Effects of Na2CO3 on seed germination, seed reserve utilizationand seedling growth in bitter vetch (Vicia ervilia L.)

Alkalinity is one of the main limiting factors of seed germination in alkaline soils. The aim of this study was to evaluate the effects of Na2CO3 levels (0, 5, 10, 15, 20 and 25 mM) on seed germination, seed reserve utilization and seedling vigour of three local bitter vetch cultivars (Buinzahra, Shahreza and Tarom). The results indicated that increasing Na2CO3 concentrations caused a decrease in germination percentage (GP), germination rate (GR), seedling length, seedling dry weight (SLDW) and seed reserve utilization efficiency (SRUE). The rate of reduction in shoot length in comparison with the control in different concentrations of Na2CO3 was detected from Buinzahra as 30-92%, Shahreza 39-97% and Tarom 13-83%. Decrease in root length was more than shoot length in all the cultivars at each Na2CO3 level, as compared to the control. The weight of mobilized seed reserve (WMSR) and seed reserve depletion percentage (SRDP) decreased with increasing Na2CO3 levels up to 10 mM in Buinzahra and Tarom cultivars and up to 15 mM in Shahreza. WMSR and SRDP then rapidly raised with higher alkalinity levels. These results suggest that reduction of seed germination and decline in seedling dry weight in response to alkalinity stress is a consequence of decline in seed reserve utilization efficiency.

Abstract A laboratory experiment regarding germination and seedling growth test was conducted with three black gram genotypes tested under three salinity levels (0, 75 and 150 mM), for 10 days, in sand culture within small plastic pot, to investigate the germination and seedling growth characteristics. Different germination traits of all black gram genotypes, like germination percentage (GP), germination rate (GR), coefficient of velocity of germination (CVG) greatly reduced, as well as mean germination time (MGT) increased with increasing salt stress. At high salt stress, BARI Mash-3 provided the highest GP reduction (28.58%), while the lowest was recorded (15.79% to control) in BARI Mash-1. Salinity have the negative impact on shoot and root lengths, fresh and dry weights. The highest (50.32% to control) and lowest reduction (36.39%) of shoot length were recorded in BARI Mash-2 and BARI Mash-1, respectively, under 150 mM NaCl saline conditions. There were significant reduction of root lengths, root fresh and dry weight, shoot length, shoot fresh and dry weight in all genotypes under saline condition. The genotypes were arranged as BARI Mash-1 > BARI Mash-3 > BARI Mash-2, with respect to salinity tolerance.

The continuous rise in global population demands more food production under limited land resources accompanied by climate change. More oversupply of quality seeds to meet ever-growing food demand is a highly challenging task. Seeds with low vigour level limit the growth and yield of crops at varied field conditions. Hence, an experiment was conducted to develop a technique for the fast and homogeneous growth of kabuli chickpea seeds. The priming treatments evaluated in the present study enhanced the various seed quality parameters of kabuli chickpea significantly when compared to control. Hydro-priming for 4 hours (at 20°C) and air drying for 72 hours showed better results compared to rest of the treatments both in fresh seeds which recorded higher first count (66.38%), germination (92.50%) root length (11.85 cm), shoot length (6.16), mean seedling length (18.01), dry seedling weight (747 mg), seedling vigour index-I (1665), higher speed of emergence (0.3992) and old seeds with higher first count (61.88%), germination (74.13%) root length (8.15 cm), shoot length (5.85 cm), mean seedling length (14.00 cm), seedling dry weight (611 mg), seedling vigour index-I (1056), higher speed of emergence (0.3814). The aged seeds have recorded 29 % increase in germination percentage in contrast to 8 percent increases in fresh seeds. Kabuli chickpea seeds could be subjected to hydropriming, an affordable, alternative, and eco-friendly technique for improving seed and seedling vigour of kabuli chickpea.

The problem of generative propagation of soursop is its seeds take a long time to germinate which are caused by the hard seed coat. The solution to solve that problem is scarification and coconut water soaking to simply the process of imbibition and seed germination and also stimulate the growth of shoot. This objective of the research was to determine the effect of the scarification position and the soaking with various concentrations of coconut water on seed germination and growth of soursop seedling. This research was held at the Faculty of Agriculture field, the University of Sumatera Utara in March - July 2017. The research used a factorial randomized block design with two factors and three replication. The first factor is the scarification position (without scarification, stomach scarification, top scarification) and the second factor is coconut water soaking (0%; 25%; 50%; 75 %). The result of the research showed that the germination rate parameter, germination percentage, crop height, number of leaves, root volume and dry weight of crop significantly higher on without scarification treatment than with scarification treatment. The interaction between the scarification position and soaking with various coconut water concentrations was not significantly affecting the seed germination and growth of soursop seedling.

