiles show no significant differences in cholesterol levels between groups; however, all treatment groups, including metformin, showed higher triglyceride levels. The APE-treated group showed significantly lower HDL and LDL, whereas CSE only showed lower LDL. The β-cell number was significantly higher after treatment with single extract CSE. The CSE and the combined extract groups showed hyperplasia adipocytes. Conclusion. The combined extract of APCSE has a moderate antihyperglycemic effect; however, a single extract may have better potential than the combined extract.

Background: Diabetes mellitus is a multifactorial metabolic disorder with several microvascular and macrovascular complications. Several plants have been used as dietary adjuvants to conventional drug therapy. Garcinia indica exhibits significant hypolipidemic and hypoglycemic activity. This study was conducted to evaluate the hypoglycemic effects of methanolic extract of seeds of Garcinia indica on blood glucose levels in Streptozotocin induced diabetic albino rats.Methods: Five groups of wistar albino rats (n=6) weighing 150-200g of either sex aged 3-4 months were obtained for the study. After overnight fasting, streptozotocin (50mg/kg) was administered intraperitoneally to induce diabetes. Five groups are: Group-1: Non diabetic control group, Group-2: diabetic control, Group-3: diabetic standard, Group-4: test group, Group-5: half of test + half of standard. Fasting blood sugar was estimated on 1, 3, 7, 14 and 28th day by capillary blood glucose method. The data obtained were subjected to statistical analysis.Results: In this study, following Streptozotocin administration the blood glucose levels increased in all groups on day 0. In group 2, blood glucose level gradually increased to 445.6±1.75mg/dl over a period of 4 weeks. Following glibenclamide administration in Group 3 - there was a gradual reduction in blood glucose levels: 269.8mg/dl - day 7 to 101.8mg/dl - week 4. Group 4 - persistent and significant (p<0.05) fall in blood glucose levels reaching upto 107mg/dl at the end of 4 weeks. Group 5 - 330mg/dl on day 1 which significantly (p<0.05) reduced to 101mg/dl on day 28. There was improvement in weight in group 4 and group 5 diabetic rats.Conclusions: The extract alone and in combination with glibenclamide showed significant hypoglycemic activity in comparison to diabetic control group.

Objectives: This study investigated the antidiabetic and antihyperlipidemic potential of the ethanol extract of the leaves and stem of Cissus gracillis on alloxan monohydrate-induced diabetic albino rats. Methods: Preliminary phytochemical screening and acute toxicity were carried out. Animals were assigned into seven groups of five rats each. Groups A and B were administered 10 mg/kg each of glibenclamide and atorvastatin respectively, C, D, and E were given 125, 250 and 500 mg/kg of ethanol extract of C. gracillis, respectively, daily for 21 days through oral gavage, group F was diabetic but untreated (diabetic control group), while group G was non-diabetic and untreated which served as the control group. Results: Phytochemical screening revealed the presence of steroids/triterpenoids and carbohydrates. LD50 was above 5000 mg/kg. The extract at 500 mg/kg showed a statistically significant (p<0.05) decrease in blood glucose level when compared with the glibenclamide group on day 21. However, gradual non- significant reduction in blood glucose levels were observed in the extract treated groups on the 7th, 14th, and 21st days of treatment. The administration of ethanol extract of C. gracillis to alloxan-induced diabetic rats produced a decrease in total cholesterol, triglycerides, and low-density lipoproteins comparable to glibenclamide and atorvastatin. Conclusion: The ethanol extract of the leaves and stem of C. gracillis possess a mildly significant antidiabetic and antihyperlipidemic activity.

