<p><strong>In vitro conservation of black pepper (Piper nigrum L.)</strong></p><p>Black pepper (Piper nigrum L.) is one of the economically im¬ potant spices. The major constraint in black pepper cultivation and conservation in ield is foot rot disease caused by Phytopthora capsici which could cause plants die. Conservation of black pepper germplasms as living collections in ield is risky due to pests and natural disaster. The experiment on in vitro cop ervation of black pepper var. LDL was conducted al the laboratory of Plant Genetic Resources and Breeding, Research Institute for Spice and Medicinal Crops (RISMC) Bogor from April 1998 to Maret 1999. Single node cuttings from sterile culture were used as explains. The explains were cultured on Murashige and Skoog (MS) medium on full and half strength concentration supplemented with paclobutrazol (paclo) (0, 1, 3 and 5 mg/1). The experiment was performed in a randomized complete block design arranged factorially with 10 replications. The result showed that the medium supplemented with paclo on both full MS and MS A medium could suppress vegetative growth until 12 months. There was no signiicant interaction between medium and paclo on shoot initiation. The effect was signiicant on shoot height, number of leaves and culture performances. Increasing paclo concen¬ tration caused higher suppression of plant growth. MS A medium supplemented with paclo 5 mg/1 showed the slowest growth with shool height 2.10 cm and number of leaves 9. Culture performance was fresh, with green leaves and vigorous. Advcntive shoots were able to regenerate on the medium supplemented with BA 0.3 mg/1. In vitro conservation of black pepper with paclo did not change plant regeneration ability. Therefore, this technique may be used as an altenative method for black pepper conservation.</p>

To improve the quality of pepper products through organic cultivation using organic growth regulators. This Research aims to investigate concentration and long immersion effect of golden snail meat (Pomacea canaliculata L.) on the growth of pepper cuttings (Piper nigrum L.). the method was experimental with a randomized block design (RAK) Factorial design with two factors namely various golden snail meat concentrations and long immersion ranging from several hours. The parameters observed in this study was Growth Percentage (%), Bud Height (cm), Number of Leaves (Strands), Root Length (cm), and Root Volume (ml). The results showed that the treatment of snail meat zpt concentration did not significantly affect the shoot height parameters but significantly affected the percentage of growth, number of leaves, root length and root volume where the effective zpt concentration was 50%. The long immersion treatment significantly affected the percentage of growth and shoot height where the best treatment at w1 was 3 hours’ immersion. The treatment combination of ZPT golden snail meat (Pomacea canaliculata L.) and long immersion did not significantly affect all observations parameters.

ABSTRACT<br /><br />The continuous supply of true-to-type seedling through in vitro technique is very important for cassava (Manihot esculenta Crantz.) as the demand for seedlings is increasing. The research consisted of two experiments and they were conducted from February 2013 to February 2014. The first experiment was in vitro shoot multiplication to evaluate the effect of culture medium and single node position on growth and shoot multiplication of cassava Jame-jame genotype. This experiment was arranged in a randomized complete block design with two factors and four replications. The first factor was culture medium (MS0 and MSB3) and the second factor was single node positions used as explant (basal, middle, and top section of the stem). The result showed that middle section of the stem cultured on MS0 medium had the highest shoot height, number of leaves, and number of nodes. The second experiment was the acclimatization to evaluate the effect of culture period on the success of the acclimatization. This experiment was arranged in a completely randomized design with culture period prior to acclimatization as treatment. The results showed that seedlings from period of 12 and 24 weeks after culture showed higher survival rates (80%) than seedling from period of 36 and 48 weeks after culture (50 and 40%). Culture period prior to acclimatization had no significant effect on plant height and number of leaves.<br /><br />Keywords: BAP, culture period, single node position, shoot multiplication

Fusarium solani f. sp. piperis (teleomorph: Nectria haematococca f. sp. piperis), causal agent of root rot and stem blight on black pepper (Piper nigrum), produces secondary metabolites with toxigenic properties, capable of inducing vein discoloration in detached leaves and wilting in transpiring microcuttings. Production of F. solani f. sp. piperis (Fsp) toxic metabolites reached a peak after 25 days of static incubation on potato sucrose broth at 25 ºC under illumination. Changes in the pH of the culture filtrate did not alter the effect of toxic metabolites. However, when the pH was changed before the medium had been autoclaved, a more intense biological response was observed, with an optimum at pH 6.0. Isolates that produced red pigments in liquid cultures were more efficient in producing biologically active culture filtrates than those which produced pink coloured or clear filtrates suggesting that these pigments could be related to toxigenic activity. Detached leaves of seven black pepper cultivars and Piper betle showed symptoms of vein discoloration after immersion in autoclaved and non-autoclaved Fsp culture filtrates indicating the thermostable nature of these toxic metabolites.

