Effects of xylazine-ketamine, xylazine-propofol and xylazine-ketamine-propofol administration on free radical generation and blood gases in sheep

This study was aimed at investigating the effects of xylazine-ketamine, xylazine-propofol and xylazine-ketamine-propofol combinations on oxidative stress, antioxidant capacity and blood gases in sheep. Excluding the control animals, the sheep included in Groups 1, 2 and 3 were administered with combinations of xylazine-ketamine, xylazine-propofol and xylazine-ketamine-propofol, respectively, by intravenous route. The comparison of the three treatment groups with the control group showed that no significant difference existed for TAS, TOS, MDA and CAT levels. The evaluation of blood gas and electrolyte levels demonstrated a significant decrease in PvO2, cSO2, Na, and Ca levels, and a significant increase in glucose levels. In result, this study showed that the three anaesthetic combinations tested did not have any adverse effect on the oxidant/antioxidant status, but caused significant alterations in blood gas levels.

Objectives: This study aims to investigate the relationship between disease activity, dietary phytochemical index (DPI), and serum total oxidant status (TOS) and total antioxidant status (TAS) in patients with ankylosing spondylitis (AS). Patients and methods: Between August 2020 and January 2021, a total of 37 patients (23 males, 14 females; mean age: 39.3±9.4 years; range, 21 to 61 years) with AS and 36 age-, sex-, and body mass index-matched healthy individuals (24 males, 12 females; mean age: 37.9±8.9 years; range, 20 to 60 years) were included. Serum TAS (μmoLTroloxEq/L) and TOS (μmoL H2O2Eq/L) measurements were performed and the oxidative stress index (OSI) was calculated. Dietary evaluation was made from a one-day dietary record and DPI was calculated. Results: Serum TAS level in AS patients was significantly lower than the healthy group (p=0.003). Serum TOS level was similar in both groups. The OSI of patients was significantly higher than the controls (p=0.035). The mean DPI, polyunsaturated fatty acid, n-3 fatty acid, and vitamin C intake of patients were significantly lower than controls (p=0.042, p=0.033, and p=0.022, respectively). A moderate positive correlation was found between the TAS level and DPI of the control group (r=0.352, p=0.035). According to medications, no significant difference was seen between the groups in terms of patients’ characteristics, DPI, and laboratory tests and there was no correlation between DPI, TAS, TOS, and OSI. Conclusion: Lower DPI and lower n-3 fatty acid and vitamin C intake in patient group demonstrated that patients with AS should pay more attention to their diet to increase serum antioxidant status.

Objective: The objective of the present study was to determine oxidant and antioxidant status in infants with hyperbilirubinemia and/or kernicterus and to find whether there is a relationship between bilirubin level and oxidant/antioxidant status. Patients: The study includes 69 full-term newborns (neonates with hyperbilirubinemia needing phototherapy [Group 1, n = 36] and neonates with kernicterus [Group 2, n = 33]) and 25 age-matched healthy newborn. Results: Plasma total antioxidant capacity (TAC) and serum total oxidant status (TOS) were significantly higher in Groups 1 and 2 than the control group. There was a significant difference between Group 1 and control cases for malondialdehyde (MDA; p < 0.001). Total free sulfhydryl group (TTHI) values were significantly elevated in Group 1 compared to Group 2 and control cases. Correlation analysis showed that the correlation between total bilirubin (TB) and TAC, TOS, MDA and oxidative stress index may be expressed by a quadratic curve. After phototherapy, a statistically significant increase in nitrite level was observed. Conclusion: We demonstrated that the relationship between serum TB and antioxidants and oxidative stress could be expressed by a quadratic correlation curve.