ABSTRAKTelah dilakukan penelitian tentang penerapan teknik kromatografi kolom vakum untuk pemisahan senyawa anti-hiperglikemik dalam ekstrak metanol kulit kayu juwet Eugenia cumini. Kondisi pemisahan optimum: fasa pendukung/adsorben silikagel G60 (63 – 200 mm), F = 3,5 cm, l = 30 cm, laju alir 1 ml per menit, dan komposisi eluen Et-OAc:Me-OH (5 : 5). Penentuan aktivitas anti-hipeglikemik dilakukan dengan menggunakan mencit normal dan diabetes induksi aloksan 50 mg/kg bb, yang terbagi menjadi 3 kelompok, kontrol, pelakuan ekstrak/fraksi, dan obat standar metformin. Sampel darah diambil pada hari ke - 0, 1, 2, 4, 6, 8, dan 10 sesaat setelah 1 jam perlakuan. Kadar glukosa darah ditentukan dengan metode orto-toluidin. Hasil yang diperoleh menunjukkan bahwa persentase penurunan kadar glukosa darah mencit setelah 8 hari perlakuan adalah: untuk kontrol normal 0%, kontrol diabetes 4%. Untuk perlakuan ekstrak dengan dosis 300 mg/kg terhadap mencit diabetes terjadi penurunan kadar glukosa darah masing-masing: heksana 9%, DCM 18%, Me-OH 38%. Untuk perlakuan fraksi Et-OAc:Me-OH (5 : 5) dengan dosis 50 mg/kg bb terjadi penurunan  sebesar 34%, dan untuk obat pembanding metformin (5mg/kg) sebesar 42%. Kesimpulan yang diperoleh dari penelitian ini adalah bahwa ekstrak metanol dari kulit kayu juwet dan fraksi Et-OAc – Me-OH yang diisolasi dengan teknik kromatografi kolom vakum memiliki aktivitas anti-hiperglikemik. Penurunan kadar glukosa darah mencit diabetes memerlukan waktu sampai 8 hari  Kata kunci: Kromatogtafi kolom vakum, Juwet,  Anti-hiperglikemik,  Glukosa DarahABSTRACT: Vacuum column chromatography technique has been developed for separation anti-hyper-glycemic compound on methanol extract from juwet Eugenia cumini barks. Optimum condition this method are: the adsorbent is silica Gel G60 (63 – 200 mm), F = 3,5 cm, l = 30 cm, elution time 1 ml menit-1, and solvent system Et-OAc:Me-OH (5 : 5). Normal and alloxan induced diabetic rats were undertaken to evaluated anti-hyperglycemic activity and divided in to three groups, control, extract/fraction, and standar drug metformin. Blood samples were collected one hour after administration on the day 0, 1, 2, 4, 6, 8, and 10. Blood glucose level was determined by the ortho-toluidine method. Treatment with the extract and fraction was isolated from methanol extract resulted in reduction in blood glucose levels after 8 days: for normal control is 0%, diabetic control is 4%. For treatment with the extract at dose 300 mg/kg to diabetic groups decrease in blood glucose is 9% (extract hexane), 18% (extract dichloro-methane), 38% (extract methanol). For treatment with Et-OAc:Me-OH (5 : 5) fraction at dose 50 mg/kg  decrease in blood glucose is 34%, and standard drug metformin at dose 5 mg/kg is 42%.The finding of this study indicated that the methanol extract juwet bark and Et-OAc – Me-OH (5:5) fraction was isolated from methanol extract with Vacuum column chromatography technique have anti-hyperglygemic effect. Objective: The present study was aimed to evaluate the antidiabetic activity of ethanolic extract of the whole aerial plant of Portulaca grandiflora Hook on streptozotocin (STZ)-induced diabetic rats. Methods: Experimental diabetes was induced by a single dose of intraperitoneal injection of STZ (150 mg/kg). Adult male Wister albino rats were divided into five groups; normal control, diabetic control, diabetic glibenclamide (5 mg/kg), diabetic P. grandiflora H. extract (200 mg/kg), and diabetic P. grandiflora H. extract (400 mg/kg) for 21 days and analyzed for body weight (BW) and blood glucose. Results: The STZ-treated diabetic control rats showed a significant increase in blood glucose with a concomitant decrease in BW. Oral administration of P. grandiflora H. extract (200 and 400 mg/kg) and glibenclamide (5 mg/kg) for 21 days showed a significant reduction in blood glucose levels and elevation in the bodyweight studies as compared to control and glibenclamide-treated rats. Conclusion: The results of the present study showed that a potent antidiabetic activity was present in the aerial part of plant P. grandiflora H. extract.

Background: Diabetic mellitus is a multifactorial disorder associated with its devastating consequences has assumed epidemic proportion in Bangladesh.Methods: The study evaluates the anti-hyperglycemic activity of the aqueous extracts of C. tamala (CTLEt) leaves in blood glucose of albino rats. Type II diabetes mellitus was induced by injecting alloxan at the concentration of 100mg/kg body weight in male albino rats. The diabetic rats were administered orally with aqueous CTLEt at the amount of 1.0ml, 1.5ml and 2.0ml with lab diet and glibenclamide (5mg/kg of body weight). Then blood glucose levels were estimated in all groups after 2 hours, 4 hours, 6 hours, 12 hours and 18 hours of the treatment with CTLEt and a known antidiabetic drug glibenclamide.Results: A comparison was made between the action of CTLEt and glibenclamide. Blood glucose levels of the CTLEt on 18th hours of the study were 8.6 to 5.1mmol/L (1ml CTLEt with lab diet), 10.4 to 4.9mmol/L (1.5ml CTLEt with lab diet), 14.7 to 4.3mmol/L (2.0ml CTLEt with lab diet) in comparison of diabetic control (9.5 to 8.5, 8.7 to 7.8, 7.7 to 7.1mmol/L) and glibenclamide (13.9 to 6.5, 16.3 to 6.1, 9.5 to 5.1mmol/L). Among the sample level, the 2.0ml CTLEt showed a higher efficiency of hypoglycemic effect on alloxan induced diabetic rats.Conclusions: Till date, there is no specific experimental work in Bangladesh about the evolution of antidiabetic activity of C. tamala plant in animal model. Further studies should be undertaken to find out the molecular mechanism of the leaf powder of C. tamala medicinal plant.

This study aimed to assess the effects of Ginkgo Biloba Extract and Troxerutin on the hippocampus of induced diabetes mellitus in adult albino rats using histological methods.50 adult male albino rats were divided into three groups; Group I (Control); Group II (diabetic): subdivided into Subgroup IIa (T1DM)), Subgroup IIb (T1DM+GBE), Subgroup IIc (T1DM+ troxerutin); Group III: subdivided into Subgroup IIIa (GBE) and Subgroup IIIb (troxerutin). The brain was removed and the cerebral hemisphere was coronally cut at the hippocampal level and used for light microscopic study (H&E staining and PCNA immunostaining). There was a statistically insignificant improvement in animal weights in subgroup IIb and subgroup IIc. Subgroup IIb showed a statistically significant reduction of blood glucose levels while the subgroup IIc showed insignificant reduction of blood glucose levels. Diabetes disturbed the light microscopic structure of the hippocampus. In subgroup IIb and subgroup IIc the hippocampus retained an apparently normal appearance and the stratum pyramidale exhibited the pyramidal cells with rounded vesicular nuclei and acidophilic cytoplasm. Diabetic hippocampal sections revealed negative PCNA immunoreactivity in all layers of DG. In subgroup IIb and subgroup IIc, hippocampal sections showed positive immunoreactivity.
