Antioxidant activity of Kappaphycus alvarezii and Sargassum polycystum seaweeds extracts

Fucoxanthin has great potential to be developed as an antioxidant. The purposes of this study were to determine the effect of acetone solvent concentration and maceration temperature on the fucoxanthin content and antioxidant activity of brown algae extract (Sargassum polycystum) and determine the best acetone solvent concentration and maceration temperature on the fucoxanthin content and antioxidant activity of brown algae extract (Sargassum polycystum) . The study was the experimental research which designed by using randomized block desin (RBD) with two factors. The first factor was solvent concentration that consists of 70, 80 and 90 percent. The second factor was maceration temperature that consists of 30±2?, 40±2? and 50±2?. The data were analyzed by analysis of varian (ANOVA) and continued with the Tukey test. The results showed that the concentration of acetone solvent was influential on the content of fucoxanthin and antioxidant activity but did not affect the yield. Maceration temperature is influential on yield, fucoxanthin content and antioxidant activity. Interactions between treatments of acetone solvent concentration and maceration temperature greatly affect the content of fucoxanthin and antioxidant activity but do not affect the yield. The treatment of acetone 80 percent solvent concentration and maceration temperature 30±2? is the best treatment to produce brown algae extract (Sargassum polycystum) with a yield of 3.24 ± 0.45 percent, fucoxanthin content 1.03 ± 0.01 g fukosantin/g and antioxidant activity 99 ± 8.49 ppm and show activity powerful antioxidant. <p>Research on the addition of antioxidants to yogurt with ethanolic extract of<br />Sargassum polycystum has been done. This study aims were to determine the effect of<br />ethanolic extract of S. polycystum into the yoghurt to the antioxidant activity and the<br />level of consumer acceptance as well as determine the effect of ethanolic extract of<br />S. polycystum of chemical composition on yoghurt. S. polycystum was extracted using<br />ethanol 96%. The ethanolic extract were inforfified into yoghurt drink and tested the<br />antioxidant activity and the chemical compositionas well as consumer acceptance test.<br />Fermented yoghurt fortified with ethanolic extract of S. polycystum of 0.00; 0.22; 0.44;<br />0.66, 0.88, and 1.10%. The results showed that the addition of ethanolic extract was not<br />significant (p&gt;0.05) on pH, but significant (p&lt;0.05) to total lactic acid, antioxidant activity,<br />chemical composition (proximate), and consumer acceptance.<br />Keywords: Antioxidant activity, ethanol cextract, Sargassum polycystum, consumer<br />acceptance, yoghurt</p>

Sargassum polycystum is a low-sodium salt raw material that met the dietary salt criteria, i.e., low Na: K ratio and NaCl content < 60%. This study aims to produce seaweed salt with high yield, dietary fiber content, and antioxidant activity according to the quality standard of dietary salt through a combination of the filtrate and salt extraction residue. The results showed that the produced S. polycystum brown seaweed salt had a mineral Na 96.97 mg/g and mineral K 247.59 mg/g. Na: K ratio of 0.39, NaCl content of 49.05%, and heavy metal residue below the Indonesian National Standard maximum standard for dietary salt. The levels of dietary fiber produced in salt samples 1:1, 1:2, and 2:1 were 34.41%, 40.16%, and 23.83%, respectively, and the iodine content (KIO3) of the control salt was 125.95 mg/kg. The antioxidant activity of seaweed salt 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) on salt K (seaweed salt without residue), residue, S1 (1:1), S2 (1:2) and S3 (2:1) was 26.41 ppm, 59.06 ppm, 12.29 ppm, 67.30 ppm and 87.50 ppm. Meanwhile, the antioxidant capacity of seaweed salt Cupric Reducing Antioxidant Capacity (CUPRAC) on K salt, residue, S1, S2 and S3 were 12.36, 19, 7.86, 8.52 and 12.27 μmol ascorbic acid/g extract, respectively. Therefore, it was concluded that increasing the yield and dietary fiber content in the seaweed salt production process is achievable by adding residues.