The effect of seed priming with calcium nitrate Ca (NO3)2 and potassium nitrate (KNO3) on germination and seedling vigour were studied in papaya varieties. Open pollinated local (gauty) papaya seeds were soaked in Ca (NO3)2 and KNO3 solutions (10000 ppm, 15000 ppm and 20000 ppm) up to 24 hours and germination percentage and seedling characteristics were recorded. The least number of days taken for seed germination (4.33 days), the highest germination percentage (82.56 % ), the highest shoot length (14.31 cm) the highest fresh biomass (1.36 g) and dry biomass (0.174 g) were recorded in 10000 ppm   Ca(NO3)2 treatment. Further, seeds of papaya varieties viz., Arka Surya, Arka Prabhat and Madhu bindhu were treated with 5000 ppm, 10000 ppm and 15000 ppm Ca (NO3)2 and observed that Arka Prabhat seeds treated with 10000 ppm Ca (NO3)2 hadtaken the least number of days for germination (4.75 days) and also the highest shoot length (25.2 cm). The results of the experiment proved the significant effect of calcium ions over potassium ions on papaya seed germination and seedling vigour.

A bioassay run was carried out in the incubator to evaluate possible allelopathic effects of water extracts of sage and white wormwood on germination and seedling growth of purslane. Results showed that the type of extract and its concentration (0, 5, 10, 15 and 20%) significantly influenced final germination percentage, germination rate and index, root and shoot length, root/shoot ratio, fresh and dry weight of seedling of purslane. The interaction between these two experimental factors was always significant, producing different results according to the different combination levels. The statistical comparison of means indicated that the maximum germination percentage and germination rate (respectively 68% and 11.4% d-1) were obtained from the untreated control (0% extract), while the lowest values for the same two characters occurred with 15% of sage (respectively 45% and 6.4% d-1) and 20% of white wormwood (38% and 7.5% d-1). Concerning germination indexes, the highest (55) and the lowest (32) values were observed respectively on the control and with 15% of sage extract or 20% of white wormwood extract. The longest (4.5 cm) and the shortest (1.3 cm) root of purslane were obtained with 5% of white wormwood and 20% of sage extracts, respectively. Changes in shoot length with sage and white wormwood extracts were similar to those in root length, even though shoot length was less affected by the concentration of extracts. Root and shoot length changes brought to maximum (22.0) and minimum (8.9) values for the root/shoot length ratio, respectively with 5% of white wormwood and 20% of sage extracts. The maximum fresh (2.111 g) and dry (0.338 g) seedling weight of purslane were obtained from untreated control, producing the same seedling weight with 5% of sage, 5 and 10% of white wormwood extract. While the minimum fresh (0.692 g) and dry (0.111 g) seedling weight were obtained from 15% of white wormwood extract, so that there is no significant differences between this value and the seedling weight produced under 10, 15 and 20% of sage and 20% of white wormwood extract.

Abstract In this study, the effect of different alkaline concentrations (0, 10, 20, 30, 40, 50, 60 mM) on germination and biochemical characteristics of the two sesame (Sesamum indicum L.) cultivares (Dashtestan and GL-13) which are registered cultivars of Iran were investigated. The experiment was carried out in a completely randomized design with three replications. Results showed that, germination percentage, germination rate, shoot length and dry weight, root length and dry weight and K+ content decreased, whereas, malondialdehyde (MDA), proline, total soluble sugars and Na+ contents increased with increasing alkalinity stress. GL-13 cultivar had the least root and shoot length, proline and K+ content than Dashtestan.

