Kappaphycus alvarezii is a type of seaweed containing numerous carbohydrate with biological activities, especially fucoidan. About chemical aspect, fucoidan is fucose polysaccharides (FCSPs) with diverse biological activities including antioxidant, anticoagulant, anti-inflammatory, antiviral, antitumor... Fucoidan is found not only in brown algae but also in K. alvarezii alga from the central coast of Vietnam. In this study, fucoidan purification, fucoidan structure and biological activities of K. alvarezii alga (collected at Dam Mon area, Khanh Hoa province) were investigated. The yield recovery of two-stage precipitation by ethanol 99% was 84.4%. The protocol of fucoidan purification by ion-exchange chromatography was applied with tris-HCl buffer, elution solvent NaCl 1M; The yield was 60.99%. The structure of fucoidan from K. alvarezii was determined by 1H-NMN spectrum with the specific characteristic properties of fucoidan. The antioxidant activity of the purified fucoidan via IC50 was 303.51±0.12 μg/mL (DPPH assay), 299.97±3.12 μg/mL (ABTS essay). The purified fucoidan was shown better ability in cases of anti-coagulant, antibacterial activities than that of the crude extract. The anti-coagulation time of the purified fucoidan was 22.52 minutes. The minimum inhibitory concentrations (MICs) of the antibacterial activity (Bacillus cereus, Escherichia coli) were 400μg/mL. MICs was 500 μg/mL (Aspergillus flavus) and 600 μg/mL (Aspergillus niger).

AbstrakTujuan penelitian adalah untuk mengetahui kandungan fitokimia dan aktivitas antioksidan pada bulir cendol rumput laut Kappahycus alvarezii dengan penambahan tepung sagu yang diekstraksi dengan pelarut methanol. Ekstrak diperoleh dengan cara maserassi terhadap bulir cendol rumput laut dan tepung sagu. Uji fitokimia dilakukan untuk mengetahui keberadaan senyawa alkaloid, flavonoid, saponin, streroid dan triterpenoid. Aktivitas antioksidan bulir cendol Kappaphycus alvarezii diuji dengan metode DPPH ( 1-1 difenil -2 pikrilhidrazil). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari tiga perlakuan yaitu perlakuan R1S1 (RL : 10% TS : 90%), R2S2 (RL : 20%: TS : 80%), R3S3 (RL 30 % TS : 70%) dan ulangan sebanyak tiga kali. Data hasil pengamatan dianalisa menggunakan ANOVA (Analysis of Variance) pada taraf  95%, apabila terdapat beda nyata (P>0,05) maka dilakukan uji lanjut dengan uji DMRT (Duncan Multiple Range Test) pada taraf nyata 95%. Hasil penelitian mempelihatkan terdeteksi senyawa alkaloid, flavonoid, streroid dan triterpenoid sedangkan pada senyawa saponin tidak terdekteksi pada ekstrak metanol cendol Kappaphycus Alvarezii. Uji aktivitas antioksidan dengan metode DPPH menunjukan nilai IC50 terendah terdapat pada perlakuan R1S1 dengan jumlah IC50 652 ppm. Hal ini menunjukan bahwa adanya aktivitas antioksidan yang sangat lemah dari cendol  Kappaphycus alvarezii yang diekstraksi. Kata Kunci :    Aktivitas antioksidan, cendol, Kappaphycus alvarezii, Metroxylon sagus Rottb, fitokimia  AbstractThe aims  of this study was to determine the phytochemical content and antioxidant  on cendol seaweed (Kappaphycus alvarezii) with additions of sago flour (Metroxylon sagus Rottb.) extracted with methanol. Phytochemical tests were performed to determine the presence of alkaloids, flavonoids, saponins, triterpenoids and steroids. Antioxidant activity of seaweed cendol. Kaphaphycus alvarezii was tested by DPPH method (1-1- diphenyl-2-picrylhydrazl). This study used a Completely Randomized Design (CRD) which consists of three treatments R1S1( RL 10% : TS 90% ), R2S2 (RL 20% : TS 80%), R3S3 ( RL 30% : TS : 70%) and three times replication. Observation data were analyzed using ANOVA (Analysis of Variance) at the level of 95%, if there were significant differences (P> 0.05) then further testing was carried out with DMRT (Duncan Multiple Range Test) at 95% significance level. Phytochemical test showed there was compounds detected with alkaloids, flavonoids, triterpenoids and steroids. While the saponins compounds was not detected in methanol extract of Kappahycus alvarezi. The antioxidant activity by DPPH method showed that the high IC50 values is 652 ppm. Oral infections (gingivitis and periodontitis) and oral cancer are under rise in developing countries. Products with antibacterial and antioxidant activity can provide a combined approach to treat oral disorders. Marine algae is a reservoir of rich bioactive phytochemicals and are considered to be potential candidates in natural pharmaceutics. Kappaphycus alvarezii is a marine algae widely cultivated for food applications. The current study investigates the phyto-pharmacological properties of K. alvarezii for oral diseases. Different polarity solvents (ethanol, ethyl acetate and chloroform) were used in the extraction of bioactive components of K. alvarezii, partially characterized by GCMS and studied for their antioxidant, antimicrobial and cytotoxic activity. All the K. alvarezii extracts exhibited good antioxidant activity and potential efficacy against oral pathogenic microbes. Although K. alvarezii extracts were found to be safe for normal Vero cells, their inhibitory activity on oral cancer cells (KB-3-1 cell lines) was found to be low. These findings have suggested the possibility of K. alvarezii using in the dental preparation/product to combat oral infections.

