Evaluation of autologous and homologous platelet rich plasma as a surgical wound healing promoter in rabbits

The present study compared the efficacy of autologous and homologous platelet rich plasma (PRP) as a surgical wound healing promoter in rabbits. Sixteen adult healthy rabbits were divided into two groups i.e. A and B. Group A was treated with autologous platelet rich plasma (APRP) and group B was treated with homologous platelet rich plasma (HPRP). Both groups were operated for skin grafting (auto grafting) at two (PRP treated experimental and control) sites. Clinical evaluation of PRP was performed by using macroscopic (edema, exudation, coloration, temperature, cosmetic appearance and healing status) and microscopic (acute inflammation, fibroblast proliferation and granulation tissue proliferation) parameters. The healing status, coloration and cosmetic appearance of APRP treated group were more satisfactory than HPRP treated group. APRP group also showed less inflammation, edema and more granular tissue proliferation as compare to HPRP group. It was concluded that autologous PRP is an efficient surgical wound healing promoter in rabbits.

Background: Diabetic foot ulcers continue to pose significant global issue despite the advances made in the management of diabetes. It causes major foot complications if they are not addressed properly. It needs multidisciplinary approach for its care. While several advancements has taken place in wound care management, platelet rich plasma and stem cell therapy promises to offer a new hope in its management, aiding in cellular and tissue regeneration. The purpose of the present study was to compare Platelet-Rich Plasma versus conventional dressing in the management of diabetic foot ulcers.Methods: This prospective study was focused on 20 diabetic foot ulcers, carried out in a surgical unit of ACS Medical College and Hospital, Chennai, Tamil Nadu, from January 2018 to June 2018. Patients were divided into two groups; Group A received conventional ordinary dressing (N=10, 50%) and Group B received PRP dressing (N = 10, 50%). The mean follow-up period was 8 weeks.Results: The estimated time of wound healing was 8 weeks and healing was found to be more effective for patients in group B compared to patients in group A; the PRP group was found to be more effective in wound healing with fewer complications, less infection, exudates and pain.Conclusions: There have been considerable advances in the use of PRP in therapeutic processes in recent years in tissue regeneration therapy. PRP is a powerful tool for the treatment of chronic wounds and very promising for diabetic foot wounds; PRP enables healing, and reduces amputation rates, infection and exudates.

<p class="BodyText1">Among wound-healing modalities, platelet-rich plasma (PRP) has been used for wound healing, through the release of multiple growth factors. The platelets mediate wound healing by initiating the clotting pathway and the subsequent matrix remodelisation. The aim of the work was to study the role of injection of platelet-rich plasma in the treatment of scars. This study was conducted on thirty patients who were divided into two groups (A and B). Each group included fifteen patients. The two groups were randomly distributed by using individual sealed envelopes. Group A was subjected to subcision of their acne scars using Nokor needle, followed by suction. Group B patients were subjected to the same treatment but followed by an injection of platelet-rich plasma once per month over three months. In Group A, 6 patients (40%) showed moderate improvement, 5 patients (33.3%) showed slight improvement and 4 patients (26.7%) showed significant improvement. In Group B, 7 patients (46.7%) showed moderate improvement, 5 patients (33.3%) showed marked improvement and 3 patients (20%) showed significant improvement. There was a statistically significant difference on the independent observer’s after-treatment assessment between the groups (P=0.014). In conclusion, subcision suction leads to a persistent improvement of acne scars in a short time, and the coupling with injection of PRP is the most effective in the treatment of depressed facial scars.</p>