Myocardial infarction (MI) is a common cause of death due to interrupted blood supply to the heart. Roselle calyx (Hibiscus sabdariffa Linn.) is rich in polyphenols and has a potential in alleviating the risk of cardiovascular disease through its antioxidant activity. This study was aimed to investigate the effects of Hibiscus sabdariffa Linn. or roselle polyphenol-rich extract (HPE) supplementation in oxidative stress and cardiac injury biomarkers as well as cardiac histological changes following isoprenaline (ISO)-induced myocardial infarction (MI). Twenty-one male rats were randomly divided into three groups: control, MI, and HPE+MI. Normal saline or HPE (20 mg/kg BW) was given to MI and HPE+MI groups, respectively, for 14 consecutive days via force feeding. On the 15th and 16th day, 85 mg/kg body weight of ISO was administered subcutaneously to induce MI. Control group was only given normal saline throughout this 16-day duration of study. The results showed that HPE reduced the oxidative stress markers malondialdehyde (MDA) and nitrite oxide (NO) in HPE+MI group when compared with MI group (p<0.05) while increased reduced glutathione (GSH) level reflexes the improvement in antioxidant status. Cardiac injury biomarkers analysis showed no significant difference in HPE+MI group when compared to MI group. Histological study showed that HPE managed to reduce cardiac muscle fibre damage and infiltration of inflammatory cells in ISO-induced MI rats. In conclusion, HPE has the potential in protecting the heart against ISO-induced MI by reducing the oxidative stress and increasing antioxidant status.

Can redox homeostasis indicators be potential non-invasive markers, crucial in the diagnosis and treatment of endometriosis? We checked if the differences in levels of serum oxidative-antioxidant balance parameters (TAS, FRAP, albumin, total bilirubin, uric acid, iron, SIRT3, SIRT5, SIRT6, telomerase, AOPP) are significant between patients with advanced endometriosis (E), healthy women (control group, C) and non-endometriosis women, but with other gynecological disorders (NE). The FRAP concentrations were significantly higher in E and NE group than in the control group (p = 0.015 and p = 0.017, respectively). The telomerase concentrations were significantly higher in the endometriosis group than in the control group (p = 0.004). Significantly higher concentrations of AOPP were observed in E (p < 0.001) and NE groups (p = 0.028) in comparison to the control subjects. Between stages III and IV of endometriosis, a significant difference existed only in concentration of iron (p = 0.013). There were no significant differences between the studied groups in the values of the remaining parameters. Based on the results of ROC curve analysis, we can conclude that the levels of serum FRAP, telomerase and AOPP may be taken into account as promising diagnostics markers that reflect the degree of oxidative stress accompanying advanced endometriosis.

Abstract The objective of this study was to determine the effects of a Saccharomyces cerevisiae yeast as a substitute to AGP on intestinal health and performance of piglets. A total of 192 piglets weaned at 28 d of age were randomly assigned into one of four treatment groups with 8 pens/ treatment and 6 pigs/pen. Piglets were supplemented with either a regular nursery diet with two phase feeding (Phase 1: d1-d14 and Phase 2: d15-d28) (CON); CON diet supplemented with 75mg/kg Chlortetracycline plus ZnO 2000 mg/kg in phase 1 and 160 mg/kg in phase 2 (AGP); Con diet supplemented with 2kg/ton live yeast in both phases (LY); Con diet plus live yeast 1kg/ton in both phases plus ZnO 2000 mg/kg in phase 1 and 160 mg/kg in phase 2 (LY-ZnO). During Phase 1, LY and LY-ZnO group had significantly higher ADG (p&lt; 0.01). In Phase 2 only the LY group has a significantly higher ADG (P=0.02). The LY and LY-ZnO group had also better feed efficiency (P&lt; 0.05) during phase 1. On the other hand, the CON group had significantly higher (P&lt; 0.01) diarrhea score. Test on antioxidant status has not shown any significant difference between the groups for most parameters measured; however, the concentration of Malondialdehyde (MDA, a marker for oxidative stress) was significantly decreased (P&lt; 0.01) in the LY and LY-ZnO group on day 28. Furthermore, superoxide dismutase (SOD, the most powerful antioxidant) was significantly increased (P&lt; 0.05) in these two treatment groups at different time points. Live yeast and Live yeast plus ZnO treatment has also significantly increased (P&lt; 0.01) concentration of IgA and IgG in the serum of piglets on d7 and d14. In conclusion, supplementation of nursery piglet diets with either live yeast or live yeast plus ZnO improved zootechnical performance of animals by improving gut health through reduction of oxidative stress and enhancement of immune response, suggesting that yeast probiotics could be a suitable alternative to substitute AGP.