Instant drink of ethanolic extract Sargassum polycystum with ginger flavor was made from seaweds which was processed into powder and have undergone processes of drying, extraction, cocrystalization, and sieving into powder. The objective of this research was to produce a formula of the seaweed instant drink which has a high antioxidant activityand is preferred by consumers. The formula that was used in this research waere ethanolic extract of S. polycystum, sugar and ginger. The compostion of formula including control (0:3:1), formula 1 (0.1:2.9:1), formula 2 (0.2:2.8:1), formula 3 (0.3:2.7:1), formula 4 (0.4:2.6:1), formula 5 (0.5: 2.5:1), and formula 6 (0.6:2.4:1). The result showed that the instant drink contained the value of antioxidant activity by 25.70-65.41%, total fenol of 27.74-100.36 mg GAE/gram, water levels of 0.44-1.99%, ash levels of 0.06-11.54% and the value of hedonic color 2,4-3,2(don’t like – like); hedonic flavor 1.29-3.66 (very don’t like – like); and hedonic taste 1.32-3.59 (very don’t like – like).Keywords: Instant drink, Sargassum polycystum, ginger, antioxidant ABSTRAKMinuman instan ekstrak Sargassum polycystum rasa jahe merupakan produk olahan rumput laut dalam bentuk serbuk yang sebelumnya telah mengalami proses pengeringan, ekstraksi, kokristalisasi dan pengayakan menjadi serbuk. Formula yang digunakan pada penelitian ini terdiri dari ekstrak etanolik S. polycystum, gula dan jahe dengan komposisi kontrol (0:3:1), formula 1 (0,1:2,9:1), formula 2 (0,2:2,8:1), formula 3 (0,3:2,7:1), formula 4 (0,4:2,6:1), formula 5 (0,5:2,5:1), dan formula 6 (0,6:2,4:1). Hasil penelitian menunjukkan bahwa minuman instanmempunyai nilai aktivitas antioksidan 25,7-65,41%, total fenol 27,74-100,36 mg GAE/gram, total gula 76,13-87,18%, kadar air 0,44-1,99%, kadar abu 0,06-11,54% dan nilai hedonis warna 2,4-3,2 (tidak suka – suka); aroma 1,29-3,66 (sangat tidak suka – suka); dan rasa 1,32-3,59 (sangat tidak suka – suka).Kata kunci: Minuman instan, Sargassum polycystum, jahe, antioksidan

Kappaphycus alvarezii is a type of seaweed containing numerous carbohydrate with biological activities, especially fucoidan. About chemical aspect, fucoidan is fucose polysaccharides (FCSPs) with diverse biological activities including antioxidant, anticoagulant, anti-inflammatory, antiviral, antitumor... Fucoidan is found not only in brown algae but also in K. alvarezii alga from the central coast of Vietnam. In this study, fucoidan purification, fucoidan structure and biological activities of K. alvarezii alga (collected at Dam Mon area, Khanh Hoa province) were investigated. The yield recovery of two-stage precipitation by ethanol 99% was 84.4%. The protocol of fucoidan purification by ion-exchange chromatography was applied with tris-HCl buffer, elution solvent NaCl 1M; The yield was 60.99%. The structure of fucoidan from K. alvarezii was determined by 1H-NMN spectrum with the specific characteristic properties of fucoidan. The antioxidant activity of the purified fucoidan via IC50 was 303.51±0.12 μg/mL (DPPH assay), 299.97±3.12 μg/mL (ABTS essay). The purified fucoidan was shown better ability in cases of anti-coagulant, antibacterial activities than that of the crude extract. The anti-coagulation time of the purified fucoidan was 22.52 minutes. The minimum inhibitory concentrations (MICs) of the antibacterial activity (Bacillus cereus, Escherichia coli) were 400μg/mL. MICs was 500 μg/mL (Aspergillus flavus) and 600 μg/mL (Aspergillus niger).

