High germination seed lots of purple coneflower [Echinacea purpurea (L.) Moench] were evaluated for laboratory germination following osmotic priming or chilling stratification. Compared to nontreated seeds, osmotic priming at 25C in salts (KNO3 + K3PO4; 1:1, w/w) or polyethylene glycol 4000 (PEG) increased early (3-day) germination percentage at 27C of all seed lots, and improved total (10-day) germination percentage of low-germination seed lots. Total germination percentage was unaffected or increased by priming for 4 days compared to 8 days, and by priming at –1.0 MPa compared to –0.5 MPa (except for one low-germination seed lot). Chilling stratification in water at 5 or 10C increased early and total germination of all seed lots, except for that same lot, compared to nontreated seeds. Total germination percentage was unaffected or increased by stratification at 10C rather than at 5C. Neither extending stratification ≥20 days nor lowering osmotic potential with PEG during stratification improved total germination percentage.

Six seed lots of purple coneflower were purchased from four commercial seed sources and evaluated for germination in either light or darkness in combination with two temperature regimes (constant 27C vs. alternating 30C for 8 hours and 20C for 16 hours). Seed lot differences accounted for the majority of variation, with two seed lots exhibiting high (81% to 91%) germination and the remaining seed lots having 39% to 66% germination. There was no effect of light on germination, regardless of the seed lot. However, alternating temperatures improved germination in one of the low-germination seed lots. Seed size and inflorescence position did not affect seed germination. Seed harvested at physiological maturity (maximum seed dry weight), but before drying had occurred, had a higher germination percentage than seeds harvested after desiccation, and they maintained a higher germination percentage even after 1 year in storage.

The germination responses of wild blue indigo [Baptisia australis (L.) R. Br.], purple coneflower [Echinacea purpurea (L.) Moench.], Maximilian sunflower (Helianthus maximiliani Schrad.), spike goldenrod (Solidago petiolaris Ait.), and Missouri ironweed (Vernonia missurica Raf.) seeds after 0, 2, 4, 6, 8, or 10 weeks of stratification at 5C were investigated. Seed viability was determined using triphenyl tetrazolium chloride staining and germination based on the percentage of viable seeds. Germination percentage (GP) increased in all five species as weeks of stratification increased. Days to first germination and germination range (days from first to last germinating seed) decreased with increasing weeks of stratification, but the effect beyond 4 to 6 weeks was minimal. The number of weeks of stratification for maximum GP was 4 for purple coneflower, 6 for Maximilian sunflower, 8 for Missouri ironweed, and 10 for wild blue indigo and spike goldenrod.

Seeds of purple coneflower (Echinacea purpurea (L.) Moench were osmotically primed (OSP) in polyethylene glycol (PEG) or matrically primed in expanded vermiculite No. 5 (solid matrix priming, SMP). With both OSP or SHP at 15C, 10-day exposure to -0.4 MPa resulted in lowered time to 50% germination (T,) and higher germination percentage than shorter exposure (5-day) or lower water potential (-1.5 MPa). SMP- and OSP-seeds performed similarly in a greenhouse trial, resulting in 80% and 34% seedling emergence at 23C and 37C, respectively, compared to 58% and 27% for non-primed seeds. Seedling emergence rate and synchrony from primed seeds were greater than from non-primed seeds at both temperatures. An incubator study established that adding 10-4M GA3 and 10 mN ethephon (2-chloroethylphosphonic acid) to the PEG or vermiculite resulted in lower T50 and higher germination percentage than priming without these growth regulators. A further incubator study established that less-expensive trade products (Pro-Gibb Pius 2X) and Florel could substitute for the reagent-grade growth regulators. Seeds primed in PEG or vermiculite containing 10-4M GA3 from Pro-Gibb Plus 2X and 10 mM ethephon from Floral had lower T50 and higher percentage emergence in a greenhouse trial than seeds primed without growth regulators. Compared to the non-primed seeds, these treated seeds had 29% greater seedling emergence and 61% less time to 50% emergence.

Increased osmotic potential from 0 to −14 bars decreased the moisture uptake and germination of mechanically scarified hemp sesbania [Sesbania exaltata(Raf.) Cory] seed. Germination percentage for osmotic potential of 0, −2, −4, −6, and −8 bars was 86%, 86%, 70%, 19%, and 1%, respectively. This response to moisture stress was modified by imbibition, or hydration-dehydration, of the seed prior to the moisture stress. Imbibing the seed for as little as 2 h significantly increased its subsequent germination against moisture stress. One hydration-dehydration cycle had little or no effect on germination, but as the number of cycles increased, the germination percentage decreased. Prolonged hydration of 3 and 7 days followed by dehydration greatly reduced subsequent seed germination.

This paper describes the dispersal–germination characteristics of seeds of 77 native tree species in a seasonal monsoon forest in Uttaranchal state, northern India. Results indicate that 50% of the species dispersed in the hot, dry summer months, 18% during the rainy season, 23% during the cold season and the remainder in late spring. Germination tests on fresh and laboratory-stored seeds revealed a relationship between morphological features of the fruit and both germination percentage and mean germination time (MGT). Highest mean germination (50%) was for dry-dehiscent fruits with winged wind-dispersed seeds, followed by dry-dehiscent fruits with non-winged seeds (38%) and seeds of dry-indehiscent fruits (37%). Lowest germination (29%) was for seeds from fleshy or pulpy fruits. MGT followed the reverse course. Germination data for seeds stored dry in the laboratory during one seeding cycle indicated six patterns of seed germination: (1) average germination percentage of fresh seeds lower than that of stored seeds, indicating an after-ripening requirement; (2) initial high germination percentage followed by low values, indicating a steep to moderate decline in viability following harvest; (3) no germination after 1 or 2 months of seed storage, due to complete loss of viability, indicating short seed longevity; (4) fresh seed germination in some species equalled the average germination value of stored seeds, indicating constant germination for one whole seeding cycle; (5) germination of both fresh and stored seeds remained consistently low throughout the season, indicating a requirement for some kind of pre-treatment or having poor quality of seeds; (6) initial high germination followed by decline and again increase, showing a seasonal rhythm of germination.

