Background: It is believed that the consumption of an adequate amount of live lactic acid bacteria (as probiotic) may improve the health of the host. Many strains of lactic acid bacteria are generally considered to be safe. However, some strains have shown some adverse effects. Objectives: This study aimed to investigate the safety of a new strain of Lactobacillus pentosus (IBRC=11143) in Wistar rats Methods:In this experimental study, male and female Wistar rats (n=6) were used. A subacute toxicity study (for 28 days) was conducted by oral administration of Lactobacillus pentosus to the animals. In each sex, one group received saline, and the other two groups received the bacteria at doses of 1×108 and 1×109 Colony-Forming Units (CFU)/rat. Results: No significant alteration in the liver and kidney tissues was seen. However, in both sexes, there were significant differences in urea and creatinine levels between the control and the experimental groups. Some blood parameters (Lymphocyte, red blood cell, hematocrit, and hemoglobin) also showed significant changes in the groups that received the bacteria. Moreover, a significant increase in alkaline phosphatase level was observed in male rats. Conclusion: The results indicate that Lactobacillus pentosus (11143) is not entirely safe like other Lactobacillus strains. Therefore, the Lactobacillus pentosus (11143) strain may not be the right choice as a probiotic for human consumption.

Abstract Introduction Study of the variations of bone characteristics with age in different animal models is important to design musculoskeletal studies. Thus, this study aimed to evaluate the bone mass, dimensions, and biomechanical parameters of the femur in young, middle-aged, and aged Wistar rats. Materials and Methods Thirty male rats (Rattus norvegicus) were divided in three groups (n = 10 per group)—3-month-old young rats, 12-month-old middle-aged rats, and 18-months-old aged rats. The right femurs were subjected sequentially to morphometric study (bone weight, cortical thickness) and biomechanical tests (maximum resistance strength and bone stiffness). Results We observed a significant increase in femur histological (cortical thickness) and biomechanical (maximum strength and bone stiffness) parameters with aging when compared with young animals. Conclusions With the advancing age, the right femoral bone of middle-aged and old animals had greater variations when compared with young animals. However, further studies with the aid of a comparison between right and left femur and other long bones in both male and female rats are needed to corroborate with our findings.

Ad libitum (AL) supply of standard chow is the feeding method most often used for rodents in animal experiments. However, AL feeding is known to result in a shorter lifespan and decreased health as compared with restricted feeding. Restricted feeding and thus limiting calorie intake prevents many health problems, increases lifespan and can also increase group uniformity. All this leads to a reduced number of animals needed. So-called standard chows are known to be prone to variation in composition. Synthetic foods have a more standard composition, contributing to group uniformity which, like diet reduction, may decrease the number of animals necessary to obtain statistical significance. In this study, we compared the effects of AL versus restricted feeding (25% reduction in food intake) on standard chow versus synthetic food of three different suppliers on body weight (BW), growth, several blood parameters and organ weights in growing female Wistar rats over a period of 61 days. Diet restriction led to a decreased growth and significantly reduced variation in BW and growth as compared with AL feeding. AL feeding on synthetic diets caused a significantly higher BW gain than on chow diets. Due to experimental design, this same effect occurred on food restriction. Blood parameters and organ weights were affected neither by diet type nor by amount. Incidentally, variations were significantly reduced on food restriction versus AL, and on synthetic diets versus chow diets. This study demonstrates that food restriction versus AL feeding leads to a significantly reduced variation in BW and growth, thereby indicating the potential for reduction when applying this feeding schedule.

Context Selenium (Se) is a trace element essential for cellular function in animals as a component of the enzymes glutathione peroxidase and iodothyronine-5-deiodinase. In many parts of Europe, Se is often deficient in livestock diets due to the low Se status of soil. Supplementation of diets with selenised yeast (predominately as seleno-methionine) or inorganic sodium selenite is common practice in most livestock systems, including ruminants. Lactic acid bacteria have been shown to convert inorganic Se into predominantly elemental nano-Se, which has been used recently in human pro-biotics as a less toxic form of Se. Therefore, silage lactic acid bacteria may provide a supplementation route of bioavailable nano-Se for ruminants. Aim Here, we report on the effect of feeding inoculated silage enriched with a supra-nutritional level of nano-Se (Selage) versus control inoculated silage (Silage) on the Se status of finishing lambs and their products, followed by a second study where blood parameters were investigated in ewes. Methods In the first study, 40 Charollais × Suffolk lambs (42 ± 1.7 kg) were paired according to weight and sex, then allocated to the two treatments for 8 or 10 weeks. Uptake of Se into wool was temporally assessed, as well as excretion of Se into faeces. Selenium concentrations in blood and muscle, carcass characteristics and meat quality are reported postmortem. In the second study, individually penned Suffolk × Mule ewes (n = 12; 76 ± 4.5 kg) were offered the same diets as in the first study. Blood parameters were assessed at the start and after 6 weeks, with intake and excretion into faeces and urine assessed temporally throughout the study. Key results In the first study, dry-matter (DM) intake was similar in both treatment groups, at 0.8 ± 0.03 kg/day, but Se concentrations of the diets were significantly different, resulting in intakes of ~0.14 and 1.60 mg/day on the Silage and Selage diets, respectively. This was reflected in higher Se concentrations in faeces (0.4 vs 2.0 mg/kg DM; P < 0.001), wool (0.11 vs 0.25 mg/kg DM; P < 0.001), blood (0.19 vs 0.46 mg/L; P < 0.001) and muscle (0.31 vs 0.41 mg/kg: P < 0.01) on the Selage than on the Silage diet. Colour (chroma) shelf life of the meat was significantly higher on the Selage treatment (8.05 vs 9.2 days; P < 0.05). In the second trial, for ewes fed Selage, blood seleno-methionine increased from 0.21 to 0.25 mg/L and seleno-cysteine from 0.25 to 0.35 mg/L after 6 weeks on the treatment, whereas there was no change in ewes fed Silage. Glutathione peroxidase increased, whereas haematocrit, haemoglobin and platelet count were decreased across time during the study, but there was no difference between the treatments. Conclusions Nano-Se provided by the Selage treatment was shown to be available to sheep and improve shelf life, with no adverse haematological effects. Implications There is potential to use silage inoculants to provide bioavailable Se to ruminants. Further research is required to determine the most appropriate dose for animal performance and product quality.