<p>Salinity stress has negative effect on seed germination; therefore, it is necessary to find technology to mitigate it. The research was conducted to study the potency of peel extract of dragon fruit to mitigate salinity stress on soybean seed germination. This research used a randomized block design with factorial patterns and three replications. The first factor was the level of salinity of NaCl (C), consisted of 3 levels (c<sub>0</sub> = 0%, c<sub>1</sub>=0.5%, c<sub>2</sub>=1%), The second factor was peel extract of dragon fruit (I), consisted of 3 levels (i<sub>0</sub> = 0% (control), i<sub>1 </sub>= 1%, and i<sub>2</sub>=2%). The parameters analyzed were germination percentage, germination rate, root length, epicotyl length, electric conductivity, and dry weight of sprout. The data were analyzed with ANOVA and Duncan’s multiple range test at α = 5 %. The result showed no interaction between salinity stress and peel extract of dragon fruit on soybean seed germination. Salinity stress significantly decreased sprout vigor with the highest decrease was shown at 1% salinity; on contrarily peel extract of dragon fruit could maintain sprout vigor. Peel extract of dragon fruit 2% was the potential to mitigate salinity stress.</p>

The influence of compost (derived from MSW and biosolids) maturity on seed germination of several weed species was evaluated. A bioassay was developed by extracting 20 g of compost of different maturities with various volumes of water, then measuring germination percentage of ivyleaf morningglory (Ipomoea hederacea) seeds placed on extract-saturated filter paper in a petri dish. A 20 g (dry weight) compost: 50 mL of water generated an extract that produced the widest percentage seed germination variation in response to composts of different maturity. Ivyleaf morningglory, barnyardgrass (Echinochloa crus-galli L.), purslane (Potulaca oleracea L.), and corn (Zea mays L) were selected as plant indicators to determine the compost maturity stage with maximum germination inhibition. Compost 8-week-old decreased percent germination, root growth, and germination index (combines germination rate and root growth), and increased mean days to germination (MDG) of each plant indicator. Immature 8 week-old compost extract effect on MDG and germination percent of 15 weed species was evaluated. Extract from 8-week-old compost inhibited germination in most weed species, except yellow nutsedge (Cyperus esculentus). Compost extracts derided from immature (3-day, 4-, and 8-week-old) compost resulted in delayed and reduced germination percent of important economic weed species.

An experiment was conducted to investigate the effect of different levels of cadmium (Cd) stress on germination, morphology, physiology and yield of rice. There were five treatments, viz. control (no Cd), 0.5 mM CdCl2, 1 mM CdCl2, 1.5 mM CdCl2 and 2 mM CdCl2, carried out by a randomized complete block design (RCBD) with three replications. Cadmium stress caused a significant reduction of plant height and tillers hill-1 in a dose-dependent manner. The highest decline of relative water content and SPAD value was found at 1.5 mM (11 and 16%) and 2 mM (14 and 19%) Cd stress compared to the control. The number of effective tillers hill-1, filled grains panicle-1, 1000-grain weight, length of panicle, grain yield and straw yield reduced significantly by 44, 32, 15, 24, 37 and 45%, respectively, over the control. Germination and seedling-related parameters were recorded 10 days after germination. The lowest germination percentage (27%), germination index (27%) and seedling vigour index (49%) were observed at the highest level of Cd stress in comparison to the control. Cadmium stress resulted in maximum abnormal seedlings and markedly reduced length of root and shoot as well as their fresh and dry weight. Root shoot ratio showed differential responses to Cd stress. Thus, it can be concluded that a higher level of Cd in soil has a detrimental effect on seed germination, morphology, physiology and yield of rice.

Abstract Seed germination and seedling growth are critical stages in the life cycle of a plant, especially under adverse abiotic stresses. Seed germination negatively affected by stress conditions. Seed priming techniques have been used to increase germination characteristics and improve germination uniformity in more field crops under stressed conditions. This experimental aimed to evaluate the effect of salinity and halo-priming on seed reserve utilization and seed germination of wheat seeds. For create salinity stress, NaCl in osmotic levels at 0 (as control), -4, -8, - 12 and -16 bar was used. Seeds of were pretreated with halo-priming at 10 °C for 24 h and water at 10 °C for 24 h. Our results showed that treatment×drought interaction on these traits: germination percentage, weight of utilized (mobilized) seed, seed reserve utilization efficiency, seedling dry weight and seed reserve depletion percentage were significant. The highest germination percentage was obtained from halo priming in control conditions. Thus priming improved study traits in wheat under salinity stress. Priming increased germination percentage and seed reserve utilization as compared to the unprimed seeds. The highest germination percentage and seed reserve utilization were as obtained from halo priming in control conditions.
