The aim of this study was to investigate lipolysis and lipid oxidation of stored eggs at different temperatures (4 and 22°C) by evaluating the changes in physicochemical index, lipid profiles, enzymatic activity, and oxidative index. The results showed that the changes in physicochemical index were more significant at 22°C than at 4°C. Weight loss, moisture content, and pH of egg yolk increased significantly (P < 0.05), whereas the yolk index decreased during storage. However, there was no significant difference in lipid profiles between 4 and 22°C storage temperature. The lipid composition analysis demonstrated that lipid hydrolysis took place during egg storage and resulted in a marked decrease of PL and increase of FFA. It was also found that the content of phosphatidylcholine (PC), phosphatidylethanolamine (PE), and phosphatidylinositol (PI) decreased significantly during storage. The correlation analysis showed that the lipid degradation is significantly positively related to lipase activity (P < 0.05), and the marked changes of lipid fractions are results of both hydrolysis and oxidation. It can be concluded that the egg physicochemical index and lipase activity were greatly influenced by temperature during storage, but the yolk lipid stability was not significantly influenced by storage temperature.

ABSTRACT Prompt refrigeration of eggs to prevent the multiplication of Salmonella Enteritidis to high levels during storage is an important practice for reducing the risk of egg-transmitted human illness. The efficacy of egg refrigeration for achieving this goal depends on the interaction among the location of contamination, the ability of contaminant strains to survive or multiply, and the rate at which growth-restricting temperatures are attained. The present study assessed the significance of several characterized genetic and phenotypic properties for the capabilities of 10 Salmonella Enteritidis isolates to multiply rapidly in egg yolk and survive for several days in egg albumen during unrefrigerated (25°C) storage. The growth of small numbers of each Salmonella Enteritidis strain (approximately 101 CFU/mL) inoculated into egg yolk samples was determined after 6 and 24 h of incubation. The survival of larger numbers of Salmonella Enteritidis (approximately 105 CFU/mL) inoculated into albumen samples was determined at 24 and 96 h of incubation. In yolk, the inoculated Salmonella Enteritidis strains multiplied to mean levels of approximately 102.6 CFU/mL after 6 h of incubation and 108.3 CFU/mL after 24 h. In albumen, mean levels of approximately 104.6 CFU/mL Salmonella Enteritidis were maintained through 96 h. The concentrations of the various Salmonella strains after incubation in either yolk or albumen were distributed over relatively narrow ranges of values. Significant (P < 0.01) differences observed among individual strains suggested that maintenance of the fimbrial gene sefD may have positive genetic selection value by improving fitness to grow inside egg yolk, whereas the antibiotic resistance gene blaTEM-1 tet(A) appeared to have negative genetic selection value by decreasing fitness to survive in egg albumen.

The article studies the possibility to use glycerol as cryoprotectant, instead of dimethylsulfoxide for cryopreservation of sperm of inconnu ( Stenodus leucichthys Gueldenstaedtii, 1772). Investigations were carried out from 2015 to 2016 in the laboratory of the Southern Scientific Center, Russian Academy of Sciences, on the basis of the Astrakhan State Technical University. The material collected on the Alexander sturgeon hatcheries (the Astrakhan region) in the spawning period. Native sperm of 6 male inconnu species was used as a control means. The semen quality was determined in terms of moving activity (life time) of sperm after its activation by water. As the cryoprotectant there were used: base solution - 80%, sucrose - 1.71 g/l, mannite - 0.98 g/l, yolk - 10%, dimethylsulfoxide - 10% and base solution - 87%, sucrose - 1.71% g/l, mannite - 0.98 g/l, yolk - 10%, glycerol - 3 variants: 3; 5 and 10%. In order to provide the most complete penetration of cryoprotectants into the cells there were used electrostimulation of cell membranes. Equilibration time was 5 and 15 minutes. Thawing semen was performed in a water bath at a temperature of 38-40°C. For removing protectors from cells there was chosen a saline solution (0.7% NaCl) as isotonic solution. In tests using dimethylsulfoxide life activity of sex cells was 2 times lower than in tests with glycerol: 78 and 186.2 s at the end of equilibration and 52.3 and 128.9 s after thawing. Sperm showed maximum activity under 5% glycerol concentration during equilibration - 15 min. Concentration of 3% was insufficient, concentration of 10% was excessive, as it suppressed activity of sperm. Egg yolk which coagulated together with glycerol, making difficulty for observing, had to be excluded from the composition of cryoprotectant.