The experiment was conducted to investigate the combined effect of different plant growth regulators and charcoal supplementation in MS medium on growth and development of plantlets regenerated from protocorm like bodies (PLBs) of hybrid orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations with charcoal supplementation was studied. The result revealed that the use of different growth regulators had significant effect on different parameters studied. The maximum weight of PLBs (5.123 g) was obtained from the combination of BAP + IBA at 1.0 mg/l each. The highest shoot height (3.239 cm) and maximum number of rooted plantlets (4.473) was obtained from 1.0 mg/l each of BAP + NAA combination. The maximum number of leaves (3.490) and the maximum length of leaves (1.946 cm) were obtained from 1.0 mg/l each of BAP + IBA and the highest leaf width (1.166 cm) was obtained from 0.5 mg/l BAP +1.0 mg/l IBA combination. The highest root length was obtained from 0.5 mg/l each of BAP + IAA and the maximum number of regenerated plantlets (20) was obtained from 0.5 mg/l IAA + 1.0 mg/l IBA combination. However, the maximum fresh weight of single shoot (0.219 g) and the maximum number of roots per plantlet (6.300) was obtained from 1.0 mg/I each of IAA + IBA combination. Abstract: The objective of this work was to select cupuaçu (Theobroma grandiflorum) tree progenies and individuals based on their agronomic traits, and, indirectly, to identify those adapted to an agroforestry system (AFS) environment in the Brazilian Amazon. For this purpose, 25 full-sib progenies were planted and tested in consortium with black pepper (Piper nigrum), banana (Musa spp.), and bacuri (Platonia insignis) trees. The experiment was carried out in a randomized complete block design, with five replicates and three plants per plot, from 2005 to 2019. For the statistical analyses, the phenotypic averages for production and incidence of witches’ broom disease, evaluated during 11 harvests, were used. Superior progenies and individuals were identified using the mixed model methodology (REML/BLUP), which led to the selection of ten plants from five families with superior agronomic traits. Cupuaçu tree progenies 6, 36, 37, 49, and 52 are the ones that best adapt to the environment of a multispecies AFS in the Amazon region because of their agronomic traits under competitive conditions. Ten matrices show agronomic potential and indirect adaptation to the AFS and can be used as clonal cupuaçu cultivars in this environment.

Tribulus (Tribulus terrestrisL.) is a medicinal plant with considerable implementation such as aphrodisiac and anti‑inflammation drugs. This research was conducted to study the effectivity of coconut water and IAA (Indole‑3‑acetic acid) application on various concentration as the growth regulator for tribulus growth on thein vitroand its effect toward observation variables. The material used were cotyledon from tribulus embryo and MS (Murashige and Skoog) medium. The research method used a Completely Randomized Design with first factor of coconut water concentration of 0 mL L-1, 50 mL L-1, 100 mL L-1, and 150 mL L-1and IAA concentration of 0 ppm, 0.15 ppm, 0.20 ppm, and 0.25 ppm as the second factor. Observed variables were shoots emergence time, number of shoots, shoot height, number of leaves, roots emergence time, number of roots and regression between observation variables. The result showed that the combination of 150 mL L-1coconut water and 0.25 ppm IAA gave the highest shoot height, roots emergence time, and number of roots. Coconut water treatment on 150 mL L-1concentration alone gave the best result on shoots emergence time, number of shoots, and number of leaves, while IAA concentration of 0.25 ppm independently gave the highest number of leaves. Regression analysis result indicate that the number of roots has a positive correlation with shoots height, number of shoots and number of leaves.

The Fall armyworm worm (FAW) (Spodopterafrugiperda) is one of the important economic pests which goes on several field crops and causes serious damage. The aim of this study was to look for efficient, less cost and environmentally friend plant extract for controlling Fall armyworm worm in cereal crops. A Complete Randomized Block Design (CRBD) experiments with three replications were conducted in the laboratory to investigate the insecticidal effects of four plant extracts consisting of ethanolic extract of Neem (Azadirachta indica) seeds, Black pepper (Piper nigrum) seeds, Usher (Calotropis procera ) leaves and water extracts of Argel (Solenostemma argel) leaves on larvae of the Fall armyworm (FAW) (Spodoptera frugiperda). Newly emerged larvae of FAW were treated topically by 4 concentrations (10, 25, 50 and 75%) of each extract, and then the larval mortalities were calculated after 24, 48 and72 hrs. The results showed the highest concentrations (75%) of the three ethanolic extracts gave higher mortality percentages (100%) after 72 hrs of exposure, compared with other concentrations. Also, these were not significantly different from the recommended dose of the standard pesticide “Spinosad”. On the other hand, Argel water extract showed no effect on the (FAW) larvae. It is recommended that this experiment to be replicated under different environments.

Ethnopharmacological Relevance: Pepper essential oils have potential immunomodulatory, anti-tumor, and anti-cancer activities. Pepper exhibits the potential to prevent or attenuate carcinogenesis as therapeutic tools. However, the related mechanism remains unelucidated.Aim of the Study: The present study aims to provide reasonable information for the explanation of the dissimilarity of the essential oils from white (WPEO) and black pepper (BPEO).Materials and Methods: WPEO, BPEO, and their single active component, as well as synthetic antioxidants, were compared by the cell model methods and chemical methods, including intracellular antioxidant activity (CAA), total antioxidant activities (TAA), superoxide radical (SR), hydroxyl radical (HR), DPPH radical (DR) scavenging activities and inhibition ability of lipoprotein lipid peroxidation (ILLP).Results: The median effective concentration (EC50) values (mg/mL) of the WPEO and BPEO of SR, HR, DR, and ILLP were 0.437 and 0.327, 0.486 and 0.204, 7.332 and 6.348, 0.688, and 0.624 mg/mL, respectively. The CAA units of WPEO and BPEO were 50.644 and 54.806, respectively. CAA, DR, and TAA of BPEO were significantly higher than those of WPEO (p &lt; 0.05). The BPEO and WPEO can be differentiated as the former have higher correlations with 3-carene, α-pinene, β-pinene, and limonene while the latter has a higher caryophyllene correlation. The WPEO and BPEO show a good intracellular scavenging ability of reactive oxygen species in HeLa cells.Conclusion: Generally, pepper oil has stronger activities than single components, indicating that pepper is a broad-spectrum natural antioxidant.