An experiment was conducted to investigate the counteracting effects of spirulina in Long Evans rats exposed to oral potentiated sulfonamide administration. 20 rats were randomly assigned into four equal groups (A, B, C and D) and were fed with standard broiler pellet (25g/rat/day) throughout the experimental period of 60 days. Rats of Group A were fed only with pellet without any experimental diet and were defined as control. Rats  of Group B were treated with potentiated sulfonamide @ 96 mg/rat/day orally whereas Group C was treated with potentiated sulfonamide @ 96 mg/rat/day plus spirulina (Spirulina maxima) @ 50 mg/rat/day orally (low dose spirulina). In Group D, potentiated sulfonamide and spirulina (Spirulina maxima) were given through feed @ 96 mg/rat/day and @ 100 mg/rat/day (high dose spirulina) respectively. Hematological parameters (TEC, Hb and absolute count of lymphocyte, neutrophil and eosinophil) and hispathological profile of liver and kidney were recorded. The investigation revealed that the oral administration of sulfonamide significantly (p<0.01) decreased the TEC (5.93±0.24) value, number of lymphocyte (581.76±3.70) and neutrophil (581.76±3.70) compared to other treated groups and control group. On the other hand significant (p<0.01) increase (422.86±2.34) in eosinophil population has been found in rats fed on sulfonamide irrespective of spirulina supplementation on the final day of experiment compared to other treated group and control group. From this experiment it is evidenced that spirulina has a potential counteracting effect against sulfonamide. We evaluate the early and late safety and efficacy of silver nanoparticle (AgNPs) in wound healing after circumcision. This multicenter prospective comparative non-randomized observational study compares wound dressing with AgNPs (group A) vs. gentamicin cream (group B). Follow-up included objective evaluation at 10 and 30 days by the Southampton Scoring System (SSS) and Stony Brook Scar Evaluation Scale (SBSES). We enrolled 392 males: 194 in group A, and 198 in group B. At 10 days follow-up, in group A, the SSS scale was grade 1 in 49.5% and grade 2 in the remaining; meanwhile, in group B, grade 1 was in 58%, grade 2 in 34.3%, and grade 4 in 7.6%. At 30 days follow-up, grade 1 healing was 97.4% and 98.4% in group A and B, respectively. At 10 days follow-up, the mean SBSES score was 3.58 and 3.69 in group A and B, respectively; while at 30 days follow-up, 4.81 and 4.76 in group A and B, respectively. Only in group B did 7.6% of males have antibiotic therapy due to pus discharge. No patients needed surgical wound revision. AgNPs led to a late but safer healing, they were non-inferior to the antibiotic cream wound dressing efficacy, and they avoided pus discharge and the need for oral antibiotics due to their polymer material.

Effect of feed supplementation to does grazed on naturally grown grasses during pre-partum on growth, reproduction and lactation performances of does and growth of the kids were investigated. Fifteen two-toothed and non-pregnant does of about one and half year of age constituted the experimental animals which were randomly allocated in the three groups of 5 and offered A) grazing ad libitum + l00g Mashkalai + 100g Wheat bran, B) grazing ad libitum + 50g Mashkalai + 50g Wheat bran and C) grazing ad libitum. The dry matter intake (DMI) between supplemented groups of A and B were almost similar (90 and 89g/kg w0.75/d respectively) and 12% higher than the control group C (80g/ kg w0.75/d). This DMI represent 4.6, 4.5 and 4.1% of live weight for group A, B and C, respectively. Does in treated group A consumed 30 and 62% more CP than group B and control group C, respectively. Similarly, group B consumed about 25% more CP than group C. There was no effect of grazing and supplementation before the pre-partum period on milk yield between treated group (A and B) and control group C (Av. 157 ml and 156 ml/d, respectively), rearing rate and growth of the suckling kids. Present study suggests that provision of grazing and supplementary feeding increases nutrients intake of does but appear to have no influence on milk yield. Higher prolificacy of Black Bengal goat may not bring any benefit unless milk production is increased.http://dx.doi.org/10.3329/bjas.v41i1.11975

Background: Lower limb ulcer is a common disease among the Indian population with a prevalence of approximately 1% to 2% which is slightly higher in the older population. There are various modalities of treatment with the main aim being early wound healing. This study is done to compare the results of negative pressure dressing and conventional dressing in lower limb ulcers.Methods: This study, done at the Department of Surgery, G. K. General Hospital and Gujarat Adani Institute of Medical Sciences, Bhuj, from October 2017 to September 2018 is a prospective study. A total of 120 patients were randomly divided in two group comprising of 60 patients each. The patients in Group A were treated with negative pressure dressing while those in Group B were treated with conventional dressing. The patients were assessed, in both test and control groups, with parameters like appearance of granulation tissue, bacterial clearance and wound healing.Results: 80% of the patients belonged to the age group of 41-60 years while others were less than 40 years. We observed that in Group A majority of the patients had wound healing in 11-40 days while in Group B majority patients took 31-60 days for their wounds to be healed.Conclusions: We conclude that negative pressure dressing is more efficient as compared to conventional dressing for healing of lower limb ulcers, enabling a shorter hospital stay and early resumption of daily activities to the patient.

<b><i>Objective:</i></b> To investigate the effect of platelet-rich plasma (PRP) injection in patients with atrophic rhinitis. <b><i>Methods:</i></b> Prepared PRP was injected into the inferior turbinate bilaterally, and nasal bacterial cultures were conducted. Improvement of symptoms was assessed with the Nasal Obstruction Symptom Evaluation (NOSE) and the Sino-Nasal Outcome Test-22 (SNOT-22). Nasal mucociliary clearance was assessed using the saccharin transit time (STT). <b><i>Results:</i></b> In the PRP-injected group (group A), NOSE (throughout the study) and SNOT-22 (1 month after injection) scores were significantly decreased during the study. However, the saline spray group (group B) showed no significant nasal symptom improvement during the study period. In group A, the STT was improved until 3 months after the injection. In contrast, group B showed STT improvement after 2 months that was maintained throughout the study. <b><i>Conclusion:</i></b> PRP injections can improve nasal symptoms and nasal mucociliary function in patients with atrophic rhinitis.