Previous studies have shown that morphine abuse during pregnancy cancause a delay in the development of the placenta and embryo and also bring about birth defects. The present study investigates the effect of the duration of maternal morphine consumption during pregnancy, as well as the impacts of morphine abuse on the development of placental layers during the three different periods of pregnancy in Wistar rats.Materials and Methodology. Female Wistar rats have been used in the present study. Experimental groups received morphine (0.05 mg/mL of drinking water) after one night of coupling with male rats for mating. On 9th, 10th, and 14th days of pregnancy, pregnant animals were killed, and placentas were removed and fixed. The cells of the placentas layers were calculated by light microscope and MOTIC and SPSS software.Results. The maternal surface thickness of the placenta was significantly increased, whereasthe fetal surface thickness of placenta was significantly decreased with morphine consumption with a time-dependent manner in experimental groups, compared to control groups. Moreover, the number of trophoblast cells increased in both maternal and fetal surfaces of placenta with respect to the duration of morphine consumption which was overt in the experimental groups compared to the control groups.Conclusion. In general, the time-dependent effects of oral morphine consumption can inhibit the development and natural functioning of cytotrophoblast and syncytiotrophoblast cells of the placental layers.

Sodium nitrate (NaNO2) widely used as food additive for coloring and preserving meat has been reported to induce oxidative stress and cause histopathologic changes, nitrosative tissue damage, and lipid peroxidation in liver and kidney. Therefore, the present study compared the curative effect of chlorophyll fromSauropus androgynus(L) Merr and Cu-chlorophyllin as antioxidant in NaNO2-induced female Wistar rats based on haematological, serum biochemical, and histological evaluation. Thirty male Wistar rats were randomly assigned into six groups of five rats each. NaNO2were given at a subacute dose of 50 mg/kg bw intraperitoneally for 10 days. Chlorophyll fromS. androgynusand Cu-chlorophyllin from K-Liquid™ were given in the following 14 days at the two doses: 0,016 mg/mL and 0.008 mg/mL. NaNO2exposure resulted in significant reductions (p<0.05) in values of packed cell volume (PCV), haemoglobin (Hb) concentration and red blood cell (RBC) count, transferrin, and ferritin and elevation in malondialdehyde (MDA) level and schistocytes percentage with insignificant reductions in serum albumin and transferrin levels. Histology of kidney and liver were changed insignificantly (p>0.05) to normal values. Chlorophyll fromS. androgynusand Cu-chlorophyllin possess antioxidant potentials to protect against toxicities induced by sodium nitrate.

An analysis of the course of infection of mixed oral infections of 12 cystacanths of Moniliformis dubius in 174 male and 179 female Wistar rats has been undertaken.There was a marked decline in the average recovery rate of worms of both sexes from hosts of both sexes during the course of the infection.Female worms from both male and female rats showed, on average, a greater power of survival than male worms from the third period (10–13 weeks) onwards.Male rats were found to retain, on average, a greater number of worms of both sexes than female rats.We wish to thank Miss Susan Arnold and Mr David Barnard for excellent technical help.

AbstractRationaleA reduced effect of a given dose of Δ9-tetrahydrocannabinol (THC) emerges with repeated exposure to the drug. This tolerance can vary depending on THC dose, exposure chronicity and the behavioral or physiological measure of interest. A novel THC inhalation system based on e-cigarette technology has been recently shown to produce the hypothermic and antinociceptive effects of THC in rats.ObjectiveTo determine if tolerance to these effects can be produced with repeated vapor inhalation.MethodsGroups of male and female Wistar rats were exposed to 30 minutes of inhalation of the propylene glycol (PG) vehicle or THC (200 mg/mL in PG) two or three times per day for four days. Rectal temperature changes and nociception were assessed after the first exposure on the first and fourth days of repeated inhalation.ResultsFemale, but not male, rats developed tolerance to the hypothermic and antinociceptive effects of THC after four days of twice-daily THC vapor inhalation. Thrice daily inhalation for four days resulted in tolerance in both male and female rats. The plasma THC levels reached after a 30 minute inhalation session did not differ between the male and female rats.ConclusionsRepeated daily THC inhalation induces tolerance in female and male rats, providing further validation of the vapor inhalation method for preclinical studies.AbbreviationsPG, propylene glycol; THC; Δ9tetrahydrocannabinol;

The knowledge of lactic acid bacteria of raw milk and the main factors affecting their variability are particularly important issues for the control of cheese processing and the bioconservation of farm raw milk food products. The present research study concerned the isolation and identification of twenty strains of the Lactobacillus genus from goat milk originating from the Oulmes region, using the API 50 CH system. All isolates found represented five species: Lactobacillus plantarum (43.75 %), Lactobacillus brevis (37.75 %), Lactobacillus pentosus (6.25 %), Lactobacillus salivarus (6.25 %), and Lactobacillus acidophilus (6.25 %). According to biochemical activities, the majority of the strains displayed weak acidification and autolysis activities in milk. In contrast, they showed high extracellular proteolytic activity. All isolates produced exopolysaccharides and most of them could metabolize citrate. The absence of hemolytic activity may suggest the use of these isolates as adjunct starters in the food fermentation process.
