<em>Carica pubescens Lenne &amp; K. Koch is one of the typical highland crops in Indonesia. This plant can be found in the region of Cangar and Bromo in East Java, as well as the Dieng Plateau, Central Java Wonosobo-Banjarnegara. Morphological, chemical content, and analysis of protein banding pattern of the Carica pubescens has been done, but more about the use of active compounds for pharmaceutical raw materials, in Diabetes Mellitus type 2 have not been studied. One therapeutic approach to lowering blood sugar levels posprandial is to slow down or inhibit the absorption of glucose through inhibition of the carbohydrates hidrolitic enzyme such as α-amylase and α-glucosidase. This study aims to identify antidiabetic compounds in silico. Antidiabetic compounds were analyzed by methods of docking. The results showed that papain and cysteine protease in C. pubescens have better binding affinity to α-amylase and α-glucosidase compared to iridoid glycosides. Papain and cysteine proteases isolated from C. pubescens are potential to be developed as an antidiabetic.</em>

Pharmaceutical industries should apply rigorous QC (quality control) to ensure the consistency, safety, and efficacy of their herbal derived drug-preparations. QC must be performed at every stage of the production line i.e. incoming raw materials, extractions, in-process control, finished products and keeping samples. Due to the complex nature of the chemical content of herbal drugs, two approaches to QC should be taken, that is quantitative determination of the selected marker(s) compound(s), and metabolite profiling. Contamination of herbal medicines by heavy metals, pesticides, toxic metabolites, microbial toxins, pathogenic microorganisms and other foreign matter should also be evaluated. A combination of chemical profiling and multivariate analysis (MVA) is recommended as the QC tool for the botanical identification method (BIM) of herbs, extracts, herb materials, and herbal drug preparations. Microscopic methods, DNA profiling or chemical marker(s) are not recommended for use as the sole BIM due to the lack of specificity. Only markers that meet certain criteria i.e. quality active (QA) markers can be utilized as a QC tool. The limit specification range of markers used as QC tools should be described in the analytical target profile (ATP). To gain reliable results of any analysis that has been performed at any QC laboratory, the analysis method must be validated according to the newest guidance. Sample detection limit of any toxic compound(s) should be lower than its cut-off value and MPL. The reliability of any results of analysis of a QC laboratory must be evaluated by using QC-samples for each series of measurements.

In modern conditions of meat production, one of the priorities of the meat processing industry is the production of products with improved consumer properties. To perform this task, it is necessary to include in the industrial circulation of physiologically functional ingredients of plant origin, which will provide the body with nutrients and expand the range of functional foods. The paper covers the issues of partial replacement of raw materials of animal origin with germinated sea-buckthorn seeds (GSBS) flour as a functional ingredient in the production of sausages. The chemical and vitamin composition of the physiologically functional ingredient has been studied. Three experimental recipes for sausages from GSBS flour has been developed. The positive influence of GSBS flour on functional-technological and physical-chemical properties of the received forcemeat is proved. Organoleptic parameters of sausages from GSBS flour are determined. The use of GSBS flour as a functional ingredient in the technology of sausages affects the appearance, taste, smell. It is established that the increase in the amount of GSBS flour introduction leads to an increase in pH by 0.4–0.8 % and finished products by 6.8 %. The improvement of functional and technological properties of experimental minced meat of sausages based on the use of GSBS flour is proved. The optimal amount of replacement of the main raw material with a physiologically functional ingredient, which is characteristic of the experimental sample № 2. The changes in the qualitative characteristics of the developed sample of sausages, which where within the regulatory requirements. The addition of 10 % GSBS flour to the minced meat of sausages leads to a slight increase in protein with a significant reduction in lipid content by 4.4 % in the finished product. As a result, the caloric content of Altaiska functional sausage is reduced by 13.2 % compared to traditional products. Production of such products will allow to expand the domestic range of functional products. Consumption of such sausages allows to consider them as foodstuff with essentially new functional properties which would correspond to modern requirements concerning food.

Introduction. All over the world, natural medicine uses species of the genus Melilotus due to their anti-inflammatory, anti-rheumatic, analgesic, spasmolytic, astringent, diuretic and anticoagulant effects. In the Polish Pharmacopoeia, only the yellow melilot herb (Meliloti herba) has a monograph. Aim. The research was aimed at a comparative analysis of the content of the sum of polyphenolic compounds, including the sum of flavonoids and phenolic acids, and antioxidant activity in water extracts of M. officinalis and M. alba herb and root. Material and methods. The research was carried out for two species of Melilotus officinalis and M. alba. The raw material was herb and melilot root, collected during flowering period from natural sites. In aqueous extracts from raw materials, the following methods were determined by colorimetric methods: the sum of polyphenolic compounds using the Folin-Ciocalteu reagent, the sum of phenolic acids using the Arnov reagent, the sum of flavonoids using the Christ-Müller method and the antioxidant activity using the 2,2-diphenyl-1-picryl-radical hydrazyl (DPPH). Results. Meliloti alba herb has a much higher content of polyphenols, including phenolic acids and flavonoids than M. officinalis. In the yellow sweet clover the sum of polyphenols in the samples ranged from 2.101 to 2.438%, while in the white sweet clover from 2.765 to 3.540%. The content of phenolic acids in the samples of yellow sweet clover herb was 0.617-0.766%, and in the samples of white sweet clover herb was 0.646-0.900%. The content of flavonoids in the samples of yellow sweet clover herb ranged from 0.748 to 0.975%, and in the case of white sweet clover herb it was at the level of 0.801-1.192%. The samples of the commercial yellow sweet clover herb had the lowest content of phenolic compounds; the sum of polyphenols was 1.579%, phenolic acids 0.361%, and flavonoids 0.441%. The determined content of total polyphenols in the roots of both melilot species was low and amounted to 0.450% in MORA1 and 0.362% in MARA2. Despite the lower content of polyphenolic compounds (2.438%), the yellow melilot showed higher antioxidant activity (IC50 = 4.35 mg/ml) compared to the white melilot herb (IC50 = 5.31) with a higher content of polyphenols (3.010%). Conclusions. A higher content of the sum of polyphenols as well as phenolic acids and flavonoids was found in the white melilot. In both the yellow and white sweet clover herb extract, significantly more polyphenolic compounds were determined compared to the content in their root extract.