This project tested rates of lignite-activated water (LAW) for its influence on seed germination, cutting propagation, and plant performance. LAW is a product of CAW Industries, Rapid City, S.D. LAW is water-activated by lignite in a process that includes the addition of sulfated castor oil, calcium chloride, magnesium sulfate, sodium meta silicate, and fossilized organics from refined lignite. LAW is reported to improve many plant performance traits. Four rates were used in this study. Seed germination trials indicated no significant differences in germination percentage with LAW applications with the two species tested, Echinacea purpurea and Hibiscus dasycalyx. In a “closed” system, LAW enhanced cutting propagation success of Aster caroliniana, Cuphea micropetala, and Verbena `Homestead Purple', as measured by percent rooting and dry weight of roots produced. Cutting propagation of two woody species, Illicium henryi and Rosa banksiae, was not improved with LAW additions. In the SFASU Arboretum, pansy performance, as measured by plant dry weight, was improved one month after establishment.

Purple coneflower seeds following priming (-0.04 MPa, 10 days, 15C, darkness) osmotically in polyethylene glycol 8000 (PEG) or matrically in expanded no. 5 vermiculite had greater germination rate and synchrony at continuous 20C or 30C than untreated seeds, but germination percentage was unaffected. Inclusion of 5.5 × 10-2 M gibberellic acid (GA3 as ProGibb Plus 2X, Abbott Laboratories, N. Chicago, Ill.) further improved germination rate and synchrony at 20C, but not at 30C. In a greenhouse study (30C day/27C night, July-August natural light), seeds primed in PEG or vermiculite containing G A3 compared to untreated seeds had 6 percentage points higher maximum emergence (ME), 3.3 fewer days to 50% ME, 1.9 fewer days between 10% and 90% ME, 116% greater shoot dry weight, and 125% longer leaves at 16 days after planting in peat-lite. Inclusion of ethephon (0.01 m, as Florel) either alone or with GA3 during priming provided no benefit to seed germination or seedling emergence. Moistened vermiculite substituted for PEG solution as a priming medium for purple coneflower seeds, the priming benefit on seedling emergence and growth being enhanced by 5.5 × 10-2m G A3 inclusion in the priming media.

Two experiments (in Petri dishes and in soil) were conducted to investigate the effects of osmopriming on seed germination and the early seedling characteristics of caraway (Carum carvi L. var. annua). The priming treatments in the Petri dish experiment were: polyethylene glycol (5%, 10% and 20%), KNO3 (0.5%, 1% and 2%) and KCL (1%, 2% and 4%) with three different soaking times (12, 24 and 36 h) along with control (non-primed seeds). Only polyethylene glycol and H2O were applied in the cell tray experiment, which were then compared with the non-primed seeds. In the Petri dish experiment, all three priming reagents significantly enhanced seedling length, with the most effective treatments being 5% PEG, 2% KNO3 and 1% KCL for 12 h. The plumule dry weights were also increased significantly after PEG (20% for 36 h), KNO3 (2% for 24 and 36 h) and KCL (1% for 12 h) treatments in comparison with the control. In the soil experiment, osmopriming with PEG significantly improved the germination rate (GR) and percentage, the plumule dry and fresh weights and the plumule length of caraway seedlings when compared with the control. A 23% higher germination percentage was recorded for the seeds treated with 5% PEG for 24 h as compared with the non-primed seeds. The PEG-primed seeds produced significantly longer seedlings when treated with 5% PEG for 24 h. All of the applied PEG treatments significantly enhanced the plumule fresh and dry weights, with the best outcomes being after 5% PEG (24 h) and 10% PEG (36 h) treatments, respectively. The 12-h hydro-priming also significantly enhanced all of the studied germination parameters when compared to the control. The results of the presented experiments show the significant positive effects of seed priming on caraway germination and how early seedling performance can easily be adopted by producers.

SummaryThis study was conducted in order to determine the appropriate treatment for breaking dormancy and the effect of salinity on seed germination of purple coneflower (Echinacea purpureacv. Magnus), in two separate experiments. In the first experiment, five levels of gibberellic acid (GA3) (0, 250, 500, 1000, and 1500 mg×L−1) with four levels of cold moist stratification period of seeds at 5°C (0, 5, 10 and 15 days) were launched. A factorial experiment was conducted in a completely randomized design with four replications. The statistical analysis showed that concentration of 250 mg×L−1GA3with 10 days of cold moist chilling significantly increased the percentage of germination of normal seedlings and reduced the mean time of germination. In the second experiment, the seeds were chilled for 10 days at 5°C and half of them treated with 250 mg×L−1GA3for 24 hours. The seeds treated with GA3, and those non-treated were subjected to NaCl for salinity stress. The experiment was conducted using five salinity levels (0, 20, 40, 60 and 80 mM NaCl) in four replications in a completely randomized design. The results showed that purple coneflower is highly sensitive to salinity in the germination stage. The results also showed that by increasing salinity levels, the percentage of germination and normal seedlings significantly decreased and the mean time to germination increased, compared to the control treatment. But the seeds treated with GA3showed higher viability and better performance under salinity stress condition.