AbstrakTujuan penelitian adalah untuk mengetahui kandungan fitokimia dan aktivitas antioksidan pada bulir cendol rumput laut Kappahycus alvarezii dengan penambahan tepung sagu yang diekstraksi dengan pelarut methanol. Ekstrak diperoleh dengan cara maserassi terhadap bulir cendol rumput laut dan tepung sagu. Uji fitokimia dilakukan untuk mengetahui keberadaan senyawa alkaloid, flavonoid, saponin, streroid dan triterpenoid. Aktivitas antioksidan bulir cendol Kappaphycus alvarezii diuji dengan metode DPPH ( 1-1 difenil -2 pikrilhidrazil). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari tiga perlakuan yaitu perlakuan R1S1 (RL : 10% TS : 90%), R2S2 (RL : 20%: TS : 80%), R3S3 (RL 30 % TS : 70%) dan ulangan sebanyak tiga kali. Data hasil pengamatan dianalisa menggunakan ANOVA (Analysis of Variance) pada taraf  95%, apabila terdapat beda nyata (P>0,05) maka dilakukan uji lanjut dengan uji DMRT (Duncan Multiple Range Test) pada taraf nyata 95%. Hasil penelitian mempelihatkan terdeteksi senyawa alkaloid, flavonoid, streroid dan triterpenoid sedangkan pada senyawa saponin tidak terdekteksi pada ekstrak metanol cendol Kappaphycus Alvarezii. Uji aktivitas antioksidan dengan metode DPPH menunjukan nilai IC50 terendah terdapat pada perlakuan R1S1 dengan jumlah IC50 652 ppm. Hal ini menunjukan bahwa adanya aktivitas antioksidan yang sangat lemah dari cendol  Kappaphycus alvarezii yang diekstraksi. Kata Kunci :    Aktivitas antioksidan, cendol, Kappaphycus alvarezii, Metroxylon sagus Rottb, fitokimia  AbstractThe aims  of this study was to determine the phytochemical content and antioxidant  on cendol seaweed (Kappaphycus alvarezii) with additions of sago flour (Metroxylon sagus Rottb.) extracted with methanol. Phytochemical tests were performed to determine the presence of alkaloids, flavonoids, saponins, triterpenoids and steroids. Antioxidant activity of seaweed cendol. Kaphaphycus alvarezii was tested by DPPH method (1-1- diphenyl-2-picrylhydrazl). This study used a Completely Randomized Design (CRD) which consists of three treatments R1S1( RL 10% : TS 90% ), R2S2 (RL 20% : TS 80%), R3S3 ( RL 30% : TS : 70%) and three times replication. Observation data were analyzed using ANOVA (Analysis of Variance) at the level of 95%, if there were significant differences (P> 0.05) then further testing was carried out with DMRT (Duncan Multiple Range Test) at 95% significance level. Phytochemical test showed there was compounds detected with alkaloids, flavonoids, triterpenoids and steroids. While the saponins compounds was not detected in methanol extract of Kappahycus alvarezi. The antioxidant activity by DPPH method showed that the high IC50 values is 652 ppm. The purpose of this study was to determine antioxidant activity of polyphenol and phlorotannin extracts from brown algae Sargassum polycystum. Polyphenol was extracted with ethanol 96% then tested total phenolic content, while phlorotannin was extracted with methanol, chloroform, aquabidest, and ethyl acetate then tested total phlorotannin content. After crude polyphenol and crude phlorotannin extracts were obtained, the studied compounds were extracted using column chromatography. The extracts were then analyzed with HPLC (High Performance Liquid Chromatograpys). Antioxidant activity was analyzed by radical scavenging activity (RSA) and ferrous ion chelating ability (FIC) for four samples, crude polyphenol, crude phlorotannin, polyphenol, and phlorotannin. The result of total phenolic content was 1.18±0.67 mg GAE/g dry sample and total phlorotannin content was 0.61±0.27 mg PGE/g dry sample. Based on RSA and FIC analysis, phlorotannin (IC50 1.12±0.02 mg/mL and 1.34±0.01 mg/mL) showed the highest activity followed by crude phlorotannin (IC50 1.20±0.01 mg/mL and 1.47±0.10 mg/mL), polyphenol (IC50 1.23±0.01 and 1.55±0.02 mg/mL), and crude polyphenol (IC50 1.27±0.01 mg/mL and 1.63±0.02 mg/mL). The results of chromatographic analysis showed that S. polycystum extract contains phenolic compounds similar to the phloroglucinol.Keywords: Sargassum polycystum, antioxidant, polyphenol, phlorotanninABSTRAKPenelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak polifenol dan florotanin dari rumput laut cokelat Sargassum polycystum. Setelah memperoleh ekstrak polifenol dan florotanin, kedua senyawa tersebut dipurifikasi dengan kromatografi kolom. Hasil kromatografi kolom dianalisis menggunakan HPLC (High Performance Liquid Chromatography). Uji aktivitas antioksidan dilakukan dengan metode radical scavenging activity (RSA) dan ferrous ion chelating ability (FIC) terhadap empat sampel, yaitu ekstrak polifenol, ekstrak florotanin, polifenol, dan florotanin. Kandungan total fenolik yang diperoleh adalah 1,18±0,67 mg GAE/g ekstrak kering, sedangkan kandungan total florotanin diperoleh 0,61±0,27 mg PGE/g ekstrak kering. Berdasarkan uji RSA dan FIC, florotanin (IC50 1,12±0,02 mg/ mL dan 1,34±0,01 mg/mL) memiliki aktivitas tertinggi diikuti ekstrak florotanin (IC50 1,20±0,01 mg/mL dan 1,47±0,10 mg/mL), polifenol (IC50 1,23±0,01 dan 1,55±0,02 mg/mL), dan ekstrak polifenol (IC50 1,27±0,01 mg/mL dan 1,63±0,02 mg/mL). Hasil analisis kromatografi menunjukkan ekstrak S. polycystum mengandung senyawa fenolik relatif seperti senyawa floroglusinol.Kata kunci: Sargassum polycystum, antioksidan, polifenol, florotanin
