Rationale: During hemosorption procedures, it is important to investigate not only the sorption and activation characteristics of hemocontact agents, but also to assess the effect of sorbents on the parameters of blood homeostasis. The intensity of hemolysis can be judged by the degree of changes in optical density of blood plasma at wavelengths corresponding to the peak absorption of hemoglobin (414, 544 and 577 nm).Aim: To assess the effect of three granular sorbents (SKT-6A, HPS, Silochrome C-120) contacting human venous blood in vitro on changes in plasma spectral characteristics.Materials and methods: The blood contact was modeled at bench conditions with the use of donated blood at rotating mode. Blood samples were drawn before the experiment and after 5, 20, 40, and 60 minutes. Spectroscopic assessment was performed in the visible light range (300–700 nm) with UNICO 2802(S) spectrophotometer.Results: The interaction of the SKT-6A sorbent with blood resulted in a 17.3% decrease in the plasma optical density at a wavelength of 540 nm, compared to baseline, as soon as at 5 minute of the experiment (p < 0.05). The decline in optical density imposed by the blood contact with HPS ranged from 2.6 to 12.1% (p < 0.05) during the observation period. The sorption activity of SKT-6A and HPS prevailed over their lytic properties. On the contrary, the percentage change in the optical density of the Silochrome C-120 sorbent during its blood contact increased from 25.6 to 38.3% (p < 0.05), indicating that this sorbent was inducing hemolysis. The sorbents tested can be arranged as follows according to their ability to induce hemolysis during their contact with blood: HPS < SKT-6A < Silochrome C-120.Conclusion: The tested SKT-6A and HPS sorbents can be used as blood-contact agents for the low volume hemoperfusion. The HPS agent seems to be the most promising for routine clinical use. The Silochrome C-120 sorbent requires some chemical modifcation to improve its properties of hemocompatibility.

Background: The relevance of the work lies in the search for new hemocontact drugs with hemocompatibility and a pronounced activation effect on the cellular and humoral blood systems for their possible use in clinical practice during low-volume hemoperfusion.The aim of this work was to assess the activation capabilities of three granular hemosorbents by the rate of adhesion of blood cellular elements to the surface of granules in vitro.Materials and methods. When using the method of low-volume hemoperfusion (LVH) in the clinic it is important to take into account the activation properties of solid-phase granular drugs. Blood-contact interaction was carried out in bench conditions with the use of donated blood in rotary mode. Blood samples were taken before the experiment and after 5, 20, 40 and 60 minutes. Changes in blood cell and subcellular populations were evaluated using the Sysmex XT 1800i hematological analyzer (26 parameters), which made it possible to indirectly judge the activation of blood cells. 30 experiments were conducted. To analyze the activation functions of the hemocontact preparations the speed-time adhesive profile of blood cells on the sorbent was used.Results. The effect of using the preparations Silochrome S-120 and SPS in comparison with SСT-6A HP as contact hemoactivators can be more pronounced, since the activation potential of these sorbents for blood cells is much higher. Silochrome S-120 has the highest activation capabilities compared to SPS and SKT-6A HP.Conclusion. Adhesion rate indicators can be indicators of the activation of blood cells upon contact with foreign surfaces and serve as a criterion for assessing the activation capabilities of these surfaces when using the LVH method in the clinic.

SummaryPaired blood samples were obtained from mothers (venous) and babies (cord venous blood) at the time of delivery by caesarean section under epidural anaesthetic. Fetal platelets failed to aggregate in response to adrenaline in vitro although adrenaline could potentiate the threshold response to adenosine diphosphate (1 μM). Fetal platelet responses to collagen and 8 Arg vasopressin did not differ significantly from maternal responses. Maternal and fetal platelets also showed similar inhibition of aggregation after activation of adenylate cyclase (PGE1 and parathormone), in contrast to the inhibition of adenylate cyclase by adrenaline.Alpha2 adrenoceptors were investigated using [3H] yohimbine binding receptor number and were reduced modestly but significantly on fetal compared to maternal platelets. The failure of fetal platelet aggregation in response to adrenaline appears to be related to a failure of receptor coupling and may represent a delayed maturation of fetal platelet alpha receptors or a response- to increased circulating catecholamines during birth.