SummaryBackground:Bell’s palsy (BP) is the most common acute mononeuropathy of unilateral facial paralysis. Immune, infective and ischaemic mechanisms are potential contributors to the development of BP, but the precise cause remains unclear. Recently, oxidative stress has been proposed as a risk factor of various idiopathic diseases. The aim of this study was to investigate the possible role of oxidative stress in patients with BP.Methods:Thirty-two patients with BP and 30 healthy controls were included in this study. Serum total oxidant status (TOS), total antioxidant status (TAS) and oxidative stress index (OSI) were measured by the Erel method.Results:Serum TOS activities and OSI values were significantly higher in patients with BP compared with the control group (P<0.003 for all comparisons), whereas there was no significant difference between the groups in terms of TAS levels (P>0.05).Conclusions:The data suggest that oxidative stress is increased in BP. These results of high oxidative stress in patients with BP may be helpful to clarify the etiopathogenesis of BP and contribute to improvement in the management or prevention of the disease.

<p><strong>Background</strong></p><p>Diabetes mellitus is a chronic disease caused by lack of insulin production in the pancreas or by insulin resistance, the disease being characterized by elevated blood glucose levels. Hyperglycemia in diabetes could lead to oxidative stress due to the rise in 8-oxo-2'-deoxyguanosine (8-oxo-dG) levels and the decrease in levels of total antioxidant status (TAS). The purpose of this study was to assess the effect <em>Aloe vera </em>extract on 8-oxo-dG level and total antioxidant status in diabetic rat testis.</p><p><strong> </strong></p><p><strong>Methods</strong></p><p>This was an experimental laboratory study with 20 rat samples which were divided into 4 groups (1 control group and 3 treatment groups). Diabetes was induced in the rats by streptozotocin (STZ) at 65 mg/kgBW. The diabetic rats were then treated for 28 days with <em>Aloe vera</em> extract at 0 mg (P0), 200 mg rind (P1), 200 mg pulp (P2), respectively. The level of 8 -oxo-dG was measured by ELISA and total antioxidant status with 2,2' -azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS). Data were analyzed with ANOVA and Least Significant Difference Advanced Test at P&lt;0.05.</p><p><strong> </strong></p><p><strong>Results</strong></p><p>8-Oxo-dG levels were significantly different between the control group and both P0 and P2, but not between the control group and P1. Among the treatment groups the 8-oxo-dG levels were significantly different. Mean total antioxidant status was significantly different between control and treatment groups, and also between treatment groups (p&lt;0.05).</p><p><strong> </strong></p><p><strong>Conclusions</strong></p><p><em>Aloe vera</em> extract reduced free radicals (level of 8-oxo-dG) and increased the total antioxidant status in diabetic rat testis.</p><p> </p>