AbstractIntroduction:Laryngopharyngeal reflux is increasingly being implicated in several otolaryngological disorders.Aims:To study a potential correlation between pre-operative laryngopharyngeal reflux and wound healing and recovery after tonsillectomy, based on subjective and objective findings.Materials and methods:A prospective, blinded study was undertaken, including 60 patients scheduled for tonsillectomy, divided into two equal groups: a study group (group A) with pre-operative laryngopharyngeal reflux documented using ambulatory 24-hour pH monitoring; and a control group (group B) without laryngopharyngeal reflux.Results:Group A had significantly higher pain scores on the seventh and 14th post-operative days (p = 0.022 and p = 0.000, respectively) and took a significantly longer time to return to normal eating (p = 0.013), compared with group B. Group A also showed significantly slower healing on the seventh and 14th post-operative days, as estimated by assessing the grade of post-operative slough formation (p = 0.016 and p = 0.029, respectively). A significant correlation between the number of pharyngeal reflux episodes and the degree of post-operative slough was also found.Conclusions:Laryngopharyngeal reflux can significantly decrease wound healing following tonsillectomy. Therefore, pre-operative recognition and management of this condition is desirable in order to eliminate its negative post-operative effect.

Objective: To evaluate the effectiveness of placing a resorbable collagen barrier in impeding the extrusion of obturation material in primary molars undergoing resorption. Study design: All the 94 canals in 47 mandibular molars were allocated to 2 groups- Group ‘A’- 47 canals with collagen barrier (Test group) and Group ‘B’- 47 canals without collagen barrier (Control group) based on randomization protocol. Pulpectomy was performed and obturation of both test and control canals were radiographically assessed. Pearson’s chi – square test was applied to analyze the results. The significance level was predetermined at p &lt; 0.05. Results: Among the test group, 93.6% of the canals showed no extrusion while, 6.4% showed visible extrusion of the material outside the apex. In the control group, 83% showed no extrusion whereas 17% of the canals showed visible extrusion outside the apex. But no significant difference was noted (p&gt;0.05). Conclusion: The placement of resorbable collagen barrier in the apical third of the canal prevented the extrusion of obturating material beyond the apex in resorbing primary molars.

Background: Obesity is a prevailing metabolic disorder that affects the functioning of the male reproductive system. Excessive adipose tissue enhances reactive oxygen species generation and is linked with male infertility. Spinach has demonstrated antioxidant effects. The present study was conducted to determine the antioxidant effects of spinach on sperm parameters in obese Sprague Dawley rats. Subjects and methods: This randomized control study was conducted at the animal house of the National Institute of Health Islamabad, Islamic International Medical College, Cosmesurge International Hospital, Rawalpindi, and Apollo lab, Islamabad, Pakistan from April 2016 to March 2017. Forty male Sprague Dawley rats having an age of 8 weeks and weight 160-200g were tagged from number 1 to 40. Every third rat was randomly allocated to control Group A (n=13) and remaining into the Experimental group (n=27). Rats of control Group A was given a standard diet while a high-fat diet was given to Experimental group rats to induce obesity for the duration of six weeks. Weight (g) was measured weekly and obesity was confirmed when rats attain more than 20% weight when compared with that of rats of control Group A. Then, after obesity induction, the experimental group was alienated into the obesity control group (Group B) and spinach treated group (Group C). For sample, rats of Group A and Group B were sacrificed, and the cauda epididymis of each rat was placed in a Petri dish containing normal saline and cut into pieces to allow the release of sperm and then sperm parameters (sperms concentration, motility, and morphology) were recorded under the microscope. Then, spinach (5% hot water extract) along with the persistence of fat diet was administered to Group C for 4 weeks and finally, sperm parameters were measured in this group. Results: Sperm concentration/ml, motility (%), and normal morphology (%) of Group B rats were significantly decreased as compared to Group A rats. However, sperm concentration/ml, motility (%), and normal morphology (%) of Group C (spinach treated group) rats was significantly increased (p<0.001) as compared to Group B (obesity control group) rats after administering spinach. Conclusion: The addition of Spinach in a normal diet regimen restores normal sperm morphology, improves sperm motility and concentration.