The main goal of the present study was to investigate the microencapsulation, in vitro release capacity and efficiency of catechin-rich Acacia catechu extract by Clinosorbent-5 (CLS-5) microparticles by in-depth detailed analyses and mathematical modelling of the encapsulation and in vitro release kinetics behaviour of the polyphenol-mineral composite system. The bioflavanol encapsulation and release efficiency on/from the mineral matrix were assessed by sorption experiments and interpretative modelling of the experimental data. The surface and spectral characteristics of the natural bioactive substance and the inorganic microcarrier were determined by Fourier Transform Infrared Spectroscopy (FTIR) and Ultraviolet/Visible (UV/Vis) spectrophotometric analyses. The maximum extent of catechin microencapsulation in acidic medium was 32%. The in vitro release kinetics study in simulated enzyme-free gastric medium (pH = 1.2) approved 88% maximum release efficiency achieved after 24 h. The in vitro release profile displayed that the developed bioflavanol/clinoptilolite microcarrier system provided sustained catechin in vitro release behaviour without an initial burst effect. Thus, the results from the present study are essential for the design and development of innovative catechin-CLS-5 microcarrier systems for application in human and veterinary medicine.

SummaryThrombin regulation in newborns remains incompletely understood.We studied tissue factor-initiated thrombin formation in cord plasma in vitro, and the effects of Factor VLeiden (FVL) heterozygosity on thrombin regulation both in vitro and in vivo in newborns. Pregnant women with known thrombophilia (n=27) were enrolled in the study. Cord blood and venous blood at the age of 14 days were collected from 11 FVL heterozygous newborns (FVL-positive) and from 16 FVL-negative newborns. Prothrombin fragment F1+2 and coagulation factors were measured. Tissue factor-initiated thrombin formation was studied in cord platelet-poor plasma (PPP) of FVL-negative and -positive newborns, and in both PPP and platelet-rich plasma (PRP) of healthy controls. The endogenous thrombin potential (ETP) in cord PPP or PRP was ∼60% of that in adult plasma, while thrombin formation started ∼55% and ∼40% earlier in cord PPP and PRP, respectively. Further, in FVL-positive newborns thrombin formation started significantly earlier than in FVL-negative newborns. Exogenous activated protein C (APC) decreased ETP significantly more in cord than in adult PRP. In FVL-negative cord plasma 5nM APC decreased ETP by 17.4±3.5% (mean±SEM) compared with only 3.5±3.8% in FVL-positive cord plasma (p=0.01). FVL-positive newborns showed similar levels of F1+2 but significantly decreased levels of factor V compared with FVL negative newborns both in cord plasma (FV 0.82±0.07 U/ml vs. 0.98±0.05 U/ml, p=0.03) and at the age of two weeks (FV 1.15±0.04 U/ml vs. 1.32±0.05 U/ml, p=0.03). In conclusion, newborn plasma showed more rapid thrombin formation and enhanced sensitivity to APC compared with adult plasma. FVL conveyed APC resistance and a procoagulant effect in newborn plasma. Lack of elevated F1+2 levels in FVL-positive infants, however, suggested the existence of balancing mechanisms; one could be the observed lower level of factor V in FVL heterozygous newborns.

1. The metabolism of incubated slices of sponge-induced granulation tissue, harvested 4–90 days after the implantation, was studied with special reference to the capacity of collagen synthesis and to the energy metabolism. Data are also given on the nucleic acid contents during the observation period. Three metabolic phases were evident. 2. The viability of the slices for the synthesis of collagen was studied in various conditions. Freezing and homogenization destroyed the capacity of the tissue to incorporate proline into collagen. 3. Consumption of oxygen reached the maximum at 30–40 days. There was evidence that the pentose phosphate cycle was important, especially during the phases of the proliferation and the involution. The formation of lactic acid was maximal at about 20 days. 4. The capacity to incorporate proline into collagen hydroxyproline in vitro was limited to a relatively short period at 10–30 days. 5. The synthesis of collagen was dependent on the supply of oxygen and glucose, which latter could be replaced in the incubation medium by other monosaccharides but not by the metabolites of glucose or tricarboxylic acid-cycle intermediates.

Background: The leaves of neem (Azadirachta indicia) is one of herbal medicine that recommended as an alternative material of root canal irrigants. The active ingredients of neem leaves such as alkaloids, tannins, saponins and flavonoids has been proven to have antibacterial activity against E. faecalis. The ideal properties of an alternative material of root canal irrigants is not only have antibacterial activity but also is not toxic to the tissues, but the toxicity of neem leaves remains unclear until now. Objective: The aims of this study to determine the toxicity effect of neem leaves extract in specific concentration. Methods: This research was an experimental laboratory in vitro study of baby hamster kidney fibroblast (BHK-21). The neem leaves extract was made by maceration method using ethanol 96% and certain dilution performed to obtain various concentrations. Cytotoxicity test was conducted by MTT assay and the optical density was measured using ELISA reader at wavelength of 620 nm. Then, the optical density values were calculated using the formula for determining the number of survival fibroblasts after tested. Results: The percentage of survival fibroblast at concentration of 50%, 55%, 60%, 65%, 70%, 75%, 80%, 85%, 90%, 95, and 100% in sequence were 71.3%, 64, 2%, 62%, 60.2%, 40.1%, 36.3%, 36.7%, 29%, 28.4%, 27.5%, and 25.6% . Conclusion: The extract of neem leaves (Azadirachta indica) has cytotoxic effects at concentration of 70% up to 100%.