Conventional techniques such as solvent extraction can be effectively used to extract free phenolic compounds in plants. However, this method is inefficient to extract bound phenolics. Solid state fermentation (SSF) approach with Aspergillus oryzae was used to enhance bioavailability of polyphenols in Kappaphycus alvarezii. A set of experiment was computed by face centered central composite design (FCCCD) to optimize the fermentation parameters based on maximum phenolic content and antioxidant activity. Four independent variables namely: time (0, 4 and 8 days), temperature (28, 30 and 32 °C), initial moisture content (60, 70 and 80 %) and inoculum level (10, 20 and 30 % (v/v)) were investigated. The experimental results for both TPC and DPPH were 9.449 ± 0.198 mg GAE/g and 87.135 ± 0.857 % of scavenging activity, respectively; where both responses were in good agreement with RSM model prediction. The RSM design used has been proven to successfully predict the total phenolic content and antioxidant activity. Fermentation condition with 70% initial moisture content, 10% (v/v) inoculum level, performed at 30oC for 4 days was found to produce maximum TPC and DPPH radical scavenging activity of Kappaphycus alvarezii.

The objective of the study was to determine the phytochemical content and antioxidant activity of Kappaphycus alvarezii seaweed extracted with methanol solvent. The extract was obtained by maceration of dried seaweed. Phytochemical tests were performed to determine the presence of alkaloids, flavonoids, saponins, triterpenoids and steroids. Kaphaphycis alvarezii seaweed antioxidant activity was tested by DPPH method (1-1-diphenyl-2-picrylhydrazyl). A phytichemical test revealed that there was not any alkaloids, flavonoids, saponins, steroids and triterpenoids content in methanol extract Kappaphycus alvarezi. The antioxidant activity by DPPH method showed the high IC50 value (163.82 ppm) It is indicated that the antioxidant activity of Kappaphycus alvarezii of dried seaweed extracted with methanol was very weak. The negative result from phytochemical test was speculated caused by the drying process of seaweed samples, the type of seaweed itself, and the insufficiency of desalting process. Hasil penelitian memperlihatkan tidak terdeteksinya senyawa alkaloid, flavonoid, saponin, steroid dan triterpenoid pada ekstrak metanol Kappaphycus alvarezi. Uji aktivitas antioksidan dengan metode DPPH menunjukkan nilai IC50 sebesar 163.82 ppm. Hal ini menunjukkan bahwa aktivitas antioksidan rumput laut Kappaphycus alvarezii kering yang diekstraksi dengan metanol sangat lemah. Tidak terdeteksinya kandungan fitokimia diduga karena sampel rumput laut dikeringkan dengan sinar matahari langsung, jenis rumput laut ini tidak memiliki aktivitas antioksidan yang tinggi dan proses desalting yang tidak maksimal. Aktivitas antioksidan yang sangat lemah dari rumput laut ini diduga berasal dari kandungan asam lemak tak jenuh

Abstract. In this study, cytoprotective and antioxidant activities of Rosa canina (RC) and Salix alba (SA), medicinal plants, were studied on mouse primary splenocytes by comparing Controlled Differential Sieving process (CDSp), which is a novel green solvent-free process, versus a conventional technique, employing hydroethanolic extraction (HEE). Thus, preventive antioxidant activity of three plant powders of homogeneous particle sizes, 50–100 µm, 100–180 µm and 180–315 µm, dissolved directly in the cellular buffer, were compared to those of hydroethanolic (HE) extract, at 2 concentrations (250 and 500 µg/mL) in H2O2-treated spleen cells. Overall, compared to HE extract, the superfine powders, i. e., fractions < 180 µm, at the lowest concentration, resulted in greater reactive oxygen species (ROS) elimination, increased glutathione peroxidase (GPx) activity and lower malondialdehyde (MDA) production. Better antioxidant and preventive effects in pre-treated cells were found with the superfine powders for SA (i. e., 50–100 µm and 100–180 µm, both p < 0.001), and with the intermediate powder for RC (i. e., 100–180 µm, p < 0.05) versus HE extract. The activity levels of catalase (CAT) and superoxide dismutase (SOD) in pretreated splenocytes exposed to H2O2, albeit reduced, were near to those in unexposed cells, suggesting that pretreatment with the fine powders has relatively restored the normal levels of antioxidant-related enzymes. These findings supported that CDSp improved the biological activities of plants, avoiding the use of organic solvents and thus it could be a good alternative to conventional extraction techniques.