Tacrolimus (TAC) is an immunosuppressive drug, optimally used for liver, kidney, and heart transplant to avoid immune rejection. In retrospect, a multitude of studies have reported effects of TAC, such as nephrotoxicity, diabetes, and other complications. However, limited information is available regarding short-term exposure of TAC on the liver. Therefore, the present study was designed to unravel the effects of short-term exposure of TAC on a rat model. The animal model was established by TAC administration for 6, 12, 24, and 48 h time points. Liver histopathological changes were observed with PAS-D, reticulin stain, and immunostaining of PCNA and CK-7 coupled with glycogen quantification in a liver homogenate. TUNEL assay was performed to evaluate the DNA damage in the liver. Concentration of GSH and activities of SOD and CAT in the serum were measured to assess the antioxidant status, whereas liver tissue MDA level was measured as a biomarker of oxidative stress. Hepatic gene expression analysis of IL-10, IL-13, SOCS-2, and SOCS-3 was performed by RT-PCR. Results revealed marked changes in liver architecture of all TAC-treated groups, as evidenced by sinusoid dilation, hepatocyte derangement, glycogen deposition, and collapsed reticulin fibers. Significant increase in PCNA and CK-7 immunostaining along with the presence of TUNEL-positive cells was revealed in treatment groups as compared to the control group. Serum antioxidant enzyme status was markedly decreased, whereas the liver MDA level was increased in TAC treatment groups indicating oxidative stress induction. The gene expression profile of cytokines was significantly upregulated in treatment groups highlighting an inflammatory response. In conclusion, results of the current study propose that even a short-term TAC exposure can induce change in antioxidant status and lipid peroxidation. Therefore, these factors should be considered to avoid and minimize immunosuppression-related issues in a prolonged course of treatment.

Background: The role of minerals in preventing the generation of oxidative stress in obese individuals has been evaluated. Magnesium is an antioxidant nutrient and a cofactor of enzymes involved in the cell membrane stabilization, attenuating the effects of oxidative stress. Objective: To evaluate the association between magnesium and concentrations of thiobarbituric acid reactive substances (TBARS) in patients with obesity and eutrophic women. Methods: A cross-sectional study was conducted with 73 women, divided into two groups: case group (patients with obesity, n=27) and control group (eutrophic women, n=46). Measurements of body mass index and waist circumference were performed. Dietary magnesium intake was assessed by the three-day food record using the NutWin software. Urinary magnesium concentration was measured by atomic absorption spectrophotometry method. Plasma concentrations of thiobarbituric acid reactive substances (TBARS) were also determined. Results: Mean values of dietary magnesium intake were 161.59 ± 60.04 and 158.73 ± 31.96 for patients with obesity and control group, respectively, with no significant difference between the groups studied (p >0.05). The value of urinary excretion of magnesium was lower than the reference values in both groups, with no significant difference between the groups studied (p >0.05). The plasma concentration of thiobarbituric acid reactive substances was significantly higher in patients with obesity compared to the control group (p <0.001). There was no correlation between levels of magnesium biomarkers and the concentration of TBARS (p >0.05). Conclusion: Patients with obesity showed a reduced dietary magnesium intake which seems to induce hypomagnesuria as a compensatory mechanism. The marker of oxidative stress evaluated in this study was not influenced by magnesium.

Background: The activation of Nrf2/HO-1 pathway has been shown to protect against cisplatin- induced nephrotoxicity by reducing oxidative stress. Berberine (Ber), an isoquinoline alkaloid, has demonstrated antioxidant, anti-inflammatory and anti-apoptotic activities in various experimental models. Aim: To check the effect of Ber on cisplatin-induced nephrotoxicity and to explore the involved mechanism. Methods: Adult male Wistar rats were divided into 6 groups: Normal, cisplatin-control, treatment groups and per se group. Normal saline and Ber (20, 40 and 80 mg/kg; p.o.) was administered to rats for 10 days. A single intraperitoneal injection of cisplatin (8 mg/kg) was injected on 7th day to induced nephrotoxicity. On 10th day, rats were sacrificed, the kidney was removed and stored for the estimation of various parameters. Results: As compared to cisplatin-control group, Ber pretreatment improved renal function system and preserved renal architecture. It also diminished oxidative stress by upregulating the expression of Nrf2/HO-1 proteins. In addition, Ber attenuated the cisplatin mediated inflammation and apoptosis. Furthermore, it also reduced the phosphorylation of p38/JNK and PARP/Beclin-1 expression in the kidney. Conclusion: Ber attenuated renal injury by activating Nrf2/HO-1 and inhibiting JNK/p38MAPKs/ PARP/Beclin-1 expression which prevented oxidative stress, inflammation, apoptosis and autophagy in renal tissue.