Various strategies have been employed in the treatment and management of Salmonella infection however, Salmonella strains have gained resistance to antibiotics. This study was to determine in vitro anti-Salmonella activity of Boerhaavia diffusa leaf extract against clinical isolate of Salmonella typhi and Salmonella typhi ATCC 14028. The aqueous and ethanol extracts of B. diffusa were studied for their antibacterial activity against pathogenic Salmonella typhi. This study was carried out between April and September 2018. The in vitro antibacterial activity was performed by agar well diffusion method and broth dilution using spectrophotometric method and the results were expressed as the average diameter of zone of inhibition of bacterial growth around the well and optical density respectively. It was observed that aqueous extract exerted slightly higher activity than ethanolic extract as revealed by the mean diameter of zone of inhibitions at a concentration of 200 mg/ml, the aqueous extract had 35.21±0.47 mm (Salmonella typhi ATCC 14028) compared with ethanol extract 26.41±0.32 mm (clinical). However, in the broth dilution method, ethanol extract significantly (p=0.05) reduced the cell, at 48 hours, the optical density of clinical isolate of S. typhi treated at concentration of 200 mg/ml of extract was 0.47±0.02 nm while at the same concentration of extract, aqueous extract had an optical density of 0.52±0.11 nm respectively. The phytochemical assay revealed that tannin (5.18±0.02 mg/g) and quinone (8.45±0.13 mg/g) in ethanol extract was significantly (p=0.05) higher than aqueous extract while saponin (14.18±0.06 mg/g) was higher in the aqueous extract. The ethanol and aqueous extracts of leaves of B. diffusa whole plant exhibited significant antibacterial activity against both clinical and typed Salmonella typhi. Therefore, the plant extract could be used for the treatment of Salmonellosis, however, the in vivo studies is needed to ascertain the safety of the extract.

Hydrogen sulfide (H2S) is rapidly emerging as a biologically significant signaling molecule. Studies published before 2000 report low or undetectable H2S (usually as total sulfide) levels in blood or plasma, whereas recent work has reported sulfide concentrations between 10 and 300 μM, suggesting it acts as a circulating signal. In the first series of experiments, we used a recently developed polarographic sensor to measure the baseline level of endogenous H2S gas and turnover of exogenous H2S gas in real time in blood from numerous animals, including lamprey, trout, mouse, rat, pig, and cow. We found that, contrary to recent reports, H2S gas was essentially undetectable (<100 nM total sulfide) in all animals. Furthermore, exogenous sulfide was rapidly removed from blood, plasma, or 5% bovine serum albumin in vitro and from intact trout in vivo. To determine if blood H2S could transiently increase, we measured oxygen-dependent H2S production by trout hearts in vitro and in vivo. H2S has been shown to mediate ischemic preconditioning (IPC) in mammals. IPC is present in trout and, unlike mammals, the trout myocardium obtains its oxygen from relatively hypoxic systemic venous blood. In vitro, myocardial H2S production was inversely related to Po2, whereas we failed to detect H2S in ventral aortic blood from either normoxic or hypoxic fish in vivo. These results provide an autocrine or paracrine mechanism for myocardial coupling of hypoxia to H2S in IPC, i.e., oxygen sensing, but they fail to provide any evidence that H2S signaling is mediated by the circulation.

This study aimed to present a literature review with data obtained in vitro and in vivo on metal free partial fixed protheses (PFP) in the posterior region, considering the following variables: ceramic material to be used; prosthesis extension; survival or longevity; main failures found, and comparison with the longevity of conventional metaloceramic PFP. A bibliographical survey was carried out using the databases: United States National Library of Medicine (PubMed) and Scientific Electronic Library Online (Sciello) for articles in English and Portuguese from 1998 to 2019. Articles should meet the inclusion criteria, which were articles that contained information that enabled the calculation of PFPs survival and success, articles with a minimum observation period of 3 years, articles that identified the reason of failures, and studies reported since 1998. Sixteen studies met the inclusion criteria and were evaluated comparatively. The survival rate of the PFP’s varies depending on the ceramic material used and the prostheses extension, no significant difference was observed in the relation between the ceramic material used and the connectors size; however, the greater the extension of the prosthesis, the higher must be the connector size. It was concluded that ceramic materials based on zirconia are the ones that have the longest survival. In addition, the main reasons that lead to decreased survival of PFP’s are secondary caries and connector fracture, however, more studies are needed to determine safely which materials and the extent of PFP’s are the most indicated.