Aim. The purpose of this work was to determine the dose of g- and X-ray pre-sowing irradiation of seeds of Hypericum perforatum L., causing an increase in biomass and the accumulation of flavonoids in medicinal raw materials. Methods. X-ray and g-irradiation of seeds, biometric methods, flavonoids extraction, quantification of flavonoid content in extracts using spectrophotometry, qualitative and semi-quantitative analysis of extracts using high-performance liquid chromatography (HPLC), common statistical methods for processing and analysis of data. Results. It was shown that acute X-ray pre-sowing irradiation of H. perforatum seeds at a dose of 20 Gy leads to an increase in the crop of medicinal raw material without losing its pharmaceutical value. Irradiation also increases concentration of quercetin, routine, hyperoside in herbal medicines, grown from seeds, that was chronically irradiated with g-radiation in a total dose of 1 Gy. The HPLC analysis of extracts confirmed that the qualitative composition of ethanol extracts of H. perforatum did not change due to the effect of irradiation on the seeds. Conclusions. The obtained results confirm the possibility of application of pre-sowing irradiation of seeds of H. perforatum in the range of 1–35 Gy in order to increase its productivity and increase the pharmaceutical value of the medicinal raw material. Keywords: Hypericum perforatum L., ionizing irradiation, productivity, flavonoids.

Abstract Diabetes mellitus continues to increase along with the increasing pattern of consuming ready-to-eat foods. The consequences of this habit will have a negative impact on the health of the body so that it can cause death. Treatment of diabetes mellitus is carried out in various ways, including administration of insulin and synthetic drug therapy. However, this medication has dangerous side effects. Therefore, research was carried out on Pelawan stems (Tristaniopsis merguensis Griff) which have the potential to be associated with secondary metabolites and bioactivity as antidiabetic so that they can be considered as raw material for herbal medicines in the future. total phenolic content of the methanol fraction of T. merguensis stems is 176.37 mg GAE/g DW. The total flavonoid content of the methanol fraction of T. merguensis stems is 9.85 mg QE/g DW. The results of the antidiabetic test for the methanol fraction of T. merguensis stems obtained an IC50 of 5.31 μg/mL. When viewed from the qualitative results of phenolics and flavonoids that have been carried out, it is likely that the activity of the α-glucosidase enzyme in this study is more influenced by polyphenolic compounds.

In today's society the continuously increasing consumption of raw materials and the associated impacts on the ecosystem tend to be a frequently discussed topic. Especially automobile companies are faced to develop new driving concepts due to the emerging energy turnaround. Usually the components of the conventional drive are replaced by an electric engine including the required energy storage. Without structural changes regarding the chassis this procedure causes an increase in the vehicle ́s weight (Conversion Design). Therefore a new approach is to integrate the battery as a load-bearing member in the vehicle structure and additionally use a weight-optimized multi-material design of the body (Purpose Design). By savings of 25% of the weight of a compact-class vehicle body, a resource-saving and energy-efficient design of the entire vehicle can be achieved. Certainly the innovative multi-material construction contributes significantly to reduce the total energy consumption of the vehicle during the use phase. Based on a Lifecycle-Assessment (LCA) the environmental sustainability of the Purpose Design will be evaluated and compared to the approach of the Conversion Design. In addition to the weight savings of the multi-material body secondary weight reductions regarding the energy storage will be taken into account. The aim is to assess the ecological advantages of the lightweight solution throughout the entire product life cycle comprising the extraction of raw materials, production of the components, use of the product and end of life including the recycling of components. However, these investigations will be carried out for the modified chassis and the lightweight constructed multi-material body. Hence, the processes of the individual life cycle phases will be collected, inventorial analysis carried out and impact assessments performed. According to the LCA it will be tested, if the additional expenses in raw material extraction, production and recycling of the lightweight body justify the expected ecological advantage in the use phase. A final overall analysis will provide information on the actual efficiency and sustainability of the Purpose Design. Due to the parallel creation of the LCA data during the development process the LCA results furthermore serve to detect and monitor significant shortcomings on component and assembly level.
